배양조건에 따른 Lactobacillus casei YIT 9018의 니산성 변화 (Influence of Culture Comditions on Acid Tolerance of Lactobacillus casei YIT 9018)

  • We studied the influence of culture conditions on the acid tolerance of Lactobacillus casei YIT 9018 in artificial gastric juice with respect to relative amount of membrane bound ATPase and their biochemical characteristics. With raising incubation temperature from 30.5$\circ$C to 40.5$\circ$C and lengthening incubation time from exponential phase to late stationary phase, the acid tolerance of L. casei YIT 9018 was increased. As acid tolerance enhanced, C$_{18:1}$ content of membrane fatty acid was reduced and C$_{19:0 cyclo}$ was enriched but the others were not changed greatly. At high ATPase activity, proton permeability was relatively low but this phenomenon did not correspond to acid tolerance. In conclusion, it was considered that changes of C$_{18:1}$ and C$_{19:0 cyclo}$ were closely related to the acid tolerance of L. casei YIT 9018.

Lcatobacillus casei YIT 9018로 부터 분리한 Prophage Cured Strain의 특성 (Characterization of Prophange Cured Strain Derivative from Lactobacillus casei YIT 9018)

  • Lactobacillus casei HY 2782, prophage cured strain was characterized to be stable as much as L casei YIT 9018, parent strain. By southern hybridization, it was confirmed that the temperate phage was incorporated in chromosomal DNA of L. casei YIT 9018 as a prophage. It was also proved that the prophage was cured from chromosomal DNA of L casei HY 2782. The growth rate, lactic acid producing ability, carbohydrates fermentation, and enzymatic activity of L. casei HY 2782 were found to be similar to those of L. casei YIT 9018. When L casei HY 2782 was used as a host, the multiplicity of infection (M.O.I.) of the temperate phage for L. casei HY 2782 was 1.0~5.0. Restriction enzyme analysis of pLC90 plasmid from L. casei HY 2782 was shown that the size was an approximately 68.22 kb. The plasmid profiles, genomic DNA patterns, and cellular fatty acids composition of L. casei HY 2782 were similar to those of L casei YIT 9018. And the major fatty acids composition of these strains were C$_{14;0}$,C$_{16;1}$, C$_{16;0}$, C$_{18;1}$ and C$_{19;cyclo-}$ 10 sets of arbitrary primer in the PCR were screened to find differentiation against two strains of L. casei. Among them, b$_{5}-1/17-1 primer was produced an approximately 1.3 kb DNA band of only L casei YIT 9018. And b$_{5}-2/17-2 primer was produced an approximately 1.0 kb DNA band of only L casei HY 2782.

Lactobacillus casei YIT 9018의 Shuttle Vector 개발을 위한 분자유전학적 연구

  • A shuttle vector, pSHvec, was constructed for Lactobacillus casei (L. casei) YIT 9018 and JM1O9 by recombinant DNA technology. This vector contained the $\beta$-lactamase II gene from Bacillus cereus as a selection marker and replication origins for both Gram(+) and Gram(-) strains. It could transform the wild type L. casei YIT 9018 as well as E. coli JM109 and transformed cells were selected based on antibiotics resistance. The ability of L. casei YIT 9018 for curd formation in 11% skim milk was maintained even after transformation with pSHvec. The vector was stable as long as antibiotics were added to the medium. These results could contribute to the study of lactic acid bacteria for the industrial purpose at a genetic level.

  • L.casei YIT 9018균주에 N-methyl-N'-nitro-N-nitrosoguanidine을 처리하여 thermoinducible mutants를 분리하였고, 이 균주를 $42^{\circ}C$에서 30분 동안 열처리하여 prophage가 cured된 균주 L.casei HYM 1213과 L.casei HYM 4024를 분리하였다. Prophage cured strain L.case HYM 1213과 L.casei HYM 4024는 temperate phage $\phi$Fsw에 대해 지시균으로서 능력을 가지고 있었으며, 어떤 온도에서도 temperate phage $\phi$FSW가 검출되지 않았다. 이들의 생리적 특성을 모균주인 L.caseiYIT 9018과 비교한 결과 L.casei HYM 1213 균주는 균의 증식, pH변화, 산생성능력, 당 발효능력에서 모균주와 유사한 것으로 나타났으나, L.casei HYM 4024균주는 pH변화와 산생성능력이 모균주에 비해 미약한 것으로 나타났다. Plasmid DNA는 두 균주 모두 모균주와 동일한 size를 보여주여, prophage가 cured되었어도 plasmid DNA에는 손실을 가져오지 않았음을 알 수 있었다.

  • L. casei Y1T 9018은 자연적으로, 또는 mutagen처리하여 plasmid DNA 7가 curing 또는 deletion되었으며, 이러한 유천적 결함이 생긴 mutants는 다음과 같은 특성을 나타냈다. 1) 탄소원으로서 lactose가 주어졌을때 mutants의 유산 생성능이 현저히 저하되었다. 2) 탄소원으로서 glucose 맺 galactose 첨가시엔 유산 생성 정도가 정상균주와 큰 차이가 없였다. 3) 5 가시 항생제에 대한 내성 맺 lactose 이외의 당에 대한 발효능 시험 갤고파 정상과 차이가 없었다.

  • L. casei YIT9018에서 자연적으로 발생하는 Lac$^-$ mutant의 발생빈도와 이러한 변이주의 특성을 검토하였다. 보존균주로부터 spontaneous Lac$^-$ mutant의 발생은 MRS 배지에서 0.5%, 10% 환원탈지유에서 0.29%, pure Lac$^+$ colony로부터의 발생은 MRS 배지에서 0.25%, 10% 환원탈지유에서 0.1%의 빈도로 나타났다. 분리된 spontaneous mutant는 대부분이 Lac$^-$ mutant였고 가끔 Lac-Gal-도 나타났다. Lac$^-$ Gal$^+$ mutant는 배양 4일째부터 partial revertant가 출현하였으나 Lac$^-$ Gal$^-$는 전혀 나타나지 않았으며 Lac$^-$ mutant는 wild type보다 우유를 응고시키는데 더 많은 시간을 요하였다.

  • 본 연구는 유산균발효유 제조에 많이 사용되는 Lactobacillus casei YIT 9018에 광범위 숙주영역을 갖고 있는 Streptococcus faecalis DS5 의 plasmid pAM $\beta_1$ DNA를 이용하여 전이, 도입하고자 유당발효능결손변이주를 이용하여 원형질체융합, 접합, 형질전환을 검토하였던 바 얻어진 결과는 다음과 같다. L.casei와 S.faecalis 균주의 conjugation 은 membrane filter mating(Milipore, 0.45$\mu \textrm m$, front side)의 경우 $6.9\times 10^7$으로 가장 높은 전이율을 나타냈으며 protoplast fusion이나 transformation에 의해서는 fusant나 transformant를 선별할 수 없었다.

  • Lactobacillus spp. 급여에 의하여 Salmonella enteritidis 감염에 의한 치사율의 저하 성향, 시험관내 억제활성, 장액 내의 총 IgA 농도변화와 Lactobacillus casei.의 16S-23S rRNA intergenic spacer region의 sequence를 측정 비교한 결과는 다음과 같다. Lactobacillus spp. 5균주는 시험관내 Salmo- nella enteritidis 억제활성 측정 결과 모든 시험균주가 억제활성을 나타내었고 그 억제활성의 차이가 통계적인 유의성을 보였으며, 억제활성의 차이는 L. helveticus CU 631 > L. rhamnosus GG ATCC 53103 > L. johnsonii C-4 > L. acidophilus ATCC 4356 > L. casei YIT 9018 인 것으로 나타났다. Lactobacillus spp. 급여에 의하여 Salmonella enteritidis challenge에 의한 치사율 저하효과는 Lb. helveticus CU 631은 대조구의 생존률 62%에 대하여 100%로서 가장 높은 치사율 저하효과를 나타내었고, L casei YIT 9018, L johnsonii C-4의 생존률이 70%와 50%를 나타내었다. Lactobacillus spp.의 16S-23S rRNA intergenic spacer region의 sequence를 측정하고 gene bank에 등록된 균주의 sequence와 비교한 결과 homology의 차이를 나타내었다.

Effects of Feeding Lactobacillus spp. on the Level of Cell Glutathione Sulphydryl and Immunoglobulin M in ICR Mice

  • Effects of feeding seven strains of Lactobacillus spp. on the level of cell glutathione sulphydryl (GSH) in spleen, liver and erythrocyte of the ICR mice and on the level of immunoglobulin M in the spleen were determined. The level of cell glutathione sulphydryl in the spleen was dependent on the strain of Lactobacilli, it was significantly higher in the mice fed with L. casei CU 001, L. rhamnosus CU 02, L. acidophilus NCFM and L. casei YIT9018 (p>0.05). The level of cell glutathione sulphydryl in the liver increased in the mice fed with L. casei YIT9018, L. acidophilus NCFM, L. casei CU 001 (p>0.05), the level of glutathione sulphydryl of the erythrocyte showed significantly higher value than control mice when fed with L. acidophilus NCFM, L. casei YIT9018, L. casei CU 001 (p>0.05). The level of immunoglobulin M in the spleen of ICR mice expressed as the plaque count revealed significantly higher value than the control mice when fed with L. casei CU 001, L. acidophilus NCFM and L.casei YIT 9018.

Phage ${\phi}$ FSV가 Lactobacillus casei의 증식에 미치는 영향 (Effect of the Phage ${\phi}$ FSV on the Growth of Lactobacillus casei)

  • In order to study effect of the phage ${\phi}$ FSV on the growth of lactic acid bacteria, Lactobacillus casei YIT 9018(wild type strain with prophage) or L. casei HYM 1213 (prophage cured strain) was infected with various concentrations of phage ${\phi}$ FSV (called Lac S21) or wild type virulent phage (called Lac J-1). When L. casei YIT 9018 was infected with Lac S21 under the concentration of $6.0{\times}10^6$ pfu/ml, the growth and lactic acid production of the strain were normal and the number of phages decreased. But L. casei HYM 1213 was susceptible to Lac S21. Regardless of the concentration of the phage infection, the number of phages increased rapidly into $10^9$ to $10^{10}$ pfu/ml at 2 day cultures and was maintained $10^7$ to $10^9$pfu/ml phage until 6 day cultures. The lactic acid production of L. casei HYM 1213 infected with Lac S21 was abnormal. Therefore, phage ${\phi}$ FSV had an evil effect on growth of L. casei HYM 1213, but not L. casei YIT 9018. On the other hand Lac J-1 caused abnormal fermentation to either L. casei YIT 9018 or L. casei HYM 1213.
