Development of a Commercial Process for Micro-Encapsulation of Lactic Acid Bacteria Using Sodium Alginate (알긴산 나트륨을 이용한 유산균 캡슐화의 상업화 공정 개발)

  • We aimed to develop commercialization process of encapsulation which is superior in productivity compared to existing methods by using sodium alginate. Also, in the same process, sodium alginate with chitosan was used to encapsulate lactic acid bacteria with the same process and then the viable cell counts of the two encapsulated lactic acid bacteria were compared. As a test result of the fluidized drying process developed by the present researchers, it was found that the drying time was shortened by 15 to 20 hours compared to the freeze drying method, but the number of viable lactic acid bacteria was about 75% as compared with freeze drying. However, considering the cost and time of drying, it can be confirmed that the commercialization process is possible by the fluidized bed drying method. When the number of viable cells of Ca-alginate capsule and Chitosan-alginate capsule were compared, it was confirmed that there were about $1{\times}10^9$ or more bacteria in the former and about $1{\times}10^3$ in the latter. The lactic acid bacterium capsules prepared by the present technique were stable for 96 hours or more at pH 4.65 and 6.01, but disappeared within 1 hour at pH 7.07 and 8.35. This suggests that the disintegration of lactic acid bacteria can be easily occurred in small and large intestine.

Study of Relationship between the Chapter of Channels in Miraculous Pivot of Emperor′s Classic of Internal Medicine and Yangmyung disease in Sanghanron (『영추ㆍ경맥편』과 『상한론』의 양명병에 대한 상관성 연구)

  • Chang Chung-ching(張仲景) in the Later Han(Eastern Han) Dynasty of Chinese history wrote the treatise on Diseases Caused by Cold Factors(傷寒論; Shang Han Lun) on the basis of the fundamental theory of Emperor's Classic of Internal Medicine(黃帝內經; ECIM) after collecting medical treatment experiences until the Han Dynasty. It had great significance that Shang Han Lun was the origin of treating six-channels(六經) and there showed the peculiar guidelines on the diagnosis and treatment of oriental medicine to divide diseases into six-channels. The oriental medical doctors who had studied Shang Han Run thought highly of meridians and until now it was generally known that the chapter of heat in the Plain Questions of ECIM(黃帝內經, 素問ㆍ熱論) was the basis of Shang Han Run. The chapter of heat in the Plain Questions of ECIM was the first text in which the basic theory on six-channels according to the types of illness was introduced. In my point of view, the theory of treating six-channels had close relation to the Chapter of Channels in Miraculous Pivot of ECIM(黃帝內經, 靈樞ㆍ經脈篇) as well as the chapter of heat in the Plain Questions of ECIM. Therefore I took a look at the origin of treating six-channels in Shang Han Lun and illuminated again the meaning to compare the parts of in Shang Han Lun with the Chapter of Channels in Miraculous Pivot of ECIM. Conclusion: The, symptoms divided into six-channels in the chapter of channels in ECIM gave the fundamental basis of diagnosis and treatment basesd on overall analysis of signs and symptoms(辨證論治) an illness in the Zangfu(臟腑) in respect of meridians. Viewed in the light of diagnosis and treatment basesd on overall analysis of signs and symptoms(辨證論治), the symptoms of YangMing-channel(陽明經) in the Chapter of Channels in Miraculous Pivot of ECIM were, for the most part, accord with those of YangMing-disease in Shang Han Lun. Furthermore, the symptoms in Shang Han Lun were explained definitely and in detail. Therefore the theory of Shang Han Lun has been developed on the basis of ECIM with the changes of the times. YangMing-disease in Shang Han Lun implied medical cases in stomach meridian(胃經) and large intestine meridian(大腸經). Therefore Shang Han Lun was the foundation of treatment basesd on overall analysis of signs and symptoms(辨證論治) in respect of meridian as well as the text in which the steps of infectious diseases(外感病) of human bodies were explained.

Effects of Dietary Yellow Clay on Growth Performance and Body Composition in Broiler Chicks (육계에서 사료내 황토 첨가가 성장 및 체조성에 미치는 영향)

  • This experiment was conducted to evaluate the effects of dietary Hwangto on growth performance and body composition in broiler chicks. A total of 216 one-day old 'Ross' broilers were assigned to 6 treatments in a completely randomized design. The six dietary treaoents were control no Hwangto added and diets containing 0.5, 1.0, 2.0, 4.0 and 8.0% of Hwangto supplementation. The weight gain of broilers tended to reduce with increasing level of Hwangto. However, there were no significant differences in feed intake and feed conversion ratio of broilers fed control and diets containing different level of Hwangto supplementation (P>0.05). There were no significant differences in TBA value of meat from broilers fed control and Hwangto supplemented diets (P>0.05). The meat cholesterol content was significantly lower in Hwangto treatment than of the control (P>.0.05). $NH_3$ concentration in feces was reduced in all Hwangto treatments (P<0.05). The Mg and Mn concentrations of meat we.e significantly higher in 8.0% Hwangto treatment than that of the control (P<0.05). The large intestine weight was significantly reduced in 2.0, 4.0 and 8.0% treatments (P<0.05).

The Effects of Korean Traditional Sauces on Quality Characteristics of Hanwoo Semitendinosus Dry-cured Ham (한국 전통장류가 한우 반힘줄모양근(M. Semitendinosus)으로 제조한 건염햄의 육질 특성에 미치는 영향)

  • This study was conducted to develop a processing method for dry-curing beef ham with Korean traditional sauces and to investigate the quality properties of the dry-cured beef hams. Semitendinosus muscle from Hanwoo was spread with sundried salt (C), sun-dried salt mixed with red pepper paste (T1), and sun-dried salt mixed with soybean paste (T2), and inserted into washed Hanwoo large intestine. It was then dried and fermented for 75 d. Moisture contents of the hams were 46-48%, without any significant differences. T1 showed the lowest pH among the three dry-cured hams, while water holding capacity (WHC) of T2 was found the lowest (p<0.05). In mechanical color determination, C showed higher CIE $a^*$ and CIE $b^*$ than T1 and T2 (p<0.05), however CIE $L^*$ did not show any significant difference. In texture, T1 demonstrated significantly lower springiness than C (p<0.05). The sensory properties of all dry-cured hams did not show any significant difference, but T1 indicated a higher taste, saltiness and overall acceptability than others, whereas C gathered the highest score in color and flavor. In conclusion, the dry-curing by combination of sun-dried salt mixed with red pepper sauce (T1) showed higher quality properties of the Hanwoo dry-cured ham.

A Study on San Jiao(三焦) stated on "Ling Shu(靈樞)" ("황제내경영추(黃帝內經靈樞)"에 기재된 삼초(三焦)에 관한 소고(小考))

  • According to the result about 'San Jiao(三焦)'recorded on "Yellow Empero's Canon Internal Medicine Ling Shu(黃帝內經 靈樞)", we achieved following results. 1. As we consider the concept of 'San Jiao' recorded on "Ling Shu", in early time, it was related to bladder(膀胱) and there was no divided concept into 'Shang Jiao(上焦)', 'Zhong Jiao(中焦)' and 'Xia Jiao(下焦)'. Later, there was appearance of body metabolism concept in anatomic way and started to emphasize that stomach(胃) is the very beginning of digestion. This point then adjusted into the concept which the core theory of digestion and water metabolism begins with 'Zhong Jiao' and processes to 'Shang Jiao' and 'Xia Jiao' then it established the 'San Jiao' theory adjusted to the functional change than property change. Later as there is set theoretical structure of 'three Yin and three Yang(三陰三陽)', it included the concept of 'San Jiao' onto meridian system theory to complete as a theory. Finally, it completed the theoretical structure that 'San Jiao' runs water metabolism of circulation, body fluid and urine for body to produce blood and Qi to protect and provide nutrition to the human body. 2. From the point of each part, 'San Jiao' means all body composition factor related to the relation to the digestion and water metabolism to produce Qi and blood. Also, the details of entire function of 'San Jiao' tells that 'Zhong Jiao' intakes food and divides the clarity and turbidity of digested substances. The origin of this digestion and water metabolism lies at Xia Jiao. The clean substance including the mood and taste climbs via Shang Jiao. The vapor like substance climbed to Shang Jiao becomes 'defensive Qi(衛氣)' and controls body temperature and sweat by supporting and spreading the Qi by Shang Jiao. The liquid substance climbed to Shang Jiao becomes blood. The blood has stronger character as substance than defensive Qi so Zhong Jiao becomes the base and the way for the blood. The turbid Qi separated at Zhong Jiao passes large intestine and the solid substance is excreted and the liquid is absorbed into bladder. The Xia Jiao that controls this process controls the liquid state of water matabolism so control the urine with bladder. Therefore, 'San Jiao' can be understood as a general concept that controls entire water metabolism as a way of food, Qi and blood.

Spermatogenesis of Black Molly and Sailfin Molly (Poeciliidae, Teleostei) (경골어류 난태생 송사리과에 속하는 black molly와 sailfin molly의 정자형성과정)

  • Black molly (Poecilia sphenops) and sailfin molly (Poecilia latipinna) are a teleost belonging to Poeciliidae. The spermatogenesis between two species were investigated by light and electron microscope. The whitish testes of both black molly and sailfin molly were located between intestine and air bladder. The size of testis was major axis 7 mm, minor axis 2 mm. The testis contained numerous testicular cysts. In both black molly and sailfin, primary spermatocytes were comparatively large ellipsoidal, and mitochondria showed a marked development. The secondary spermatocyte was smaller than that of primary spermatocyte, highly condensed according to their development. The nucleus with electron-dense was round shape and flagella started to be formed. In spermiogenesis, chromatin was more condensed. The mitochondria were rearranged along the tail. The number of mitochondria was 2 to 4 in cross section and 8 to 10 in longitudinal section. The head of mature sperm was long cone shape and had not acrosome. The microtubules of flagella were arranged 9+2 structure. Also, the tail of sperm have lateral fins. In conclusion, spermatogenesis and sperm morphologies of these two species were same. These morphological similarity seems to be an indication of the Poeciliidae.

Analysis of Trans Fatty Acid Content in Retort Food, Powdered Milk, Biscuit and Pizza Products (레토르트식품, 분유, 비스킷 및 피자 내에 함유되어 있는 트랜스지방산 함량 분석)

  • The consumption of foods containing trans fatty acids (TFAs) is a matter of concern at present. According to many studies, trans fatty acids (TFAs) may cause illnesses such as the coronary heart disease, diabetes mellitus, large intestine cancer, and breast cancer. They can also raise low density lipoprotein (LDL) cholesterol and reduce high density lipoprotein (HDL) cholesterol. TFAs can also inhibit the synthesis of phospholipids containing polyunsaturated fatty acids in arterial cells. As a consequence the Food and Drug Administration has deemed that saturated fatty acid, cholesterol and trans fatty acid levels be listed on food labels as of 2006. The Korea Food and Drug Administration also has required the listing of trans fatty acid content on food labels since 2007. The aim of this study was to determine the total lipid and trans fatty acid (TFA) contents in retort food, powdered milk, biscuit and pizza products. The number of samples examined were 2 retort food, 6 powdered milk, 7 biscuit and 3 pizza products. The extraction of total lipids in retort food and powdered milk followed the chloroform methanol method. The extraction of total lipids in biscuit and pizza was by the acid digestion method. All samples were analyzed by gas chromatography (GC) using a SP-2560 capillary column and a flame ionization detector. The TFA contents per 100g of sample were 1-2.8% (1.9%) in retort foods, 0.4-2.4% (1.37%) in powdered milk products, 0-2.9% (1.23%) in biscuits, and 2.8-3.45% (3.03%) in pizzas.

LIFE CYCLE AND CHEMOTHERAPEUTIC CONTROL OF A FILARIAN WORM, PHILOMETROIDES CARASSII PARASITIC IN CARASSIUS AURATUS (붕어에 기생하는 철사충 Philometroides carassii의 생활사와 치료에 관하여)

  • Philometroides carassii (ISHII, 1931) is a parasitic nematode of Carassius auratus and gold fish. Recently a large number of parasitized fishes by this nematode have been reported by fish farmers in Korea. The present investigation was aimed to find out the life cycles of the parasites and effects of chemotherapeutic medicines on the emerged larvae from the adult parasites. Results of the study are summarized as follows : 1. Intermediate host of p. cararsii is a freshwater copepod, Cyclops sp. and the invaded lavae in the body cavity of copepod are capable for parasitizing the fish host after 3 hours of invasion. 2. The lavae eaten directly by the fish host are digested or excreted. 3. The larvae invaded in the fish intestine through the intermediate host pass into the body cavity in 1 to 10 days, and become subadult stage. After 18 to 20 months the subadults parasitic in the body cavity of the Carassius auratus, penetrate through muscle fissile into tile caudal fin where they become adults. Whereas those parasitic in the body cavity of the gold fish, 7 to 8 months the sutadults penetrate through muscle tissue into the caudal fin where they become adults. 1. Male subadults found in the body cavity of the Carassius auratus and gold fish, were larger than females, This is the first record of male subadults occurring in the fish host. 5. Among four different :drugs tested, most of all larvae died in 4 ppm of dipterex in 5 days. But no adults were harmed even in 6 ppm dipterex in 11 days.

Gizzard Impaction with Bile Stasis in Captive Oriental White Storks (Ciconia boyciana) (사육하는 황새에서 담즙정체를 동반한 사낭경색)

  • The Korea Institute of Oriental White Stork Rehabilitation Research (KIOWSRR) is breeding oriental white storks for residential rehabilitation in South Korea. Three oriental white storks were referred with anorexia and depression. Two storks were died before examinations and one stork was died after 1 day of fluid therapy. On necropsy, salmon-colored subcutaneous and visceral fats were examined in all storks. Multiple focal necroses on liver surface were also examined. In gizzards, many gravels and grasses were severely tangled; therefore, it produced a large mass that blocked digestive system of the storks. Small intestinal segments of the stork, which was dead after the fluid therapy, were severely adhesive. On histopathology, diffuse necroses with/without inflammation were examined in the liver, gizzard and intestine in all storks. Bile stasis was additionally examined in the liver. According to the results of the examinations, all storks were diagnosed as gizzard impaction caused by parorexia.

The regional distribution and relative frequency of gastrointestinal endocrine cells of the ICR mice: An immunohistochemical study (ICR 마우스 위장관 내분비 세포의 부위별 분포 및 출현 빈도 : 면역조직화학적 연구)

  • The regional distributions and relative frequencies of some gastrointestinal endocrine cells in the 8 portions (fundus, pylorus, duodenum, jejunum, ileum, cecum, colon and rectum) of the gastrointestinal tract of ICR mouse (ICR) with immunohistochemical method using 7 types of specific antisera against somatostatin, serotonin, glucagon, cholecystokinin (CCK)-8, secretin, pancreatic polypeptide (PP) and gastrin. In this study, somatostatin-, serotonin-, glucagon-, CCK-8-, secretin- and gastrin-immunoreactive (IR) cells were identified. Most of these IR cells in the intestinal portion were generally spherical or spindle in shape (open-typed cell) while cells showing round in shape (close-typed cell) were found in the stomach regions occasionally. Their relative frequencies were varied according to each portion of gastrointestinal tract. Somatostatin-IR cells were demonstrated throughout whole gastrointestinal tract except for large intestine. Serotonin-IR cells were detected throughout whole gastrointestinal tract and they were most predominant endocrine cell types in this species of mouse. Glucagon-IR cells were restricted to the fundus and rectum with moderate and a few frequencies, respectively. CCK-8-IR cells were observed in the pylorus, duodenum and ileum with numerous, moderate and rare frequencies, respectively. Secretin-IR cells were restricted to the duodenum and ileum with a few and rare frequencies, respectively. Gastrin-IR cells were restricted to the pylorus with numerous frequency. However, no PP-IR cells were found in this study. In conclusion, some peculiar distributional patterns of gastrointestinal endocrine cells were found in the ICR mouse compared to those of other mammals.
