Resistance Screening to Pepper mild mottle virus Pathotypes in Paprika Cultivars (고추약한모틀바이러스 병원형에 대한 파프리카 품종의 저항성 스크리닝)

  • The Paprika plant infected with Pepper mild mottle virus (PMMoV) do not produce commercial fruit as causing necrotic spots symptom on the fruit. Ten cultivars of paprika were analyzed to select the resistance cultivars against PMMoV pathotypes, $P_{1.2}$ and $P_{1.2.3}$, using bioassay and genetic markers. $L^1$, $L^3$, and $L^4$ genotypes expressing resistance to the pathotypes existed in those cultivars but $L^2$ genotype did not. $L^4L^4$ in cvs. Easy and Magnifico, $L^4L^3$ in cvs. Scirocco and Orange glory F1, $L^4L^1$ in cv. Special F1, $L^3L^3$ in cvs. Fiesta, Piero and Derby, and $L^3L^1$ in Cupra and Mazzona F1 were identified with SCAR and CAPS markers. The resistant cvs. to the 2 pathotypes were Magnipico, Easy, Scirocco F1, Orange glory and Special F1 and the susceptible cvs. were Fiesta, Piero, Derby, Cupra and Mazzona F1. The susceptible cvs. of the absence of $L^4$ genotype showed systemic infection when inoculated with PMMoV-$P_{1.2.3}$. However, those cvs. despite the presence of $L^3$ genotype showed vein necrosis on the inoculated leaf and hypersensitive necrosis symptom on the upper parts when inoculated with PMMoV-$P_{1.2}$.

  • When a $CH_2Cl_2$ solution of the dipyridyl species L' (N,N'-bis-(1-pyridin-4-yl-ethylidene)-naphthalene-1,5-diamine) was layered onto the top of a MeOH solution of $Co(NO_3)_2{\cdot}6H_2O$, a molecular cobalt compound [$CoL_2(MeOH)(NO_3)_2$] (1), not a coordination polymer, was formed. X-ray structural analysis of compound 1 revealed that it contains the pyridyl-amine ligand L (N1-(4-imino-1-methyl-but-2-enylidene)-naphthalene-1,5-diamine), instead of L'. Structure of compound 1 strongly suggests that the original ligand L' has been hydrolyzed to ligand L during the reaction.

Antimicrobial Effect of Chlorine Dioxide on Vase Life of Cut Rose 'Beast' (Chlorine Dioxide가 절화장미 수명연장에 미치는 항균효과)

  • This study was conducted to investigate the antimicrobial effect of chlorine dioxide ($ClO_2$) on the vase life of cut rose 'Beast' (Rosa hybrida L.). Postharvest treatments to extend the vase life of cut roses were divided into two: holding solution treatment and pulsing solution treatment. In holding solution treatment, the cut roses were treated with preservative solutions containing tap water (TW, control), distilled water (DW), $ClO_2$ 2, 4, 6, and $8{\mu}L{\cdot}L^{-1}$, and compared with a commercialized antimicrobial compound of 8-HQS $200{\mu}L{\cdot}L^{-1}$. In pulsing solution treatment, cut roses were dipped into the $ClO_2$ solutions of 50, 100, 150, 200, and $250{\mu}L{\cdot}L^{-1}$ for 60 seconds and were placed in DW. The air temperature was $18.4^{\circ}C$, RH 51.5%, and light (photosynthetically active radiation, PAR) $3.6{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ with 12 hour day length. The longest vase life of cut roses was observed in the holding solution with $ClO_2$ $4{\mu}L{\cdot}L^{-1}$ as 13.8 days and pulsing with $200-250{\mu}L{\cdot}L^{-1}$ as 13.5-13.7 days, where vase life were extended four days longer than TW. Whereas, the inclusion of 8-HQS $200{\mu}L{\cdot}L^{-1}$ in vase solution resulted in phytotoxicity. The relative fresh weight and water uptake have similar tendencies. Bacteria inhibition by $ClO_2$ and 8-HQS were very effective. But bacteria at TW and DW treatments on cut flower with stem were detected in $3.7{\times}10^5CFU{\cdot}L^{-1}$ and $6.3{\times}10^5CFU{\cdot}L^{-1}$, respectively (without stem in DW $1.4{\times}10^4CFU{\cdot}L^{-1}$). The $ClO_2$ contents in holding solution of all treatments were scavenged in two-four days after treatment. This study indicated that $ClO_2$ $4{\mu}L{\cdot}L^{-1}$ holding solution treatment and $200-250{\mu}L{\cdot}L^{-1}$ pulsing solution treatment can be applied to extend the postharvest life of cut roses.

  • Four strains of Sporobolomyces genera were isolated from soy sauce mashes at the fermentating and aging periods and identified. Their salt-resistance and color formation were examined in order to improve the color density and nutrition of red pepper paste preparations. The results obtained were as follows; a) The strain L1, L2, L3 and L4 isolated were identified as Sporobolomyces gracilis, Sporobolomyces roseus, Sporobolomyes gracilis and Sporobolomyces roseus respectively. b) The yeasts isolated were grown on the media containing $0{\sim}10$ percent of sodium chloride but their growth were almost restrained on the media containing $15{\sim}18$ per cent of sodium chloride. And the strain L2 and L3 were more salt-resistant as compared with L1 and L4. c) The strain L3 were the best among the 4 strains of yeasts comparison with their salt-resistance, color and flavour formation.

Callus Induction and Somatic Embryogenesis from Sicyos angulatus L. (야생식물 Sicyos angulatus L.로부터 캘러스 유도 및 체세포배 발생)

  • In order to investigate the possibility of in vitro mass propagation via somatic embryogenesis from Sicyos angulatus L., effects of plant growth regulators and carbon sources on callus induction and somatic embryogenesis were evaluated. Optimal combinations of plant growth regulator for callus induction from cotyledon and inflorescence explants were 2,4-D 2.0 mg/L + BA 0.1 mg/L and 2,4-D 1.0 mg/L + BA 0.1 mg/L in MS basal medium supplemented with sucrose 30 g/L,, respectively. Somatic embryogenesis was observed from cultured inflorescence explants, but it could not be achieved from leaf or cotyledon explants. The most effective plant growth regulators for somatic embryogenesis from callus was NAA 1.0 mg/L + kinetin 10 mg/L in the half strength of MS basal medium supplemented with 20 g/L sucrose.

Effect of Medium Compositions on the Mycelial Growth of Inonotus obliquus (차가버섯의 균사체 성장에 대한 배지성분의 영향)

  • Effect of temperature($22-32^{\circ}C$), pH(5-7) and medium composition on the mycelial growth for the submerged culture of Inonotus obliquus. The concentrations of glucose, starch, peptone, yeast extract, $K_2HPO_4$, $MgSO_4{\cdot}7H_2O$ and $CaCl_2$ were examined in the ranges of 30-120 g/L, 0-10 g/L, 0-20 g/L, 0-15 g/L, 0-2 g/L, 0-1.5 g/L and 0-0.5g/L, respectively. The maximum mycelial growth of Inonotus obliquus was obtained for $26-27^{\circ}C$ and pH 6. The concentrations of glucose, yeast extract and $CaCl_2$, which gave the maximum mycelial growth of Inonotus obliquus, were 70 g/L, 5 g/L and 0.1 g/L, respectively. In the cases of starch, peptone and $K_2HPO_4$, the mycelial growth of Inonotus obliquus increased with increasing the concentrations. However, as the concentration of $MgSO_4{\cdot}7H_2O$ increased, the mycelial growth of Inonotus obliquus decreased. The medium for maximum mycelial growth of Inonotus obliquus consisted of (per 1 L): glucose, 70 g; peptone, 5-20 g; starch, 10 g; yeast extract, 5 g; $K_2HPO_4$, 2 g and $CaCl_2$, 0.1 g.

  • The structures of dioscin(1) and saponin P-d(2), isolated from the subterranean part of Allium fistulosum, were determined as diosgenin 3-O-$\alpha$-L-rhamnopyranosyl (1longrightarrow2)-[$\alpha$-L-rhamnopyranosyl (1longrightarrow4)]-$\beta$-D-glucopyranoside (1, ) and diosgenin 3-O-$\alpha$-L-rhamnopyranosyl (1longrightarrow4)-$\alpha$-L-rhamnopyranosyl(1longrightarrow4)[$\alpha$-L-rhamno-pyranosyl (1longrightarrow2)]-$\beta$-D-glucopyranoside (2) by spectroscopic and chemical degradational methods. The prosa-pogenin(4), which was provided on partial hydrolysis of 2, was not reported in previous literature.

The Effect of Pyroligneous Liquor and Coconut Water on Plantlet Multiplication and in Vitro Flowering of Dendrobium moniliforme (목초액 및 코코넛액이 석곡(Dendrobium moniliforme)의 유묘 증식과 기내 개화에 미치는 영향)

  • This experiment was carried out to clarify the effect of pyroligneous liquor and coconut water on plantlet multiplication and in vitro flowering of Dendrobium moniliforme. Plantlet growth and multiplication was good in 1.0 ml/L pyroligneous liquor treatment which was added to the basal media of 3 g/L hyponex and 4 g/L peptone ($H_{3} P_{4}$) containing 0.1 mg/L NAA and 1.0 mg/L kinetin. In the treatment of 30 ml/L coconut water showed good results on plantlet growth and multiplication. In vitro flowering showed highest rate in the treatment of 1.0 ml/L pyroligneous liquor and 30 ml/L coconut water which were added to the basal media of $H_{3} P_{4}$ containing 0.1 mg/L NAA and 1.0 mg/L kinetin.

Studies on Leptospiral Antibody in Korean Cattle and Pigs (Leptospira 속균(屬菌)에 대한 한우(韓牛)와 돈(豚)의 혈중항체조사(血中抗體調査))

  • Investigation of leptospiral antibody in Korean cattle and pigs was carried out from February to October, 1971. Ten different living antigens, namely L. icterohaemorrhagiae, L. canicola, L. antumnalis, L. hebdomadis, L. australis A, L. pomona, L. pyrogenes, L. grippotyphosa, L. bataviae and L. javanica, were used. A total of 590 Korean cattle and 460 pig blood samples collected from Seoul Majang-dong slaughterhouse were tested by the rapid microscopic agglutination test. Throughout the studies the following results were obtained and summarized. 1. Of 590 serum samples of Korean cattle 51 were positive(8.64%). 2. Of 460 serum samples of pigs 27 were positive (5.87%). 3. Of 51 positive cattle samples, 29(4.92%) showed antibody to a serotype of L. icterohaemorrhagiae and 18(3.0%) to L. canicola, and 4 (0.68%) to L. pomona. Eight of L. icterohaemorrhagiae positive samples showed a cross reaction to L. canicola. 4. Of 27 positive pig samples, 14(3.04%) showed antibody to L. ictereohaemorrhagiae and 7(1.52%) to L. grippotyphosa. 4(0.87%) to L. canicola, 2(0.43%) to L. pomona. Two of L. canicola positive samples showed a cross reaction to L. grippotyphosa. 5. Serum samples of seven pigs, showing antibody positive to L. grippotyphosa were first observed in Korea. 6. Infection rate of bovine and porcine leptospirosis, in Korea, appeared to be lower than that of Japan, Taiwan, Thailand and the Philippines.

Optimal Design of L1B4 Linear Ultrasonic Motor using Evolutionary Strategy Algorithm (진화 전략 알고리즘을 이용한 L1B4 선형 초음파 모터의 형상 최적 설계)

  • 본 논문에서는 진화 전략 알고리즘(Evolution Strategy Algorithm)를 이용한 L1B4 선형 초음파 모터(L1B4-USM)의 최적 설계 기법을 제시하고자 한다. 유한요소법(Finite Element Method)을 정식화 하였고, 2차원 유한요소법을 L1B4-USM의 임피던스와 모드의 해석을 통해 검증 하였다. 검증된 2차원 유한 요소 해석을 통한 선형 초음파 모터의 임피던스 해석, mode 해석 및 최적 모드의 탐색 프로그램, 자동 요소분할 프로그램 그리고 진화 전략 알고리즘을 수행하였다. 이를 통해 선형 초음파 모터의 L1모드, B4 모드 각각이 발생하는 공진주파수를 일치시키며, 최대 속도를 얻기 위한 최적 설계기법을 완성 하였고, 최적화된 형상의 L1B4-USM를 설계하였다.
