To prepare oligouronic acids from high-molecular-weight alginates, sea tangle Laminaria japonicus alginates were hydrolyzed at $80^{\circ}C$ for various hydrolysis times (HT). The effects of hydrolysis time on the distribution ratios (DR) of the molecular weights (MW) of the hydrolyzed alginates were in-vestigated. As HT increased, the DR of the alginates with MW>500 kDa decreased ex-ponentially, while those with MW=50-100 kDa or MW<50 kDa increased exponentially. For MW=300-500 kDa, DR increased exponentially until HT reached 60 min, and then decreased exponentially. Similarly, for MW=100-300 kDa, DR increased exponentially until HT reached 90 min, and then decreased.
In order to study the utilization of brown seaweed Laminaria japonica as an alternative renewable feedstock for bioethanol production, the properties of acid hydrolysis and ethanol fermentation were investigated. The acid hydrolysis enhanced the final yield of fermentable sugars, which led great increase of ethanol productivity. The maximum yield of reducing sugars reached 135 mg/g-dry Laminaria japonica after 1.0N sulfuric acid-hydrolysis at $130^{\circ}C$ for 6 h. The Saccharomyces cerevisiae (ATCC 24858) could ferment $C_6$-sugars like glucose, galactose and mannose into ethanol, but not $C_5$-sugars like arabinose and xylose. Optimal fermentation time varied with sugars; 48 h for glucose, 72 h for galactose, and 96 h for mannose. Nevertheless, the ethanol yield from the hydrolysate reached 242 mg/g-dry Laminaria japonica after fermentation by the S. cerevisiae at $35^{\circ}C$ for 96 h, which corresponds to approximately 4 times more than the theoretical yield from total reducing sugars in the hydrolysates. It indicates that the non-reducing sugars or oligosaccharides dissolved in the hydrolysate played an important role in producing bioethanol. The ethanol concentration linearly increased from 2.4 to 9.2 g/L, while the ethanol yield per dry weight of biomass decreased from 242 to 185 mg/g, with increasing the ratio of biomass to acid solution from 1 to 5% (w/v). The bioethanol yield estimated was approximately 7,400~9,600 kg/ha/year, and indicated that Laminaria japonica is a promissing feedstock for bioethanol production.

Laminaria roots have not been practically used in Korea. In this study, the extraction process of carbohydrates and minerals from Laminaria roots was investigated and the properties of extracted components were measured. Hydrochloric acid generally used in carbohydrate extraction from seaweeds in order to obtain high extraction yield. But in this work, to utilize extracted components as a functional food material, organic acids such as citric acid were used. Organic acid as extraction solvent has low extraction yield compared to strong acids. Therefore optimum condition for maximum yield was investigated in carbohydrate extraction from Laminaria roots using organic acid. We measured the extraction yields of carbohydrate with variation of extraction temperature, extraction time, concentration of organic acid and particle size of samples. The extraction yield increased as the particle size decreased and temperature became high. The extraction yield was 19.0 wt% after 4.0 hours extraction with 0.2 wt% citric acid at $100^{\circ}C$. Potassium concentration was high compared other minerals in extraction solution, that is, the ratio of K/Na was about 3.0. Fucoidan from Laminaria roots had same carbohydrate composition and lower molecular weight compared that of Undaria pinnatifida.
This study was designed to investigate the effects of sea tangle (Laminaria japonica) extract and fucoidan components on chronic degenerative diseases. Sprague-Dawley(SD) male rats (210$\pm$5g) were fed experimental diets: Dasi-Ex group: dasima extract powder of 4.0% added to control diet; Fuco-I, II and III groups: fucoidan powder of 1, 2 and 3% added to Dasi-Ex group for 45 days. Triglyceride (TG) levels in serum were significantly lower (10~15%) in Fuco-I, II and III groups compared with control group. Total cholesterol levels were significantly decreased (7~10% and 15~ 35%) in brain mitochondria and microsomes of Fuco-II and III group compared with control group. LDL-cholesterol levels were remarkably decreased (20~30%) in Dasi-Ex and Fuco-I, II, III groups, but HDL-cholesterol levels were significantly increased (10~12%) in Fuco-II and III groups only compared with control group. The ratios of HDL/total cholesterol resulted in a marked increase (3 5~55%) in Dasi-Ex and Fuco-I, II, III groups, but atherogenic indices were remakably decreased (40~50%) in Dasi-Ex and Fuco-I, II, III groups compared with control group. Membrane fluidities were remarkably increased (45~70% and 38~42%, respectively) in brain mitochondria and microsomes of Fuco-II and III groups compared with control group. Administrations of fucoidan added to dasima effectively decreased TG, total and LDL-cholesterol, and atherogenic index, while also effectively increased HDL-cholesterol, HDL/total cholesterol ratio, and membrane fluidity, suggesting chronic degenerative diseases were very effectively prevented by the administration of fucoidan component.

The re-examination of morphological and anatomical characters of Laminaria appressirhiza and L. inclinatorhiza collected from different localities in the Sea of Okhotsk was performed. Despite their commercial and ecological importance to the region they have not been comprehensively reviewed since their first description in 1970. Our results show that some original diagnostic key characters (e.g., shape of holdfast, shape of sporangial sori, and dissection of blade) are not stable and have deviations from the type morphology when plants grow in different environments. In L. inclinatorhiza, the sporangial sori development occurred differently to the pattern indicated in original species description as they did not develop simultaneously on both sides of the blade. Instead, the sporangial sori outlines on both sides of the blade did not coincide at first and only became coincident later. Also, a deep-water population of L. inclinatorhiza with an unusual and interesting morphology, growing at depths of 15-25 m on opened rocky coasts in Taujskaya Bay (northern part of the Sea of Okhotsk) was found. The stable diagnostic key characters to distinguish these two species are the cone-like, multilayered, very thick and massive holdfast (in L. inclinatorhiza) and rolled margins of blades, lamellar rosette-like part of thallus, and sporangial sori developing only on one side of the blade (in L. appressirhiza). The ecological characteristics, distribution, and abundance of both species in the Sea of Okhotsk are discussed. Both species are perennial and widely distributed in the region. L. appressirhiza is more often found as a subdominant species among other kelps, forming maximum biomass and density of 7-9 kg and 8-25 plants per $1m^2$, respectively. L. inclinatorhiza sometimes forms local mono-species communities with maximum biomass and density of 10-12 kg and 10-15 plants per $1m^2$, respectively.

This study examined the effects of the dietary inclusion of Porphyra and sea tangle Laminaria japonica on the growth, feed utilization, body composition, and plasma chemistry of juvenile Korean rockfish Sebastes schlegeli. Eight hundred and forty juvenile fish averaging 5.0 g were allocated 40 fish per tank to 21 180-L flow-through tanks. Seven experimental diets were prepared: control (Con) without additive, 0.5 and 1% Porphyra extract (PE), 3% Porphyra powder (PP), 0.5 and 1% sea tangle extract (STE) and 3% sea tangle powder (STP), referred to as PE-0.5, PE-1, PP-3, STE-0.5, STE-1, and STP-3, respectively. Each additive was included in the experimental diet at the expense of the same amount of wheat flour. Each experimental diet was fed to triplicate groups of fish. The experimental diets had no effect on the survival, weight gain or specific growth rate of the fish, feed consumption, feed efficiency ratio, protein efficiency ratio, protein retention, hepatosomatic index, condition factor, moisture or crude protein content of the entire body excluding the liver or moisture, crude protein or crude lipid content of the liver. None of the plasma parameters were affected by the experimental diets. Based on these results, the dietary inclusion of Porphyra and sea tangle did not affect the growth, feed utilization, body composition or plasma chemistry of juvenile Korean rockfish.
This study was designed to investigate the effects of sea tangle (Laminaria japonica) extract (Dasi-Ex group: dry base 4.0%) and fucoidan-added (Fuco-I, II, III group: fucoidan of 1.0%, 2.0%, 3.0% added to Dasi-Ex) beverages on the anti-stress action. ICR male mice (20$\pm$2g) were fed basic experimental diets and given free through water bottle filled with these beverages instead of water for 18 days including sociopsychological stress. Body weight gains were consistently lower in Dasi-Ex and Fuco-I,II,III groups compared with control group, expecting in a inhibitory effect of obesity. Dasi-Ex group resulted in a significant decrease of 25% in serum corticosterone (CS) secretion, while Fuco-I,IIand IIIgroups resulted in a marked decreases of 45~55% in serum CS secretion compared with control group. Noradrenaline (NA) secretions were significantly increased about 15% in Dasi-Ex group, and 20~22% in Fuco-I,II,III groups compared with control group. Significant differences in brain MHPG-SO4 levels of Dasi-Ex group could not be obtained, but Fuco-I,II,III groups resulted in a marked decreases of 20~25% in brain MHPG-SO4 ratio of brain, but Fuco-I,II and III groups resulted in a marked increases of 45~60% in NA/MHPG-SO4 ratio of brain compared with control group. These results suggest that fucoidan beverage may play a effective role in a ridding of the sociopsychological stress by pivotal anti-stress effect of fucoidan.
Laminaria japonica is a type of brown algae widely consumed in Asian countries and contains many essential nutrients and exhibits anti-obesity, antioxidant, and anti-inflammatory effects. In this study, the antioxidant and immunomodulatory effects of a Laminaria japonica water extract (LJE) were investigated using an in vitro model. Mean total polyphenol content of LJE was 2.16±0.11 ㎍ GAE/mg, and LJE dose-dependently inhibited ABTS radical activity but did not scavenge DPPH radicals. In addition, LJE enhanced nitric oxide (NO) production and upregulated the mRNA expressions of proinflammatory cytokines (i.e., tumor necrosis factor-α and interleukin-6) in RAW 264.7 cells. On the other hand, LJE inhibited NO production and downregulated proinflammatory cytokine mRNA levels in endotoxin-stimulated RAW 264.7 cells. Thus, our data show that LJE has moderate antioxidant activity and biphasic immunomodulatory effects on RAW 264.7 cells. In summary, the study indicates that LJE has potential therapeutic use as a novel biphasic immuno-modulator.
Fucoidan has been shown to exhibit a host of biological activities, including anti-coagulant, anti-thrombotic, anti-tumourigenic, anti-inflammatory, anti-viral, anti-complementary and neuroprotective effects. In the present study, we attempted to determine the effects of Fucoidan on both apoptosis and cell proliferation in the hippocampal CA1 region and the dentate gyrus of gerbils after the induction of transient global ischemia. This experiment involved the use of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) assay as well as immunohistochemisty for caspase-3 and 5-bromo-2'-deoxyuridine (BrdU). The monosaccharide composition of the purified Fucoidan which had been extracted from Laminaria religiosa was utilized in this study. The present study clearly induces that apoptotic cell death and cell proliferation in the gerbil's hippocampal regions increased significantly following the induction of transient global ischemia and the results of this study also indicate that Fucoidan exerted a suppressive effect on this observed ischemia-induced increase in apoptosis within the CA1 and dentate gyrus, and also suppressed cell proliferation in the dentate gyrus.
The sea tangle, Laminaria japonica has long been used in Korea as a folk remedy to promote health. Gamma-amino butyric acid-enriched (5.56% of dry weight) sea tangle was obtained by fermentation with Lactobacillus brevis BJ-20 (FLJ). A suppressive effect of FLJ on carbon tetrachloride-induced hepatotoxicity has been shown previously. Alcohol administration to Sprague-Dawley rats leads to hepatotoxicity, as demonstrated by heightened levels of hepatic marker enzymes as well as increases in both the number and volume of lipid droplets as fatty liver progresses. However, FLJ attenuated alcohol-induced hepatotoxicity and the accumulation of lipid droplets following ethanol administration. Additionally, FLJ increased the activities and transcript levels of major alcoholmetabolizing enzymes, such as alcohol dehydrogenase and aldehyde dehydrogenase, and reduced blood concentrations of alcohol and acetaldehyde. These data suggest that FLJ protects against alcohol-induced hepatotoxicity and that FLJ could be used as an ingredient in functional foods to ameliorate the effects of excessive alcohol consumption.
