육계 생산성 및 맹장내 미생물에 대한 유산균의 첨가 효과 (Effects of Supplemental Lactobacillus spp. on Performance and Cecum Microflora in Broiler)

  • This study was carried out to examine the effects of Lactobacillus spp. on performance and cecal microflora in broiler chicks. Eight diets consisting of basal diet(C), C+Lactobacillus crispatus BC7 (T2), C+Lactobacillus reuteri BC9 (T3), C+L.reuteri BC5+L.crispatus BC9 (T4), C+L. reuteri BC5 (T1)+L.reuteri BC9 (T5), C+L.crispatus BC7+L.reuteri BC9 (T6) and C+L.reuteri BC5+L.crispatus BC7+L.reuteri BC9 (T7), were fed to Ross male broiler chicks for 5 weeks. The level of supplemented Lactobacillus spp. was 107 cfu/g diet. Body weight and feed intake were measured every week, and cecal microfla was counted at 1 and 5 weeks for Lactobacillus and yeast. Body weight increased signigicantly in supplemental Lactobacillus treatments from 2 weeks of age (p<0.05). Chicks in T2, T5 and T5 of treatments were heavier than those of other treatments at 5 weeks of age(p<0.05). Viability was not different significantly. Feed intake and feed conversion also were not different, although feed conversion improved slightly in supplemental Lactobacillus treatments. Lactobacillus spp. of cecal content was increased in supplemental Lactobacillus treatments at 1 week, but no significance was found. The number of yeast in cecum was not different from that of supplemental Lactobacillus. In conclusion, Lactobacillus, L.crispatus BC7 and L.reuteri BC9 from broiler cecum could contribute to the increase in body weight with supplemented mono-or mixing of di-lactobacilli. The results indicate that Lactobacillus feeding can benefit to intestinal lactobacillus at early growing broiler.

In vivo Antagonistic Effect of Lactobacillus helveticus CU 631 against Salmonella enteritidis KU101 infection

  • In vivo antagonistic effect of Lactobacillus helveticus CU 631 and Lactobacillus spp. against typical enteritis causing pathogen Salmonella enteritidis KU 101 have been determined, which showed an increase in survival rate and the decline in viable cell numbers of pathogen in liver and spleen at sacrifice. A signifcant difference in the antagonistic effect against KU 101 were observed, which was species and/or strain dependent of Lactobacillus (p<0.01), the survival rate of the mice in the Salmonella infection by feeding L. helveticus CU 631 has been shown to be 157%, whereas those of L. rhamnosus GG ATCC 53103, L. acidophilus ATCC 4356, L. johnsonii C-4 were 137%, 132%, 119% respectively on the basis of lactobacilli non-associated control KU101 fed mice to be 100%. Viable cells of S. enteritidis KU101 in the liver and in the spleen at sacrifice were decreased in Lactobacillus spp. fed group with no significant difference. The higher level of total secretory IgA concentration in the intestinal fluid of lactobacilli fed mice than control mice have been observed. In vitro antagonistic activity of Lactobacillus spp. against KU101 have been determined, a prominent antagonistic activity of CU 631 against KU 101 were demonstrated.

유산균의 급여가 육계의 성장능력에 미치는 영향 (Effects of Supplemental Lactobacillus on Broiler Performance)

  • This study was conducted to investigate the effects of feeding various Lactobacillus on performance, nutrients digestibility, intestinal microflora, and fecal noxious gas of broiler chicks. One thousand eighty one dat old male broiler chicks were fed into Lactobacillus crispatus avibrd 1(LCB), Lactobacillus reuteri avibro 2(LRB), Lactobacillus crispatus avihen 1(LCH), and Lactobacillus vaginalis avehen 2(LVH) at the level of $10^{4}$ and 10(sup)7cfu/g diet. There were four replicates with thirty chicks per pen. Diets contained ME 3,100, 3,100kcal/kg, and CP 22.0, 20.0% for starting and finishing period, respectively. Weight gain of checks fed Lactobacillus spp. tended to increase from the first week and was higher from 50 to 100g in Lactobacillus treatments than control. Feed intake and feed conversion were not statistically different of all treatments. Dry Matter(DM) digestibility of Lactobacillus treatments was tended to improve compared to that of control, but was not significantly different. Protein and Ca digestibility were also tended to improve in Lactobacillus treatments relative that of control. Lactobacillus treatments showed improved tendency in crude ash and fat compared to those of control, whereas phosphorus digestibility was not consistency. Nutrients digestibilities of bird fed LCH were superior to those of other treatments. It showed significantly higher in Ca and P digestibility than control(P〈0.05). Total Lactobacillus spp. of birds fed various lactobacillus was significantly higher in ileum for five weeks(P〈0.05), but was not different at cecum. Yeast was thought to be not completely attached to intestinal lumen for one week. However, total number of yeast was significantly increased in cecum and ileum from three weeks old chicks(P〈0.05). The number of anaerobes exhibited to tendency the increase in Lantobacillus treatments from one week old of age at both ileum and cecum. Moisture contents of birds fed Lactobacillus spp. was shown from 27 to 30% at five weeks old, whereas it was 38.2% in control. It decreased approximately 25% in Lactobacillus treatments relative to that of control and showed significance between them(P〈0.05). Fecal $NH_{3}$ gas emission was significantly decreased in Lactobacillus treatments and maintained $\frac{1}{3}$to $\frac{1}{2}$ of control(P〈0.05). LCH and LVH treatments were most effective to decrease fecal $NH_{3}$ gas and especiallt impreved ti significantly at the level of the level of 10(sup)7cfu/g diets covpared to that of $10^{4}$ cfu/g diets and control(P〈0.05). In conclusion, feeding Lactobacillus spp. increase performance and nutrients digestibilities of broiler chicks and improve environments of house. Proper supplemental level of Lactobacillus spp. is considered more than $10^{4}$ cfu/g diet.

Effect of Lactobacillus rhamnosus hsryfm 1301 on the Gut Microbiota and Lipid Metabolism in Rats Fed a High-Fat Diet

  • Accumulating evidence indicates that lactic acid bacteria could improve host physiology and lipid metabolism. To investigate the effect of the gut microbiota on host lipid metabolism, a hyperlipidemic rat model was established by feeding rats a high-fat diet for 28 days, and the gut microbiota of the rats was analyzed using real-time PCR before and after administration of Lactobacillus rhamnosus hsryfm 1301 and its fermented milk for 28 days. The findings showed that the Lactobacillus spp., Bifidobacterium spp., Bacteroides spp., and Enterococcus spp. content in the hyperlipidemic rats gut was increased significantly (p < 0.05), while the Clostridium leptum and Enterobacter spp. content was decreased significantly after intervening with L. rhamnosus hrsyfm 1301 and its fermented milk for 28 days (p < 0.05). Furthermore, the lipid levels of the serum and the liver were decreased significantly (p < 0.05) and the fecal water content was increased significantly (p < 0.05) in the hyperlipidemic rats after the intervention, and hepatocyte fatty degeneration of liver tissues was also prevented. A positive correlation was observed between the Clostridium leptum content and the level of serum cholesterol, triglycerides, low-density lipoprotein, and high-density lipoprotein, and a negative correlation was observed between the Enterobacter spp. content and the Lactobacillus spp. and Bifidobacterium spp. content in the hyperlipidemic rats gut. These results suggest that the gut microbiota and lipid metabolism of hyperlipidemic rats could be improved by supplementation with L. rhamnosus hsryfm 1301 and its fermented milk.

양돈용 생균제 균주개발을 위한 유산균주 선발 (In vitro selection of lactic acid bacteria for probiotic use in pig)

  • In order to develop probiotic strain for pigs, Lactobacillus spp. (527 isolates), Streptococcus spp. (95 isolates) and Bifidobacterium spp. (25 isolates) were isolated from the feces of 35 pigs. These isolates were tested through in vitro experiment such as acid tolerance at pH 2.0 (Lactobacillus spp. and Streptococcus spp.) or pH 3.0 (Bifidobacterium spp.), bile tolerance in MRS broth containing 0.3% (w/v) Oxgall, heat resistance at $70^{\circ}C$ and $80^{\circ}C$ for 5 min, antibiotic resistance, antimicrobial activity against pathogenic bacteria and Caco-2 cell adherence assay. Finally ten most superior strain (5 Lactobacillus spp. strain, 3 Bifidobacterium spp. strain and 2 Streptococcus spp. strain) were selected as potential candidate for probiotic use in pig industry. It could be used as an alternative to antibiotics in feed additives.

쑥 추출물의 포도상구균과 질 유산균에 대한 선택적 저해효과 (Differential Inhibitory Effect of Artemisia Extract between Staphylococcus aureus and vaginal Lactobacillus spp)

  • 본 연구에서는 우리나라 민속약이나 한방의 약재로 널리 사용되어 온 참쑥 (Artemisia mongolica fishcher)으로부터 추출한 쑥 추출물이 포도상구균 (Staphylococcu aurues)과 유산균 (Lactobacillus spp.)에 대해 선택적 저해 효과를 갖는지 조사하였다. 우선 항균 활성 실험을 통해 포도상구균을 저해하는 우수한 항균력을 가진 유산균 7균주를 선별하였다. 그리고 쑥을 추출한 용매의 종류, 농도 그리고 추출시간을 달리하여 쑥 추출물의 최적 농도 조건을 결정하였다. 생존율을 측정하기 위해 배양액에 각각 3일 동안 용매 $100\%$ (아세톤, 에탄올, 메탄올)로 추출한 쑥 추출물을 $17\%$ 첨가하였고, 이때 항균 활성 있는 유산균 KLB 224은 배양 48시간까지 생존하였다. 그러나 포도상구균은 동일한 조건에서 모두 사멸되었다. 또한 주사전자현미경 관찰을 통해 쑥 추출물이 세포 형태에 미치는 영향을 확인하였다. 그 결과, 포도상구균은 세포 형태가 심하게 손상되었지만 유산균은 저해를 받지 않고 세포형태를 유지함을 관찰하였다.

Probiotic균주의 Pathogenic Organism에 대한 억제 활성과 송아지분변 분리균주의 억제활성 특성 (Inhibition Activity Against Pathogenic Organism of Probiotic Bacteria and Characterization of Inhibition Activity of Isolated Bacteria from Calf Dejecta)

  • 본 실험은 Lactobacillus spp.와 Bacillus spp. 의 Salmonella typhimurium, E. coli 및 Listeria monocytogenes에 대한 억제활성과 송아지 분변 분리균주의 생장 및 억제활성의 특성을 알아보기 위해 실시되었다.Lactobacillus spp.와 Bacillus spp.의 병원성 세균 Salmonella typhimurium에 대한 억제활성은 Lactobacillus helveticus CU631이 가장 높았고, Bacillus spp.는 활성이 약하였다. 송아지 분변 분리균주를 동정한 결과 Lactobacillus pentosus CU13과 CU05, Pediococcus pentosaceus CUR02, Lactococcus lactis ssp lactis CUM14로 확인되었다. Lactobacillus rhamnosus CU02와 Lactobacillus pentosus CU13의 Listeria monocytogenes에 대한 억제활성에 영향을 미치는 배지성분과 첨가수준은 Tween 80 1.0%, peptone 3.0%, yeast extract 3.0%, glucose 3.0% beef extract 3.0%, NaCl 1.0~3.0%이며, whole cell과 세포벽 물질은 Listeria monocytogenes에 대하여 억제활성을 나타내었다. 억제 성향을 강하게 보인 균체 배양액을 80℃로 열처리한 결과 억제력은 나타나지 않았으나 catalase 및 Proteinase-K 처리는 억제활성에 영향을 미치지 않았다. 이 결과 억제활성 물질은 유기산에 의한 것으로 사료된다. Lactobacillus pentosus CU13과 Lactobacillus rhamnosus CU02의 21균주에 대한 억제 능력을 측정한 결과 병원성균주를 포함한 16균주에서 억제활성을 보였으나 5균주에서는 억제성향을 나타내지 않았다. Escherichia coli O157:H7을 감염시킨 mouse 중 Lactobacillus pentosus CU13을 접종한 경우 다소 체중 회복 현상이 나타났으나, Lactobacillus rhamnosus CU02을 접종한 경우 체중 회복이 빠르게 나타났다.

Lactobacill spp. 의 황산화 효과 및 활성산소에 대한 내성 (Antioxidant Activity and Tolerance to Reactive Oxygen Species of Lactobacillus spp.)

  • 본 연구에서는 4종류의 Lactobacillus spp.의 항산화 효과와 활성산소에 대한 내성을 측정하였다. 그 결과 L. casei KCTC 3260의 항산화 효과가 intact cell에 서 36.9%, cell lysate에 서 79.8%로서 높은 항산화 효과를 나타내었다. 또 한 lmM 농도의 hydrogen peroxide에 서 생존성에 직접적으로 영향을 받지 않는 등 활성산소에 대해서 높은 내성을 나타내었다. 또한 높은 GPX 활성이 활성산소 하에서 L. casei KCTC 3260의 생존성에 중요한 역할을 하는 것으로 생각된다.

감마선 조사된 저염 오징어젓갈 발효의 미생물균총 특성 (Microbiological Characteristics of Gamma Irradiated and Low-Salted Fermented Squid)

  • 염 농도와 감마선 조사 선량을 달리한 오징어젓갈의 미생물 균총 변화를 살펴보았다. 총균, Lactobacillus spp., Staphylococcus spp.는 발효기간에 따라 지속적으로 생장하여 발효 40일째에 염도 5%와 10%에서는 $10^8\;CFU/mL$까지, 염도 20%에서는 $10^7\;CFU/mL$까지 생장하였으며 5 kGy 이상 감마선 조사구는 초기 균수 감소로 발효 후기의 미생물 밀도가 낮아졌다. 염도가 증가할수록 감마선 조사가 미생물 생장에 미치는 영향이 컸으며, 감마선 조사에 의한 오징어젓갈 관련 발효 미생물의 생장억제는 염도 10%에 10 kGy의 감마선 조사 조건이 적합하였다. 발효미생물의 동정 결과, L. plantarum이 전 실험구에 걸쳐 가장 일반적인 발효균으로 분리되었으며 Lactobacillus sp. 2, Mic. varians, Streptococcus sp. 1 등은 염도 5%의 실험구에만, Mic. morrhuae 등은 20%의 염농도에서만 분리되어 염도에 따른 microflora의 차이를 나타내었다. 또한, L. brevis, Ped. halophilus, Pse. diminuta 등은 방사선 감수성이 큰 미생물군이었으며, L. plantarum, Mic. morrhuae, Pseudomonas sp. 3 등은 방사선에 대한 저항성이 높았다. 전체적으로 보아 염도 20% 실험구에서의 분리 미생물이 15종으로 가장 다양한 분포를 보였으며 염도가 낮아질수록, 감마선 조사선량이 높아질수록 종의 다양성은 낮았다.

세가지 색상차이를 보이는 착색제를 이용한 치아 우식 관련 균에 관한 연구 (Study of Bacteria Associated with Dental Caries Using a 3 Tone Disclosing Agent)

  • 본 연구는 치태의 성숙도에 따라 치태를 서로 다른 색상으로 염색하는 GC Tri Plaque ID $Gel^{TM}$(GC corporation, Tokyo, Japan)을 이용하여 치아 우식 위험도를 평가하고자 하였다. 치아 우식의 발생 및 진행과 연관된 균인 Streptococcus mutans, Streptococcus sobrinus, Lactobacillus spp.의 수를 Quantitative real-time polymerase chain reaction (qRT-PCR)로 측정하여 치아 우식 위험도를 보았다. 본 실험은 강릉원주대학교 치과병원 임상시험 심사위원회의 심의를 받고 진행하였다. 강릉원주대학교 치과병원 소아치과에 내원한 전신질환이 없는 건강한 9 - 12세의 초등학생 15명의 치면을 착색제로 염색하였다. 치태의 성숙도에 따라 서로 다른 세가지 색상으로 염색되었으며 색상별로 3개의 실험군인 I군(pink/red), II군(blue/purple), III군(light blue)으로 나누었다. 3개의 실험군에서 각각 DNA를 추출한 후, qRT-PCR을 이용하여 S. mutans, S. sobrinus, Lactobacillus spp.의 수를 측정하였다. 3개의 실험군 사이에 S. mutans, S. sobrinus와 Lactobacillus spp. 균 수의 유의한 차이가 관찰되었으며 3종류의 균 모두 III군에서 가장 많이 관찰되었다(p < 0.05). GC Tri Plaque ID $Gel^{TM}$는 기존 착색제와는 달리 치태의 성숙도에 따라 서로 다른 세가지 색상으로 염색되며, 치태 염색 색상의 차이는 치아 우식 관련 균 수의 차이를 보여주었다. GC Tri Plaque ID $Gel^{TM}$이 치아 우식 위험도를 평가하는 하나의 지표로서 사용될 수 있는 가능성을 확인하였다.
