Production of Polyphenols and Flavonoids and Anti-Oxidant Effects of Lactic Acid Bacteria of Fermented Deer Antler Extract

  • The deer antler has been used as a major drug in oriental medicine for a long time. Recently, the demand for easy-to-take health functional foods is increasing due to economic development and changes in diet. As part of research on the development of functional materials for antlers, lactic acid fermentation of antler extract was performed. It was intended to develop a functional material with enhanced total polyphenol and flavonoid content and enhanced antioxidant activity. Lactic acid bacteria fermentation was performed by adding 4 types of lactic acid bacteria starter products, B. longum, Lb. Plantarum, Lb. acidophilus and mixture of 8 types of lactic acid bacteria to the antler water extract substrate, respectively. During the fermentation of lactic acid bacteria, the number of proliferation, total polyphenol and total flavonoid content, DPPH radical scavenging and antioxidant activity were quantified and evaluated. As a result of adding these four types of lactic acid bacteria to the antler water extract substrate, the number of lactic acid bacteria measured was 2.04~5.00×107. Meanwhile, a protease (Baciullus amyloliquefaciens culture: Maxazyme NNP DS) was added to the antler extract to decompose the peptide bonds of the contained proteins. Then, these four types of lactic acid bacteria were added and the number of lactic acid bacteria increased to 2.84×107 ~ 2.21×108 as the result of culture. The total polyphenol contents were 4.82~6.26 ㎍/mL in the lactic acid bacteria fermentation extracts, and after the reaction of protease enzyme and lactic fermentation, increased to 14.27~20.58 ㎍/mL. The total flavonoid contents were 1.52~2.21 ㎍/ml in the lactic acid bacteria fermentation extracts, and after the protease reaction and fermentation, increased to 5.59 ~ 8.11 mg/mL. DPPH radical scavenging activities of lactic acid bacteria fermentation extracts was 17.03~22.75%, but after the protease reaction and fermentation, remarkably increased to 32.82~42.90%.

Effects of Starter Cultures on Physicochemical Properties of Fermented Sausages

  • Fermented sausages prepared by inoculation with different starter cultures were analyzed for their physicochemical characteristics. Three types of fermented sausages were processed separately, without starter culture (control), with a commercial culture mix, and culture mix plus Lactobacillus plantarum (LP). On proximate analysis, two inoculated sausages showed an increase in moisture and fat contents (p<0.05). The inoculated sausages showed lower hardness and gumminess values (p<0.05) than control. The combination of starter culture with LP displayed the lowest chewiness and cohesiveness values and showed a more intensive red color (p<0.05). Two inoculated batches showed significantly lower pH values and water activity than control, in accordance with the increase in lactic acid bacteria (p<0.05). The inoculated sausages reduced the extent of lipid oxidation (p<0.05) and induced an increase in lauric acid, linoleic acid, eicosadienoic acid, and arachidonic acid, as well as they had a higher polyunsaturated fatty acid content and ratio of n-6 and n-3 fatty acids (p<0.05). The addition of LP to the starter culture in a suitable combination resulted in a positive effect on the physicochemical and microbiological attributes of fermented sausages.

단백분해효소 전처리 및 starter culture의 종류에 따른 frozen soy yogurt의 품질특성 (Quality Characteristics of Frozen Soy Yogurt Prepared with Different Proteolytic Enzymes and Starter Culture)

  • 분리대두단백을 단백분해효소로 처리하여 가수분해시킨 후 B. bifidum으로 단독배양하거나 B. bifidum과 L. acidophilus 또는 B. bifidum과 L. bulgaricus로 혼합배양하여 frozen soy yogurt를 제조한 다음 그 품질특성 및 관능적 특성을 측정하였다. 혼합배양시에 단독배양보다 생균수는 더 많았고 담즙산 내성은 더 우수하였으나 내산성은 큰 차이가 없었다. 그러나 ${\alpha}-chymotrypsin$으로 처리하고 B. bifidum과 L. bulgaricus 혼합배양한 시료가 비교적 내산성 및 담즙산 내성이 우수하였다. 시료의 overrun은 B. bifidum과 L. bulgaricus 혼합배양시에 ${\alpha}-chymotrypsin$ 처리군에서 가장 높았으며 아이스크림 제조기에서 30분 조작으로 73.45%를 형성하였다. ${\alpha}-chymotrypsin$ 처리군을 제외한 모든 시료에 있어서 단독배양시에 혼합배양보다 녹아내리는 정도가 더 컸다. 시료의 관능평가 결과 bromelain 처리군과 ${\alpha}-chymotrypsin$ 처리군의 경우 B. bifidum과 L. bulgaricus 혼합배양시에 단독배양보다 콩 비린내와 쓴맛은 적었고 신맛은 더 좋았으며 전반적으로 바람직하다고 평가되었다. 특히 ${\alpha}-chymotrypsin$ 처리군의 B. bifidum과 L. bulgaricus 혼합배양시에 신맛이 좋았고 쓰지 않았으며 떫지 않았다고 평가되어 가장 바람직하다고 평가되었다.

Sucrose, Aspartame 및 Oligo당의 첨가가 Yoghurt Starter의 발효에 미치는 영향 (Effect on Sucrose, Aspartame and Oligosaccharide Added as Sweeteners for the Fermentation of Yoghurt Starter)

  • 요구르트 starter의 발효에 미치는 sucrose, aspartame 및 올리고당의 영향을 알아보기 위하여 탈지유배지에 sucrose를 2~10%, aspartame을 0.01~0.05%, 올리고당을 3.6~11.6% 첨가하고 배양시간에 따른 산도, pH 및 유산균의 증식상태를 조사한 결과는 다음과 같다, 1. 요구르트 starter 발효에 있어 Lac. bulgaricus, Str. thermophillus는 sucrose 4% 첨가구와 올리고당 5.6% 첨가구에서 산도가 크게 증가하였고, pH의 저하가 빨랐으며 sucrose 8% 첨가구와 올리고당 9.6% 이상의 첨가구에서는 산도의 증가와 pH의 저하가 둔화되었다. 2. 요구르트 starter 발효에 있어 Lac. bulgaricus 및 Str. thermophillus의 증식은 sucrose 4% 첨가구와 올리고당 5.6% 첨가구에서 촉진되었고 그 이상의 첨가구에서는 둔화되었다. 3. 요구르트 starter 발효에 있어 aspartame의 첨가는 산도, pH 및 유산균 증식에 영향을 주지 못하였다. 4. Lac. bulgaricus와 Str. thermophillus의 균주에 비하여 혼합균주는 균주의 증식, 및 pH의 변화에서 sucrose와 올리고당의 농도에 따른 영향을 크게 받지 않았다.

Investigation of the Microbiological and Biochemical Properties of Kimchi in the Submerged Model System Designed for Fermented Sausages

  • The objective of this study was to investigate the potential of the application of lactic acid bacteria (LAB) from kimchi as a starter culture in the production of fermented sausages. To achieve this, a submerged model medium that contained LAB as part of a complex system of kimchi (0.5, 1.0, 1.0, 3.0, and 5.0%) and lyophilized kimchi powder (0.2 and 0.5%) was fermented for 120 h. During the fermentation period, the growth of total viable organisms and LAB, and the changes in the pH and the titratable acidity, were investigated. The initial LAB counts ranged from 6.4 to 7.7 Log CFU/mL for the kimchi media, and from 6.9 to 6.9 Log CFU/mL for the kimchi powder media. In all the kimchi batches, the LAB increased logarithmically, and the highest LAB counts (around 9 Log CFU/mL) were reached in 24 h. An evident lag phase of the LAB was observed in the kimchi powder samples and reached 8.8 Log CFU/mL in 8 h. The decrease in the pH and the formation of lactic acid were rapid in the kimchi batches, and reached pH values of 3.4-3.5 in 12 h. With these results, the LAB that was integrated with the addition of kimchi or kimchi powder demonstrated its potential utility as a substitute for starter culture.

Improvement in the Quality of Kimchi by Fermentation with Leuconostoc mesenteroides ATCC 8293 as Starter Culture

  • To investigate the effect of the predominant microorganisms in kimchi on quality, Leuconostoc mesenteroides ATCC 8293 was used as starter culture during kimchi fermentation. A higher number of lactic acid bacteria and lower initial pH were observed in starter kimchi than in non-starter kimchi in the early stage of fermentation. The concentrations of the main metabolite, lactic acid, were 69.88 mM and 83.85 mM for the non-starter and starter fermented kimchi, respectively. The free sugar concentrations of starter kimchi decreased earlier than those of non-starter kimchi, and the levels of free sugars in both kimchi samples decreased during fermentation. At the end of fermentation, non-starter kimchi had a softer texture than starter kimchi, suggesting that L. mesenteroides is useful in extending shelf life. Sensory evaluation showed that starter kimchi had higher sourness and lower bitterness and astringency values, resulting in high sensory quality. These results suggest that the L. mesenteroides ATCC 8293 strain could be a potential starter culture in kimchi.

  • 본 연구에서는 메타분석을 통하여 heterofermentative과 homofermentative LAB가 사일리지의 품질과 호기적 안정성에 대한 효과를 분석하였다. 일련의 분석결과 접종균으로 사용된 젖산균의 종류에 따라서 사일리지의 품질이 다르게 나타남을 확인할 수 있었다. 특히 호기적 안정성에 있어서 heterofermentative LAB의 사용이 매우 중요함을 알 수 있었다. 본 연구결과는 사일리지의 제조 목적 및 사용방법에 따라서 접종균주를 다르게 선발하여야 하며, 사용방법에 따라서 사일리지 내 유기산 조성을 변화시킬 수 있는 발효조건이 필요함을 시사하고 있고, 또한 보다 정밀한 사일리지 발효기술에 대한 기초자료를 제공한다.

매실(Prunes mime) 착즙액이 Yoghurt Starter의 산생성 및 증식에 미치는 영향 (The Effect of Maesil(Prunus mime) Extract on the Acid Production and Growth of Yoghurt Starter)

  • Skim milk에 매실 착즙액을 수준별로 첨가하고 4종(Streptococcus thermophilus, Lactobacillus acidophilus, Lactobacillus bulgaricus, Lactobacillus casei)의 젖산균을 단독균주 또는 혼합균주로 접종하여 매실 착즙액의 첨가가 젖산균의 산생성 및 생육에 미치는 영향과 저장성을 조사하였고, 매실 착즙액으로 사용한 매실의 일반 성분을 분석하였다. 매실의 일반 성분은 조회분 0.4%, 식이섬유 4.1%, 구연산 4.66%, 총당 0.264%, vitamin C 405.34mg%이었고, 매실 착즙액의 첨가는 젖산균의 증식을 촉진시켰으며 산생성도 증가하였다. 실험구 중 Str. thermophilus, Lac. acidophilus와 Lac. casei의 혼합균주에 3% 매실 착즙액을 첨가한 실험구가 가장 많은 양의 젖산(1023%)을 생성하였고, 가장 높은 생성균수(3.6$\times$$10^{11}$ cfu/mL)를 나타내었다. 매실 착즙액 3% 첨가 호상 요구르트를 대조구와 함께 4$^{\circ}C$와 2$0^{\circ}C$에서 30일 동안 저장한 결과, 세 개의 혼합균주(Str. thermophilus, Lac. acidophilus)를 사용한 호상 요구르트가 4$^{\circ}C$에서 pH는 4.25, 생균수는 4.1$\times$$10^{9}$ cfu/mL 이었고, 2$0^{\circ}C$에서 pH는 3.76, 생균수는 8.7$\times$$10^{8}$ cfu/mL으로 대조구인 매실 무첨가 호상 요구르트에 비하여 높은 저장성을 나타내었다. 이상의 결과로 보아 매실 착즙액을 첨가한 호상 요구르트의 제조가 가능할 것으로 사료된다.

발효 소시지 제조를 위한 기능성 유산균의 선발 (Screening of Lactic Acid Bacteria as Starter Culture for Making Fermented Sausage)

  • 발효 소시지의 제조에 starter로 사용한 균주를 선발하기 위해 각 유산균 균주들의 ACE 저해 활성, 콜레스테롤 흡착능력, 발효 육제품 저장 중 S. aureus에 대한 저해 황성을 평가하였다. ACE 저해 활성은 L. plantarum L167이 가장 우수하였고(58.75%), 콜레스테롤 흡착 능력이 우수한 균주로는 L. plantarum L155를 선발하였다. 박테리오신을 생산하는 P. damnosus를 starter로 이용하여 제조한 발효 소시지에 S. aureus를 접종하여 상온 저장 중 생균수를 측정한 결과 저장 35일째에 대조구에 비하여 1 log의 균수 감소 효과가 나타났다.

Fermentative characteristics of wheat bran direct-fed microbes inoculated with starter culture

  • This study was conducted to determine the fermentative characteristics of wheat bran inoculated with a starter culture of direct-fed microbes as a microbial wheat bran (DMWB) feed additive. Wheat bran was prepared with 1% (w/w, 0.5% Lactobacillus plantarum and 0.5% of Saccharomyces cerevisiae) starter culture treatment (TW) or without starter culture as a control (CW). Those were fermented under anaerobic conditions at $30^{\circ}C$ incubation for 3 days. Samples were taken at 0, 1, 2, and 3 days to analyze chemical composition, microbial growth, pH, and organic acid content. Chemical composition was not significantly different between CW and TW (p > 0.05). In TW, the number of lactic acid bacteria and yeast increased during the 3 days of fermentation (p < 0.05) and the population of lactic acid bacteria was significantly higher than in CW (p < 0.05). After 3 days, the number of yeast in TW was $7.50{\pm}0.07log\;CFU/g$, however, no yeast was detected in CW (p < 0.05). The pH values of both wheat bran samples decreased during the 3 days of fermentation (p < 0.05), and TW showed significantly lower pH than CW after 3 days of fermentation (p < 0.05). Contents of lactic acid and acetic acid increased significantly at 3rd day of fermentation in TW. However, no organic acids were generated in CW during testing period. These results suggest that 3 days of fermentation at $37^{\circ}C$ incubation after the inoculation wheat bran with starter culture makes it possible to produce a direct-feed with a high population of lactic acid bacteria at more than $10^{11}CFU/g$.
