Characterization of Paraplantaricin C7, a Novel Bacteriocin Produced by Lactobacillus paraplantarum C7 Isolated from Kimchi

  • A Lactobacillus paraplantarum strain producing a bacteriocin was isolated from kimchi using the spot-on-the lawn method and named L. paraplantarum C7 [15]. The bacteriocin, paraplantaricin C7, was found to inhibit certain Lactobacillus strains, including L. plantarum, L. pentosus, and L. delbrueckii subsp. lactis. It also inhibited Enterococcus faecalis, yet did not inhibit most of the other LAB (lactic acid bacteria) tested. The maximum level of paraplantaricin C7 activity was observed under the culture conditions of $25^{\circ}C$ and a constant pH of 4.5. Paraplantaricin C7 retained 90% of its activity after 10 min of treatment at $100^{\circ}C$ and remained stable within a pH range of 2-8. Based on a culture supernatant, paraplantaricin C7 was purified by DEAE-Sephacel column chromatography and $C_{18}$ reverse-phase HPLC. SDS-PAGE and activity staining were then conducted using the purified paraplantaricin C7, and its molecular mass determined to be about 3,800 Da. The 28 N-terminal amino acids from the purified paraplantaricin C7 were determined, and the structural gene encoding paraplantaricin C7, ppnC7, was cloned by PCR using degenerate primers based on the N-terminal amino acid sequence. The nucleotide sequences for ppnC7 and other neighboring orfs exhibited a limited homology to the previously reported plantaricin operon genes. Paraplantaricin C7 is a novel type II bacteriocin containing a double glycine leader sequence.

Preventive Effects of a Probiotic Mixture in an Ovalbumin-Induced Food Allergy Model

  • Although there has been a steady increase in the prevalence of food allergies worldwide in recent decades, no effective therapeutic strategies have been developed. Modulation of the gut microbiota composition and/or function through probiotics has been highlighted as a promising target for protection against food allergies. In this study, we aimed to investigate the allergy-reducing effects of a probiotic mixture (P5: Lactococcus lactis KF140, Pediococcus pentosaceus KF159, Lactobacillus pentosus KF340, Lactobacillus paracasei 698, and Bacillus amyloliquefaciens 26N) in mice with ovalbumin (OVA)-induced food allergy. Administration of P5 significantly suppressed the oral OVA challenge-induced anaphylactic response and rectal temperature decline, and reduced diarrhea symptoms. Moreover, P5 also significantly inhibited the secretion of IgE, Th2 cytokines (interleukin (IL)-4, IL-5, IL-10, and IL-13), and Th17 cytokines (IL-17), which were increased in mice with OVA-induced food allergy, and induced generation of CD4+Foxp3+ regulatory T cells. These results revealed that P5 may have applications as a preventive agent against food allergy.

Influence of Extracellular Products from Haematococcus pluvialis on Growth and Bacteriocin Production by Three Species of Lactobacillus

  • The effects of Haematococcus pluvialis extracellular products on microbial growth and bacteriocin production were investigated to improve bacteriocin synthesis during the growth cycle of Lactobacilli. Lactobacillus pentosus KJ-108, L. plantarum KJ-10311, and L. sakei KJ-2008 were cultured in MRS and enriched medium (ERM) with or without supplement of the extracellular products obtained from a late exponential phase culture of Haematococcus pluvialis in modified Bold's basal medium (MBBM). In both MRS and ERM, the extracellular products strongly enhanced the growth as well as the bacteriocin production of all the lactic acid bacteria tested. The enhancing effect was observed in ERM with pH adjusted at 5 and 6. In addition, some difference in growth effects with the extracellular products of H. pluvialis was observed between pH 5 and 6 in ERM, but no effect was observed in the minimal medium. The final biomass and the final concentration of bacteriocin activity were associated with the cell growth that was promoted by the extracellular products of H. pluvialis, and the enhanced cell growth of the three lactic acid bacterial strains induced the increase of the specific bacteriocin production. Therefore, bacteriocin production and activity were influenced by the addition of the extracellular products of H. pluvialis in the culture medium.

  • 차세대식품으로 주목받고 있는 스피룰리나의 항균성 유산균(Probiotics: Lactobacillus Pentosus) 발효를 통해 얻어지는 항균성 발효스피룰리나의 국민건강증진 방편으로 이를 이용, 소비자 및 수요자의 접근을 용의하기위하며, 주 소비층인 환자, 노약자, 임산부, 어린이층의 Target content의 개발과 발효스피룰리나에 대한 시험결과 즉, 2010년 3월 락토바실러스 펜토수스 K34를 이용해 발효한 결과 발효공정을 통해 락트산; 아세트산; 구연산; 숙신산등의 유기산이 적어도 3종 이상 생성되었으며, 단백질 함량은 10% 이하로 변화를 보이지 않았고, 탄수화물의 거의 분해되어 락트산 및 프로피온산 등의 유기산으로 변화됨에 따라, 개발된 새로운 식품원료의 다양한 식품개발은 건강증진을 위해 소비수요층의 용의한 접근의 한 방편으로, 만화나 일러스트레이션을 이용한 접근을 통한 홍보 전략이 필요하다.

Innovative and practical conditioning beverages for public health and athletic performance: Focus on immunopotentiation by lactic acid bacteria B240

  • [Purpose] Functional beverages are a protective or enhancing factor influencing not only public health but also athletic performance. The purpose of this study was to highlight the new conditioning beverage of the Lactobacillus pentosus strain b240 (B240) with electrolytes or proteins, which strengthens immune functions to improve the quality of life. [Methods] ISeveral related studies systematically reviews three main issues associated with conditioning beverages: (a) utilization and availability of the functional beverage; (b) significance of B240 in immune strengthening; and (c) availability and application of conditioning drinks in the daily life and sports field. [Results] Intake of B240 led to greater enhancements, including blood T-helper, NK cell, IgA and IgG level in conjunction with strengthen immune functions. These results speculated that the practical application of B240 contained beverages on physiological health and performance. [Conclusion] BODYMAINTÉ, this novel conditioning beverage is expected biological utility responsible for improved sports performance as a functional drink and has potential health-related implications.

소금의 종류와 농도가 배추김치에서 분리한 유산균의 생육에 미치는 영향 (Effect of Salt Type and Concentration on the Growth of Lactic Acid Bacteria Isolated from Kimchi)

  • 소금의 종류에 따른 유산균 생육저해 효과를 보면, 김치초기 생육균인 W. confusa가 3.0%의 저염소금, 제재염에서 5시간의 유도기 후 급격히 증식했으나 3.0%의 천일염, 죽염첨가시 유도기간이 12시간으로 늘어나 이 소금들에 의해 생육이 약간 저해됨을 알 수 있었다. 중기 생육균인 Lactobacillus delbruekii ss lactis나 Lactobacillus pentosus와 말기 생육균인 L. hamsteri는 3.0%의 사용된 소금 모두에서 12시간의 유도기 후 증식이 확인되었다. 소금 농도를 달리하여 배지에 첨가했을 때 저염소금인 농도를 3.0%, 5.0%로 증가시킨 경우 W. confusa를 제외한 다른 균들의 유도기는 12시간으로 늘어났고 이 효과는 5.0%에서 더 컸다. 천일염, 죽염, 재제염도 같은 경향으로 농도가 커질수록 생육저해 효과가 뚜렷했으며 김치 초기 생육균 W. confusa보다는 후기생육균인 L. hamsteri가 소금에 의한 생육저해효과가 커서 5.0% 천일염, 죽염, 재제염에서는 24시간까지 증식이 억제된 것을 볼 수 있었다.

Polyphasic Microbial Analysis of Traditional Korean Jeung-Pyun Sourdough Fermented with Makgeolli

  • Jeung-pyun, a fermented rice cake, is prepared by fermenting rice sourdough using makgeolli, a traditional Korean rice wine, in the presence of yeast and lactic acid bacteria (LAB). The goal of this study was to conduct biochemical and microbial analyses of five different rice sourdoughs, each fermented with a different commercial makgeolli, using culture-dependent and culture-independent approaches. All sourdough samples fermented with different makgeolli for 6.5 h showed different profiles in pH, total titratable acidity, organic acid concentration, and microbial growth. LAB belonging to different genera were identified based on colony morphology on modified MRS and sourdough bacteria agar medium. PCR-denaturing gradient gel electrophoresis analyses of the five sourdoughs showed different bands corresponding to LAB and yeast. 16S/26S rRNA gene sequence analyses of the samples confirmed that the predominant LAB in the five fermented rice doughs was Lactobacillus plantarum, Lb. pentosus, and Lb. brevis. Various other Lactobacillus spp. and Saccharomyces cerevisiae were common in all five fermented samples. This study provides comprehensive and comparative information on the microflora involved in fermentation of rice sourdough and signifies the need to develop effective starters to enrich the quality of jeung-pyun.

A Synergistic Effect of Chitosan and Lactic Acid Bacteria on the Control of Cruciferous Vegetable Diseases

  • Two lactic acid bacteria (LAB) designated J02 and J13 were recovered from fermented vegetables based on their ability to suppress soft rot disease caused by Pectobacterium carotovorum subsp. carotovorum (Pcc) on radish. J02 and J13 were identified as Lactobacillus pentosus and Leuconostoc fallax, respectively. The ability of J02 and J13 to suppress plant diseases is highly dependent on chitosan. LAB alone has no effect and chitosan alone has only a moderate effect on disease reduction. However, J02 or J13 broth cultures plus chitosan display a strong inhibitory effect against plant pathogens and significantly reduces disease severity. LAB strains after being cultured in fish surimi (agricultural waste) and glycerol or sucrose-containing medium and mixed with chitosan, reduce three cruciferous vegetable diseases, including cabbage black spot caused by Alternaria brassicicola, black rot caused by Xanthomonas campestris pv. campestris, and soft rot caused by Pcc. Experimental trials reveal that multiple applications are more effective than a single application. In-vitro assays also reveal the J02/chitosan mixture is antagonistic against Colletotrichum higginsianum, Sclerotium rolfsii, and Fusarium oxysporum f. sp. rapae, indicating a broad-spectrum activity of LAB/chitosan. Overall, our results indicate that a synergistic combination of LAB and chitosan offers a promising approach to biocontrol.

생약 추출물이 유산균의 생육 및 동결건조 보호제로서의 효과 (Enfluence of Herbal Extract on Lactic Acid Bacteria Growth and Cryoprotectants)

  • 가시오가피, 구기자, 당귀, 오미자의 열수 추출물이 유산균의 생육과 동결건조시 보호제로서의 역할 및 동결건조된 유산균원료의 저장 시 생존율에 미치는 영향을 조사하였다. MRS 배지에 생약 추출물을 첨가한 경우 유산균주 16종에 대하여 가시오가피, 구기자, 당귀 및 오미자 추출물을 MRS broth에 각각 50%(v/v)씩 첨가한 배지에서의 생육상태를 조사하였다. 가시오가피 추출물을 첨가한 경우 L. acidophilus, L. bulgaricus, L. paracasei, L. casei, L. pentosus 균주가 생육이 양호하였으며 구기자 추출물을 첨가한 경우 L. acidophilus, L. bulgaricu L. paracasei 등이 양호하였고 당귀 추출물에서는 L. acidophilus, L. bulgaricus, L. casei, L. plantarum, L. lactis 등이 잘 생육하였으며 오미자 추출물에서는 L. acidophilus, L. rhamnosus, L. pentosus 등이 양호하였다. 기본 동결보호제로 조성으로 구성된 skim milk 10%(w/v)와 sucrose 5%(w/v)에 생약 추출물 2.5%(v/v)를 첨가한 경우 동결건조 직후 생존율은 L. acidophifus, L. bulgaricus, L. paracasei, L. casei가 평균 각각 81.7%, 63.8%, 73.2%, 78.1%를 나타냈다. 동결건조한 유산균 원료를 온도 4$0^{\circ}C$, 습도 70% 유지하는 항온항습기에서 한달간 방치시킨 후 생존율은 L. acidophilu의 경우 대조군에 비해 약 6%, L. paracasei와 L. bulgaricu의 경우 약3%의 생존율 증가를 보여 가혹조건에서 균의 생존율을 연장시키는데 역할을 하는 것으로 보이며, 구기자 추출물을 2.5% 첨가하여 동결건조 한 균 원료의 경우 L. paracasei에서 약 1.5% 정도 증가를 나타냈으며 L. aoidophilus와 L. bulgariolis에서는 약 1% 정도 감소하는 경향을 보였다. 특히, 구기자 추출물의 경우 L. acidophilus와 L. bulgaricus에서 당귀 추출물의 경우 L. bulgaricu와 L. casei에서 동결 보호제로서 이용할 수 있을 것으로 사료된다.

굴 효소 가수분해물 첨가 요구르트의 제조 및 특성 (Preparation and Characterization of Enzymatic Oyster Hydrolysates-added Yogurt)

  • 양식산 굴에 대하여 Protamex(1단 가수분해)와 Neutrase (2단 가수분해)로 가수분해하고, 한외여과장치(MWCO, 3 kDa)로 분획한 획분을 동결건조한 다음, 이의 효율적 이용을 위하여 굴 효소가수분해물(OHP) 첨가 기능성 강화 요구르트의 제조를 시도하였고, 아울러 그 특성에 대하여도 살펴보았다. pH, 적정산도, 점도, 생균수 및 관능평가를 통해 OHP 첨가 요구르트의 최적 starter는 lactobacillus bulgaricus와 Streptococcus thermophilus의 1:1의 비율로 혼합한 균주로 선정되었다. OHP의 첨가량별 최적 발효시간은 무첨가 제품 8시간, OHP 0.5 제품이 6.5시간, OHP 1.0 제품은 5.5시간, OHP 1.5 제품은 5.0시간, PHP 무첨가 대조군 및 시판 요구르트에 비해 우수하였고, 관능검사 결과 OHP를 1.0 g 첨가한 제품이 가장 적합하였다. OHP 1.0 제품의 일반성분, pH 4.31, 적정산도(1.07%) 및 생균수 $4.9{\times}10^8\;CFU/mL$는 시판 요구르트에 비하여 조단백질의 함량만이 다소 높은 반면, pH, 적정산도 및 생균수는 차이가 없었다. 저온저장 중 시제 요구르트의 pH, 적정산도 및 생균수는 차이가 없었다. 저온저장 중 시제 요구르트의 pH 적정산도 및 생균수의 변화 결재로 미루어 보아 OHP 첨가 요구르트는 $5^{\circ}C$에서 15일간 유통 가능할 것으로 판단되었다.
