Somatic Embryogenesis and Plant regeneration with Embryogenic Tissue Lines in Larix leptolepis (낙엽송 (Larix leptolepis) 배발생조직 라인에 따른 체세포배 유도 및 식물체 재분화)

  • This study was conducted to evaluate various effects of kinds of culture medium, concentrations of abscisic acid (ABA) or /kinds of osmotica on maturation of somatic embryos (SEs) with four (LL-L, LL-K, LL-P and LL-N) embryogenic tissue lines (ETLs) in Japanese larch (Larix leptolepis). In comparison of two culture medium, the LL-P produced the highest number of the cotyledon-staged SE (134.9/90 mg tissue) in 1/2LM medium. In contrast, no SEs were obtained except the LL-P (32.9) in medium of BLG. Effects of two concentrations of ABA in the medium with four ETL for SEs maturation were also compared. In the test of 60 or 100 ${\mu}M$ ABA, the highest result was obtained in 60 ${\mu}M$ ABA (142.9). However, the influence of ABA had little on SEs production except the LL-N regardless of concentrations of ABA. In comparison of different kinds/concentrations of osmotica, the best response was obtained from the treatment of 0.2 M maltose, the LL-K (540.5). In conclusion, the effects of production of SEs were greatly rely on the ETLs, rather than kinds of medium, concentrations of ABA or osmotica which were used in maturation of SEs.

  • The effect of Ligustrum Lucidum(LL) on the production of nitric oxide (NO) and superoxide by murine peritoneal macrophages were investigated. Stimulation of the cells with LL in the presence or absence of interferon-r(IFN-r) resulted in the increased accumulation of nitrite in the medium. To further examine the mechanism of LL induced. NO Synthesis, we evaluated the secretion of tumor necrosis $factor-{\alpha}(TNF-{\alpha})$ by LL in murine macrophages. Treatment of LL increased the secretion of bioactive $TNF-{\alpha}$ in cultured medium. In addition, LL induced NO production was decreased by the treatment of anti-murine $TNF-{\alpha}$. neutralizing antibodies, indicating that LL induced superoxide production was decreased by the treatment of anti-murine $TNF-{\alpha}$ neutralizing antibodies. These data suggested that LL induced superoxide production was related to $TNF-{\alpha}$ secretion. In conclusion, our results indicates that LL may enhance innate immune response and be applied as a immunoregulating drug improving phagocytosis.

Effects of Ginseng on Textural and Sensory Properties of Long Life Noodles (인삼첨가 Long Life 면의 조직감과 관능적 특성)

  • The influence of ginseng on the paste or gelatinization properties by amylograph and mixing properties by farinograph of wheat flour and on quality properties color cooking quality textural and sensory properties and reducing microbial population of LL(Long Life) noodles was studied. The contents of ginseng used were from 5% to 10% based on flour weight. The viscosity property of wheat flour with ginseng was increased the initial pasting temperature but the amylograph peak viscosity were decreased in vice versa. The farinograph absorption stability and breakdown were increased by ginseng. The whiteness of Ll nodles manufactured with ginseng was lower than that of control The shear extrusion force and hardness of LL noodles manufactured with ginseng were shown much higher value than those of control. At cooking quality examination of LL noodles manufactured with ginseng weight of cooked LL noodles was decreased but volum was appeared in ice versa,. Extraction amounts of LL noodles manufactured with ginseng during cooking were much smaller than those of control Total count of microorganism of Ll noodles manufactured with ginseng were decreased during storage at 3$0^{\circ}C$ Sensory properties of cooked LL noodles which was manufactured with ginseng showed quite acceptable. Based on the cooking and sensory evaluation test addition of 7.0% ginseng to wheat flour may be suitable for processing LL noodles.

  • The influence of organic acid dips on the quality properties, color, cooking quality, textural and sensory properties, and reducing microbial population of LL(Long Life) noodles was studied. The contents of organic acid used were 0.2% based on flour weight and LL noodles were treated by dipping in pH $2.5{\pm}0.1$ for $60{\sim}90sec$. The whiteness of LL noodles treated with dl-malic acid was higher than that of others. The shear extrusion force and hardness of LL noodles treated with dl-malic acid were shown much higher value than those of others except treated with dl-malic acid. acetic acid(=1:1). At cooking quality examination of LL noodles treated with organic acids, weight of cooked LL noodles treated with dl-malic acid was decrease but volume was appeared in vice versa. Extraction amounts of LL noodles treated with dl-malic acid, dl-malic acid : acetic acid(=1:1) during cooking were much smaller than those of others. Total counts of microorganism of LL noodles treated with dl-malic acid,dl-malic acid. acetic acid(=1:1) were disappeared during storage at $30^{\circ}C$ but treated with latic acid, acetic acid were increase during storage. Sensory properties of cooked LL noodles which was treated with dl-malic acid showed quite acceptable.

Effects of Lotus (Nelumbo nucifera) Leaf on Quality and Antioxidant Activity of Yogurt during Refrigerated Storage

  • The supplementation with natural ingredients that are rich in polyphenols could improve the quality and functionality of yogurt. Lotus leaf (LL) are abundant in phenolic compounds. We aimed to investigate the effects of LL powder on the quality properties, total phenolic content (TPC), and antioxidant activity of yogurt. Yogurt was supplemented with four different concentrations (0%, 0.2%, 0.5%, and 1%) of LL powder and evaluated for 14 d refrigerated storage. During storage, the titratable acidity (TA) of all LL yogurt groups was higher than that of the control (p<0.05). The $L^*$ and $a^*$ values decreased by LL addition to the yogurts, while the $b^*$ values increased. All LL yogurt groups indicated better viscosity than that of control, and among them, 0.2% LL yogurts had the highest viscosity without significant decrease until the end of the storage duration. The addition of LL into yogurt enhanced the water-holding capacity (WHC) by at least 1.5-fold than that of control for the entire storage duration. The TPC of yogurts gradually increased with the addition of LL (p<0.05) and continued to increase during storage; the 1% LL yogurt at 14 d showed the highest value of $61.94{\pm}1.68{\mu}g$ GAE/g. The DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity and reducing power of the yogurts were also significantly enhanced with increasing LL concentrations (p<0.05). These results suggest that lotus leaf may be useful as a natural ingredient for improving the quality and antioxidant activity of yogurt.

  • ${\ll}$Xiang Ming Suan Fa${\gg}$ written by Ahn Ji-Jae, a scholar of Yuan Dynasty, is a very important mathematics text in development of mathematics in Joseon Dynasty. Also, ${\ll}$Xiang Ming Suan Fa${\gg}$ in possession of Keimeung university was designated as a Korean National Treasure on February 25, 2011. In this paper, we analyzed the structure and contents of ${\ll}$Xiang Ming Suan Fa${\gg}$. Also, we studied the influences of ${\ll}$Xiang Ming Suan Fa${\gg}$ on Joseon Dynasty's mathematics according to the comparing with mathematics books such as ${\ll}$Mook Sa Jib San Bub> and ${\ll}$San Hak Yib Moon${\gg}$.

  • Objective : The purpose of this study is to research for the formation of Ohaeng-acupuncture. Methods : I refered to ${\ll}$Classic on Difficulty${\gg}$ (難經), ${\ll}$Ling Shu${\gg}$ (靈樞), ${\ll}$Zhen Jiu Ju Ying${\gg}$ (針灸聚英), ${\ll}$Yi Xue Ru Men${\gg}$ (醫學入門) and annotations were excerpted and record that notied the Ohaeng-acupuncture. Results : The results obtained as follows. 1. ${\ll}$Ling Shu${\gg}$ "Sheng Ze Xie Zhi, Xu Ze Bu Zhi"(盛則瀉之, 虛則補之) united with "Ying Sui Bu Xie"(迎隨補瀉), developed the principle of "Qu Xue" in ${\ll}$Classic on Difficulty${\gg}$. 2. ${\ll}$Classic on Difficulty${\gg}$ explained the interdependent relations, interrestraining relations, the relations of subjugation and reverse restriction in illness condition between the five viscera according to the theory of generation, restriction, subjugation and reverse restriction in five elements. ${\ll}$Classic on Difficulty${\gg}$ united five shu points (五兪穴) with five elements. 3. Zi jing Bu xie according to Xiang Sheng theory is develped from ${\ll}$Classic on Difficulty${\gg}$ "Xu Ze Bu Qu Mu, Shi Ze Xie Qi Zi"(虛則補其母 實則瀉其子) to ${\ll}$Classic on Difficulty${\gg}$ , ${\ll}$Zhen Jiu Ju Ying${\gg}$. 4. Ta jing Bu xie according to Xiang Sheng theory is develped from ${\ll}$Tu Zhu Nan Jing${\gg}$ to ${\ll}$Yi Xue Ru Men${\gg}$. 5.   • Background : IGF-I is an important mitogen in many types of malignancies. Tumors also express many IGF binding proteins, which modulate IGF action. The propose of this study was to evaluate the effect of IGF-I and IGFBP on cell proliferation in mouse lung cancer cells (3LL). Methods : The cellular proliferation of 3LL with the treatment of growth factors was evaluated using MTT assay. Western ligand blot was performed in order to determine whether 3LL cells secrete IGFBPs and we evaluated the effect of IGFBP on cellular proliferation. Results : The treatment of 3LL cells with IGF-I increased cellular proliferation in a serum free media. Western ligand blot of conditioned medium of 3LL with $^{125}I$-IGF-I demonstrated one single major band with an estimated molecular mass of 24 kDa. This band was identified as IGFBP-4 with immunoblot analysis using antisera. The addition of anti-IGFBP-4 antibody to abrogate the effect of IGFBP-4 resulted in increased cellular proliferation suggesting that IGFBP-4 inhibits cell growth. Conclusion : IGF-I increases cellular proliferation, however the secreted IGFBP-4 has an inhibitory function on cell growth in 3LL. These findings suggest that IGF-I and IGFBP are involved in the cell proliferation.

  • Background : Stroke occurrences are prevalent and require intensive care for its treatment and rehabilitation. Acupuncture has been widely used in Korea throughout the history and provides an efficient method in the treatment of it. Objectives : To establish a distinctive and efficient acupuncture method for the treatment of stroke based in literature research. Method : We reviewed four Korean medical literature, $ll$治腫指南Guide to Swollen Sore Treatment$gg$, $ll$東醫寶鑑Treasured Mirror of Eastern Medicine$gg$, $ll$鍼灸經驗方Experiential Prescriptions of Acupuncture and Moxibustion$gg$, and $ll$舍岩鍼法Essential Rhymes on Acupuncture and Moxibustion by Master Sa-am$gg$, and analyzed the therapeutic characteristics in the treatment of stroke. Result : 1. In $ll$治腫指南Guide to Swollen Sore Treatment$gg$, various types of treatment can be found. Besides acupuncture, cupping, moxibustion with moxa tube, and bath therapy using Duchesneae Indicae Herba and Sal was applied. Needling in sublingual and vocal region, and also points like GB20, GB31, LI15, BL60, GV20, GV20, TE4, GB39, LU5, ST36, GB30 were frequently inserted. 2. In $ll$東醫寶鑑Treasured Mirror of Eastern Medicine$gg$, moxibustion treatment were in top priority. Points like GV20, LI15, LI11, GB31, ST36, GB39, LI4, GB20 pertinent to Governor, Conception, Gallbladder, Large Intestine, and Stomach meridian were most frequently needled. Selection of adjacent point was widely applied. 3. $ll$鍼灸經驗方Experiential Prescriptions of Acupuncture and Moxibustion$gg$ has some similarity compared to $ll$東醫寶鑑 Treasured Mirror of Eastern Medicine$gg$ in considering the moxa in top priority. But selected points far from the disease site. Main points were LI4, ST36, GB39, PC5, GV20, LI11, LR3, BL40, HT7. 4. $ll$校勘 舍岩道人鍼法Essential Rhymes on Acupuncture and Moxibustion by Master Sa-am$gg$ having the same content orders with $ll$鍼灸經驗方Experiential Prescriptions of Acupuncture and Moxibustion$gg$, it may had some influence from it. But the differences are also apparent. It emphasized in using mother-supplementing child-draining method, experiential prescriptions and visceral pattern identification. Conclusions : We could find various efficient methods through literature research of medical classics. This will not be limited in stroke alone but also will be applied in other diseases. This study will concurrently result in establishing distinctive therapeutic method characteristic of Korea.
