Analysis of Growth and Flowering of Thymus quinquecostatus Using Smart Farming System (스마트 재배시스템을 활용한 백리향 생장 및 개화 분석)

  • 백리향(Thymus quinquecostatus)은 꿀풀과의 낙엽반관목으로 국내에서 자생하는 허브 식물 중 하나이다. 백리향은 특유의 좋은 향기 및 항산화, 항염증, 항균, 미백 등의 효능을 가지는 각종 폴리페놀 성분을 함유하고 있어 의약품이나 기능성 식품, 화장품의 천연 소재 원료로 활용되고 있다. 국내에서 백리향은 재배 환경에 맞추어 주로 고산지대의 노지에서 재배되고 있다. 노지 재배는 지역, 시기, 기후 등의 외부환경에 영향을 받아서 백리향의 유효성분, 품질 및 생산성을 안정적으로 유지하기 어렵다는 한계를 가지고 있다. 따라서 본 연구에서는 스마트팜 시스템을 활용하여 백리향의 유효 성분 등을 안정적으로 얻기 위한 생장조건을 탐색하기 위해 4종의 Light Emitting Diode(LED) 광원과 4종의 토양 조성에 따라서 백리향의 생육조건을 수행하였다. LED는 white, purple, RGB1, RGB2를 사용하였으며, 토양은 상토:펄라이트 비율(상토, 5:1, 3:1, 1:1)로 조성하여 백리향 묘목을 이식한 뒤 생장과 개화시기를 분석하였다. 재배환경은 백리향 재배지의 기상 데이터를 참고하여 동일하게 설정하였으며, 총 8주 동안 생육상태를 관찰하였다. 연구 결과 백리향 재배 4주차에 일부 개체에서 봉오리가 올라오며 개화를 시작하였으며, 8주차에는 대부분의 조건에서 개화를 관찰할 수 있었다. 백리향의 지상부 면적을 비교한 결과 가장 우수한 생장을 보이는 조건은 토양은 3(상토):1(펄라이트) 비율로 분석되었다. 따라서 이번 연구 결과를 바탕으로 백리향 재배에 스마트팜 농업 기술을 활용한다면 기존 노지 재배 한계를 보완하여 안정적이고 지속적인 백리향을 생산할 수 있을것으로 기대된다.

  • This study was carried out to find prioritization and evaluation of importance factor for plant factory development direction. According to the results, experts have put profitability secure at the top of development of plant factory industry. It is urgently needed in economic sector that products from plants factory secure the market and adjust the shipping date through contract cultivation and year-round production. Also, illuminant technology is also important in technology development section and R&D. On the other hand, consumers do not yet fully understand the benefits of the plant factory, and this will lead to continued slow sales. Therefore, the government needs to draw up measures to support producers who cultivated agricultural products from plant factory.

Improvement of Canopy Light Distribution, Photosynthesis, and Growth of Lettuce (Lactuca Sativa L.) in Plant Factory Conditions by Using Filters to Diffuse Light from LEDs (LED 식물공장에서 산란 유리 이용에 의한 상추(Lactuca Sativa L.)의 군락 광분포, 광합성 및 생장 향상)

  • Plant factories with artificial lights require a large amount of electrical energy for lighting; therefore, enhancement of light use efficiency will decrease the cost of plant production. The objective of this study was to enhance the light use efficiency by using filters to diffuse the light from LED sources in plant factory conditions. The two treatments used diffuse glasses with haze factors of 40% and 80%, and a control without the filter. For each treatment, canopy light distribution was evaluated by a 3-D ray tracing method and canopy photosynthesis was measured with a sealed acrylic chamber. Sixteen lettuces for each treatment were cultivated hydroponically in a plant factory for 28 days after transplanting and their growth was compared. Simulation results showed that the light absorption was concentrated on the upper part of the lettuce canopy in treatments and control. The control showed particularly poor canopy light distribution with hotspots of light intensity; thus the light use efficiency decreased compared to the treatments. Total light absorption was the highest in the control; however, the amount of effective light absorption was higher in treatments than the control, and was highest in treatment using filters with a haze factor of 80%. Canopy photosynthesis and plant growth were significantly higher in all the treatments. In conclusion, application of the diffuse glass filters enhanced the canopy light distribution, photosynthesis, and growth of the plants under LED lighting, resulting in enhanced the light use efficiency in plant factory conditions.

  • A new technique, which can increase the channel bandwidth in an optical wireless orthogonal frequency division multiplexing (OFDM) link based on a light emitting diode (LED), is proposed. The technique uses adaptive sampling to convert an OFDM signal to a sparse waveform. In compressive sensing (CS), a sparse signal that is sampled below the Nyquist/Shannon limit can be reconstructed successively with sufficient measurements. The data rate of the proposed CS-based visible light communication (VLC)-OFDM link increases from 30.72 Mb/s to 51.2 Mb/s showing an error vector magnitude (EVM) of 31 % at the quadrature phase shift keying (QPSK) symbol.

Exhibition Hall Lighting Design that Fulfill High CRI Based on Natural Light Characteristics - Focusing on CRI Ra, R9, R12 (자연광 특성 기반 고연색성 실현 전시관 조명 설계 - CRI Ra, R9, R12를 중심으로)

  • To faithfully represent the intention of the work in the exhibition space, lighting that provides high color reproduction like natural light is required. Thus, many lighting technologies have been introduced to improve CRI, but most of them only evaluated the general color rendering index (CRI Ra), which considers eight pastel colors. Natural light provides excellent color rendering performance for all colors, including red and blue, expressed by color rendering index of R9 and R12, but most artificial lighting has the problem that color rendering performance such as R9 and R12 is significantly lower than that of natural light. Recently, lighting technology that provides CRI at the level of natural light is required to realistically express the colors of works including primary colors but related research is very insufficient. Therefore this paper proposes exhibition hall lighting that fulfills CRI with a focus on CRI Ra, R9, and R12 based on the characteristics of natural light. First reinforcement wavelength bands for improving R9 and R12 are selected through analysis of the actual measurement SPD of natural and artificial lighting. Afterward virtual SPDs with a peak wavelength within the reinforcement wavelength band are created and then SPD combination conditions that satisfy CRI Ra≥95, R9, and R12≥90 are derived through combination simulation with a commercial LED light source. Through this, after specifying two types of light sources with 405,630nm peak wavelength that had the greatest impact on the improvement of R9 and R12, the exhibition hall lighting applied with two W/C White LEDs is designed and a control Index DB of the lighting is constructed. Afterward experiments with the proposed method showed that it was possible to achieve high CRI at the level of natural light with average CRI Ra 96.5, R9 96.2, and R12 94.0 under the conditions of illuminance (300-1,000 Lux) and color temperature (3,000-5,000K).

Establishing Optimal Conditions for LED-Based Speed Breeding System in Soybean [Glycine max (L.) Merr.] (LED 기반 콩[Glycine max (L.) Merr.] 세대단축 시스템 구축을 위한 조건 설정)

  • Plant breeding is a time-consuming process, mainly due to the limited annual generational advancement. A speed breeding system, using LED light sources, has been applied to accelerate generational progression in various crops. However, detailed protocols applicable to soybeans are still insufficient. In this study, we report the optimized protocols for a speed breeding system comprising 12 soybean varieties with various maturity ecotypes. We investigated the effects of two light qualities (RGB ratio), three levels of light intensity (PPFD), and two soil conditions on the flowering time and development of soybeans. Our results showed that an increase in the red wavelength of the light spectrum led to a delay in flowering time. Furthermore, as light intensity increased, flowering time, average internode length, and plant height decreased, while the number of nodes, branches, and pods increased. When compared to agronomic soil, horticultural soil resulted in an increase of more than 50% in the number of nodes, branches, and pods. Consequently, the optimal conditions were determined as follows: a 10-hour short-day photoperiod, an equal RGB ratio (1:1:1), light intensity exceeding 1,300 PPFD, and the use of horticultural soil. Under these conditions, the average flowering time was found to be 27.3±2.48 days, with an average seed yield of 7.9±2.67. Thus, the speed breeding systems reduced the flowering time by more than 40 days, compared to the average flowering time of Korean soybean resources (approximately 70 days). By using a controlled growth chamber that is unaffected by external environmental conditions, up to 6 generations can be achieved per year. The use of LED illumination and streamlined facilities further contributes to cost savings. This study highlights the substantial potential of integrating modern crop breeding techniques, such as digital breeding and genetic editing, with generational shortening systems to accelerate crop improvement.

Effects of Light Quality Using LEDs on Expression Patterns in Brassica rapa Seedlings (LED 광원의 다양한 광질이 배추 유묘의 유전자 발현에 미치는 영향)

  • Light with two faces, beneficial and harmful effects is an important signal for every living cell. Optimal adaptation to light environment enhances the fitness of an organism and survival in nature. Understandings of light quality and plant growth provide with the economical guides for artificial light sources like LEDs. Compared with those under white light, the 1 week seedlings of Chinese cabbage (Brassica rapa) under monochromic red and blue light showed normal development and growth. In contrast to extremely long and etiolated hypocotyls of the seedlings under dark, those under far-red etiolated were extremely short. Based on the microarray analysis, blue light induced the vigorous development and growth and two fold changes of transcripts than red light condition. To have insight of gene products under different light qualities conditions, GO term enrichments were calculated and each gene according to their GO terms were categorized. The blue and red lights affected the expressions of genes related to biological process. Especially, the genes related to metabolic process and developmental process and plastid and chloroplast in the cellular component category were induced under blue light. This study provided the molecular biological evidence for various light qualities on the growing process of B. rapa.

Nonthermal Sterilization of Pathogenic Escherichia coli by Intense Pulsed Light Using a Batch System (회분식 광펄스 처리에 의한 병원성 대장균의 비가열 살균)

  • Intense pulsed light (IPL), a nonthermal technology, has attracted increasing interest as a food processing technology. However, its efficacy in inactivating microorganisms has not been evaluated thoroughly. In this study, we investigated the influence of IPL treatment on the inactivation of Escherichia coli O157:H7 depending on light intensity, treatment time, and pulse number. Increased light intensity from 500 V to 1,000 V, raised the inactivation rate at room temperature. At 1000 V, the cell numbers were reduced by 7.1 log cycles within 120 s. In addition, increased pulse number or decreased distance between the light source and sample surface also led to an increase in the inactivation rate. IPL exposure caused a significant increase in the absorption at 260 nm of the suspending agent used in our experiments. This indicates that IPL-treated cells were damaged, consequently releasing intracellular materials. The growth of IPL-irradiated cells were delayed by about 5 h. The degree of damage to the cells after IPL treatment was confimed by transmission electron microscopy.

  • 농산물의 품질 및 성분을 측정하는데 있어 기존의 화학적 분석 방식은 정밀도가 높으나 측정에 소요되는 시간과 비용이 많이 들어, 현장 적용하기에는 한계가 있다. 일반적으로 근적외선 분광 분석(Near Infra Red Spectroscopy, NIRS) 방법은 가공 과정에 따라 빠르게 변화되는 단백질 조성 및 수분함량 측정 등에 이용되고 있다. 분석에 소요 시간이 많이 걸리는 켈달법(Kjeldahi method)에 비해 NIR 분광 분석을 통한 보정으로 연속적인 모니터링이 가능하다. 본 연구에서 사용된 시료를 고정시키기 위한 프레임을 제작한 후 NIR센서와 광원인 LED의 각도를 고정시키고 측정 대상체인 사절된 감자 크기에 따라 시료를 고정시킬 수 있는 프레임을 반사면에 위치시켰다. 확산 반사법을 이용하여 프레임에 씨감자 시료를 고정 시킨 후 백색 LED를 이용하여 감자 표면에 빛을 반사시켜 3일 동안 12시간 마다 해당 시료들(열처리, 비누용액 침지, 생감자)의 스펙트럼을 측정하였다. 해당 시료들은 측정 기간 동안 저온상태($4^{\circ}C$)와 실온상태($20^{\circ}C$)에서 보관되었다. 실험 결과는 파장대 145nm에서 저온상태에서 보관된 생감자는 시간경과에 따른 흡광도의 결정 계수값($r^2$)은 0.98 이었다. 이는 감자가 저온에서 생감자의 상태 변화가 일어나고 있다는 것을 의미하고 파장대 145nm에서 시간에 따른 저온상태에서 보관한 감자의 상태 변화 예측이 가능함을 의미한다. 비누용액에 침지시킨 후 실온에 보관한 감자는 시간이 경과함에 따라 파장이 증가함에 따라 흡광도가 증가하였다. 이는 감자에 들어있는 Polyphenol Oxidase 함량 변화로 갈변 현상이 일어난 것을 알 수 있다. 또한 실온에서 보관한 생감자도 시간 경과에 따라 갈변 현상이 일어났지만 용액에 침지시킨 감자보다는 갈변 현상이 36시간 이후로 발견되었다. 열처리 후 실온에서 보관한 감자의 경우에는 갈변현상이 나타나지 않았다. 저온상태에서 보관한 감자시료들 모두 갈변형상이 나타나지 않았지만, 24시간이 지난 후 용액에 침지시킨 감자는 갈변 현상이 발생되었다. 생감자와 열처리한 감자는 시간 경과에 따른 갈변현상이 일어나지 않았으므로, 감자의 갈변현상은 감자의 표면 처리 방법에 국한되지 않고 온도에 영향을 더 많이 받는다는 것을 나타내고 있다. 본 연구는 향후 감자의 품질 및 성분 측정에서 간편하게 사용될 수 있는 감자의 품질 계측 기술에 기여할 것으로 판단된다.

Development of Optical Molecular Imaging System for the Acquisition of Bioluminescence Signals from Small Animals (소동물 발광영상 측정을 위한 광학분자영상기기의 개발)

  • Purpose: Optical imaging is providing great advance and improvement in genetic and molecular imaging of animals and humans. Optical imaging system consists of optical imaging devices, which carry out major function for monitoring, tracing, and imaging in most of molecular in-vivo researches. In bio-luminescent imaging, small animals containing luciferase gene locally irradiate light, and emitted photons transmitted through skin of the small animals are imaged by using a high sensitive charged coupled device (CCD) camera. In this paper, we introduced optical imaging system for the image acquisition of bio-luminescent signals emitted from small animals. Materials and Methods: In the system, Nikon lens and four LED light sources were mounted at the inside of a dark box. A cooled CCD camera equipped with a control module was used. Results: We tested the performance of the optical imaging system using effendorf tube and light emitting bacteria which injected intravenously into CT26 tumor bearing nude mouse. The performance of implemented optical imaging system for bio-luminescence imaging was demonstrated and the feasibility of the system in small animal imaging application was proved. Conclusion: We anticipate this system could be a useful tool for the molecular imaging of small animals adaptable for various experimental conditions in future.
