Antioxidant Properties of Different Polysaccharides Extracted with Water and Sodium Hydroxide from Rice Bran

  • The antioxidant properties of water extracted polysaccharides (PW), 1%(w/v) NaOH extracted polysaccharides (PN1), and 5%(w/v) NaOH extracted polysaccharides (PN5) were assessed in this paper. PW showed good capability of scavenging $H_2O_2$, anti-lipid peroxidation, reduction power, and scavenging 1,1-diphenyl-2-picrylhydrazyl (DPPH). The maximum values were all observed at the dose of 1 mg/mL sample. As far as the antioxidant activities of PN1 were concerned, the capability of scavenging superoxide radical, chelating metal, and total antioxidation showed higher than those of PN5. When polysaccharide concentration was 1 mg/mL, the 3 index described above were attached 71.8%, 51.6%, and $446.3{\mu}M$ Trolox equivalent, respectively. With respect to PN5, higher capabilities of scavenging hydroxyl free radicals were obtained while the peak of 84.8% was observed at the concentration of 1.0 mg/mL. Moreover, the concentration-dependent influences were characterized in all cases.

The Influences of Addition of Sugar with or without L. buchneri on Fermentation and Aerobic Stability of Whole Crop Maize Silage Ensiled in Air-stress Silos

  • The whole plant of crop maize was chopped and ensiled in double-layered polyethylene bags to determine the influence of residual sugar on the fermentation of lactic acid and aerobic stability by L. buchneri in whole crop maize silage made in airstress condition. There were a total of six treatments used in this experiment as follow: added 25 g de-mineralised water per kg chopped maize serving as control (con), 37.5 g glucose solution containing 12.5 g glucose ($g_1$), 75 g glucose solution containing 25 g glucose ($g_2$), 25 g, L,.buchneri suspension intended for $10^6$ cfu $g^{-1}$ (L.b.), $g_1$+L.b. and $g_2$+L.b. All silos were opened at day 91 after ensiling for measuring the pH values, microbiological enumeration, fermentative products and aerobic stability. The dry matter loss increased significantly (p<0.01) due to inclusion of sugar or L. buchneri. The lower lactic acid concentrations were observed (p<0.01) in silages inoculated with L. buchneri only or in combination with sugar addition than the correspondent uninoculated silages. Compared with control silage, ethanol production was about 3 or 6-fold higher due to addition 12.5 or 25 g glucose per kg chopped maize at ensiling. The silages added with sugar contained less acetic acid concentration (p<0.01) than control, but silages inoculated with L. buchneri showed the contrary effects (p<0.01) at different sugar levels. No butyric acid was found in uninoculated silages, silages inoculated with L. buchneri. producted more propionic acid, 1-propanol and butyric acid. Lactic acid bacteria counts increased markedly (p<0.01) due to inoculation with L. buchneri, whereas it was reduced (p<0.01) by added sugar. No significant difference was observed in count of yeast, but inoculation with L. buchneri shows a decreasing trend. Mould count in all silages was less than 2 (log cfu $g^{-1}$). The added sugar had negative effects on aerobic stability of maize silage made under air-stress conditions, whereas inoculation with L. buchneri improves (p<0.01) the aerobic stability.

문형 공작기계의 Zig-Zag 오차 분석 (Analysis of Zig-Zag Error in Gantry Type Machine)

  • 대형 5축 문형공작기계는 Gantry가 bed를 따라 절삭, 급송 이송하면서 가공을 수행 한다. 가공을 수행하는 중에 발생하는 오차(Zig-Zag)원인으로써 1) 양쪽 X축 이송모터의 위치오차 제어차이 2) 양쪽 Column의 무게차이에 의해 발생하는 마찰력에 대한 오차 3) spindle 회전력에 따른 오차 3가지의 원인이 있다. 3가지 오차원인을 분석하여 문형공작기계성능을 향상시키기 위해 연구를 진행 하였다. 연구결과로는 첫 번째로 양쪽 Column이 동일한 무게를 갖는 Gantry 구조에서 한쪽 축의 무게를 변경함으로써 마찰력이 미치는 영향을 확인하였다. 두 번째로 우리는 스핀들 회전이 미치는 영향을 해석 및 실험하였다. 그리고 우리는 스핀들 회전이 Gantry이송에 미치는 영향을 확인하였다.

Temperature Dependent Characteristics of a Combined Discharge Type Ozonizer (CDO)

  • A combined discharge type ozonizer was designed and manufactured. The increase or decrease of temperature greatly influences the characteristics of ozone concentration ( $O_{3con}$), ozone generation ( $O_{3g}$) and ozone yield ( $O_{3Y}$) of a discharge type ozonizer. The characteristics of ozone concentration, ozone generation and ozone yield rate were investigated by varying the gas flow rate (Q), the discharge power ( $W_{d}$) and the temperature (T). At T=25[$^{\circ}C$] the values of $O_{3con}$ were found as 5632, 4200, 2500 and l800[ppm] at Q = 1, 2, 4 and 6[l/min] respectively. At the same temperature the corresponding values of $O_{3g}$ were found as 662, 988, 1176 and l270[mg/h] and those of $O_{3Y}$ were found as 67, 102, 119 and 135[g/kWh] respectively. When the temperature is decreased to -50[$^{\circ}C$], the values of $O_{3con}$ were found as 9000, 6700, 4000 and 2800[ppm] respectively at Q = 1, 2, 4 and 6[l/min]. At the same value of temperature the corresponding values of $O_{3g}$ were found as 1220, 1576, 1882 and 2050[mg/h] and those of $O_{3Y}$ were found as 120, 159, 188 and 202[g/kWh] respectively. Hence as the temperature was decreased from 25 to -50[$^{\circ}C$], the efficiencies of ozone generation were increased by 79, 55, 58 and 49[%] respectively at Q = 1, 2, 4 and 6[l/min].]. 6[l/min].].

Relation among Food Consumption, Conversion and Cocoon Production in Silkworm, Bombyx Mori L. Reared at Different Temperatures

  • Rearing temperature influences food consumption, conversion and cocoon production in silkworm Bombyx mori L. Young age $({1^st}to{3^rd} instar)$ worms of Pure Mysore, a multivoltine were reared at 26, 28, 30 and $32^{\circ}C$ followed by combinations of these temperature levels during late age $({4^th} and {5^th} instar)$. Food consumption, conversion, larval duration and growth for each instar were recorded. The influence of combinations of these temperature levels during rearing on survival and cocoon production was assessed. Food consumption increased with the temperature up to $4^th$ instar and decreased during $5^th$ instar. Temperature beyond $28^{\circ}C$ during young age followed by higher temperature ($30^{\circ}C$ and above) has shown deleterious effect on cocoon production. The paper discusses the rearing performance in relation to food intake and conversion under the tested temperature levels.

황산염용액으로부터 전기도금 된 FCCL용 Cu 필름의 특성에 미치는 Cu 이온농도 및 첨가제의 영향 (Effect of Cu2+ Concentration and Additives on Properties of Electrodeposited Cu Thin Films for FCCL from Sulfate Baths)

  • Nanocrystalline Cu thin films were electrodeposited from sulfate baths and investigated systematically the influences of $Cu^{2+}$ concentration and additives on current efficiency, residual stress, surface morphology, and XRD patterns of electrodeposited Cu film. Current efficiency was nearly 100% at from 0.2M to 1.0 M $Cu^{2+}$ concentration. but it was linearly increased with $Cu^{2+}$ concentration at less than 0.2M. The residual stress was observed in range of 7.9 to 18.4 MPa and tensile stress mode. Dendritic and powdered form was obtained at below 0.1 M. As increased with $Cu^{2+}$ concentration in solution, the main peak in the XRD pattern shifted (111) and (220) from (200). In the other hand, all about 100% current efficiency observed in all additive concentration systems, and residual stress observed in range of 20.4 to 26.3 MPa tensile stress. The condition 5(Ultra make-up - 10 ml/l, Ulta A - 0.5ml/l, Ultr B - 0.5 ml/l) was good surface morphology, and fcc(111) peak in XRD patterns increased with increasing additive concentration.

Candida mogii ATCC 18364를 이용한 D-Xylose로부터 Xylitol 생산 (Xylitol Production from D-Xylose by Candida mogii ATCC 18364)

  • 볏짚 가수분해물로부터 xylitol 생산능력이 가장 우수하다고 알려진 Candida mogii ATCC 18364의 xylose 발효특성에 관한 기초연구를 수행하였다. 상업적인 자일로즈로부터 C. mogii게 의해 xylitol을 생산할 때 수율과 속도에 미치는 여러 영향을 검토한 결과, 포도당을 함유한 기질에서 12시간 배양한 종균을 3.0 g/L의 농도로 접종하여 발효시켰을 때 가장 좋은 결과를 얻었다. 또한 xylose 발효에서 가장 중요한 요인인 용존산소 농도의 영향을 알아보기 위해, 2 L 발효조에서 통기량과 교반속도를 조절해 가면서 xylitol 생산을 비교 검토하였다. 농축균과 10% 접종균을 사용했을 경우 모두, 1 vvm의 통기량과 200 rpm의 교반속도에서 xylitol 생산이 가장 우수하였다.

Potentiation of T Cell Stimulatory Activity by Chemical Fixation of a Weak Peptide-MHC Complex

  • The stability of peptide-MHC complex (pMHC) is an important factor to shape the fate of peptide-specific T cell immune response, but how it influences on T cell activation process is poorly understood. To better understand that, we investigated various T cell activation events driven by $L^d$ MHCI loaded with graded concentrations of P2Ca and QL9 peptides, respectively, with 2C TCR Tg T cells; the binding strength of P2Ca for $L^d$ is measurably weaker than that of QL9, but either peptides in the context of $L^d$ interact with 2C TCR with a similar strength. When their concentrations required for early T cell activation events, which occur within several minutes to an hour, were concerned, $EC_{50}s$ of QL9 were about 100 folds lower than those of P2Ca, which was expected from their association constants for $L^d$. When $EC_{50}s$ for late activation events, which takes over several hours to occur, were concerned, the differences grew even larger (> 300 folds), suggesting that, due to weak binding, $L^d/P2Ca$ dissociate from each other more easily to lose its antigenicity in a short time. Accordingly, fixation of $L^d/P2Ca$ with paraformaldehyde resulted in a significant improvement in its immunogenicity. These results imply that binding strength of a peptide for a MHC is a critical factor to determine the duration of pMHC-mediated T cell activation and thus the attainment of productive T cell activation. It is also suggested that paraformaldehyde fixation should be an effective tool to ameliorate the immunogenicity of pMHC with a poor stability.

해양환경용 알루미늄 합금의 플라즈마 전해 산화 시 표면 특성에 관한 불화칼륨(KF)의 영향 (Influences of Potassium Fluoride (KF) Addition on the Surface Characteristics in Plasma Electrolytic Oxidation of Marine Grade Al Alloy)

  • In this study, we investigated the influences of potassium fluoride(KF) addition on the surface characteristics of plasma electrolytic oxidation(PEO) coating produced on Al alloy. The PEO of marine grade Al alloy(5083 grade) was conducted in KOH 1g/L solution adding different concentrations of KF(0, 1 and 2 g/L) under a galvanostatic regime. With KF addition, unusual behavior was observed on the voltage-time characteristic curves, which can be characterized by the following process: (i) initial rapid increase in voltage (ii) a short plateau after 1st breakdown (iii) gradual increase in voltage (iv) intermittent fluctuation of voltage after 2nd breakdown. The SEM observation revealed irregular surface morphology with KF addition, as compared with one formed without KF addition, which had a reticulate structure. The XRD analysis detected the formation of aluminium hydroxide fluoride hydrate($H_{4.76}Al_2F_{3.24}O_{3.76}$) on surface grown by PEO process with KF. Particularly, at very early stage of the process (~ 120 s), thin film was formed having nanoporous structure, and F element was confirmed on surface by EDS analysis. The thickness and surface roughness of the coating increased with increasing KF concentration. As a result, KF addition was found to be less beneficial influences on PEO of marine grade Al alloy, and therefore needs further research to improve its capability.

개량 12Cr-1Mo강에서 탄소 함량 및 응고속도에 따른 응고 조직 형성 거동 (Solidification Microstructures with Carbon Contents and Solidification Rates in Modified 12Cr-lMo Steels)

  • The influences of solidification rates and carbon contents on the formation of the $\delta$-ferrite were studied by directional solidification in modified 12%Cr-l %Mo steels. Directional solidification experimental results showed that solidification microstructure depended on solidification rate and carbon content and chromium equivalent. The length of the mushy zone increased and the dendrite arm spacings decreased as the solidification rate increased. The volume fraction of the 8-ferrite decreased with increasing the solidification rate and carbon content. The volume fraction of the ferrite showed much higher at low solidification rates with planar and cellular interfaces than that at high solidification rates with dendritic interface. It is expected that macro-segregation of C causes lower C content at the lower solidification fraction in the directionally solidified sample, where lower C results in higher volume fraction of the ferrite. In order to estimate solidification microstructure in modified 12Cr-l%Mo steels, various solidification conditions, such as solidification rate, cooling rate, segregation, alloy composition, should be considered.
