꿩의비름(Sedum erythrostichum Miq.)의 잎과 줄기 절편으로부터 식물체의 재분화에 미치는 생장조절제의 영향 (Effect of Plant Growth Regulators on Plant Regeneration from Leaf and Stem Explant Cultures of Sedum erythrostichum Miq.)

  • 꿩의비름(Sedum-erythrostichum Miq.)의 잎과 줄기의 절편을 몇 가지의 호르몬이 첨가된 MS 배지에 배양하였다. 배양 2주후 2.0 mg/L. 2,4-D와 1.0 mg/L BA를 혼합한 배지에서 잎의 절편으로부터 100%의 캘러스가 형성되었다. 이 캘러스를 2.0 mg/L 2,4-D와 1.0 mg/L BA를 혼합한 배지로 계대배양하였을때 캘러스의 생장이 왕성하였다. 또한 2.0 mg/L NAA와 1.0 mg/L BA를 혼합한 배지로 계대배양한 캘러스로부터 부정아의 분화가 왕성하였다. 생장호르몬이 없는 배지에서 분화된 신초는 1.0 mg/L IAA를 첨가한 배지에서 발근하여 완전한 식물체로 생장하였다.

하향주 누룩으로부터 분리한 젖산균의 동정 및 발효 특성 (Identification and Fermentation Characteristics of Lactic Acid Bacteria Isolated from Hahyangju Nuruk)

  • 하향주의 제조에 이용되는 누룩으로부터 젖산균을 분리하고, 배양학적 특성과 생리적 특성을 검토하였다. 분리된 총 11균주를 이용하여 알코올 10%와 pH 4.0에서 생육이 억제되는 양조학적으로 적합한 5균주의 젖산균을 분리하였고, 16S rRNA의 염기서열을 분석하여 동정한 결과 Lactobacillus plantarum 1균주, Lactobacillus sakei 1균주 및 Lactobacillus curvatus 3균주로 나타났으며 모두 젖산 간균으로 확인되었고, 이들을 각각 L. plantarum L-3, L. sakei L-10 그리고 L. curvantus L-8, 11, 12로 명명하였다. L plantarum L-3이 배양 초기 pH 4.0일 때 0.78%의 산도를 나타내어 나머지 4균주에 비해 약 10배정도 높은 산을 생성하였다. 분리된 5균주는 모두 $30^{\circ}C$에서 가장 산생성능이 우수하였고, L. sakei L-10을 제외한 모든 균주는 $45^{\circ}C$에서 생육이 정지되었으나, L. sakei L-10은 $40^{\circ}C$에서 생육이 저해되었다. 그리고 에탄을 농도가 6%일 때 산의 생성이 급격히 감소되었으며 L. curvatus L-8, 11, 12는 8% 에탄올에서 거의 생육이 이루어지지 않아 에탄올에 대한 내성이 낮았으며, L. plantarum L-3과 L sakei L-10은 10% 에탄올에서 생육이 정지되었다.

  • 에리스리톨을 산업적으로 대량생산하기 위해서는 에리스리톨의 생산성을 높이고 부산물인 글리세롤의 생성을 억제하는 배지 및 발효 공정의 최적화가 필요하다. 에리스리톨 생성을 위한 최적 탄소원은 포도당이고 최적 농도는 400 g/L이며 최적 온도는 3$0^{\circ}C$였으며 이 때 산소는 과잉으로 공급해 주어야 함을 알았다. 에리스리톨의 수율과 생산성을 향상시키기고 배지가격을 줄이기 위해 효모 추출물의 사용을 5 g/L에서 3 g/L로 줄이고 urea 2.72g/L$K_2HPO_4$ 1.79 g/L, MgSO$_4$. $7H_2O$ 0.18 g/L를 첨가해 줌으로써 에리스리톨 수율을 31.4%에서 45.2%로 에리스리톨 생산성을 0.747 g/L/h에서 1.071 g/L/h로 향상시켰다. 또한 글리세롤의 생산량도 96.6 g/L에서 45.7 g/L로 줄었다. 이 최적 배지를 바탕으로 5L 발효기에서의 재현성과 pH에 대한 영향을 알아보기 위해 실험을 한 결과 pH를 조절한 경우에 에리스리톨 생산성이 낮아졌다. 5L 발효에서는 다량의 거품이 발생되는 것을 관찰하였다.

Antibacterial activity of Chamaecyparis obtuse extract and Profile of Antimicrobial Agents Resistance for Metallo-β-lactamase-Producing Pseudomonas aeruginosa

  • In vitro antimicrobial activities of hot water extracts of Chamaecyparis obtuse, for clinical metallo-β-lactamase-Producing Pseudomonas aeruginosa (MBLPA.) was compared to commonly used conventional antimicrobial agents. All MBLPA was susceptible to colistin or amikacin, but also to imipenem 88.6%, meropenem 100%, piperacillin 85.7%, ceftazidime 97.1%, gentamicin 97.1%, and ciprofloxacin 100% were non-susceptible. MIC range to imipenem, meropenem, cefotaxime, ceftazidime, gentamicin, and ciprofloxacin for MBLPA were each 1 - >128 ㎍/mL, 4 - >128 ㎍/mL, 4 - >128 ㎍/mL, 8 - >128 ㎍/mL, 4 - >128, and 2- >128 ㎍/mL. MIC range to aztreonam for MBLPA were 1 - 128 ㎍/mL. MIC90 to imipenem, meropenem, cefotaxime, ceftazidime, gentamicin, and ciprofloxacin for MBLPA were each 32 ㎍/mL, >128 ㎍/mL, >128 ㎍/mL, >128 ㎍/mL, >128 ㎍/mL, and 128 ㎍/mL. MIC90 to colistin and amikacin were each 1 ㎍/mL and 64 ㎍/mL. The hot water extracts of C. obtuse leaf had the lowest MIC range (0.25 - >0.5 μL/mL), MIC50 (>0.5 μL/mL), and MIC90 (>0.5 μL/mL) of the clinical MBLPA tested, and it was possible more potent than various conventional antimicrobial agents for MBLPA infection patients. Therefore, it suggested the possibility of using extract components of C. obtuse or their derivatives to treat MBLPA infection patients.

  • The addition reactions of cysteine without blocking amino and carboxyl groups to substituted and unsubstituted ${\beta}$-nitro-styrene derivatives were investigated. ${\beta}$-Nitrostyrene(1a), p-methyl-${\beta}$-nitrostyrene(1b), 3,4,5-trimethoxy-$[\beta}$ -nitrostyrene(1c), $[\varpi}$-3,4-methylenedioxy-${\beta}$ -nitrostyrene(1d), o-, m- and p-chloro-${\beta}$ -nitrostyrene (1e, 1f, 1g) and o-, m- and p-methoxy-${\beta}$-nitrostyrene (1h, 1i, 1j) easily undergo addition reactions with cysteine to form S-(2-nitro-1-phenylethyl)-L-cysteine(3a), S-[2-nitro-1-(p-methyl)phenyl-ethyl]-L-cysteine(3b), S-[2-nitro-1-(3',4',5'-trimethoxy) phenylethyl]-L-cysteine(3c), S-[2-nitro-1-($[\vatpi}$ -3',4'-methylenedioxy)phenylethyl]-L-cysteine(3d), S-[2-nitro-1-(o-chloro)phenylethyl]-L-cysteine(3e), S-[2-nitro-1-(m-chloro)-phenylethyl]-L-cysteine(3f), S-[2-nitro-1-(p-chloro)phenylethyl]-L-cysteine(3g), S-[2-nitro-1-(o-methoxy)phenylethyl]-L-cysteine(3h), S-[2-nitro-1-(m-methoxy)phenylethyl]-L-cysteine(3i) and S-[2-nitro-1-(p-methoxy)phenylethyl]-L-cysteine(3j), respectively. The structure of adducts were confirmed by means of UV-spectrum, IR-spectrum, molecular weight measurement and elemental analysis. The various factors effecting the yield of cysteine adducts to ${\beta}$-nitrostyrene derivatives were also studied.

  • 조화 단진동자 파동함수를 써서 원자핵의 LS에너지 행열요소를 계산하였다. 범위는 1$_1$= $l_{s}$ , $l_2$=lp, $l_3$=ld, 2s, $l_4$=1f, 2p, $l_{5}$ =1g, 2d, 3s라 ( $l_{i}$ $l_{i+1}$$l_{i}$ $l_{i+1}$)의 배치에 대한 것이었다. 계산결과는 Talmi적분 $I_1$과 Slater 적분 $F^{k}$ 를 써서 표시하였다. 또 여러가지 합법칙을 유도하고 이를 써서 계산의 결과를 검산하였다.하였다.

  • Six-coordinate molybdenum(Ⅴ)-oxo complexes (PyH)[MoOCl2L] and (R4N)-[MoOCl2L] (R=CH3 and C2H5) with N-salicylidene-2-aminophenol(L1) and its derivatives(L2=5-CH3, L3=3-CH3O, L4=5,6-C4H4 and L5=5-NO2) as ONO-donor ligands have been synthesized and the spectral and electrochemical properties of the complexes by elemental analysis, molar conductivity, UV-vis, IR, 1H NMR and CV have been studied.

Listeria monocytogenes Serovar 4a is a Possible Evolutionary Intermediate Between L. monocytogenes Serovars 1/2a and 4b and L. innocua

  • The genus Listeria consists of six closely related species and forms three phylogenetic groups: L. monocytogenes-L. innocua, L. ivanovii-L. seeligeri-L. welshimeri, and L. grayi. In this report, we attempted to examine the evolutionary relationship in the L. monocytogenes-L. innocua group by probing the nucleotide sequences of 23S rRNA and 16S rRNA, and the gene clusters lmo0029-lmo0042, ascB-dapE, rplS-infC, and prs-ldh in L. monocytogenes serovars 1/2a, 4a, and 4b, and L. innocua. Additionally, we assessed the status of L. monocytogenes-specific inlA and inlB genes and 10 L. innocua-specific genes in these species/serovars, together with phenotypic characterization by using in vivo and in vitro procedures. The results indicate that L. monocytogenes serovar 4a strains are genetically similar to L. innocua in the lmo0035-lmo0042, ascB-dapE, and rplS-infC regions and also possess L. innocua-specific genes lin0372 and lin1073. Furthermore, both L. monocytogenes serovar 4a and L. innocua exhibit impaired intercellular spread ability and negligible pathogenicity in mouse model. On the other hand, despite resembling L. monocytogenes serovars 1/2a and 4b in having a nearly identical virulence gene cluster, and inlA and inlB genes, these serovar 4a strains differ from serovars 1/2a and 4b by harboring notably altered actA and plcB genes, displaying strong phospholipase activity and subdued in vivo and in vitro virulence. Thus, by possessing many genes common to L. monocytogenes serovars 1/2a and 4b, and sharing many similar gene deletions with L. innocua, L. monocytogenes serovar 4a represents a possible evolutionary intermediate between L. monocytogenes serovars 1/2a and 4b and L. innocua.

벨트를 이용한 도수견인이 허리뼈 척추사이원반 탈출증 환자의 허리뼈 척추사이 공간에 미치는 효과 (The Effects of Manual Traction Using Belt on Lumbar Intervertebral Space of Lumbar HIVD)

  • Background: Manual traction with a belt is a physiotherapy treatment method that reduces disk pressure and widens the disk space. In clinical settings, it is applied to numerous patients with herniated intervertebral disk (HIVD). This study aimed to identify the effects of manual traction with a belt on the intervertebral space in patients with lumbar HIVDs. Methods: The intervention was performed on 17 patients with lumbar HIVDs who were divided into two groups: one with eight patients having HIVD at L4~L5 and another group with nine patients having HIVD at L5~S1. The participants received manual traction with a belt twice a week for 12 weeks, and radiographic imaging was used to visualize the intervertebral space and compare it before and after treatment. Results: Manual traction with a belt increased the lumbar intervertebral space at L4~L5 and L5~S1 in patients with L4~L5 HIVD. A significant difference was observed in the L4-L5 distance (p<.01); however, no significant difference was observed in the L5~S1 distance (p>.05). The intervertebral space significantly increased at both L4~L5 and L5~S1 in patients with L5~S1 HIVD (p<.05). Conclusion: Thus, manual traction with a belt increased the intervertebral space in patients with L4~L5 and L5~S1 HIVDs. These results are expected to guide studies on manual traction with belts in clinical settings in the future. Further studies using the present research as an objective study method are anticipated.

부분적(部分的)인 균일전단하중(均一傳達荷重)을 받는 평판(平板)에서의 응력분포(應力分布) (Stress Distributions in a Plate due to Shear Loading Uniformly Distributed on the End Portions of its Side Boundary.)

  • The plate under shear loading umformly distributed on the end portions of its side boundary was considered. Infinite hyperbolic serieses and Fourier serieses were combined as a stress function and from which exact solutions for the 15 cases for the parameters of b/L=0.25, 0.5, 1.0 and l/L=0.2, 0.4, 0.6, 0.8, 1.0 are obtained. In each cases the first 5 terms of the infinite series at the 36 points as shown in Fig. 3. The results are presented in Fig. 4-1, 4-2, and 4-3. The conclusions are as follows: 1) The stresses ${\sigma}_x$ increase very slightly as $\chi$ increases in the range of 0<x<L-l 2) When the parameters satisfy the conditions b/L<0.25 and l/L<0.2, the stresses in the region of 0<x<L-l can be obtained by replacing the uniform shear loading by the equivalent uniform shear loading by the equivalent uniform tensile force and pure bending moment at x=l. 3) The stress ${\sigma}_y$ is negligible throughout the region. 4) When the parameter b/L varies, the stresses ${\sigma}_x$ and u vary as L/b, while strain $\upsilon$ varies as $(L/b)^2$.
