유산균이 생성하는 세포외 다당류의 최적 생성조건에 관한 연구 (Study on the Optimum Cultural Conditions for the production of Extracellular Polysaccharide by Lactic Acid Bacteria)

  • 유산균이 생성하는 다당류에 관한 연구의 일환으로서 발효유 제품의 종균으로 널리 사용되고 있는 Str. themophilus, Lb. bulgaricus, Lb. casei 등을 10% 환원멸균 탈지유에 배양하면서 다당류 생성에 따른, 우유 배양액 점도의 경시적 변화 및 다당류의 최적 생성 조건에 대하여 조사하였다. 1. 시험균 중에서 Str. thermophilus 510이 가장 높은 점도를 나타냈으며 $41^{\circ}C$ 배양 5일째에 5,000 CP로서 다른균에 비해서 1,000 CP 이상 높았다. 2. 인공합성배지를 사용하여 흡광도를 측정한 결과 $41^{\circ}C$에서의 흡광도가 가장 높았으며 다당류 합성을 위한 탄소원으로서는 Sucrose의 이용성이 1~5% 농도에서 가장 높았다. 3. 다당류 합성효소의 금속염에 대한 영향을 살펴본 결과 $Mg^{++}$의 효과가 인정되었다.

Comparison of Probiotic Characteristics in Lactobacillus acidophilus Strains

  • Twelve strains of Lactobacillus acidophilus isolated from feces of human or animal sources were tested for probiotic properties such as cholesterol assimilation, bile and acid tolerances, and CLA production. Although the cultures showed some variation with respect to each test, the 12 strains could be classified into 3 groups based on their ability to assimilate cholesterol. The cholesterol assimilation showed positive correlation with bile tolerance and negative correlation with acid tolerance. The cholesterol assimilation of L. acidophilus strains may not be related to the deconjugation activity, but may in fact be attributed to its bile tolerance. CLA production by lactic acid bacteria (LAB) exhibited a wide variation that ranged from 2.69 to 7.64 mg/g fat. CLA production of Bifidobacterium longum ATCC 15707 was the highest among the LAB tested, but there was no evidence for differences in CLA production between genus and species.

Optimization of Streptococcus macedonicus MBF10-2 Lysate Production in Plant-based Medium by Using Response Surface Methodology

  • Bacterial lysates have become a common ingredient for natural health care. Lactic acid bacteria (LAB) could serve as potential candidates for lysate production: the lactic acids produced by LAB have been utilized for their moisturizing, antimicrobial, and rejuvenating effects, while other substances provide topical benefits and health effects for the skin. Our study aimed to obtain lysate from a LAB S. macedonicus MBF 10-2 through an optimized fermentation using the Response Surface Methodology. Strain MBF10-2 was cultivated in a 2L fermenter tank in de Man Rogosa and Sharpe (MRS) medium and in plant-based peptone modified MRS, i.e. Soy-peptone and Vegitone. The duration and the medium composition (dextrose and soy peptone or proteose peptone) were adjusted to obtain an optimum production of cell lysate. Central Composite Design was employed for Design Expert 7.0.0 by adjusting 3 factors: dextrose (1%, 1.5%, 2%, 2.5%, 3%), soy or proteose peptone (0.5%, 0.75%, 1%, 1.25% and 1.5%), and duration of fermentation (8, 10, 12 14, and 16 h for MRS-Soy peptone and 15, 17, 19, 21, and 23 h for MRS Vegitone). Bacteriocin-Like Inhibitor Substance activity of lysate and pH were used as indicators. The optimum condition for lysate production using MRS Soy Peptone and Vegitone are as follows: dextrose concentration 2.5%, plant-based peptone 1.25%, while optimum fermentation duration were 11.18 h (MRS Soy Peptone) and 17 h (MRS Vegitone) with a starter concentration of 10% at $OD_{600nm}$ $0.2{\pm}0.05$. However, the standard MRS medium produced better quality lysate compared to MRS plant-based peptones.

녹각 및 목이버섯의 젖산발효를 통한 GABA 생산 및 항염증활성 효과 (Anti-inflammatory effects and GABA production of old antler and Auricularia auricula-judae extract fermented by Lactobacillus plantarum)

  • 본 연구에서는 젖산발효를 통해서 얻어진 목이버섯과 녹각 추출액 혼합물의 항산화 및 생리활성평가를 수행하였다. 녹각 추출액의 GABA생성 최적조건에서 probiotics 및 기능성 강화를 위해 목이버섯을 2.5% 첨가하여 $30^{\circ}C$에서 7일간 젖산발효를 하였다. 발효 7일 pH 5.06, 산도 0.77%로 나타났으며 $1.3{\times}10^8CFU/mL$로 높은 균수를 유지하였고 GABA를 1.4% 생성하는 것으로 나타났다. 목이버섯을 첨가했을 때 젖산균 발효물의 물성이 개선되며 단기간에 고농도의 GABA를 생성하는 것으로 나타났다. 녹각 추출액의 젖산발효물의 세포 생존율을 실험한 목이버섯 2.5% 조건에서 발효 전 6 mg/mL에서 독성이 나타났으나 발효 후 독성이 완화되는 것으로 나타났다. 발효 후 6 mg/mL 농도에서는 $5.58{\mu}M$로 발효 후 NO 생성이 감소되는 것으로 나타났다. 결론적으로 녹각 추출액과 목이버섯 혼합물의 젖산균을 이용한 정치배양을 통해 단기간에 고농도의 GABA 생산이 가능하였으며, 젖산 발효물은 세포독성 완화 효과 및 NO 생성을 저해하는 것으로 나타났다. 따라서 발효물은 GABA, probiotic, 식이섬유 등을 함유하여 기능성 식품소재로 이용이 가능할 것으로 기대된다.

The Effects of Two Inoculants Applied to Forage Sorghum at Ensiling on Silage Characteristics

  • Whole forage sorghum (saccharatum) cultivar FS5 was harvested at the soft dough ($303{\pm}9g\;kg^{-1}$ DM) stage of maturity. The sorghum was chopped into approximately 20 mm pieces and ensiled under laboratory conditions in 1.5 L Weck glass jars. At ensiling, it was treated with two commercial silage inoculants: Pioneer 1188 (Inoculant A) and Eco-corn (Inoculant B). The inoculant A and B was applied at ca $2{\times}10^5$ or $2{\times}10^4$ colony forming units $g^{-1}$ DM., respectively. Silage with no additives served as a control. Three jars per treatment were opened on days 2, 4, 8, 15 and 60 post-ensiling to study fermentation dynamics. After 60 days of ensiling the silages were analyzed and subjected to an aerobic stability test lasting 5 days. Results showed that both inoculants caused a more rapid rate of pH decrease and a higher amount of lactic acid production. All the silages were well preserved and were stable upon exposure to air. Inoculants did not influence (p>0.05) the ash and total N contents, but tended to reduce acetic acid (p<0.05), butyric acid (p<0.01) and propionic acid (p<0.01) contents, and to increase the lactic acid content (p<0.01). The lower DM content of silages treated with Inoculant A agrees with the greater gas loss resulting from the DM loss, which was in good agreement with the higher yeast counts upon aerobic exposure. Silage treated with inoculant B had the highest DM (p<0.05) and lactic acid contents (p<0.01), and the lowest acetic acid content (p<0.05), which agrees with the rapid reduction of pH and smaller gas loss. Inoculant B reduced the ADF (p<0.01), ADL and NDF (p<0.05) contents, which also indicates smaller losses of organic soluble material. The control silages contained the highest levels of volatile fatty acids but no lactic acid, indicating secondary fermentation. It was concluded that both inoculants may improve the fermentation process, since silages from all treatments were stable upon aerobic exposure, noadvantage could be attributed to any of the inoculants used.

RUMINAL ACID CONCENTRATIONS OF GOATS FED HAYS AND SILAGES PREPARED FROM ITALIAN RYEGRASS AND ITS PRESSED CAKE

  • From the same harvest of Italian ryegrass (Lolium multiflorum, Lam.), hay(H), wilted silage(WS), pressed cake hay (PCH) and pressed cake silage (PCS) were prepared. These four preserved roughages were restrictedly fed to four goats attached with rumen fistula by $4{\times}4$ Latin square design to determine the effect of different physical and chemical properties of the roughages on the ruminal acid concentrations. Each goat was given a diet at 2% of the body weight daily in dry matter basis by separating into two equal portions. Half was given at 9 AM and the other half at 5 PM. Ruminal pH was reduced to around 5.5 within 30 minutes after feeding PCS and it was recovered above 6 in 1-2 hours after feeding. By feeding WS, ruminal pH was also reduced but never fell below 6. The two hays rather increased ruminal pH after feeding. The reduction of ruminal pH in the silage feedings was due to the high lactic acid content of the silages, because the highest ruminal lactic acid concentration was observed 30 minutes after feeding when the lowest ruminal pH was attained. While the ruminal VFA concentrations became the highest 1-2 hours after feeding. The ruminal acetic acid concentration fluctuated so much that no significant tendency was observed among the four dietary treatments. The ruminal propionic acid concentration was higher in feeding silages reflecting the initial high lactic acid concentration. As the result, acetic/propionic acid ratio was lower in the silage feedings than in hay feedings. Higher ruminal butyric acid concentration was observed in WS than in others.

Production and Characterization of GABA Rice Yogurt

  • Yogurt containing high ${\gamma}$-aminobutyric acid (GABA) was developed using lactic acid bacteria and germinated brown rice. Lactobacillus acidophilus, L. plantarum, and L. brevis OPY-1 strains were inoculated into Lactobacillus MRS broth for use as yogurt starter. After treatment with 5% monosodium glutamate in MRS broth, L. brevis OPY-1 strain isolated from Kimchi produced GABA concentration of 8,003.28 nmol/mL. Starter was inoculated into fermentation substrate mixture containing germinated brown rice extract and blend of powdered whole milk and skim milk. Samples were incubated, and viable cell colonies were counted. Highest number of lactic acid bacteria was reached between 16 and 20 hr. Concentrated rice milk fermented with high GABA-producing strain contained GABA concentrations of $137.17\;{\mu}g/g$ D.W., whereas concentrated fermented milk prepared by conventional method contained GABA of $1.29\;{\mu}g/g$ D.W. Sensory evaluation panelists gave favorable ratings to fermented rice milk containing high GABA concentration.

비타민 K와 골 건강 (Vitamin K and Bone Health)

  • Vitamin K, which is a nutritional factor, play a role in the regulation of bone metabolism. Vitamin K exists naturally in 2 forms, namely, vitamin $K_1$ (phylloquinone) in green plants and vitamin $K_2$ (menaquinone) in animals and bacteria. Vitamin $K_1$ has an effect on bone metabolism. Vitamin $K_2$ is essential for the ${\gamma}$-carboxylation of osteocalcin, a bone matrix protein containing ${\gamma}$-carboxyglutamic acid (Gla) which is synthesized in osteoblast of bone tissues. This paper is to provide the basic information of vitamin K by supplying function and biological activity of vitamin K together with vitamin K producing lactic acid bacteria and it's utilization for the dairy products production.

Effects of Maturity Stages on the Nutritive Composition and Silage Quality of Whole Crop Wheat

  • The changes in yields and nutritive composition of whole crop wheat (Triticum aestivum L.) during maturation and effects of maturity stage and lactic acid bacteria (LAB) inoculants on the fermentation quality and aerobic stability were investigated under laboratory conditions. Whole crop wheat harvested at three maturation stages: flowering stage, milk stage and dough stage. Two strains of LAB (Lactobacillus plantarum: LAB1, Lactobacillus parafarraqinis: LAB2) were inoculated for wheat ensiling at $1.0{\times}10^5$ colony forming units per gram of fresh forage. The results indicated that wheat had higher dry matter yields at the milk and dough stages. The highest water-soluble carbohydrates content, crude protein yields and relative feed value of wheat were obtained at the milk stage, while contents of crude fiber, neutral detergent fiber and acid detergent fiber were the lowest, compared to the flowering and dough stages. Lactic acid contents of wheat silage significantly decreased with maturity. Inoculating homofermentative LAB1 markedly reduced pH values and ammonia-nitrogen ($NH_3$-N) content (p<0.05) of silages at three maturity stages compared with their corresponding controls. Inoculating heterofermentative LAB2 did not significantly influence pH values, whereas it notably lowered lactic acid and $NH_3$-N content (p<0.05) and effectively improved the aerobic stability of silages. In conclusion, considering both yields and nutritive value, whole crop wheat as forage should be harvested at the milk stage. Inoculating LAB1 improved the fermentation quality, while inoculating LAB2 enhanced the aerobic stability of wheat silages at different maturity stages.

Silage preparation and fermentation quality of natural grasses treated with lactic acid bacteria and cellulase in meadow steppe and typical steppe

  • Objective: In order to improve fermentation quality of natural grasses, their silage preparation and fermentation quality in meadow steppe (MS) and typical steppe (TS) were studied. Methods: The small-scale silages and round bale silages of mixed natural grasses in both steppes were prepared using the commercial lactic acid bacteria (LAB) inoculants Chikuso-1 (CH, Lactobacillus plantarum) and cellulase enzyme (AC, Acremonium cellulase) as additives. Results: MS and TS contained 33 and 9 species of natural grasses, respectively. Stipa baicalensis in MS and Stipa grandi in TS were the dominant grasses with the highest dry matter (DM) yield. The crude protein (CP), neutral detergent fiber and water-soluble carbohydrate of the mixed natural grasses in both steppes were 8.02% to 9.03%, 66.75% to 69.47%, and 2.02% to 2.20% on a DM basis, respectively. All silages treated with LAB and cellulase were well preserved with lower pH, butyric acid and ammonia-N content, and higher lactic acid and CP content than those of control in four kinds of silages. Compared with CH- or AC-treated silages, the CH+ AC-treated silages had higher lactic acid content. Conclusion: The results confirmed that combination with LAB and cellulase may result in beneficial effects by improving the natural grass silage fermentation in both grasslands.
