Effects of the enhanced level of L-ascorbic acid (AA) on the water deficit-induced oxidative damage were studied in rice (Oryza sativa L.) seedlings. The seedlings sprayed with 20 to 80 mM L-galactono-${\gamma}$-lactone (GL), a putative precursor of AA, showed 2 to 5-fold higher levels of AA compared with controls. Pretreatment of the seedlings with GL prior to water stress imposition caused virtually no effect on dehydration of tissues during water deficit but substantially mitigated oxidative injury, as accessed by 2-thiobarbituric acid-reactive substances, ${\alpha}$-tocopherol, chlorophylls and ${\beta}$-carotene. Proline accumulation during water stress was also significantly lowered in the treated seedlings. In a complementary experiment, AA retarded photodegradation of ${\alpha}$-tocopherol in isolated thylakoids far more efficiently than glutathione. GL in itself did not show any noticeable reactivity toward ${\alpha}$-tocopheroxyl radical. The results demonstrate the antioxidative function of AA in rice seedlings encountering water-limited environments, suggesting a critical role of AA as a defense against oxidative stress in plants.
The purpose of this Study was to investigate effect of TakliSodokSan(TSS) on the anti-tumor, immunocytes and nitric oxide(NO) production from mice peritoneal macrophages. This Study estimated the proliferation of L1210 cell lines, A431 cell lines, Hep-G2 cell lines, K562 cell lines, 3T3 cell lines, mouse thymocytes and mouse splenocytes and NO production from pcritoneal macrophages in vitro, and estimated the proliferation of L1210 cells, thymocytes and splenocytcs, NO production from peritoneal macrophages and body weight in L1210 cells-transplanted mice in vivo. The results were obtained as follows; 1. TSS inhibited significantly the proliferation of L1210, A431, Hep-G2, K562 cell lines in vitro. 2. TSS accelerated the proliferation of mice thymocytes and splenocytes in vitro. 3. TSS was not increased the nitric oxide production from mice peritoneal macrophages in vitro. 4. TSS inhibited significantly the proliferation of L1210 cells in Ll210 cells∼transplanted mice. 5. TSS accelerated the proliferation of mice thymocytes and splenocytes In L1210 cells-transplanted mice. 6. TSS was increased significantly the nitric oxide production from mice peritoneal macrophages in L1210 cells-transplanted mice. 7. TSS was increased the body weight as comparing with control group in Ll210 cells-transplanted mice.
We investigated the effects of CP-2 extracted from the mixture of Copis and Croton tiglium L, which showed very high cytotoxic effect to the various tumor cells, on the growth and steroidenesis of primary and transformed cell lines PA-GS6 and PO-GRS1 by cotransfectionwith SV40 and Ha ras oncogenes. CP-2 inhibited the growth of PA-GS6 and PO-GRS1 in a dose dependent manner when the growth of them was measured by cell number and by protein content, while CP-2 did not affect the growth of primary granulose cells. Productions of progesterone ofprimary and transformed granulosa cells were stimulated by forskolin, but this stimulatory effect was blocked by treatment of CP-2. Clinical application of CP-2 asa new anti-cancer drug and utilization of transformed granulosa cells as a model system for the screening of anti-cancer drug were also discussed.
This experimental study was performed to investigate the antifungal effects of mixture of Sophorac Subprostratae Radix, Aconti Radix, and Hibisci Syriaci Cortex by the three types of extraction in the dermatophytes such as Trichophyton rubrum, Trichophyton mentagrophytes, and Epidermophyton floccosum. The following results were obtained by using Minimum Inhibition Concentration(MIC) and Inhibition Zone. 1. MIC on Trichophyton mentagrophytes in Sample S-2, Sample S-3, and Sample-Y were 10${\mu}l$, respectively, showing the highest antifungal effect. 2. MIC on Epidermophyton floccosum in Sample S-2, Sample S-3, and Sample-Y were 10㎕ respectively, showing the highest antifungal effect. 3. MIC on Trichophyton rubrum in Sample S-3, Sample-Y were 10${\mu}l$ respectively, showing the highest antifungal effect. 4. The size of inhibition zone on Trichophyton mentagrophytes for Sample S-3 were 22.3mm in 10${\mu}l$, showing the highest antifungal effect. 5. The size of inhibition zone on Epidermophyton floccosum for Sample S-3 was 34.3mm in 10${\mu}l$, showing the highest antifungal effect. 6. The size of inhibition zone on Trichophyton rubrum for Sample-Y was 26.5mm in 10${\mu}l$, showing the highest antifungal effect.
Nineteen flavonoids, five phenolics, two coumarins, maltol and L-ascorbic acid were tested as scavenger of nitrite which is believed to participate in the formation of N-nitroso compounds. Many were found to be potent scavenger and the five most potent ones were (+)-catechin, (-)-epicatechin, phloroglucinol, caffeic acid and L-ascorbic acid. The nitrite scavenging effect was higher at pH 1.2 than pH 3.0 and increased when the incubation time was longer. The possible relationship of structures to scavenging effect of the flavonoids tested was discussed.
This study explored how Korean L2 learners of English utilize multiple acoustic cues (VOT and F0) in perceiving and producing the English alveolar stop with a voicing contrast. Thirty-four 18-year-old high-school students participated in the study. Their English proficiency level was classified as either 'high' (HEP) or 'low' (LEP) according to high-school English level standardization. Thirty different synthesized syllables were presented in audio stimuli by combining a 6-step VOTs and a 5-step F0s. The listeners judged how close the audio stimulus was to /t/ or /d/ in L2 using a visual analogue scale. The L2 /d/ and /t/ productions collected from the 22 learners (12 HEP, 10 LEP) were acoustically analyzed by measuring VOT and F0 at the vowel onset. Results showed that LEP listeners attended to the F0 in the stimuli more sensitively than HEP listeners, suggesting that HEP listeners could inhibit less important acoustic dimensions better than LEP listeners in their L2 perception. The L2 production patterns also exhibited a group-difference between HEP and LEP in that HEP speakers utilized their VOT dimension (primary cue in L2) more effectively than LEP speakers. Taken together, the study showed that the relative cue-weighting strategies in L2 perception and production are closely related to the learner's L2 proficiency level in that more proficient learners had a better control of inhibiting and enhancing the relevant acoustic parameters.
An objective of local exhaust hood design is to design the hood to operate as efficiently as possible. The greatest loss normally occurs at the entrance to the duct, due to the vena contracta in the throat of the duct. This can be accomplished by minimizing the loss that results from the vena contracta. There have been little studied to be cost-effective approach as installing simple instrument inside the throat of the hood. The aims of this paper were to minimize entry loss using inner square, and to measure the effect of inner square when installed inside hood throat. The results of this study were as follows; First, the magnitude of vena contracta could be considered as the difference between direct measured velocity and calculated velocity, which is from Bernoulli theory. In circle hood, calculated velocity and direct measured velocity were 10.7m/sec and 10.3n/sec, respectively. And the calculated velocity and direct measured velocity in square hood were 7.7m./sec and 6.5m/sec, respectively. Second, effect of inner square by width was carried out. The widths of inner square were L/1(18cm), L/2(9cm), L/3(6cm) and L/6(3cm). In case inner square was installed with 3cm width, the entry of coefficient was 0.93, comparing with 0.85 of entry of coefficient of general hood. Third, effect of inner square by distance from hood inside surface to inner square was carried out. The distances were L/3(6cm), L/6(3cm), L/9(2cm) and L/l8(1cm). In case the distance was 3cm the best efficiency was shown (Ce= 0.93). Fourth, effect of inner square by location from hood entry to duct inside was carried out. The locations of inner square were entry(0cm), L/6(3cm), L/3(6cm), L/2(9cm) and L/l(12cm). In case the location was 0cm, 3cm and 6cm the entry of coefficients were 0.93, 0.92 and 0.90, respectively.
The lactic acid bacteria, including Latilactobacillus sakei and Latilactobacillus curvatus, have been widely studied for their preventive and therapeutic effects. In this study, the underlying mechanism of action for the antioxidant and immunostimulatory effects of two strains of heat-treated paraprobiotics was examined. Heat-treated L. sakei KU15041 and L. curvatus KU15003 showed higher radical scavenging activity in both the 2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and 2,2-diphenyl-1-picryl-hydrazyl (DPPH) assays than the commercial probiotic strain LGG. In addition, treatment with these two strains exhibited immunostimulatory effects in RAW 264.7 macrophages, with L. curvatus KU15003 showing a slightly higher effect. Additionally, they promoted phagocytosis and NO production in RAW 264.7 cells without any cytotoxicity. Moreover, the expression of tumor necrosis factor-α, interleukin (IL)-1β, and IL-6 was upregulated. These strains resulted in an increased expression of inducible nitric oxide synthase and cyclooxygenase-2. Moreover, the nuclear factor-κB and mitogen-activated protein kinase signaling pathways were stimulated by these strains. These findings suggest the potential of using L. sakei KU15041 and L. curvatus KU15003 in food or by themselves as probiotics with antioxidant and immune-enhancing properties.
