Secretion and Localization of Pseudomonas auratiaca Levansucrase Expressed in Saccharomyces cerevisiae (Saccharomyces cerevisiae에서 발현된 Pseudomonas aurantiaca Levansucrase의 분비국재성)

  • Levansucrase gene(lscA) from Pseudomonas aurantiaca was subcloned downstream of GAL1 promoter in pYES 2.0 and pYInu-AT [GAL10 promoter + exoinulinase signal sequence of Kluyveromyces marxianus], resulting pYES-lscA and p YInu-lscA, respectively. The two expression plasmids were introduced into an invertase-deficient strain, Sacchromayces cerevisiae SEY2102, and transformants with high activity of levansucrase were selected. When each yeast transform ants was cultivated in medium containing galactose, the extracellular and intracellular activities of levansucrase reached about 8.62 U/ml with the strain harboring pYES-lscA and 5.43 U/ml with the strain harboring pYInu-lscA. The levansucrase activity of 80% was detected in the periplasmic space and cytoplasm. The levansucrase activity in the medium of SEY2102/pYInu-lscA was 0.87 U/ml whereas that of SEY2102/pYES-lscA was 0.47 U/ml, which implying the exoinulinase signal sequence didn't enhance the secretion efficiency of levansucrase. Furthermore, the recombinant levansucrase expressed in yeast seems to be produced as a hyper-glycosylated form.

Characteristics of Biosurfactants produced by Bacillus sp. LSC11 (Bacillus sp. LSC11가 생산하는 biosurfactant의 특성)

  • Several bacterial strains producing biosurfactants were isolated from polluted marine and soil by oil. One of the strains named LSC11 showed strong production activity of biosurfactants. This strain was identified as a Bacillus sp. LSC11 based on the morphological, biochemical, and physiological characteristics. The biosurfactant, produced by the strain, emulsified crude oil, vegetable oil, and hydrocarbons. The surface tension of the culture broth of Bacillus sp. LSC11 decreased to 32 mN/m. The crude biosurfactant was obtained from the culture broth by acid precipitation, freeze drying, solvent extraction, and evaporation. The emulsifying activity of the biosurfactant showed better than the chemically synthesized surfactant (SDS, Span40, Span 85). The biosurfactants had strong properties as an emulsifying agent and as an emulsion-stabilizing agent.

  • This study was carried out to compare the characteristics of hydraulic architecture by measurement of leaf specific conductivity(LSC) in the twenty-one deciduous broad-leaved species. The LSC's of stems were higher than in branches except Acer mono, A. triflorum, Betula schmidtii, Fraxinus rhynchophylla and Zelkova serrata, and lowest in junction parts as compared with stems and branches. In diffuse-porous species, the LSC was much higher in B. plalyphylla var. iaponica, B. schmidtii, Cornus controversa, Tilia amurensis than those of other species and especially lowest in A. triflorum. In ring-porous species, the LSC was much higher in Maackia amurensis, Paulownia tomentasa, Quercus aliens, Q. serrata than those of other species and lowest in Ulmus davidiana var. japonica. The LSC's of 1-year-old terminal shoots of stem(the leader and adjacent laterals) in A. triflorum, C. controversa. F. mandshurica, Q. mongolica and Z. serrata ranged from 22 to $139{\mu}{\ell}/g$ in the leaders, and 11 to $73{\mu}{\ell}/g$ in the adjacent laterals. The LSC's of leaders were usually greater than the adjacent laterals because of the hydraulic dominance of the leader shoot.

Review of Land Suitability Classification in Japan and Its Application to Korea (일본의 토지적성구분론과 우리나라의 적용성 고찰)

  • Land suitability classification(LSC) is an appraisal and grouping(or the process of appraisal anti grouping) of specific tracts(of land) in terms of their relative land suitability for a definEd use, and is one of the land use planning techniques. This paper reviews the selected studies on LSC whose purposes are to especially contribute land use planning in case of Japan, So, this study examines the LSC's application to Korea based on Japanes LSC studies with a view to development of the methods on LSC for rational land use planning in our rural area. The result resolves itself into Table 2. However, it is undesirable to borrow from Japanes LSC studies like that, because of the difference of administrative,geographical,traditional,social and economical conditions. Therefore, it is necessary that the many case studies and examinations should be carried out in order to develop the methods on LSC suitable to Korean actual circumstances.

  • A study on the measuring and evaluation of LSC(Least Square Center) roundness was carried out. The experimental set-up was made by index head and indicator, and the measuring data were compensated by a developed computer program. The results obtained are as follows : 1) An index head can conveniently be used to measure LSC roundness. 2) A program for calculating LSC roundness is developed. 3) Without a high quality roundnes measuring apparatus, LSC roundness can be measured and calculated by using index head and the developed program in machine shop as well as in a measuring room

Calculation of the Least Significant Change Value of Bone Densitometry Using a Dual-Energy X-ray Absorptiometry System

  • Purpose: The precision error of a bone density meter reflects the equipment and reproducibility of results by an examiner. Precision error values can be expressed as coefficient of variation (CV), CV%, and root mean square-SD (RMS-SD). The International Society for Clinical Densitometry (ISCD) currently recommends using RMS-SD as the precision error value. When a 95% confidence interval is applied, the least significant change (LSC) value is calculated by multiplying the precision error value by 2.77. Exceeding the LSC value reflects a significant difference in measured bone density. Therefore, the LSC value of a bone density equipment is an essential factor for accurately determining a patient's bone density. Accordingly, we aimed to calculate the LSC value of a bone density meter (Lunar iDXA, GE) and compare it with the value recommended by the ISCD. We also assessed whether the value measured by the iDXA equipment was below the LSC value recommended by ISCD. Material and Methods: The bone densities of the lumbar spine and thighs of 30 participants were measured twice, and the LSC values were calculated using the precision calculation tool provided by the ISCD (http://www.iscd.org). To check the reproducibility of the measurement, patients were asked to completely dismount from the equipment after the first measurement; the patient was then repositioned before proceeding with the second measurement. Results: The LSC values derived using the CV% values recommended by the ISCD were 5.3% for the lumbar spine and 5.0% for the thigh. The LSC values measured using our bone density equipment were 2.47% for the lumbar spine and 1.61% for the thigh. The LSC value using RMS-SD was 0.031 g/cm2 for the lumbar spine and 0.017 g/cm2 for the thigh. Conclusion: that the findings confirm that the CV% value measured using our bone density meter and the LSC value using RMS-SD were maintained very stably. This can be helpful for obtaining accurate measurements during bone density follow-up examinations.

Comparison between FFT and LSC Method for the Residual Geoid Height Modeling in Korea (한국의 잔여지오이드고 모델링을 위한 FFT 및 LSC 방법 비교)

  • In this study, we performed the residual geoid modeling using the FFT and LSC methods in context of application of R-R (Remove and Restore) technique as a general technique for gravimetric geoid model in order to propose the effective way of geoid determination in Korea. For this, a number of data compiled for residual geoid modeling by the multi-band spherical FFT method with Stoke's formula and LSC method as known as statistical method. The geometric geoidal heights obtained from 503 GPS/Levelling data were used for inducing the various elements and proper computation process which should be considered for improving the accuracy of residual geoid modeling. Finally, we statistically compared the results of residual geoid heights between FFT and LSC methods and reviewed then the proper way of residual geoid modeling to the region of Korea. As the results of comparison, LSC method is not suitable for residual geoid modeling in Korea due to the noise and lack of gravity observations and the effects of local characteristics, while FFT method by applying Stokes' integral with proper cap size and modified kernel which provides the better accuracy of residual geoid heights up to 10 cm more than those of LSC method.

Investigation of the La1-x(Ca or Sr)xCrO3x=0 and 0.25) Interconnect Materials for High Temperature Electrolysis of Steam (고온수증기전기분해용 La1-x(Ca or Sr)xCrO3(x=0 and 0.25) 연결재 재료 연구)

  • The $La_{1-x}(Ca\;or\;Sr)xCrO_3$(x=0 and 0.25) interconnect materials for high temperature electrolysis of steam were investigated in views of sinterability and electrical conductivity. $LaCrO_3$, $La_{0.75}Ca_{0.25}CrO_3$ (LCC), and $La_{0.75}Sr_{0.25}CrO_3$ (LSC) powders were synthesized by coprecipitation method. Crystal structure was confirmed by X-ray diffraction (XRD). The sintering characteristics were analyzed by relative density and scanning electron microscopy. The electrical conductivity was measured by a DC four probe method. From the analyses of relative densities, it was found that the doped $LaCrO_3$ showed better sinterability than $LaCrO_3$ and the those sinterability increased with decrease of those particle sizes. The XRD results at different sintering temperatures for LCC and LSC revealed that the sinterability is closely related to the second phase transformation, that is, the second phase melting above $1,300^{\circ}C$ for LCC and $1,400^{\circ}C$ for LSC significantly promotes the sinterability. In case of electrical conductivities of LCC and LSC, which had a similar relative density, LCC showed better electrical conductivity than LSC.

Defining Semantics of Live Sequence Chart Specification (Live Sequence Chart 명세언어의 의미론적 정의)

  • While developing a complex reactive software system. it is very important to analyze the user requirement and reflect it to the developed system. Therefore understanding the need of users precisely and promptly is the key to the successful software system development. Among several requirement specification languages, message sequence charts (MSCs), also known as sequence diagrams in UML are the most widely used scenario notation. Live Sequence Charts (LSCs) are a variant ot MSCs, characterized by its message abstraction facility and the modality of scenarios. In this paper, I define the formal semantics of LSC specification including the essential language constructs such as pre-charts, variables, assignment and conditions. The range of the formalized LSC language has been broadened, and the scope of the formalized semantics is much closer to the complete LSC specification.

  • 본 논문에서는 구간선형 모델을 적용하여 낮은 복잡도를 가지는 LSC(Lens-Shading Correction) 알고리즘을 제안한다. 제안한 알고리즘은 각 화소와 렌즈 중심점으로부터 거리를 정수형으로 계산하고, 이 정수를 거리에 대한 LSC 이득값이 저장된 LUT(Look-Up Table)에 대한 주소로 적용하여, 입력 화소 값에 곱함으로써 LSC를 수행한다. 거리를 구하려면 제곱근 회로가 추가되어야 한다. LUT에 저장된 이득값은 원점으로부터의 거리에 대한 평균 이득값을 저장하고 있기 때문에, 제곱근 계산에 높은 정밀도를 할애하여도 LSC 보상된 영상의 화질에 미치는 영향은 높지 않으므로 정수형 제곱근 연산을 수행한다. 제곱근 계산은 구간 선형화하여 단지 덧셈과 쉬프트 연산만으로 제곱근 연산을 완료할 수 있도록 간략화 하였다. 제안한 알고리즘을 양산 중인 일반 카메라 모듈에 적용한 결과, 카메라모듈 제조업체의 LSC 평가 기준을 상회하는 수준으로 나타나며, 구현될 하드웨어 복잡도가 매우 낮아서 모바일 카메라 구현에 매우 적합하다.
