$H_{\infty}$ Control of Seeker Scan-Loop using LSDP (LSDP를 이용한 탐색기 주사루프의 $H_{\infty}$ 제어)

  • $H_{\infty}$ Controller of seeker scan-loop is designed using LSDP proposed by McFarlane. The performance and robustness of $H_{\infty}$ controller are analyzed using robustness theorems by Lehtomaki and compared with those of the LQG/LTR controller. Especially, structured singular value .mu. -test of Doyle is used to evaluate robust performance of seeker scan-loop. It is demonstated that seeker scan-loop by $H_{\infty}$ controller is very robust to model uncertainties described by additive and multiplicative perturbations.

Application of Transposable Elements as Molecular-marker for Cancer Diagnosis (암 진단 분자 마커로서 이동성 유전인자의 응용)

  • Until now, various oncogenic pathways were idenfied. The accumulation of DNA mutation induces genomic instability in the cell, and it makes cancer. The development of bioinformatics and genomics, to find the precise and reliable biomarker is available. This biomarker could be applied the early-dignosis, prediction and convalescence of cancer. Recently, Transposable elements (TEs) have been attracted as the regulator of genes, because they occupy a half of human genome, and the cause of various diseases. TEs induce DNA mutation, as well as the regulation of gene expression, that makes to cancer development. So, we confirmed the relationship between TEs and colon cancer, and provided the clue for colon cancer biomarker. First, we confirmed long interspersed nuclear element-1 (LINE-1), Alu, and long terminal repeats (LTRs) and their relationship to colon cancer. Because these elements have large composition and enormous effect to the human genome. Interestingly, colon cancer specific patterns were detected, such as the hypomethylation of LINE-1, LINE-1 insertion in the APC gene, hypo- or hypermethylation of Alu, and isoform derived from LTR insertion. Moreover, hypomethylation of LINE-1 in proto-oncogene is used as the biomarker of colon cancer metastasis, and MLH1 mutation induced by Alu is detected in familial or hereditary colon cancer. The genes, effected by TEs, were analyzed their expression patterns by in silico analysis. Then, we provided tissue- and gender-specific expression patterns. This information can provide reliable cancer biomarker, and apply to prediction and diagnosis of colon cancer.

Eruption Styles and Processes of the Dongmakgol Tuff, Cheolwon Basin, Korea (철원분지 동막골응회암의 분출유형과 분출과정)

  • The Dongmakgol Tuff is divided into 8 lithofacies based on their grain size and depositional structures: massive tuff breccia(TBm), welded tuff and lapilli tuff(LTw), rheomorphic tuff and lapilli tuff(LTr), massive lapilli tuff(LTm), stratified lapilli tuff(LTs), gradedly bedded lapilli tuff(LTg), crudely bedded lapilli tuff(LTb) and massive fine tuff(Tm). They can be divided into 3 pyroclastic rock group based on their constituents of the lithofacies. The lower group(LI) is composed of LTm, LTw and LTr, which are interpreted to have resulted from emplacement of voluminous pyroclastic flows due to ignimbrite-form eruption to boiling-over eruption. The middle group(LT+MI) consists of LTs, LTg and LTm associated with Tm in the lower part, and of LTm, LTw and LTr in the middle and upper parts; these suggest that started with deposition of pyroclastic surges from phreatoplinian eruption by poor eternal water, passed through emplacement of pyroclastic flows from ignimbrite-form eruption and ended with deposition of voluminous pyroclastic flows from boiling-over eruption. The upper group(lUT+uUT+UI) is composed of LTs, LTg and Tm in the lowermost, TBm, LTb, LTb and Tm in the lower part, and LTm and LTw in the middle and upper part, suggesting that began with deposition of surges from phreatoplinian eruption, passed through deposition of pumice- and ash-fallouts from plinian eruption and transformed into emplacement of pyroclastic flows due to boiling-over eruption. As result, eruptive processes in the Dongmakgol Tuff approximately began with phreatoplinian or/and plinian eruption, transformed into ignimbrite-forming eruption and proceeded into boiling-over eruption in each volcanism, but proceeded presumably without phreatoplinian or plinian eruption in the earlier stage of 1st volcanism.

A Study on the Transmission of a Transgene in the Offspring of Transgenic Mice (형질전환 생쥐의 후손에서 외래 유전자의 유전성에 대한 연구)

  • It is known that the incorporation of genes into transgenic mice is generally stable and is p passed on to succeeding generations in a Mendelian fashion. In this report, transgenic mice were set as a model to evaluate whether the transgenes are transmitted in a Mendelian principle in a successive generations and how they are tran s smitted into their offspring. A 3.0 kb linear DNA fragment, containing the MMTV LTR, bovine aSI casein cDNA and SV 40 splicing and polyadenylation site; was microinjected into fertilized mouse embryos. The tail DNAs of the resulting pups were subjected to dot and Southern hybridizations to screen transgenic founders. The DNAs of their offspring were anlyzed by PCR to confirm the transmission of the transgene from F0. Out of 72 live pups four pups (5.6%), 3 males and 1 female, were positive for the transgene. The rates of transmission from F0 into F1 were 33.3, 7.7, 0, and 62.5%. Those from F1 into F2 were 63.6, 5.9, and 68.8% and those from F2 into F3 were 85.7, and 88.2%. In this report, the transmission pattern of transgenes in transgenic mice into their offspring was demonstrated. It either follows or does not follow in a Mendelian fashion. Deletion or loss of the transgenes from F0 in some lines became apparant to the succeeding generations.

Analysis of Asthma Related SNP Genotype Data Using Normalized Mutual Information and Support Vector Machines (정규상호정보와 지지벡터기계를 이용한 천식 관련 단일염기다형성 유전형 자료 분석)

  • Introduction: There are two types of asthma according to aspirin hypersensitivity: aspirin intolerant asthma (AIA) and aspirin tolerant asthma (ATA). The genetic risk factors that are related with asthma have been investigated intensively and extensively. However the combinatory effects of single nucleotide polymorphisms (SNPs) have hardly been evaluated. In this paper we searched the best set of SNPs that are useful to diagnose the two types of asthma. Methods: We examined 246 asthmatic patients (94 having aspirin intolerant asthma and 152 having aspirin tolerant asthma) and analyzed 25 SNPs typed in them, which are suspected to be associated with asthma. Normalized mutual information values of combinations of typed SNPs are calculated, and those with high normalized mutual information values are selected. We use support vector machines to evaluate the prediction accuracy of the selected combinations. Results: The best combination model turns out four-locus and consists of ALOX5_p1_1708, B2ADR_q1_46, CCR3_p1_520, CysLTR1_p1_634. Its normalized mutual information value is 0.053 and the accuracy in predicting ATA disease risk among asthmatic patients is 71.14%.

Integration of a target gene into chromosomal genome of BF-2 cells using UV-inactivated snakehead retrovirus (SnRV)

  • Integration and expression of a target gene into chromosomal genomes of host cell by retrovirus mediated gene transfer system usually require complicate and laborious procedures. In the present study, we investigate a simple method to integrate a target gene into genome of BF-2 cells using ultraviolet (UV)-inactivated snakehead retrovirus (SnRV), a fish retrovirus. First of all, an optimization of transfection condition was determined with BF-2 cells using Lipofectamine 2000 and Transome. Using 0.5 $\mu\ell$ Lipofectamine 2000 resulted in 33.8, 40.6 and 40.2% of transfection efficacy with high survival rate (minimum 80%) in 0.5, 1 and 2 $\mu{g}$ DNA, respectively, and those of Transome were all less than 5%. It was confirmed that UV-treatment for 5 min was enough to inactivate infectivity of SnRV. Next, a cassette composed of GFP (green fluorescent protein) gene flanked by LTR (long terminal repeats) sequences derived from SnRV was constructed and transfected into BF-2 cells followed by treatment with UV-inactivated SnRV for optimization of integration and expression of the cassette gene. As the results, the fluorescence was expressed in BF-2 cells treated with UV-inactivated SnRV 3 and 5 times, while there was no expression in BF-2 cells with once and non treatment. Accordingly, it was confirmed that GFP gene was integrated into chromosomal genome of BF-2 cells with UV-inactivated SnRV.

  • Transposable elements (TEs) occupy approximately 45% of the human genome and can enter functional genes randomly. During evolutionary radiation, multiple copies of TEs are produced by duplication events. Those elements contribute to biodiversity and phylogenomics. Most of them are controlled by epigenetic regulation, such as methylation or acetylation. Every species contains their own specific mobile elements, and they are divided into DNA transposons and retrotransposons. Retrotransposons can be divided by the presence of a long terminal repeat (LTR). They show various biological functions, such as promoter, enhancer, exonization, rearrangement, and alternative splicing. Also, they are strongly implicated to genomic instability, causing various diseases. Therefore, they could be used as biomarkers for the diagnosis and prognosis of diseases such as cancers. Recently, it was found that TEs could produce miRNAs, which play roles in gene inhibition through mRNA cleavage or translational repression, binding seed regions of target genes. Studies of TE-derived miRNAs offer a potential for the expression of functional genes. Comparative analyses of different types of miRNAs in various species and tissues could be of interest in the fields of evolution and phylogeny. Those events allow us to understand the importance of TEs in relation to biological roles and various diseases.

A Study on the Gaze Flow of Internet Portal Sites Utilizing Eye Tracking (아이트래킹을 활용한 인터넷 포털사이트의 시선 흐름에 관한 연구)

  • This study investigated through eye tracking what gaze path the audience searches through portal sites (Naver, Daum, Zoom, and Nate). As a result of the layout analysis according to the gaze path of the search engine, the four main pages, which can be called to be the gateway to information search, appeared in the form of a Z-shaped layout. The news and search pages of each site use an F-shape, which means that when people's eyes move from top to right in an F-shape, they read while moving their eyes from left to right(LTR), which sequentially moves to the bottom. As a result of analyzing through the heat map, gaze plot, and cluster, which are the visual analysis indicators of eye tracking, the concentration of eyes on the photo and head copy was found the most in the heat map, and it can be said to be of high interest in the information. The flow of gaze flows downward from the top left to the right, and it can be seen that the cluster is most concentrated at the top of the portal site. The website designer should focus on improving the accessibility and readability of the information desired by the user in the layout design, and periodic interface changes are required by investigating and analyzing the tendencies and behavioral patterns of the main users.

An OTHBVS Cell Line Expresses the Human HBV Middle S Protein

  • An OTHBVS cell line from HepG2 was established. This cell line stably expresses the human hepatitis B virus (HBV) middle S protein that includes the preS2 region which is important for HBV particle entry into the hepatocyte. To establish this cell line, the middle S open reading frame (ORF), with a promoter located in the 5' region and enhancer located in the 3' region, was cloned downstream from the metallothionine (MT) promoter of the OT1529 vector. In this vector, expression of the middle S protein was constructed to be regulated by its own promoter and enhancer. Expression of the large S protein which contains the preS1 region in addition to the middle S protein was designed to be regulated by the MT promoter. When extracts of OTHBVS cells were examined with an S protein detection kit (RPHA, Korea Green Cross Co.), an S protein was detected. Total mRNA of OTHBVS cell examined by northern blot analysis with an S ORF probe revealed small/middle S transcripts (2.1 kb). When the MT promoter was induced by Zn, large S transcripts (2.4 kb) were detected. The GP36 and GP33 middle S proteins were presumably detected, but large S proteins were not detected by immunostain analysis using anti-preS2 antibody.

Immediate Effects of Strain-Counterstrain Technique on Pressure Pain Threshold and Muscle Activity in Male Adults With Upper Trapezius Latent Trigger Point

  • The aim of this study was to determine the immediate effects of single treatment of strain-counter strain (SCS) on pressure pain threshold (PPT) and muscle activity during scapular plane abduction with 3% body weight load. Fifteen asymptomatic male adults with upper trapezius latent trigger point (LTrP) (PPT<2.9 $kg/cm^2$) participated in this study. Pressure algometer was used to measure PPT and surface electromyography was used to record upper, middle arid lower trapezius, serratus anterior, infraspinatus and middle deltoid muscle activity and relative ratio during scapular plane abduction between pre- and post-intervention. There was a significant increase in upper trapezius PPT after a 90-second SCS (p<.05). The activity of the upper trapezius and middle deltoid was significantly decreased (p=.014, p=.001), coupled with a decreased muscle activity ratio between the upper and lower trapezius (p<.05). These results indicate that the SCS may effectively deactivate upper trapezius activity, thereby alleviating muscle balance and reducing pain sensitivity.
