Quantitative Analysis of Microcystins, Cyanobacterial Toxins in Soyang Lake (소양호에서 남조류 독소, 마이크로시틴의 정량 분석)

  • It is very difficult to analyze the microcystins, cyanobacterial toxins quantitatively since it exists in a trace level in lakes. In this paper, two different analytical methods were tried to analyze the microcystins, cyanobacterial toxins quantitatively in water samples collected in Soyang lake. The first method was solid phase extraction method fol-lowed by High Performance Liquid Chromatography(HPLC), and the second method was Enzyme-Linked Immu-nosorbent Assay(ELISA) using the monoclonal antibody of microcystin.

Identification of Autumn Phytoplankton in the Lakes of Han River system (한강수계 호수에 출현하는 가을철 식물플랑크톤의 생태적 현황 연구)

  • Han River is very important as a source of drinking water in metropolitan area. This study was conducted to figure out diversity and dynamics of plankton community in the seven lakes (lakes Paro, Chunchon, Soyang, Uiam, Chungpyung, Paldang, Chungju) of Han Riversystem. A total of 76 genera and 121 species were investigated by taxonomic identification in November 2008. Cyclotella sp. and Microcystis sp. was a dominant species in lake Paro and Chungju respectively. Aulacoseira granulata was dominant in lakes Chunchon, Paldang and Chungpyung. And Fragilalia crotonesis was a dominant species in lake Soyang and Uiam. Our results can be useful when compared to the results using molecular biological method to supplement taxonomic identification.

Estimation of Atmospheric Mercury Wet-deposition to Lake So-yang (대기 중 수은의 습식 침적 평가: 소양호를 중심으로)

  • The important source of the mercury in water-column is the influx of atmosphere mercury, via dry and wet deposition. In this study, wet deposition of mercury was estimated to be $14.56{\mu}g/m^2$ during 15 months at the Lake Soyang, which is a little higher than those observed in the several rural US Mercury Deposition Network (MDN) sites with similar precipitation depth. The mercury concentration in precipitation did not show a positive correlation with atmospheric RGM (reactive gaseous mercury) concentration, while maintaining good correlation with atmospheric $PM_{2.5}$ at Soyang Dam. This result suggests that the contribution of particulate Hg to the total Hg wet deposition should be more significant than that of RGM. In this study, both precipitation depth and precipitation type affected the amount of wet deposition and the concurrent mercury levels in precipitation. There was generally an inverse relationship between precipitation depth and Hg concentration in precipitation. Precipitation type was another factor that exerted controls on the Hg concentration in precipitation. As a result, the highest concentration of Hg was observed in snow, followed by in mixture (snow+rain) and in rain.

A Study on the Ozonation of Aquatic Humic Substances(AHS) Extracted from Soyang Lake Water (소양호에서 추출한 수중 부식질(AHS)의 오존처리에 대한 연구)

  • Aquatic humic substance (AHS) was extracted from Soyang lake water using XAD-8 resin. and its concentration distribution in the lake from May to October. 1997 was determined. Ozonation characteristics of the AHS were studied as factors of pH, carbonate ion concentration. and the biodegradability and structural changes of the AHS were also investigated after ozonaticn. DOC distribution in Soyang Lake water was 1~3 mg/L. and the concentrations of AHS ranged between 0.2~0.8 mg/L. which was corresponding to 20~30% of DOC. AHS was composed of around 20% of HA and 80% of FA. The optimum pH value for AHS ozonation was in range of pH 7~9. When carbonate ions were added for AHS ozonation as a ladical scavenger. it was found that DOC removals were decreased. and the absorbance decreases were increased slightly. Biodegradability of the ozonized AHS was 50% higher than that of unozonated AHS. $^{13}C-NMR$ analysis showed that the aromatic compounds of AHS, after ozonation, were decreased from 49% to 17%. and the aliphatic and carboxylic compounds were increased from 34% to 51% and from 17% to 32%. respectively.

The Distribution of POC and DOC in Four Reservoirs on the North Han River and the Relationship with Algal Density (북한강수계 호수의 POC와 DOC 분포와 조류밀도의 관계)

  • Spatial and temporal distributions of POC and DOC were surveyed in the North Han River system, Korea The proportion of algal cells was calculated in four reservoirs (Lakes Soyang, Paro, Chunchon, and Uiam). Monthly average DOC concentrations ranged from 1.5 to 2.3 mg C/L, and POC showed larger variation than DOC (range 0.3 to 1.9 mg C/L). The average proportion of POC in TOC was higher than those of typical natural lakes. Due to the influence of the Asian summer monsoon, the seasonal variation in POC concentration depended on heavy rain events occurring during the summer. POC concentrations increased during the summer monsoon season due to turbid storm runoff laden with debris, while DOC concentrations did not increase. The highest POC concentrations were observed in Lake Soyang in 2006 when a severe rain event occurred. In two deep stratified reservoirs (Lake Soyang and Paro) storm runoffs formed an intermediate turbidity layer with high POC and chlorophyll concentrations which is thought to originate from terrestrial debris and periphyton transported by inflowing streams. The proportion of algal cells in total POC was much lower than for most natural lakes, and it varied with season; low in the monsoon season and high in dry seasons with algal blooms. An analysis of POC concentration and chlorophyll a concentration showed that the ratio of POC/Chl.a varied from 24 to 80.

Genomic Analysis of a Freshwater Actinobacterium, "Candidatus Limnosphaera aquatica" Strain IMCC26207, Isolated from Lake Soyang

  • Strain IMCC26207 was isolated from the surface layer of Lake Soyang in Korea by the dilutionto-extinction culturing method, using a liquid medium prepared with filtered and autoclaved lake water. The strain could neither be maintained in a synthetic medium other than natural freshwater medium nor grown on solid agar plates. Phylogenetic analysis of 16S rRNA gene sequences indicated that strain IMCC26207 formed a distinct lineage in the order Acidimicrobiales of the phylum Actinobacteria. The closest relative among the previously identified bacterial taxa was "Candidatus Microthrix parvicella" with 16S rRNA gene sequence similarity of 91.7%. Here, the draft genome sequence of strain IMCC26207, a freshwater actinobacterium, is reported with the description of the genome properties and annotation summary. The draft genome consisted of 10 contigs with a total size of 3,316,799 bp and an average G+C content of 57.3%. The IMCC26207 genome was predicted to contain 2,975 protein-coding genes and 51 non-coding RNA genes, including 45 tRNA genes. Approximately 76.8% of the protein coding genes could be assigned with a specific function. Annotation of the IMCC26207 genome showed several traits of adaptation to living in oligotrophic freshwater environments, such as phosphorus-limited condition. Comparative genomic analysis revealed that the genome of strain IMCC26207 was distinct from that of "Candidatus Microthrix" strains; therefore, we propose the name "Candidatus Limnosphaera aquatica" for this bacterium.

Comparison of TOC and DOC Distribution and the Oxidation Efficiency of BOD and COD in Several Reservoirs and Rivers in the Han River System (한강수계 주요하천과 호수내 TOC와 DOC분포 및 BOD와 COD의 산화율 비교)

  • The seasonal variation of organic matter and the oxidation efficiency of BOD and COD were investigated in several reservoirs and rivers located in the Han River system from March 2005 to April 2006. The concentrations of TOC in the Dong River, the Myeongjicheon stream and the Han-River varied with season. Mean concentration of TOC in the lotic environment ranged from 1.9 to 6.8 mgC/L. Oxidation efficiencies calculated from carbon were approximately 12.6 to 27.1% for BOD and 27.2 to 75.0% for COD, respectively. DOC to TOC ratio in the rivers ranged from 64.3 to 79.0%. Epilimnetic TOC concentrations in two clear and deep lakes (Lake Soyang and Paro) ranged from 2.0 to 2.8 mgC/L. TOC concentrations in the lower three dams (Lake Chunchon, Uiam, and Chungpyung) were relatively high that ranged from 3.4 to 3.7 mgC/L.Oxidation efficiencies in five lakes ranged from 10.6 to 17.2% for BOD and 32.6 to 49.5% for COD, respectively. The range of DOC to TOC in Lake Soyang and Paro was approximately 69. 0 to 73.4%. The ratios of lower three dams ranged from 48.2 to 62.3%. The differences among the lakes seemed to be asociated with limnological and hydrological characteristics of the lakes.

Water Quality and Phytoplankton Distribution Pattern in Upper Inflow Rivers of Lake Paldang (팔당호 상류 유입하천의 수질 및 식물플랑크톤 분포 패턴)

  • The distribution pattern of water quality parameters and phytoplankton biomass in upper inflow rivers of lake Paldang had surveyed. In North-Han river system, nutrient concentrations and algal biomass was below the standard of mesoand oligotrophy from lake Soyang to lake Paldang, maintaining good water quality, except the N5 site located near the Chuncheon-Si, showing high nutrients concentrations and algal biomass. The algal biomass of the South-Han river system showed oligotrophic level in the upstream near the lake Chungju, and increased along the flow direction showing eutrophic level in the downstream within the lake Paldang area. On the other hand, the highest concentrations of nutrients were detected in the middle stream near the Yeoju-Si and Yangpyung-Gun rather than in the downstream suggesting algal biomass in lake Paldang would not come from the upper river area but come from the growth within the lake area using nutrients from the upper inflow river.

Infection status of pond smelts, Hypomesus olidus, and other freshwater fishes with trematode metacercariae in 6 large lakes

  • In order to determine the current infection status of pond smelts, Hypomesus olidus, and other freshwater fishes with trematode metacercariae, a total of 4,861 pond smelts and 18 other freshwater fishes, collected from the Soyang and Uiam Lakes in Gangwon-do, the Unam Lake in Jeollabuk-do, the Jangseong Lake in Jeollanam-do, the Uirim-ji (lake) in Chungcheongbuk-do, and the Andong Lake in Gyeongsangbuk-do, were individually digested with 1 % pepsin-HCI and examined under a dissecting microscope. In all pond smelts caught from the 6 lakes, we were unable to detect any known human infectious trematode metacercariae in Korea. However, in other freshwater fishes, such as, Squalidus japonicus coreanus (Unam Lake), and Zacco platypus (Jangseong Lake) and Opsariichthys uncirostris amurensis (Jangseong Lake), metacercariae of human-infecting trematodes, i.e., Clonorchis sinensis and Metagonimus sp. were detected, respectively.

The Seasonal Variation of Active Bacterial Abundance in Lake Soyang (소양호에서 활성세균수의 계절적.수직적 변화)

  • Vertical and temporal variations of active and total bacterial abundance were monthly estimated in Lake Soyang from April 1999 to January 2000. The number of total and respiring bacteria was determined directly under microscope by AODC and CTC methods, respectively. The number of total and active bacteria varied from $2.1{\times}10^5 to 3.1{\times}10^6 $,$cells{\cdot}ml^{-1}$ and $1.8{\times}10^4 to 8.0{\times}10^5 $,$cells{\cdot}ml^{-1}$, respectively. The proportions of respiring bacteria to total cell ranged from 3.7 to 44.2% : The proportions was the highest in November 1999 and the lowest in December 2000. The specific activity of${\beta}$-glucosidase divided by total bacteria was$1.6{\times}10^5\;amol{\cdot}cell^{-1}{\cdot}hr^{-1}$in August and$1.4{\times}10^5\;amol{\times}cell^{-1}{\times}hr^{-1}$in September while the specific activity divided by CTC active bacteria was about$3.6{\times}10^5\;amol{\cdot}cell^{-1}{\cdot}hr^{-1},\;24.0{\times}10^5\;amol{cdot}cell^{-1}{cdot}hr^{-1}$. The specific activity of active bacteria in September was 6.7 times higher than that of August. By these data of active bacteria, the new information of aquatic ecosystem was unveiled.
