  • Polarizer와 Analyzer의 Rotation 각에 의해 광의 luminance를 조절할 수 있다. Contrast Sensitivity에 대응한 luminance 값 $L_{max}$, $L_{min}$은 평균 luminance($L_m$)의 회전각 ${\theta}_m$ 최대 진폭과 최소 진폭에 대응하는 회전각 (${\theta}_{max}$, ${\theta}_{min}$)로 부터 구할 수 있다. $$L_{max}=I(0)e^{-2at}{\cdot}cos^2{\theta}_m(1+C_s^{-1})$$ $$L_{min}=I(0)e^{-2at}{\cdot}cos^2{\theta}_m(1-C_s^{-1})$$ 평균 luminance($L_m$)의 회전각 ${\theta}_m$과 측정할 Contrast Sensitivity($C_s$)로부터 polarizer와 analyzer의 회전각(${\theta}_{max}$, ${\theta}_{min}$)을 얻었다. $${\theta}_{max}=cos^{-1}[cos{\theta}_m{\cdot}(1+C_s^{-1})^{1/2}]$$ $${\theta}_{min}=cos^{-1}[cos{\theta}_m{\cdot}(1-C_s^{-1})^{1/2}]$$.

  • 본 연구에서는 액상성장법(LPE법)으로 $Bi_2$-x(sub)$LSr_2$Ca(sub)1+x(sub)$LCu_2$O(sub)8+d (x(sub)L=0, 0.05, 0.1, 0.2) 막(film)을 작성하여 초전도특성을 알아보았고, XPS 측정으로 얻은 Sr3d과 Ca2p의 피크분해 결과와 EPMA 측정 결과를 통하여 각 layer에 있어서 원자들의 분포상태에 관하여 조사하였다. 작성한 막(film)의 c-축의 길이는 x(sub)L(융해조성비)의 증가에 따라 단조로이 증가하였고, 임계온도 T(sub)c는 x(sub)L=0.1 부근에서 최고치를 나타내고 있었다. x(sub)L의 변화에 따른 임계온도 T(sub)c와 c-축의 길이의 변화는 BiO-layer에 있는 과잉산소의 변화에 의한 것이며, SrO-layer에서 원소들의 분포상태와 결손이 초전도특성 및 결정구조에 영향을 미치고 있음을 알 수 있었다.

R.F. sputtering 방법에 의해 c-BN 표면처리된 316L 오스테나이트계 스테인리스 강의 내마모특성 향상 (Wear Resistance of c-BN Surface Modified 316L Austenitic Stainless Steel by R.F. Sputtering)

  • Cubic boron nitride (c-BN) is a promising material for use in many potential applications because of its outstanding physical properties such as high thermal stability, high abrasive wear resistance, and super hardness. Even though 316L austenitic stainless steel (STS) has poor wear resistance causing it to be toxic in the body due to wear and material chips, 316L STS has been used for implant biomaterials in orthopedics due to its good corrosion resistance and mechanical properties. Therefore, in the present study, c-BN films with a $B_4C$ layer were applied to a 316L STS specimen in order to improve its wear resistance. The deposition of the c-BN films was performed using an r.f. (13.56 MHz) magnetron sputtering system with a $B_4C$ target. The coating layers were characterized using XPS and SEM, and the mechanical properties were investigated using a nanoindenter. The friction coefficient of the c-BN coated 316L STS steel was obtained using a pin-on-disk according to the ASTM G163-99. The thickness of the obtained c-BN and $B_4C$ were about 220 nm and 630 nm, respectively. The high resolution XPS spectra analysis of B1s and N1s revealed that the c-BN film was mainly composed of $sp^3$ BN bonds. The hardness and elastic modulus of the c-BN measured by the nanoindenter were 46.8 GPa and 345.7 GPa, respectively. The friction coefficient of the c-BN coated 316L STS was decreased from 3.5 to 1.6. The wear property of the c-BN coated 316L STS was enhanced by a factor of two.

의사위성의 펄싱 방법에 대한 GPS L2C 신호획득 성 소능프분트석웨어 수신기의 L1, L2C 신호획득 성능분석 (GPS L1, L2C Signal Acquisition Performance of GPS Software Receiver with respect to Pseudolite Pulsing Scheme)

  • 의사위성은 우주상공의 GPS 위성과는 달리 지상이나 비행체에 설치되어 GPS 위성과 같이 거리를 측정할 수 있는 ranging 신호를 전송하는 전송기이다. 의사위성은 정확성, 가용성, 무결성을 향상시킬 수 있으나 GPS 위성 신호에 대해 신호의 포화 또는 간섭을 일으킨다. 의사위성의 근원거리 문제를 해결하기 위해 의사위성의 신호를 펄싱이 있을 때만 내보내는 기법을 주로 사용한다. 본 논문에서는 의사위성의 정적 펄싱, 이동 펄싱, 랜덤 펄싱 기법과 의사위성의 개수가 GPS 소프트웨어 수신기의 L1 및 L2C 신호획득에 미치는 영향을 분석하였다. GPS L1신호의 경우 의사위성이 1개일때는 정적펄싱에 대한 GPS 소프트웨어 수신기의 신호 획득 및 추적 성능이 가장 좋았고 2개 이상일 때는 랜덤펄싱에 대한 성능이 가장 좋았다. L2C 신호는 세가지 펄싱에 대한 성능이 비슷하게 안정적으로 나왔는데 정적펄싱에 대한 성능이 약간 좋게 나왔다. L1, L2C 모두 위사위성 3개까지는 모든 펄싱에 대해서 측위가 가능한 것으로 나왔다.

미세조류 4종의 성장, CO2 동화 및 지질 생성 특성 (Characterization of Cellular Growth, CO2 Assimilation and Neutral Lipid Production for 4 Different Algal Species)

  • 미세조류는 효율적으로 바이오매스를 증가시킬 수 있으며 유용한 생물학적 자원들을 축적할 수 있기 때문에 에너지 및 식품 생산 등 다양한 분야에서 유망한 자원으로써 주목받고 있다. 본 연구에서는 4종의 미세조류(Chlorella vulgaris, Mychonastes homosphaera, Coelastrella sp., Coelastrella vacuolata)를 선정하여 이들의 성장, CO2 동화, CO2 농도에 따른 미세조류의 지질 생성 특성을 분석하였다. 각 미세조류의 크기는 C. vulgaris가 가장 작았으며, M. homosphaera, C. vacuolata, Coelastrella sp. 순으로 큰 크기를 나타냈다. C. vulgaris는 다른 3종의 미세조류와 비교해서 크기가 가장 작으며 성장과 CO2 동화 속도가 가장 빠르게 나타났다. 또한, 초기 바이오매스가 증가함에 따라 CO2 동화 속도는 최대 9.62 mmol·day-1·l-1를 나타냈으며, 다른 3종의 미세조류(약 3 mmol·day-1·l-1)보다 3배 이상 높은 CO2 동화 속도를 보여주었다(p < 0.05). M. homosphaera를 제외하고 3종의 미세조류는 CO2 농도와 CO2 동화 비속도 사이에 양의 상관관계(positive correlation)를 나타냈다. 특히, C. vulgaris는 다른 3종의 미세조류와 비교해 더 높은 CO2 동화 비속도를 보여주었다(14.6 vs. ≤ 11.9 mmol·day-1·l-1). 4종의 미세조류는 CO2 농도가 증가함에 따라 지질 함량이 증가했으며 그 중에서 C. vulgaris는 최대 18 mg·l-1를 나타내 다른 3종의 미세조류(최대 12 mg·l-1)보다 최소 50% 이상 높은 지질 함량을 보여주었다. 4종의 미세조류 중 C. vulgaris가 효율적으로 CO2를 동화하며 다른 미세조류보다 높은 바이오매스와 지질 생산이 가능함을 시사한다.

  • Erro-prone PCR에 의해서 열안정성이 향상된 Streptomyces coelicolor A(3) 유래의 L-threonine aldolase를 Corynebacterium glutamicum R에서 과잉발현시키기 위하여 Corynebacterium용 vector plasmid인 pCRB1의 SD배열과 개시코든사이의 1염기를 제거한 고발현용 vector plasmid인 pCG-H44(2)를 구축하였다. pCG-H (2)에 의해서 형질전환된 C. glutamicum R 균주(CGH44-2)에서 L-TA를 발현시킨 결과, 기존의 Corynebacterium용 vector plasmid인 pCRB1(CGH44-1)보다 L-TA의 발현량이 높았다. L-threo-DOPS의 합성을 위한 최적조건은 $30^{\circ}C$, 0.1 M cirtric acid buffer(pH 7.0)이었으며, 0.1% TritonX-100를 첨가하였을 경우 보다 높은 활성을 보였다. 최적조건하에서 CGH44-2를 whole cell biocatalyst로 이용한 반복회분식반응에서 재조합대장균을 숙주로 이용한 경우보다 재조합Corynebacterium을 이용하였을 경우, 목적하는 L-threo-DOPS의 합성이 안정적으로 이루어졌다.

Ca2+/calmodulin-dependent regulation of polycystic kidney disease 2-like-1 by binding at C-terminal domain

  • Polycystic kidney disease 2-like-1 (PKD2L1), also known as polycystin-L or TRPP3, is a non-selective cation channel that regulates intracellular calcium concentration. Calmodulin (CaM) is a calcium binding protein, consisting of N-lobe and C-lobe with two calcium binding EF-hands in each lobe. In previous study, we confirmed that CaM is associated with desensitization of PKD2L1 and that CaM N-lobe and PKD2L1 EF-hand specifically are involved. However, the CaM-binding domain (CaMBD) and its inhibitory mechanism of PKD2L1 have not been identified. In order to identify CaM-binding anchor residue of PKD2L1, single mutants of putative CaMBD and EF-hand deletion mutants were generated. The current changes of the mutants were recorded with whole-cell patch clamp. The calmidazolium (CMZ), a calmodulin inhibitor, was used under different concentrations of intracellular. Among the mutants that showed similar or higher basal currents with that of the PKD2L1 wild type, L593A showed little change in current induced by CMZ. Co-expression of L593A with CaM attenuated the inhibitory effect of PKD2L1 by CaM. In the previous study it was inferred that CaM C-lobe inhibits channels by binding to PKD2L1 at 16 nM calcium concentration and CaM N-lobe at 100 nM. Based on the results at 16 nM calcium concentration condition, this study suggests that CaM C-lobe binds to Leu-593, which can be a CaM C-lobe anchor residue, to regulate channel activity. Taken together, our results provide a model for the regulation of PKD2L1 channel activity by CaM.

몰약(沒藥) 에탄올 추출물의 항비만에 관한 연구 (The Study on anti-obesity of Myrrh ethanol extract)

  • Objectives : The objective of this study was to investigate the effect of Myrrh 80% ethanol extract on adipocyte differentiation and adipogenesis in 3T3-L1 cell.Methods : Myrrh was prepared by extracting with 80% ethanol. Cell viability was assessed by MTT assay using 3T3-L1 cells. Anti-obesity activity was measured in lipid droplets and triglyceride (TG) accumulation in 3T3-L1 cells. We also analyzed the expression of C/EBPβ, C/EBPα, PPARγ, SREBP1c, and aP2 by reverse transcriptase polymerase chain reaction (RT-PCR). In addition, we observed the production of fatty acid, acetyl-CoA carboxylase and Oil-red O stainingResults : No cytotoxicity from Myrrh 80% ethanol extracts was observed at the concentration of 1, 10, 100 (㎍/㎖) in 3T3-L1 cells. Treatment with Myrrh significantly suppressed the terminal differentiation of 3T3-L1 in a dose-dependent manner, as confirmed by a decrease in triglyceride and Fatty acid and Acetyl-CoA carboxylase. Also, Myrrh exhibited potential adipogenesis inhibition and downregulated the expression of pro-adipogenic transcription factors, such as sterol regulatory element binding protein-1c (SREBP-1c), peroxisome proliferator-activated receptor-γ (PPARγ), CCAAT/enhancer binding proteins α (C/EBPα) and C/EBPβ, and adipocyte expressed genes, such as adipocyte fatty acid binding protein (aP2) and Fas. In addition, lipid accumulation determined by Oil-red O staining showed that Myrrh extract had inhibitory effects on lipid accumulation in 3T3-L1 cells.Conclusions : These results suggest that Myrrh suppresses obesity factors in 3T3-L1 cells. Myrrh may be a useful medical herbs for attenuating metabolic diseases such as obesity.

Increased Production of Ginsenoside Compound K by Optimizing the Feeding of American Ginseng Extract during Fermentation by Aspergillus tubingensis

  • The ginsenoside compound K (C-K) is widely used in traditional medicines, nutritional supplements, and cosmetics owing to its diverse pharmacological activities. Although many studies on C-K production have been conducted, fermentation is reported to produce C-K with low concentration and productivity. In the present study, addition of an inducer and optimization of the carbon and nitrogen sources in the medium were performed using response surface methodology to increase the C-K production via fermentation by Aspergillus tubingensis, a generally recognized as safe fungus. The optimized inducer and carbon and nitrogen sources were 2 g/l rice straw, 10 g/l sucrose, and 10 g/l soy protein concentrate, respectively, and they resulted in a 3.1-fold increase in the concentration and productivity of C-K (0.22 g/l and 1.52 mg/l/h, respectively) compared to those used before optimization without inducer (0.071 g/l and 0.49 mg/l/h, respectively). The feeding methods of American ginseng extract (AGE), including feeding timing, feeding concentration, and feeding frequency, were also optimized. Under the optimized conditions, A. tubingensis produced 3.96 mM (2.47 g/l) C-K at 144 h by feeding two times with 8 g/l AGE at 48 and 60 h, with a productivity of 17.1 mg/l/h. The concentration and productivity of C-K after optimization of feeding methods were 11-fold higher than those before the optimization (0.22 g/l and 1.52 mg/l/h, respectively). Thus, the optimization for the feeding methods of ginseng extract is an efficient strategy to increase C-K production. To our knowledge, this is the highest reported C-K concentration and productivity via fermentation reported so far.
