  • Both bark and wood sections obtained, by cutting with a razor blade, from living tissue of Cronartium ribicola cankers of Korean white pine (Pinus koraiensis) were transferred to a mixture of 2 parts of chloroform and 1 part of methanol, and the sections were stained using a modified lactophenol cotton blue. The formula for this staining is as follows: Lactic acid 20gm, phenol crystal 20gm, cotton blue 0.05gm, and 60% EtOH 100ml. The rust hyphae and haustoria were stained blue, and the wide hyphae with straight or curved haustoria could be distingwished from the pine tissue.

  • Fungus generally doesn't produce toxic or harmful substances so it has low chances to cause food poisoning. However it leads to change appearance, odor and characteristics of the contaminated foods and result in sanitary risk problems. Therefore the contamination of fungi should be prevented since they are not proper for human consumption. Green fungi with white outline raised from the air cell of cooked eggs which were collected by Gyeongi Livestock Veterinary Service in August, 2006. The results came out after the cultivation using Sabouraud's Dextrose Agar(SDA). The conidium appeared white and monospore, the shape of colony was round and oval. Conidiophore was brown and granulated and wrinkles and formed. It was confirmed as Aspergillus nidulans based on the dying using Lactophenol cotton blue, the observation of septum and vesicle from the grown spores, and rDNA sequencing.

  • The dermatophytes was isolated from skin of the 175 healthy dogs and 22 dogs with pathological skin lesions at Pohang and Kyongju. The isolates were identified by the morphological appearance after cultivation and lactophenol cotton blue staining. 1, The isolation rates of dermatophytes were 8.5%(15/175) in dog with healthy skin and 27% (6/22) in dogs with pathological skin lesions. 2. From asymptomatic dogs, the isolation rates of dermatophytes in female dogs were higher than those in male dogs and those in young dogs were higher than those in old dogs. 3. Isolation rates of microsporum canis and trichophyton spp from asymtomatic dogs were 14 (93%) and 1(6.7%), respectively. 4. The causative agents of 6 dogs with fermatophytosis were identified as all M canis.

Concentrations and environmental influences of airborne fungi at university laboratories, hospital diagnostic laboratories (대학실험실과 병원진단검사실에서의 부유진균 농도와 환경영향인자)

  • This study evaluated the airborne concentrations of fungi in university laboratories, hospital diagnostic laboratories in Seoul. The incubated fungi was identified by lactophenol cotton blue (LPCB) staining method. Variables such as types of ventilation, temperature and relative humidity were investigated to explain laboratory airborne fungal concentrations. A total of 97 air samples were collected from 10 facilities in two institutions. Aspergilus spp., including Aspergilus niger, Aspergillius flavos and Penicillium spp. were found as predominant species. Airborne fungal concentrations ranged from not detected (ND) to 1,890 CFU/$m^3$. Airborne fungal concentrations were high in general-ventilated facilities and in laboratories where relative humidity ( > 60 %) were high ( p < 0.001). Therefore, we suggest that relative humidity should be maintained to properly reduce the concentration of fungal in university and hospital laboratories.

Concentrations of Airborne Fungi and Environmental Factors in the Subway Stations in Seoul, Korea (서울지하철 일부 역사 내 부유 곰팡이 농도 및 환경요인)

  • Objectives: We measured the concentrations of culturable airborne fungi (CAF) in enclosed environments at 16 underground subway stations of the Seoul Metro in 2013, and investigated the effect of environmental factors, including temperature, relative humidity, the number of passengers, and distance from the platform. Methods: The cultured fungi were identified by the lactophenol cotton blue (LPCB) staining method and were classified by observing the form, shape, and color of colony. A nonparametric analysis was used to determine if the differences in the concentrations of CAF were statistically significant. Results: The concentrations of CAF at the stations were the highest in station p ($367CFU/m^3$) with arange between 3 and $437CFU/m^3$. There was a significant correlation between CAF concentration and the distance from platform (r = 0.544, p < 0.01). Geotrichum spp. and Penicillium spp. were the predominant species. Conclusion: It is recommended that special attention be given during rush hour, which is in the morning (08:00-10:00) and in the early evening (18:00-19:00) to improve the indoor air quality of the subway stations.

An Investigation on Concentration of Airborne Microbes in a Hospital (병원내 공기중 미생물의 농도에 관한 조사연구)

  • A survey was conducted to measure concentration of airborne microbe in a hospital using RSC air sampler during October~November 1991.The result was as follows: 1) In an agar strip GK-A media for total counts of microbial particles. The highest count were 1384 CFU/m$^3$ in the main lobby, followed by 912 CFU/m$^3$, in the obstetric room, 688 CFU/m$^3$ in 1CU. By gram staining, the distribution for organisms in the air were shown 74.1% in gram possitive cocci followed by 16.8%, in gram possitive bacilli 6.7% in gram negative bacilli and 4.7% in yeast, but low organism was detected in recovery room with 194 CFU/m$^3$. 2) In agar strip S media for Staphylococci the count at the main lobby was detected in the recovery room with 92 CFU/m$^3$, Tests of coagulase were negative Staphylococci with 78%, and positive Staphylococci with 22%. The Staphylococci were highly resistance to penicillin, ampicillin and sensitive to amikacin, cefazolin, gentamycin and chloramphenicol. 3) In agar strip C media for coliform bacteria the colony counts at the main lobby was 139 CFU/m$^3$ and treatment room was 190 CFU/m$^3$, most frequently isolated microorganisms were non fermentative bacilli. 4) In agar strip HS media for yeast and molds. Most frequently colony counts 17~76 CFU/m$^3$, 0.5% lactophenol cotton blue stains were shown unidentified 77.2%, 8.1%, in Penicillium 8.1% in Aspergillus, and 3.8% in mucor.
