Evaluation of Lactic Acid Bacterial Community in Kimchi Using Terminal-Restriction Fragment Length Polymorphism Analysis (Terminal-Restriction Fragment Length Polymorphism 분석을 이용한 김치발효 관련 유산균 군집의 평가)

  • Terminal-restriction fragment length polymorphism (T-RFLP) analysis, one of rapid culture-independent microbial community analysis methods, was used to determine the lactic acid bacterial complexity and dynamics during kimchi fermentation at $15^{\circ}C$ and $4^{\circ}C$. At both temperatures, the common presence of Leuconostoc mesenteroides, Lc. inhae, Lc. kimchi, Weissella koreensis, W. cibaria, Lactobacillus sakei, Lb. curvatus, Lb. plantarum, Lb. paraplantarum, Lb. pentosus, and Lb. brevis was predicted. Lc. citreum and Enterococcus faecalis were detected at $15^{\circ}C$ and $4^{\circ}C$, respectively. W. koreensis predominated during the mid stage of kimchi fermentation whereas lactobacilli were dominants during later stage. Lb. sakei and Lb. curvatus became dominants regardless of fermentation temperature but the growth of Lb. plantarum, Lb. paraplantarum, Lb. pentosus, and Lb. brevis was restricted at psychrophilic temperature. Some species of leuconostocs were maintained until the later stage of kimchi fermentation.

Effects of Feeding a Combination of Probiotics Containing Lactobacillus plantarum and Bacillus Subtilis on Immune Response and Diarrhea Incidence in Post-weaning Piglets (이유자돈에 대한 Lactobacillus plantarum과 Bacillus subtilis 합제 투여에 따른 면역반응과 설사발생에 미치는 효과)

  • A study investigated the effects of a mixture of Lactobacillus plantarum (L. plantarum) and Bacillus subtilis (B. subtilis) on diarrhea incidence, immune response, and fecal microflora counts in post-weaning piglets. One hundred 28-day-old piglets were randomly assigned to five treatment groups: negative control (NC), free of antibiotics; positive control (PC), 0.03% chlortetracycline; LB 1, a mixture of L. plantarum and B. subtilis (LB) 0.5 kg/ton feed; LB 2, LB 1.0 kg/ton feed; and LB 3, LB 2.0 kg/ton feed. Diarrhea scores for LB 2 and LB 3 from the 2nd week were significantly reduced compared to NC (P<0.05). In terms of the level of IgG and IFN-${\gamma}$, all treatment groups were significantly higher than NC (P<0.05), and the IgG level of LB 3 was significantly higher than that of PC (P<0.05). Furthermore, fecal lactic acid bacteria (LAB) counts for LB 2 and LB 3 were significantly higher than those of NC and PC (P<0.05). In addition, fecal Enterobacteriaceae (ENT) counts for PC, LB 2 and LB 3 were significantly lower than those of NC (P<0.05). Based on the results from this study, it was concluded that a combination of L. plantarum and B. subtilis strains could be used as potential alternatives to antibiotics to prevent diarrhea incidence in piglets.

  • The permeability of oxygen and nitrogen was investigated from the polymeric LB films containing imidazole-metal ion complexes and compared with its corresponding cast films on porous membrane filters. The amphiphilic polymer, poly(N- (2-(4-imidazolyl)ethyl)-maleimide-alt-1-octadecene) (IM-O), was synthesized by reaction of poly(maleic anhydride-alt-1-octadecene) with histamine. The IM-O nonolayer showed high stability on Fe (III) ion-containing subphase. The molecular structure in the LB films was investigated by means of FT-IR spectroscopy. The metal ion concentration incorporated into the LB films was determined by means of XPS measurements. The mechanical stability and uniformity of the LB films on porous substrates were indirectly evidenced by SEM observation. The LB and cast films showed more or less higher selectivity toward nitrogen, and high permeability was found to both the oxygen and nitrogen.

  • We have studied a property of arachidic acid Langmuir-Blodgett films at room temperature with two different electrodes ; Al/LB/Al and Au/LB/Au. Since a natural oxide layer is formed on top of the Al electrode, the appropriate structure of AL/LB/Al is Al/Al$_2$O$_3$/LB/Al. To obtain a property of Pure LB film, Aua/LB/Au structure was made. In Al/Al$_2$O$_3$/LB/Al structure, a conductivity of 3.7${\times}$10$\^$-14/ S/cm was obtained by using current-voltage(I-V) characteristics. In Au/LB/AU structure, however, I-V curve was not able to be measured because of short current even in 51 layers of the LB film. A status of defects in the film was confirmed by copper decoration method. We have clearly seen a rather big difference of defect in those two above structures.

Organic Gas Response Characteristics for Horizontal Direction of Fatty Acid LB Ultra-thin Films (지방산 LB초박막의 수평방향에 대한 유기가스 반응특성)

  • Langmuir-Blodgett(LB) films which have high ordered orientation and ordering structure are fabricated by LB method which deposit the ultra-thin films of organic materials at a molecular level. The electrical characteristics of stearic acid LB ultra-thin films for the horizontal direction were investigated to develop the gas sensor using LB ultra-thin films. The optimal deposition condition to deposit the LB ultra-thin films was obtained from $\pi-A$ isotherms and the deposition status of stearic acid LB ultra-thin films was verified by the measurement of deposition ratio, UV-absorbance, and electrical properties for LB ultra-thin films. The conductivity of stearic acid LB ultra-thin films for horizontal direction was about $10_{-8}[S/cm]$. The activation energy for LB ultra-thin films with respect to variation of temperature was about 1.0[eV], which was correspond to semiconductor material. The response characteristics for organic gas were confirmed by measuring the response time, recovery time, and reproducibility of the LB ultra-thin to each organic gas. Also, the penetration and adsorption behavior of gas molecule were confirmed through the organic gas response characteristics of LB ultra-thin films with respect to temperature.

Intestinal Colonization Characteristics of Lactobacillus spp. Isolated from Chicken Cecum and Competitive Inhibition Against Salmonella typhimurium

  • Probiotics are live microbial feed supplements which beneficially affect the host animal by improving its intestinal microflora. To select the best Lactobacillus spp. as a chicken probiotic, probiotic characteristics of 10 selected Lactobacillus strains isolated from chicken cecum or obtained from KCTC were investigated. The strains were examined for resistance to pH 2.0 and 0.3% oxgall, and adhesion to cecal mucus and cecal epithelial cells. All strains grew in MRS containing 0.3% oxgall. However, Lb. plantarum AYM-10, Lb. fermentum YL-3, AYM-3, and Lb. paracasei YL-6 showed relatively high resistance to 0.3% oxgall. Lb. fermentum YL-3, YM-5, AYM-3, and Lb. paracasei YL-6 survived 4 hours of incubation at pH 2.0. Lb. fermentum YL-3, KCTC 3112, and Lb. plantarum AYL-5 were strongly adhesive to cecal mucus, while the rest showed moderate or low adhesion. Lb. plantarum AYM-10, AYL-1, and AYL-5 had good adhering properties to cecal epithelial cells (30.7$\pm$10.82, 40.2$\pm$20.90, and 14.5$\pm$4.22, respectively). Lb. fermentum YL-3, AYM-3, and KCTC 3547 showed Intermediate adhesion ability, and Lb. plantarum showed better adhesion ability to cecal epithelial cells than Lb. fermentum. Attached Lb. fermentum YL-3 to cecum after 60 min incubation was confirmed using CLSM. Lb. fermentum YL-3 attached to a matrix which was composed of a mucus layer adjacent to intracrypts and pericryptal region. Some Lb. fermentum YL-3 bound to mucosal epithelial cells. From these results, Lb. fermentum YL-3 was selected as a chicken probiotic. In vivo trials of chicks inoculated with Lb. fermentum YL-3 had decreased Salmonella population in cecal contents and livers (p<0.5).

  • The electrical properties of the Langmuir-Blodgett (LB) films layered with arachidic acid were studied at the room temperature. The sample was formed with 2 different structure ; One was Al/LB/Al and the other was Au/LB/Au. The precise structure of Al/LB/Al was considered as Al/Al$_{2}$O$_{3}$/LB/Al, because the natural oxide layer was formed on surface of lower Al electrode. The electrical conductivity of Al/Al$_{2}$O$_{3}$/LB/Al structure was determined the value of 3.5 * 10$^{-14}$ S/cm from the measurement of current-voltage (I-V) characteristics. The sample with the structure of Au/LB/Au was made to eliminate the influence of oxide layer in the electrical properties of the LB films. The short circuit current was observed in this sample from the I-V characteristics. To verify the reason of short circuit current generation, copper decoration method was employed to the 15 layers of LB films deposited on the Al and Au electrode each. The defects were shown on the films deposited with Au electrode. This results means that the defects on the LB films which layered with the Au electrode were contributed to the short circuit current. Several films (15, 31, 51, 71L) were deposited on the Au electrode and measured the size of defects with the copper decoration method. The size of defects becomes smaller as the film layer was increased. We conclude that the existence of defects affects the short circuit current generation.

Expression of Canavalia Iineata Leghemoglobin cDNA in Transgenic Nicotiana tabacum (형질전환된 담배에서 해녀콩 Leghemoglobin cDNA의 발현)

  • Tobacco (Nicotiana tahacum L. cv. Wisconsin 38) leaf discs were cocultivated with Agrohacterium carrying a leghemoglobin (Lb) cDNA from Canavalia lineata. Seven plants were regenerated from the transformed leaf discs on MS media supplemented with 0.5 mg/L BAP, 0.1 mg/L ${\alpha}-NAA$, 200 mg/L kanamycin and 500 mg/L carbenicillin. Southern hybridization and PCR of genomic DNA from transgenic plants showed that the Lb cDNA was stably integrated into the genome of the tobacco. Total RNA from the transgenic tobacco showed northern hybridization signal at 1,000 nt and PCR of the first strand cDNA synthesized from the total RNA amplified 0.5 kb Lb cDNA. Furthermore, western hybridization using a polyclonal antibody against soybean Lb showed a 15.8 kD LB-like band on SDS-PAGE of proteins from the transformed tobacco. These results demonstrated that the Lb cDNA of C. lineata was not only incorporated into the genome of tobacco, but also transcribed into mRNA and translated into Lb protein in the transformed tabacco.

A Study on the Structural Behavior of LB-DECK Panel Considering Rebar-Arrangement in Site (현장 배근이 LB-DECK 패널의 구조거동에 미치는 영향)

  • The objective of this study is to verify whether the composite action is sufficiently strong to withstand at the interface and the structural behavior of LB-DECK panel with field concrete slab strengthened with main reinforcing bars. Static and fatigue tests are performed for LB-DECK panels with varied shapes and amounts of rebars, and the results are compared with those of field concrete panel(FCP). The test results indicate that the LB-DECK panel with 1.5 times of more rebars inside significantly increase the overall stiffness. LB-DECK penel usually shows on average 52.1 percent of improved stiffness compared with the FCP. The fatigue test results also show that the LB-DECK panel can withstand two-million cycles of repeated loads without any damage.

  • Makgeolli is made from rice or flour, yeast, and nuruk, a fermentation starter. The flavor of makgeolli is affected by sugars, amino acids, organic acids and volatile flavor compounds produced by various microorganisms. In this study, lactic acid bacteria (LAB) were isolated from unsterilized makgeolli samples collected from several provinces in Korea, and then later identified. Under anaerobic conditions, LAB density ranged from $5.0{\times}10^6$ to $1.5{\times}10^8CFU/mL$; yeast density ranged from $2.5{\times}10^7$ to $1.5{\times}10^8CFU/mL$. Of the LAB isolated from makgeolli, 1,126 were analyzed using restriction fragment length polymorphism (PCR-RFLP) analysis of 16S rRNA, which allowed for classification into five groups. Of the 1,126 LABs tested, 130 produced ${\gamma}$-aminobutyric acid (GABA).
