A tissue cultured wild ginseng (TCWG) suspension was inoculated with lactic acid bacteria and fermented to improve the functionality of TCWG. The utilization of TCWG was increased directly using the freeze-dried powder. The optimal ratio of TCWG powder and water for fermentation was 1:19 (5%), which was selected by measuring the fluidity and viable cell count according to concentration. The effects on ADH activation and immune cell activation by each ferments with 10 kinds of Lactobacillus sp. strains were examined. The ferments with the Lactobacillus casei KFRI 692 strain showed 5.4 times higher ADH activity and 1.3 times higher ALDH activity than the non-fermented TCWG powder (control). The level of NO production and cytotoxicity was also measured by Raw 264.7 cells. The ferment with the Lac. casei KFRI 692 strain showed the highest level of NO production and lower cytotoxicity than the others. Therefore, the Lac. casei KFRI 692 strain was selected as a strain for fermentation of a TCWG suspension to maximize its functionality. To identify the optimal fermentation time of the selected Lac. casei KFRI 692 strain on the 5% TCWG suspension, the viable cell count of lactic acid bacterial and the changes in pH were observed for 72 hours. 24-hrs was found to be the optimal fermentation time. In this way, fermented TCWG with lactic acid bacteria showed higher ADH activation efficacy and immune cell activation than non-fermented TCWG.
In the genome annotation of Escherichia coli MG1655, the orf382 (1,149 bp) is designated as a gene encoding an alcohol dehydrogenase that may be Fe-dependent. In this study, the gene was amplified from the genome by PCR and overexpressed in Escherichia coli BL21(DE3). The recombinant $6{\times}$His-tag protein was then purified and characterized. In an enzymatic assay using different hydroxyl-containing substrates (n-butanol, $\small{L}$-threonine, ethanol, isopropanol, glucose, glycerol, $\small{L}$-serine, lactic acid, citric acid, methanol, or $\small{D}$-threonine), the enzyme showed the highest activity on $\small{L}$-threonine. Characterization of the mutant constructed using gene knockout of the orf382 also implied the function of the enzyme in the metabolism of $\small{L}$-threonine into glycine. Considering the presence of tested substrates in living E. coli cel ls and previous literature, we believed that the suitable nomenclature for the enzyme should be an $\small{L}$-threonine dehydrogenase (LTDH). When using $\small{L}$-threonine as the substrate, the enzyme exhibited the best catalytic performance at $39^{\circ}C$ and pH 9.8 with $NAD^+$ as the cofactor. The determination of the Km values towards $\small{L}$-threonine (Km = $11.29{\mu}M$), ethanol ($222.5{\mu}M$), and n-butanol ($8.02{\mu}M$) also confirmed the enzyme as an LTDH. Furthermore, the LTDH was shown to be an ion-containing protein based on inductively coupled plasma-atomic emission spectrometry with an isoelectronic point of pH 5.4. Moreover, a circular dichroism analysis revealed that the metal ion was structurally and enzymatically essential, as its deprivation remarkably changed the ${\alpha}$-helix percentage (from 12.6% to 6.3%).
Effects of extracts obtained from the flowers of Elsholtzia splendens on the serum lipid profile and hepatotoxicity in mice were investigated. Female ICR mice were given E. splendens ethanolic extract (ESEs) orally at a dose of 10 or 50 mg/kg BW for 50 days. Significant dose-dependent decreases in triglyceride and low-density lipoprotein (LDL)-cholesterol of serum were observed. In addition, ESEs prolonged the lag-time of LDL oxidation in vitro. In the serum of ICE mice given ESEs orally at 10 and 50 mg/kg BW, the serum levels of aspartate aminotransferase (AST) and lactic dehydrogenase (LDH) increased significantly, while total protein, albumin, creatinine, alanine aminotransferase (ALT), and total bilirubin did not change. Therefore, ESEs may be beneficial to human health, although it has some hepatotoxicity.
Alterations in serum enzymes were studied in twenty-five patients who underwent open heart surgery in N.M.C. during the period from June 1979, to Feb. 1980. There were fifteen congenital and ten acquired heart diseases. In all patients, Rygg-Kyvsgaard five head roller pump and Polystan bubble oxygenator were used and serial determination of total level of Creatine phosphokinase [CPK], Lactic dehydrogenase [LDH], Glutamic oxaloacetic transaminase [SGOT] were made preoperatively, operation day [just after aortic clamp release, 2 hrs later, 4 hrs later, 6 hrs later], and postoperative days up to 5th day. Immediate postoperative clinical courses were also evaluated. Twenty-five patients were divided into two groups: Group A[13] was cardiopulmonary bypass time more than 95 minutes and aortic clamp time more than 45 minutes. Group B[12] was cardiopulmonary bypass time less than 95 minutes and aortic clamp time less than 45 minutes. The peak levels of SGOT, LDH in Group A were more significantly elevated than Group B [P<0.05]. But peak levels of CPK were not significant between two groups. In the view of clinical evaluation, poor clinical courses were more frequent in Group A [54%] than Group B[8%].
Sodium arsenate and Yeonlyeonggobon-dan (nianlinggubendan) extract (YGD), a herbal restorative were treated p.o. 20 mg/kg and 500 mg/kg, respectively, and concurrently to rats, and examined the biochemical parameters in blood. The values of white blood cell (WBC), red blood cell (RBC), hemoglobin (Hgb) and hematocrit (Hct) in each group did not show significant variance. The value of aspartate aminotrasferase (AST) of arsenic-treated group was increased for 2 weeks significantly while that of the group of concurrent administration with YGD became low significantly compared with arsenic-treated group and the value of alkaline phosphatase (ALP) of arsenic-treated group was decreased while that of the group of concurrent administration with YGD was increased significantly compared with arsenic-treated group. In arsenic-treated groups, the value of glucose (Glu), and those of lactic dehydrogenase (LDH), blood urea nitrogen (BUN) and triglyceride (TG) were decreased at first but increased later while the groups of concurrent administration with YGD showed significant recovery from the toxicity of arsenic.
The present study was carried out to investigate the serum enzyme activities of racehorses. The enzymes investigated were aspartate and alanine aminotransferase (GOT, GPT), ${\gamma}$-glutamyl transpeptidase(${\gamma}$-GTP), Lactic dehydrogenase(LDH), creatine phosphokinase(CPK), and alkaline phosphatase(ALP). Animals used were 30 healthy racehorses(♀17, ♂13) average weighing 435kg and were from 2 to 8 years of age. LDH activity was progressively decreased with age and next in the order of GOT and CPK activities. GOT and ${\gamma}$-GPT activities were not changed with age but ALP activity tended to be decreased wish age. Activities of GOT, GPT, ${\gamma}$-GTP and ALP were higher in female than in male. There was no difference in CPK activity by sex, and no difference was found out among breeds.
KJST is the oriental herbal medicine of hyperlipemia which consisted of a herb of 17. We induced hyperlipemia to Spontaneous Hypertensive Rat(SHR) and studied the treatment effect by KJST. Hyperlipidemia was induced by hyper-lipidemic diet fed for 4weeks. Total cholesterol, LDL-cholesterol, HDL-cholesterol, Triglyceride, Total bilirubin, Lactic dehydrogenase(LDH), Glucose and Total protein were measured on the serum after an oral administration of KJST. Total cholesterol, LDL-cholesterol, Glucose and LDH was significantly lower in the KJST treated animals, and HDL was significantly raised in the KJST treated animals. These results suggest that KJST is effective for the treatment of hyperlipidemia.
The changes of serum creatine phosphokinase [CPK], serum lactic dehydrogenase [LDH], serum glutamic oxaloacetic transaminase [GOT] and serum glutamic pyruvic transaminase [GPT] were studied in a total of 82 cases who underwent open heart operation under the cardiopulmonary bypass at the Department of Thoracic and Cardiovascular Surgery, Chungnam National University Hospital. The results were as follows 1. The average values of CPK and COT after the operation were reached to the maximal values on the first postoperative day, which were returned to the normal range until the fifth postoperative day. The average values of LDH and CPT after the operation were reached to the maximal values on the first and second postoperative day respectively, which were significantly increased until the seventh postoperative day. 2. In the relationship of the serum enzymes and duration of the extracorporeal circulation, the values on the group over 90 minute of the extracorporeal circulation were more increased than on the group below 90 minute of the extracorporeal circulation. 3. In the relationship of the changes of the serum enzymes and congenital heart diseases and acquired heart diseases, there were no significantly differences in the values between the two groups.
To evaluate the protective effects of extracts of silkworm on carbon tetrachloride-induced hepatotoxicity, serum glutamic-oxaloacetic transaminase, glutamic-pyruvic transaminase and lactic dehydrogenase activities, malondialdehydes values and glutathione-S-transferase activity were measured in ICR mice. Extracts of silkworm was administered orally at 30min after the administration of CCl4 Mice were sacrificed at 24h after the administration of extracts of silkworm. The activities of serum aminotransferase and the hepatic content of lipid peroxide after carbon tetrachloride treatment were markedly increased than normal control but those levels were decreased by the treatment of butanol soluble fraction of silkworm methanol extract. Glutathione S-transferase activity was decreased by carbontetrachloride than control, but also inhibited by the treatment of butanol soluble fraction of silkworm methanol extract.
Comparative LDH isozyme studies on normal, malignant and virus-transformed derivatives of Chinese and Armenian hamsters were illustrated.
