Analysis of Physical Properties for Various Compositions of Reusable LMG and LCV Micelle Gel

  • In this study, we evaluated the reusable leuco malachite green (LMG) micelle gel properties dependent on various components of chemical concentration and compared with leuco crystal violet (LCV). The gels were delivered to 10, 20, 30, 40 and 50 Gy at 6 MV photon beam from linear accelerator and analyzed using spectrophotometry. We confirmed that the reusable LMG and LVC absorbance wavelength peak were made up at 630 nm and 600 nm respectively. The transparency of reusable LMG decreased with higher amount of trichloroacetic acid (TCAA) and lower reusable LMG dyes. 1 mM reusable LMG was the lowest transparency. The sensitivity was increased depending on lower trichloroacetic acid (TCAA) concentrations and the amount of suitable surfactant (Triton X-100), which was found to be 7 mM. However, we were not able to investigate sensitivity effects factor from reusable LMG dyes. The gel dosimeter containing 16 mM TCAA, 7 mM Triton X-100 gel dosimeter showed the highest sensitivity at $0.0021{\pm}0.0001cm^{-1}.Gy^{-1}$. The sensitivity of LCV was found to be higher than reusable LMG at $0.0037{\pm}0.0005cm^{-1}.Gy^{-1}$. The reusable LMG and LCV dose responses were shown to be $R^2=0.997$, $R^2=0.999$ respectively, as stable measurement results. Future research is necessary to improve dose sensitivity, dose rate dependency and gel fading with extensive chemical formulations.

신속 FOB(분변 잠혈) 검사 키트를 이용한 혈흔 검출 및 인혈 검사 (Identification of human blood using Rapid FOB (Fecal Occult Blood) Test Kit)

  • 본 연구는 분변 잠혈 검출 키트 (one-step FOB(Fecal Occult Blood) kit)가 법생물학적으로 매우 중요한 혈흔 검출 및 사람 혈흔 판정에 적용 가능한지 알아보고자 하였다. 먼저 FOB 키트의 민감도를 결정하고 기존의 혈흔 검사법인 LMG 검사와 비교한 결과 1,000,000배 희석된 혈액까지도 검출 되었는데 이는 LMG 검사에 비해 약100배 정도 예민한 것이었다. 다른 동물 혈액에 대한 교차 반응 여부를 실험한 결과 FOB 키트는 사람의 혈액과만 반응하여 높은 특이성을 보여주었다. 또한 혈액의 보관 온도 및 경과 시간의 영향, 고온 처리의 영향을 알아보았으며, LMG 및 Luminol 시약의 영향에 대해서도 실험하였다. 사람 혈액 특이 항원은 매우 높은 안정성을 보여주었으며, 전통적인 혈흔 검사 시약인 LMG 및 Luminol에 대해서도 영향을 받지 않았다. 따라서 FOB 키트는 LMG 검사 및 Luminol 검사와 병행하여 사용하면 보다 신속하고 정확하게 사람 혈흔 여부를 판정할 수 있어 법과학 실험실에서는 물론 사건 현장에서의 혈흔 검사에 크게 기여할 수 있을 것으로 사료된다.

Lactobacillus plantarum LMG 7945가 생산하는 bacteriocin의 항균력과 그 특성 (Antibacterial Activity and Characteristics of Bacteriocin Produced by Lactobacillus plantarum LMG 7945)

  • Bacteriocins from lactic acid bacteria have attracted much attention in recent years because of their useful worth in increasing safety and extending shelf life of foods. These substances show an inhibitory effect against some food spoilage bacteria and food-borne pathogens. The inhibitory effect fo the bacteriocin produces by lactic acid bacteria against Listeria monocytogenes(L. monocytogenes) was examined in this study. The culture supernatants of 5 kinds of bacteria among the 10 kinds of testes lactic acid bacteria had the inhibitory activity against Listeria sp., various Gram positive and Gram negative bacteria. Bacteriocin produced by Lactobacillus plantarum(Lact. plantarum) LMG 7945 was the most active toward L. monocytogenes. Bacteriocin production of the Lact. plantarum LMG 7945 cultured on MRS broth was increased late logarithmic phase over early stationary phase. This bacteriocin was stable at heat treatment and acidic pH relatively; The activity was retained after heating at 121$^{\circ}C$ for 15min and was active in the pH range of 2~4 but was lost above pH 5.

한지형 잔디의 종자발아에 미치는 Ethephon,ABA,BA,NAA 침지처리 효과 (The Effect of Ethephon, ABA, BA and NAA Soaking Treatment of Seed Germination of Cool Season Turfgrass -es)

  • The study is aimed to understand effects of ethephon(lmg/1), ABA(0.lrng/l), BA(0.lmg/l). and NAA (0.lmg /1) soaking treatment on the seed germination of perennial ryegrass 'Dandy', creeping bentgrass 'Penlinks', tall fescue 'Arid' and kentucky bluegrass 'Nasaw'. These experiments are carried in vitro and their results are summarized as the followings ; In case of ethephon (lmg /1) soaking treatment of seed, the early germination of tall fescue 'Arid' and perennial ryegrass 'Dandy' was good without the soaking times. creeping bentgrass 'Penlinks' and kentucky bluegrass 'Nasaw' obtained the best results at 12 hr. soking treatment, and the longer the soaking time, the higher the germination percentage. The early germination percentage of tall fescue 'Arid' and perennial ryegrass 'Dandy' increased more highly at ABA(0. tmg /1) soaking treatment than at the control. The germination percent-age of creeping loentgrass 'Penlink' and kentucky bluegrass 'Nasaw' had respectively the best results at 6hr. and l2hr. soaking treatment. The early germination percentage of tall fescue 'Arid' and kentucky bluegrass 'Nasaw'increased more highly at BA (0.lmg /1) soaking treatment than at the control, and especially had the best results in 6hr. soaking treatment. perennial ryegrass 'Dandy' had the best results at l0min. soaking treatment, and the longer the soaking time, the lesser the germination percentage. The early germination of perennial ryegrass 'Dandy'. creeping bentgrass 'Penlinks' and tall fes- cue 'Arid' increased more highly at NAA (0.lmg /1) soaking treatment than at the control, and especially had the best results at l2hr. soaking treatment. However, the germination of Kentucky bluegrass 'Nasaw' seeds inhibited at NAA (0.lmg /1) soaking treatment. Key words: Ethephon, ABA, BA, NAA, Soaking treatment, Seed germination.

Accurate Determination of Malachite Green and Leucomalachite Green in Fish using Isotope Dilution Liquid Chromatography/Mass Spectrometry (ID-LC/MS)

  • Malachite green (MG) has been used world-widely in aquaculture as a parasiticide or fungicide. Although MG performed successfully, it has not been permitted for use in aquaculture from European Union, USA, and Canada because of its carcinogenicity and mutagenicity. We developed a sensitive and specific method to determine MG and its principal metabolite, leucomalachite green (LMG), respectively by isotope dilution liquid chromatography mass spectrometry (ID-LC/MS). To enhance the extraction recovery of MG and LMG from fish tissue, an additional step, saponification, was introduced in sample preparation process to remove fat in sample extract, which hampered the performance of SPE columns. The residue of MG and LMG in fish was analyzed using liquid chromatography mass spectrometry in the selected ion monitoring (SIM) mode by monitoring at m/z 329 and 334 for MG and $d_5$-MG and at m/z 331 and 337 for LMG and $^{13}C_6$-LMG, respectively. This method was validated by comparing with the value of the reference material provided by Laboratory Government Chemistry (LGC). The results agreed within the measurement uncertainty and the accuracy was much improved than the provided reference value by LGC.

감초로 배양한 표고버섯 균사체 추출물이 항암 효과 및 알레르기 억제 효과 검증 (Anti-Cancer and Anti-Allergy Activities of Mycelia Extracts of Lentinus edodes Mushroom-Cultured Glycyrrhiza radix)

  • This study investigated the effects of mycelia of Lentinus edodes mushroom-cultured Glycyrrihiza radix(LMG) on cancer cell lines and sarcoma 180(S-180), as well as on human mast cells. In an anti-cancer tests using Hep3B(hepatic cancer cell), MCF-7(breast cancer), and HeLa(uterine cancer) cells, LMG extract exhibited greater anti-proliferation effects than Glycyrrihiza glabra(GG) extract. LMG extract multiplication restraining effects were 60% that of ethanol at 3 mg/mL extract also displayed tumor suppressive effects in mice injected with S-180 cells. The growth-inhibition rates against tumor cells were 56% for LMG and 37% for GG. When LMG was added to human mast cells, the Intensity of RT-PCR products using primers($FC{\varepsilon}RI\;c-kit$) decreased. significantly compared with that of control. These results suggest that Lentinus edodes Mushroom-Cultured Glycyrrhiza glabra has an anti-proliferation effects against cancer cell lines(Hep3B, MCF-7 and HeLa) and S-180 tumors and will be also beneficial in treating allergic reactions.

키토산이 치주인대 섬유아세포에 미치는 영향 (The effects of chitosan on the human periodontal ligament fibroblasts in vitro)

  • Periodontal therapy has dealt primarily with attempts at arresting progression of disease, however, more recent techniques have focused on regenerating the periodontal ligament having the capacity to regenerate the periodontium. The effect of chitosan(poly-N-acetyl glucosaminoglycan), a carbohydrate biopolymer extracted from chitin, on periodontal ligament regeneration is of particular interest. The purpose of this study was to evaluate the effect of chitosan on the human periodontal ligament fibroblasts(hPDLFs) in vitro, with special focus on their proliferative properties by M'IT assay, the synthesis of type I collagen by reverse transcription-polymerase chain reaction(RT-PCR) and the activity of alkaline phosphatase(ALP). Fibroblast populations were obtained from individuals with a healthy periodontium and cultured with ${\alpha}MEM$ as the control group. The experimental groups were cultured with chitosan in concentration of 0.01,0.1, 1,2mg/ml. The results are as follows; 1. Chitosan-induced proliferative responses of hPDLFs reached a plateau at the concentration of O.lmg/ml(p<0.05). 2. When hPDLFs were stimulated with 0.lmg/ml chitosan, mRNA expression of type I collagen was up-regulated. 3. When hPDLFs were stimulated with 0.lmg/ml chitosan, ALP activity was significantly up-regulated(p<0.05). In summary, chitosan(0.lmg/ml) enhanced the type I collagen synthesis in the early stage, and afterwards, facilitated differentiation into osteogenic cells. The results of this in vitro experiment suggest that chitosan potentiates the differentiation of osteoprogenitor cells and may facilitate the formation of bone.

Immediate Effect of Spinal Mobilization on Lower Limb Strength in Healthy Individuals: A Pilot Study

  • Background: Spinal Mobilization is one of the manual therapy technique that clinicians have used to treat pain, however, there is still a lack of research on changes in strength in healthy people. Objectives: To investigate the effect of posterior-anterior lumbar mobilization on lower limb strength in healthy individuals. Design: Two-group pretest-posttest design. Methods: In this study, 23 healthy subjects aged 20 years were assigned to 12 lumbar mobilization group (LMG) and 12 sham group (SG) to perform intervention and measurement through pre- and post-design. Intervention was performed in LMG with grade III~IV on L3-5 of the lumbar spine, and lumbar mobilization was performed for each segment. After intervention, knee flexion and extension strength were measured. To measure the main effect on muscle strength, a comparative analysis was conducted using paired t-test and independent t-test. Results: In LMG, knee flexor and extensor strength were increased significantly at 60°/s (P<.05). In addition, the extensors of LMG and SG were significantly different only at 60°/s, and the flexors were significantly different between groups at both 60°/s and 180°/s (P<.05). Conclusion: In healthy individuals, lumbar mobilization results in improvement of strength of knee flexor and extensor, and additional experiments on the effect of mobilization on the lumbar spine on functional changes in the lower limbs will be needed.

나노 구조 Double Gate MOSFET의 핀치오프특성에 관한 연구 (A study on the pinch-off characteristics for Double Cate MOSFET in nuo structure)

  • 본 논문에서는 main gate(MG)와 side gate(SG)를 갖는 double gate(DG) MOSFET를 디자인하고 TCAD를 이용하여 시뮬레이션하였다. MG와 SG의 길이(LMG, LSG)는 각각 50nm, 70nm로 하였으며, MG와 SG의 전압(VMG, VSG)이 각각 1.5V, 3.0V일 때 드레인전압(VD)을 0에서 1.5V까지 변화시키면서 핀치오프특성을 조사하였다. LMG가 아주 작음에도 불구하고, 핀치-오프특성이 아주 좋게 나타났다. 이것은 DG MOSFET의 VMG가 게이트를 제어하는 역할을 잘 수행하여 나노 구조에서 유용한 구조임을 알 수 있었다.
