The role of Lactobacillus reuteri DSM 17938 for the absorption of iron preparations in children with iron deficiency anemia

  • Purpose: To determine whether Lactobacillus reuteri DSM 17938 plays a role in absorption of iron preparations given to children with iron deficiency anemia (IDA). Methods: We performed a quasi-experimental study involving pre- and postintervention tests using a control group in North Sulawesi province, Indonesia, between July and September 2017. We conducted a single-blind controlled trial that included primary school children who were diagnosed with IDA based on reticulocyte hemoglobin equivalent (Ret-He) levels <27.8 pg/L. Results: A total of 66 children were randomized into 2 groups. Thirty-four children received iron preparations with the addition of L. reuteri DSM 17938 (group 1), whereas the other 32 received iron preparations alone (group 2). The baseline Ret-He levels before intervention were similar in both groups. After 14 days of intervention, mean Ret-He level in group 1 changed from $24.43{\pm}1.64$ to $28.21{\pm}1.72pg/L$ (P=0.000). Mean Ret-He level in group 2 changed from $24.31{\pm}1.42$ to $27.03{\pm}2.14pg/L$ (P=0.000). Statistical analysis showed a significant increase in Ret-He levels in both groups; Ret-He levels were significantly higher in the experimental group than in the control group (P<0.05). Conclusion: Children with IDA receiving iron preparations with L. reuteri DSM 17938 for 14 days show higher Ret-He levels than those receiving iron preparations alone.

Anti-Candida Activity of YH-1715R, a New Triazole Derivative

  • YH-1715R, (2R,3R)-2-(2,4-difluorophenyl)-3-(3-methoxy-1,2,4-isothiazol-3-yl-thio)-1-( 1H-1,2,4-triazol-l-yl)-2-butanol, a new triazole derivative obtained by the structural modification of fluconazole, was found to exhibit potent anti-Candida activity against a wide variety of Candida albicans (C. albicans) (MIC: 0.4-12.5 mg/l). To investigate the mode of action of YH-1715R, its effect on ergosterol biosynthesis in cell-free extracts and whole cells of C. albicans was examined. The inhibitory activity of YH-1715R was approximately ten-fold higher than that of fluconazole. To determine the primary action mechanism of YH-1715R, its inhibitory activity against lanosterol $14\alpha$-demethylase (14$\alpha$-DM), a major target for azole, was measured using gas-liquid chromatography. YH-1715R and fluconazole were found to inhibit 14a-DM with an $IC_{50}$ of 0.015 $\mu$M and 0.01$8\mu$M, respectively, plus the mode of inhibition of YH-1715R and fluconozole was noncompetitive with a $K_i$ of 0.0533$\mu$M and 0.0975$\mu$M.

  • Claviceps purpurea PRL 1980 produces a fluorescent reddish brown pigment in the alkaloid production medium. When D,L-tryptophan $[side\;chain-3-^{14}C]$ was administered into the production medium, the radioactive pigment and 5-hydroxytryphan were isolated from the cultures. Conversion of tryptophan to 5-hydroxytryptophan in vivo was shown by an isotopic trapping procedure. 5-hydroxytryptophan isolated from the cultures contained appreciable radioactivity and was recrystallized to constant specific radioactivity. The injection of the $^{14}C-labelled$ 5-hydroxytryptophan showed an incorporation of radioactivity into brown pigment significantly higher than that of tryptophan. The brown pigment produced by Claviceps purpurea PRL 1980 seems to be derived from tryptophan through 5-hyrdroxytryptophan.

GNSS Antenna PCO/PCV and Position Changes due to the Switch IGS08/igs08.atx to IGS14/igs14.atx

  • For precise GNSS applications, the antenna phase center correction (PCC) is absolutely required. The PCC magnitude can reach the centimeter level with the antenna structure. In the present study, we first investigate the phase center offset (PCO) and phase center variation (PCO) of three different antenna models in two different reference frames, IGS08/igs08.atx and IGS14/igs14.atx. Clear L1 and L2 PCO differences were found between IGS08 and IGS14. In addition, the PCV showed characteristics that is dependent upon the signal direction (azimuth and elevation angle). The remarkable thing is that the changes of a Dorne Margolin choke-ring antenna model (AOAD/MT DOME) was very small in two reference frames. In order to analyze changes in positions according to different reference systems, GNSS data obtained from DAEJ, SUWN, and TSKB stations were processed by the precise point positioning (PPP) method. We suggest that an antenna PCO/PCV can affect the precise GNSS positioning on the order of several millimeters in two different reference frames.

A Study on the Location of Measurements in the Abdominal Fat using CT (In asymptomatic Korean individual) (CT를 이용한 복부 지방측정에서 측정 위치에 대한 고찰 (무증상 한국인을 대상으로))

  • This study was performed to compare the differences between total fat area, subcutaneous fat area and visceral fat area according to L1-S1 inter vertebral disc level in asymptomatic Korean patients with non-contrast abdominal CT scan. The location of the maximum fat ratio of the total fat area is the total fat area measured in male L2-3, L3-4, L4-5, female L4-5, and L5-S1. The location showing the maximum fat ratio of the subcutaneous fat area is the subcutaneous fat area measured in male L4-5, and the woman is the subcutaneous fat area measured in L4-5 and L5-S1. The location of the maximum fat ratio of visceral fat area was the visceral fat area measured in L1-2 and L2-3 males, and there was no difference in visceral fat area according to the measured position in females. Although abdominal fat measurement is common at the L4-5 inter vertebral disc level, fat distribution varies widely depending on nationality, race, and gender. Therefore, it is necessary to analyze through the research data which location to choose according to the research purpose, and to mention why the location is appropriate.

The Influence of the Application of NEES to GV20 and L14, on HSP27 and HSP70, after the Ischemic Brain Injury

  • This research was attempted to seek for a positive approach within the framework of physical therapy instead of the drug treatment in the past, with regard to the ischemic brain injury in the early stage. Accordingly, the aim of this research is to observe the change of HSP27 and HSP70, the genes that are expressed in the early stage of brain injury and to investigate the effects of needle electrode electrical stimulation(NEES), upon applying NEES after ischemia. The experimental method is to give rise to global ischemia and apply NEES to 27 SD-Pat rats with the particulars of being eight-week-old, male, around 300g, and adapted to laboratory environment for more than a week, and divide them into three groups, that is, GV20 NEES group(n=9), L14 NEES group(n=9), no applied NEES global ischemia(GI) group(n=9), and then observe their changes of HSP27 and HSP70 at the time lapse of 6, 9 hr and 12 hr after ischemia, using immunohistochemistry methods. Upon observing through the immunohistochemistry method, it was noticed that there was a significant difference between the GV20 NEES group and the L14 NEES group as for HSP27 and there were significant differences among all groups as for HSP70(p<.05). Accordingly, it is supposed that the application of NEES after the outbreak of cerebral ischemia delay the apoptosis in the early ischemic part of forebrain or protect neurons against apoptosis.

Studies on Leptospiral Antibody in Korean Cattle and Pigs (Leptospira 속균(屬菌)에 대한 한우(韓牛)와 돈(豚)의 혈중항체조사(血中抗體調査))

  • Investigation of leptospiral antibody in Korean cattle and pigs was carried out from February to October, 1971. Ten different living antigens, namely L. icterohaemorrhagiae, L. canicola, L. antumnalis, L. hebdomadis, L. australis A, L. pomona, L. pyrogenes, L. grippotyphosa, L. bataviae and L. javanica, were used. A total of 590 Korean cattle and 460 pig blood samples collected from Seoul Majang-dong slaughterhouse were tested by the rapid microscopic agglutination test. Throughout the studies the following results were obtained and summarized. 1. Of 590 serum samples of Korean cattle 51 were positive(8.64%). 2. Of 460 serum samples of pigs 27 were positive (5.87%). 3. Of 51 positive cattle samples, 29(4.92%) showed antibody to a serotype of L. icterohaemorrhagiae and 18(3.0%) to L. canicola, and 4 (0.68%) to L. pomona. Eight of L. icterohaemorrhagiae positive samples showed a cross reaction to L. canicola. 4. Of 27 positive pig samples, 14(3.04%) showed antibody to L. ictereohaemorrhagiae and 7(1.52%) to L. grippotyphosa. 4(0.87%) to L. canicola, 2(0.43%) to L. pomona. Two of L. canicola positive samples showed a cross reaction to L. grippotyphosa. 5. Serum samples of seven pigs, showing antibody positive to L. grippotyphosa were first observed in Korea. 6. Infection rate of bovine and porcine leptospirosis, in Korea, appeared to be lower than that of Japan, Taiwan, Thailand and the Philippines.

Free Radical Scavenging Activity and Protective Ability of Methanolic Extract from Duchesnea indica Against Protein Oxidation and DNA Damage

  • The antioxidant potency of methanolic extract of Duchesnea indica (MDI; Indian strawberry) was investigated by employing various established in vitro systems, such as total phenolic content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity, reducing power assay, metal chelating assay, superoxide radical scavenging activity and protective ability of DNA damage and protein oxidation. MDI inhibited metal chelating by 75.57% at 2 mg/mL, scavenged 50% DPPH free radical at 29.13 ${\mu}$g/mL, and eliminated approximately 46.21% superoxide radical at the concentration of 1 mg/mL. In addition, MDI showed strong ability on reducing power, DNA damage protection and protein oxidation protection. Overall, results suggested that MDI might be beneficial as a potent antioxidant and effectively employed as an ingredient in food applications.

Improving 3'-Hydroxygenistein Production in Recombinant Pichia pastoris Using Periodic Hydrogen Peroxide-Shocking Strategy

  • 3'-Hydroxygenistein can be obtained from the biotransformation of genistein by the engineered Pichia pastoris X-33 strain, which harbors a fusion gene composed of CYP57B3 from Aspergillus oryzae and a cytochrome P450 oxidoreductase gene (sCPR) from Saccharomyces cerevisiae. P. pastoris X-33 mutants with higher 3'-hydroxygenistein production were selected using a periodic hydrogen peroxide-shocking strategy. One mutant (P2-D14-5) produced 23.0 mg/l of 3'-hydroxygenistein, representing 1.87-fold more than that produced by the recombinant X-33. When using a 5 L fermenter, the P2-D14-5 mutant produced 20.3 mg/l of 3'-hydroxygenistein, indicating a high potential for industrial-scale 3'-hydroxygenistein production.

Continuous Production of Gluconic Acid and Sorbitol from Glucose and Fructose using Perrneabilized cells of Zymomonas mobilis (투과화된 Zymomonas mobilis 균체를 이용한 Glucose와 Fructose로 부터 Gluconic Acid와 Sorbitol의 생산)

  • Continuous and simultaneous production of gluconic acid and sorbitol from glucose and fructose was carried out by using glucose-fructose oxidoreductase and glucanolactonase of Zymomonas mobilis. In order to utilize the enzymes without purification, Zymomonas mobilis was permeabilized with toluene. Optimum conditions for permeabilization and reaction kinetics of permeabilized Zymomonas mobilis were studied. In batch operation with the permeabilized cells immobilized in alginate beads, about 90% conversion was obtained within 35 h reaction. Continuous production of gluconic acid and sorbitol using the immobilized permeabilized cells was carried out. Optimum conditions for continuous operation with the imn~obilized cells were; pH 6.2 and temperature $40^{\circ}C$. Maximum productivities for gluconic acid and sorbitol were about 14.5 g/l/h and 14.8 g/l/h respectively at the dilution rate of 0.075 $h^{-1}$ when 300 g/l each of substrates was fed.
