Evaluation of Liver Function and Blood Exam including hs-CRP in Adults with Nonalcoholic Fatty Liver Findings (비알코올성 지방간 소견을 보이는 성인에 대한 간 기능 및 hs-CRP 혈액 검사 항목 평가)

  • As a test for diagnosing fatty liver, recently, ultrasound and blood exam are being performed simultaneously. In particular, in the case of high-sensitivity C-reactive protein in blood exam, it is used as an index indicating the level of inflammation in various parts of the body as well as cardiovascular diseases. Thus, this study was conducted to analyze the association between metabolic syndrome components, liver function, and high-sensitivity C-reactive protein levels according to the degree of nonalcoholic fatty liver, and use it as a clinical indicator for fatty liver diagnosis. Metabolic syndrome components, liver function and high-sensitivity C-reactive protein blood test values analyzed from 1,139 men and women over 20 years of age with nonalcoholic fatty liver in abdominal ultrasonography from March 2021 to August 2021 at the Korea Association of Health Promotion, Gwangju-Jeonnam Branch. Analyzed for all men and women, the blood test values for subjects with mild fatty liver were AST 30 U/L, ALT 32.1 U/L, γ-GTP 41.2 IU/L, and hs-CRP 0.14 mg/dL. These values were lower than the blood test values of subjects with moderate fatty liver (AST 38 U/L, ALT 47.6 U/L, γ-GTP 54.9 IU/L, hs-CRP 0.22 mg/dL) and was statistically significant (p<0.001). In this case of high-sensitivity C-reactive protein test, it is statistically significant, showing higher values in Subjects with moderate fatty liver than Subjects with mild fatty liver. thus, it is considered that hs-CRP can be used as clinical data for the prevention and management of fatty liver.

Growth of Creeping Bentgrass after Application of Microbial Fertilizer Containing Saccharomyces cerevisiae HS-1 and Streptococcus thermophiles HS-2 (Saccharomyces cerevisiae HS-1와 Streptococcus thermophiles HS-2 함유 복합 미생물비료 처리 후 크리핑 벤트그래스의 생육)

  • This study was conducted to evaluate the effects of soil microbial fertilizer (SMF) containing Saccharomyces cerevisiae HS-1 and Streptococcus thermophiles HS-2 on the growth of creeping bentgrass. For the pot experiment, the treatments were as follows: no fertilizer (NF), control (3 N g/m2/month), SMF-1 (control+SMF 2 mL/m2/time), and SMF-2 (control+SMF 4 mL/m2/time). For the plot experiment, the treatments were as follows: NF, control, SMFp-1 (control+SMF 1 mL/m2/time), SMFp-2 (control+SMF 2 mL/m2/time), and SMFp-3 (control+SMF 4 mL/m2/time). In the pot experiment, visual turfgrass quality and the uptake amount of nitrogen (N) and potassium (K) were increased under the SMF treatments, whereas the content of chlorophyll (a, b, and a+b) and clipping yield were not considerably different compared with the control. In the pot experiment, the amount of SMF positively correlated with visual turfgrass quality and uptake amount of N and K. In the plot experiment, turfgrass density was increased by 12.9-19.2% under SMFp treatments compared with the control. These results indicated that the application of SMF containing Sa. cerevisiae HS-1 and St. thermophiles HS-2 improved the quality, density, and growth of creeping bentgrass via prompting the uptake of N and K.

Novel Discovery of LINE-1 in a Korean Individual by a Target Enrichment Method

  • Long interspersed element-1 (LINE-1 or L1) is an autonomous retrotransposon, which is capable of inserting into a new region of genome. Previous studies have reported that these elements lead to genomic variations and altered functions by affecting gene expression and genetic networks. Mounting evidence strongly indicates that genetic diseases or various cancers can occur as a result of retrotransposition events that involve L1s. Therefore, the development of methodologies to study the structural variations and interpersonal insertion polymorphisms by L1 element-associated changes in an individual genome is invaluable. In this study, we applied a systematic approach to identify human-specific L1s (i.e., L1Hs) through the bioinformatics analysis of high-throughput next-generation sequencing data. We identified 525 candidates that could be inferred to carry non-reference L1Hs in a Korean individual genome (KPGP9). Among them, we randomly selected 40 candidates and validated that approximately 92.5% of non-reference L1Hs were inserted into a KPGP9 genome. In addition, unlike conventional methods, our relatively simple and expedited approach was highly reproducible in confirming the L1 insertions. Taken together, our findings strongly support that the identification of non-reference L1Hs by our novel target enrichment method demonstrates its future application to genomic variation studies on the risk of cancer and genetic disorders.

Rapid Determination of Volatile Organic Compounds in Human Whole Blood Using Static Headspace Sampling with Gas Chromatography and Mass Spectrometry

  • Headspace (HS) and headspace solid-phase microextraction (HS-SPME) were studied for extracting volatile organic compounds (VOCs) from whole blood, with chemical and instrumental variables being optimized for maximum sensitivity: incubation at $60^{\circ}C$, equilibration for 30 min, pH 11, and 2 mL injection volume. Both techniques provided accurate analyses, with detection limits of 0.05-0.1 ng $mL^{-1}$ and 0.05-0.5 ng $mL^{-1}$. HS showed better sensitivity, reproducibility, and analysis times than HS-SPME. Overall levels of chloroform in whole blood were found to be 0.05-5.84 ng $mL^{-1}$; detected levels of benzene were 0.05-2.20 ng $mL^{-1}$.

Development of Analytical Technology Using the HS-SPME-GC/FID for Monitoring Aromatic Solvents in Urine

  • Headspace solid phase micro-extraction gas chromatography/flame ionization detection (HS-SPME-GC/FID) method was compared with headspace gas chromatography/mass selective detection (HS-GC/MS). Organic solvent-spiked urine as well as urine samples from workspace was analyzed under optimal condition of each method. Detection limit of each compound by HS-SPME-GC/FID was $3.4-9.5{\mu}g/L$, which enabled trace analysis of organic solvents in urine. Linear range of each organic solvent was $10-400{\mu}g/L$, with fair correlation coefficient between 0.992 and 0.999. The detection sensitivity was 4 times better than HS-GC/MS in selected ion monitoring (SIM) mode. Accuracy and precision was confirmed using commercial reference material, with accuracy around 90% and precision less than 4.6% of coefficient of variance. Among 48 urine samples from workplace, toluene was detected from 45 samples in the range of $20-324{\mu}g/L$, but no other solvents were found. As a method for trace analysis, SPME HS GC/FID showed high sensitivity for biological monitoring of organic solvent in urine.

Biological Control of Diamondback Moth (Plutella xylostella L.) by Lysobacter antibioticus HS124 (Lysobacter antibioticus HS124를 이용한 배추좀나방 (Plutella xylostella L.)의 생물학적 방제)

  • Lysobacter antibioticus HS124 was isolated from rhizosphere soil in previous experiments, which produced lytic enzymes such as chitinase, gelatinase, lipase and protease. In addition, HS124 released an antibiotic compound, 4-hydroxyphenylacetic acid (4-HPAA). When larvae of P. xylostella was treated with HS124 culture broth, its body was destroyed, and degraded with the increase of incubation time, yielding glycine which was detected from HS124 culture broth. When 4-HPAA produced from HS124 was sprayed, larvae mortality increased with increasing concentration of 4-HPAA. When HS124 culture supplemented with Tween 80 was sprayed, its insecticidal activity against larvae was approximately 1.4 times higher compared to the culture without Tween 80. Insecticide (IS), HS124 culture broth (HS124), Magic-pi (MP) and HS124 culture broth+Magic-pi (HS124+MP) were each treated against larvae of P. xylostella to investigate their insecticidal effect where sterile diluted water (SDW) was used as a control. The highest mortality of larvae was found in HS124+MP, followed by IS, MP, HS124 and SDW respectively. Mortality of larvae in HS124 was 31% higher than that in SDW, but 41% lower than that in HS124+MP, meaning that both enzymes and antibiotics produced from HS124 may synergistically act as active agents with plant extract containing neem oil and turmeric in HS124+MP treatment. These results suggested that L. antibioticus HS124 together with plant extract can be one of candidates for biocontrol agents against Plutella xylostella.

Isolation of Antifungal Compound and Biocontrol Potential of Lysobacter antibioticus HS124 against Fusarium Crown Rot of Wheat

  • Fusarium graminearum is the main cause of substantial economic loss in wheat production. The aim of this study is to investigate biocontrol potential of Lysobacter antibioticus HS124 against F. graminearum and to purify an antifungal compound. In preliminary study, n-butanol crude extract revealed destructive alterations in the hyphal morphology of F. graminearum and almost degraded with $1,000{\mu}g\;mL^{-1}$ concentration. For further study, the antifungal compound extracted from the n-butanol crude extract of L. antibioticus HS124 was identified as N-Butyl-tetrahydro-5-oxofuran-2-carboxamide ($C_9H_{16}NO_3$) using NMR ($^1H-NMR$, $^{13}C-NMR$, $^1H-^1H\;COSY$, HMBC, and HMQC), and HR-ESI-MS analysis. To our knowledge, N-Butyl-tetrahydro-5-oxofuran-2-carboxamide may be a novel compound with molecular weight of 186.1130. The minimum inhibitory concentration value of antifungal compound was $62.5{\mu}g\;mL^{-1}$ against F. graminearum. In an in vivo pot experiment, crown rot disease from F. graminearum was inhibited when wheat seeds were treated with both HS124 culture and F. graminearum. Growth of wheat seedling was enhanced by treatment of HS124 compared to control. Our results suggest that L. antibioticus HS124 characterized in this study could be successfully used to control F. graminearum and could be used as an alternative to chemical fungicides in modern agriculture.

Comparison of serum amyloid A protein and C-reactive protein levels as inflammatory markers in periodontitis

  • Purpose: The purpose of this study was to compare serum amyloid A (SAA) protein levels with high-sensitive C-reactive protein (hs-CRP) levels as markers of systemic inflammation in patients with chronic periodontitis. The association of serum titers of antibodies to periodontal microbiota and SAA/hs-CRP levels in periodontitis patients was also studied. Methods: A total of 110 individuals were included in this study. Patients were assessed for levels of hs-CRP and SAA. Nonfasting blood samples were collected from participants at the time of clinical examination. The diagnosis of adipose tissue disorders was made according to previously defined criteria. To determine SAA levels, a sandwich enzyme-linked immunosorbent assay was utilized. Paper points were transferred to a sterile tube to obtain a pool of samples for polymerase chain reaction processing and the identification of Porphyromonas gingivalis, Aggregatibacter actinomycetemcomitans, and Tannerella forsythia. The serum level of IgG1 and IgG2 antibodies to P. gingivalis, A. actinomycetemcomitans, and T. forsythia was also determined. Results: SAA and hs-CRP levels were higher in periodontitis patients than in controls (P<0.05). In bivariate analysis, high levels of hs-CRP (>3 mg/L) and SAA (>10 mg/L) were significantly associated with chronic periodontitis (P=0.004). The Spearman correlation analysis between acute-phase proteins showed that SAA positively correlated with hs-CRP (r=0.218, P=0.02). In the adjusted model, chronic periodontitis was associated with high levels of SAA (odds ratio [OR], 5.5; 95% confidence interval [CI], 1.6-18.2; P=0.005) and elevated hs-CRP levels (OR, 6.1, 95% CI, 1.6-23.6; P=0.008). Increased levels of serum IgG2 antibodies to P. gingivalis were associated with high levels of SAA (OR, 3.6; 95% CI, 1.4-8.5; P=0.005) and high concentrations of hs-CRP (OR, 4.3; 95% CI, 1.9-9.8; P<0.001). Conclusions: SAA and hs-CRP concentrations in patients with chronic periodontitis are comparably elevated. High serum titers of antibodies to P. gingivalis and the presence of periodontal disease are independently related to high SAA and hs-CRP levels.

A Study on the Removal of Fluorine from Leachate of Reclaimed Mine Waste Dump Site Using Alum Coagulation (알럼 응집을 이용한 광산폐기물 적치장 침출수 내 불소 제거 연구)

  • This study was conducted to remove fluorine (F) (initial concentration of 9.5 mg/L) from leachate of reclaimed mine waste dump site via different methods: (1) co-precipitation using Ca-based materials; (2) adsorption using activated carbon and fly ash; and (3) coagulation and sedimentation using alum. The F removal efficiencies of each case were estimated as 65.6% (Ca co-precipitation), 27.9% (adsorption of activated carbon), 71.5% (adsorption of fly ash), and 96.6% (alum coagulation and sedimentation). In addition, the applicability of the continuous treatment process using alum coagulation was evaluated by lab-scale experiments using simulated mine drainage containing F of lower (6.4 mg/L) and higher (15.7 mg/L) concentrations, and it was confirmed that the treatment of both cases met the domestic standard (below 3 mg/L) for discharged water in clean areas. Furthermore, the results of bench-scale field tests indicated that the water quality standard of discharged water could be satisfied with the proper operation and management of the process.

Association between Employment Status and hs-CRP Level in Korean Wage Workers (국내 임금근로자에서 고용형태에 따른 hs-CRP level과의 연관성)

  • Objectives: We aimed to find the relationships between employment status and hs-CRP level among Korean wage workers using the 2016 Korean National Health and Nutrition Examination (KNHANE, revised). Methods: This study was conducted on 1,937 daytime wage workers over the age of 19 and within a normal weight range (18.5≤BMI≺25). Regular workers were defined as those granted an employment guarantee until reaching retirement age, and non-regular workers were defined as temporary, non-typical, dispatched, short-term workers and contractors. For hs-CRP, three divisions were classified as recommended by the Centers for Disease Control and Prevention (CDC) and American Heart Association (AHA) with less than 1.0 mg/L indicating low risk of cardiovascular disease, above 1.0 mg/L and below 3.0 mg/L considered moderate risk, and more than 3.0 mg/L indicating high risk. To find the relationship between work type and hs-CRP level in Korean wage workers, multinominal logistic regression analysis was performed. Results: For non-regular workers, the odds ratio of the cardiovascular moderate-risk group and cardiovascular high-risk group was statistically significant compared to regular workers. After adjusting for factors such as gender, age, subjective health status, income, education, smoking, and physical labor, the odds ratio of the cardiovascular high-risk group was statistically significant. Conclusions: In this study, the relationship between non-regular workers and high hs-CRP level was examined. Based on this, institutional strategies should be pursued to prevent and manage cardiovascular disease among non-regular workers.
