Lactobacillus spp.와 Bacillus coagulans의 Glutathione Sulphydryl 함유율과 황산화 활성 (Glutathione Sulphydryl Contents and Antioxidant Activities of Lactobacillus spp. and Bacillus coagulans)

  • 활성산소의 세포막 파괴 효과로부터 보호작용과 항산화 활성이 강한 균주를 선별하기 위하여 33균주에 대한 세포네 glutathione 함유율과 linoleic acid 산화억제활성을 측정하였다. Lactobacillus casei CU4114 25.15 umol/g으로 가장 높은 함유율을 보이고 있으며Lactobacillus rhamnosus CU4201, Lactobacillus plantarum CU4203이 14.0umol/g으로 통계적인 유의성을 나타내는 수준으로 높은 함유율을 나타내었다. 포자형 성균주중의 glutathione 함유율 측정 결과 Lactobacillus sporogenes CU4109, Sporolactobacillus inulinus CU4110 은 Bacillus coagulans 균주의 GSH 함유율보다 유의성이 인정되는 높은 함유율을 나타내었다. L. acidophilus CU4111, L. casei CU4114, L. brevis CU4206, L. helveticus Cu4231은 Lactobacillus spp. 중 높은 항산화 활성을 나타내었고 Bacillus coagulans의 항산화 활성범위는 6.02${\sim}$51.72%로 나타났으며 Bacillus coagulans 균주의 항산화 활성이 Lactobacillus spp.의 산화억제활성보다 높은 성향을 나타내었다.

Antimutagenic Activities of Cell Wall and Cytosol Fractions of Lactic Acid Bacteria Isolated from Kimchi

  • Cell wall (lactic acid bacteria-sonicated precipitate ; LAB-SP) and cytosoll(lactic acid bacteria-sonicated supernatant ; LAB-SS) fractions were prepared from kimchi fermenting lactic acid bacteria such as Leuconostoc mesenteroides, Lactobacillus brevis, Lactobacillus fermentum , Lactobacillus plantarum and Pediococcus acidilactici, with Lactobacillus acidophillus isolated from yogurt. Using the Ames mutagenicity test and SOS chormotest system, the antimutagenic acitivity of those cell fractions was studied . One hundered eighty $\mu$l of LAB-SP from lactic acid bacteria isolated from kimchi, excepting Pediococcus acidilactici, supressed the mutagenicity of 4-nitroquinoline-1-oxide(4-NQO) in Ames mutagenicity test and SOS chromotes system , by above 90% and 60% , respectively. LAB-SP from lactic acid bacteria also inhibited the mutagenicity mediated by 3-amino-1-methyl-5H-pyrido [4,3-b]indole (Trp-P-2). Lactobacillus fermentum, Lactobacillus plantarum, and Lactobacillus acidphillus had higher antimutagenicity against Trp-P-2). Lactobacillus fermentum , Lactobacillus plantarum , and Lactobacillus acidphillus had higher antimutagenicity against Trp-P-2 than the other lactic acid bacteria. However, LAB-SS of lactic acid bacteria did not show any mutagenic activity against 4-NQO in Ames mutagenicity test and SOS chromotest systems. On the mutagenicity of MEIQ and Trp-P-2 , LAB-SS of lactic acid bacteria from kimchi or dairy products exhibited a weaker inhibitory effect than LAB-SP of those bacteria. These results represent that, whether the lactic acid bacteria from kimchi are viable or nonviable, antimutagenic acitivity was still effective. We suggest that the strong, antimutaganic activity of lactic acid bacteria might be found in the cell wall fraction , rather than in the cytosol fraction.

Immunostimulation Effects of Cell Wall Components Isolated from Lactobacillus plantarum

  • Immunostimulation effects of the cell wall components isolated from Lactobacillus plantarum were investigated by studying the macrophage s tumorcidal activity, splenocyte proliferation, anticomplementary activity and the inhibition of peritoneal tumor cell growth measured with ICR mice inoculated with sarcoma 180. The immunopotentiating cell wall components were a complex of peptidoglycan and exopolysaccharides. The tumorcidal activity of macrophage against Yacl and B16 tumor cells was enhanced when the cell wall components were added into the macrophage s culture medium. They also stimulated splenocytes to proliferate up to the same level as when the concanavalin A was added into the splenocyte's culture medium. The complementary activity was inhibited by 50% when the cell wall components were incubated with the sheep red blood cells treated with hemolysin and guinea pig complement. This result confirmed that the cell wall components had an antitumor effect, because the anticomplementary activity is usually accompanied by an antitumor activity at the same time. This fact was confirmed again by the inhibition of the growth of sarcoma 180 when the cell wall components were injected intraperitoneally into ICR mice inoculated with sarcoma 180. As a result, it is concluded that the cell wall components isolated from Lactobacillus plantarum had multifunctional immunostimulation effects in vitro and in vivo.

Evaluation of Optimum Conditions for Bacteriocin Production from Lactobacillus sp. JB-42 Isolated from Kimchi

  • Bacteriocin-producing microorganism was isolated from Kimchi. The microorganism was identified as a Lactobacillus sp. and named Lactobacillus sp. JB-42. The optimum conditions for the bacteriocin production from the isolated microorganism were evaluated. For the maximum yield of bacteriocin from Lactobacillus sp. JB-42, the cell should be harvested at the early stationary phase and the temperature, pH and NaCl concentration should be $30^{\circ}C$, pH 7 and without the addition of NaCl, respectively. Sucrose, glucose or fructose should be used as a carbon source and organic nitrogen such as tryptone should be used as a nitrogen source for the best yield. The production of bacteriocin was related to the cell growth of Lactobacillus sp. JB-42 indicating the role of Lactobacillus in the Kimchi fermentation process.

  • The optimal conditions for protoplast fusion between the cells of kanamycin resistant Lactobacillus bulgaricus IFO 13593 and those of lincomycin resistant Lactobacillus helveticus IAM 12090 were investigated in this study. The highest fusion frequency of 9.1*10-4 was obtained when protoplast fusion was carried out for 0.5 min using 40% PEG 4000 solution as a cell fusion stimulant and subsequent post-PEG-incubation was undergone at 30 for 30 min in the PPI medium.

김치 유래 젖산균의 Cell-envelope Proteinase 존재 확인 (Identification of the Cell-envelope Proteinase of Lactic Acid Bacteria Isolated from Kimchi.)

  • 김치로부터 분리된 젖산균들의 16S rDNA 부분 염기서열 결정 결과, 6균주 중 2균주는 Leuconostoc mesenteroides로, 나머지 균주는 Lactobacillus속으로 재동정되었다. 이들 김치유래 젖산균들은 유제품 유래 젖산균과 마찬가지로 cell-envelope proteinase (CEP)로 추정되는 세포외 proteinase 활성을 보유하고 있는 것으로 나타났고, 젖산균의 CEP 특이적 primer를 제작하여 PCR을 수행한 결과, Leu. mesenteroides로부터는 예상 크기의 PCR 산물이 증폭되지 않았지만 Lactobacillus속 균주들로부터는 예상되는 1.2 kb 의 DNA 단편이 증폭되었다. Lactobacillus pentosus KFR1821의 genomic DNA로부터 증폭된 PCR 산물의 추정 아미노산서열은 Lactococcus lactis subsp. cremoris의 PrtP와 95%, Labtobacillus paracasei subsp. paracasei이 PrtP와 92%의 높은 상동성을 보였다. 증폭된 PCR 산물을 probe로 하여 Southern hybridization을 수행한 결과, Lb. pentosus KFR1821의 CEP 유전자는 chromosomal DNA에 암호화 되어있는 것으로 확인되었다.

뽕잎 장아찌로부터 분리된 Lactobacillus plantarum 균주의 유해균 증식 억제 활성 (Antipathogenic Activity of Lactobacillus plantarum Isolated from Pickled Mulberry Leaf)

  • 전국 각지에서 수집된 발표식품으로부터 젖산균을 분리하고 유해균인 B. cereus, L. monocytogenes, S. Typhimurium, S. aureus 및 S. epidermidis 균주에 대하여 생육 억제효능이 강한 L2167 균주를 선발하고, 16S rDNA 분석법을 이용하여 L. plantarum로 동정하였다. L2167 균주의 단백질 분획물을 제작하여 S. aureus 등 5종의 유해균 배양액에 첨가한 후, 전자현미경으로 관찰하였다. L. plantarum L2167 균주의 균체 단백질 분획물은 유해균의 생육을 억제하였으며, 생육 억제활성은 균체 내부에 함유된 단백질이 유해균의 세포막을 파괴하는 것에서 기인하는 것으로 추정되었다. L. plantarum L2167은 pH 5.5로 조절한 MRS 배지에서 35℃로 배양하였을 때, 가장 우수한 비성장속도를 나타냈으며, 대조구로 사용된 L. plantarum KCTC21004보다 NaCl에 대하여 민감한 특징을 나타내었다. 본 연구를 통하여 선발한 L. plantarum L2167 균주는 추가연구를 통하여 유해균의 생육을 효율적으로 억제할 수 있는 스타터 균주로서 개발될 수 있을 것으로 기대된다.

혐기적 조건하에서 젖산균의 cadmium 축적 (Accumulation of Cadmium in Lactic Acid Bacteria under the Anaerobic Conition)

  • In this study, authors investigated the cadmium tolerance, the accumulation of cadmium, and the cellular distribution of accumlated cadmium in lactic acid bacteria under the anaerobic condition. Lactic acid bacteria grew fairly well in modified EG medium containing 10 ppm of cadmium but could hardly grow at 50 ppm of cadmium. Tolerance to cadmium of genus Lactobacillus was greater than that of genus Streptococcus, and Lactobacillus acidophilus showed the higest cadmium tolerance amomg the bacteria tested. The capacity of cadmium accumlation (9.304-12.428 mg/g wet cell) of lactic acid bacteria was higher than that (6.775 mg/g wet cell) of Escherichia coli. Lactobacillus casei of them took up the largest amount of cadmium. The cadmium elimination amount (28.46-29.25%) of lactic acid bacteria from modified EG medium containing cadmium were also higher than that (14.43%) of Escherichia coli. Accumulated cadmium in Lactobacillus acidophilus was distributed by 42.41% at cell wall, 28.97% at cytoplasm, and 28.62% at plasma membrane, respectively.

세라믹막을 이용한 Lactobacillus cell의 농축 공정의 특성 (Characteristics of the Concentration Process of Lactobacillus Cell Using a Ceramic Membrane)

  • 본 연구에서는 유기질막에 비해 안정성이 우수하고 사용수명이 긴 세라믹 막을 이용하여 혐기성 Lactobacillus 균주의 농축에 관해서 연구하였다. Cell harvesting (CH)에 영향을 주는 인자로 막투과 압력, 온도, 선속도 등에 대해 조사하였으며 세라믹 막을 이용하여 농축율 변화에 따른 플럭스와 TMP (transmembrane pressure)의 변화, 일정 VCF(volumetric concentration factor)에서 TMP변화 등의 최적조건에 관해 알아보았다. 그 결과 플럭스는 TMP, 선속도, VCF가 증가함에 따라 투과수량도 증가함을 알 수 있었다. 온도가 증가할수록 점도에 의해 초기 플럭스도 증가하지만 시간이 경과함에 따라 막 표면의 겔층의 형성으로 온도의 영향은 미미하였다. 또한 농축비 이하의 일정한 VCF에서 운전할 경우 플럭스가 안정적임을 알 수 있었다. 선속도 5 m/s, 공급액의 온도 $37^{\circ}C$, TMP 1 bar에서 6∼8 h 운전할 경우 PS 406 원액의 생균수는 4.9{\times}10^9$으로 약 8배 농축됨을 알 수 있었다.

Lactobacillus crispatus 생균 생산을 위한 whey 배지 최적화 (Production of Viable Lactobacillus crispatus by Using Whey Based Medium)

  • Whey based medium was optimized for the production of viable Lactobacillus crispatus KLB 46 isolated from the vagina of Korean women. Among the various nitrogen sources such as yeast extract, beef extract, and proteose peptone no. 3 supplemented to whey, beef extract showed the highest viable cell production. The addition of Tween 80 to the whey based medium increased viable cell concentration. As beef extract supplementation is not economically attractive, corn steep liquor was added as a supplementary nitrogen sources. When corn steep liquor was supplied with beef extract with the ratio 5 : 1, the viable cell count was $3.11{\times}10^9$ CFU/mL. Also, the addition of mineral salts containing sodium acetate (5 g/L), potassium phosphate dibasic (2 g/L), magnesium sulfate (0.1 g/L) and manganese sulfate (0.05 g/L) to the whey medium increased viable cell count further ($5.00{\times}10^9$ CFU/mL).
