A Study of the Antioxidant Activities and Whitening Activities of Areca semen Extracts as Cosmetic Ingredient (화장품 소재로서 빈랑자의 항산화 및 미백활성에 관한 효과)

  • Herbal plant extracts are good resources to find functional compounds for cosmetic ingredient. In this study, the extract of Areca semen (A. semen) was studied for melanogenesis inhibition and antioxidant activity. The results showed that ethyl acetate fraction of A. semen contained phenolic contents, $301.35{\pm}0.88{\mu}g/mg$, and exhibited potent antioxidant activity with $IC_{50}$ value of $1.02{\pm}0.07{\mu}g/mg$. Further, FRAP value exhibited potent antioxidant activity with $9.07{\pm}0.36mM$. Disk diffusion assay was performed for antibacterial activity. Ethyl acetate fraction of A. semen showed antibacterial activity against Staphylococcus aureus (S. aureus) and Staphylococcus epidermidis (S. epidermidis) at $80{\mu}g/mL$, whereas it showed no significant antibacterial activity against Escherichia coli (E. coli). The results of cell viability indicated that ethyl acetate fraction did not show cytotoxicity to B16/F1 cells at $80{\mu}g/mL$ and showed significant cytotoxicity at $100{\mu}g/mL$ of concentration and showed inhibition of melanin synthesis inhibitory, $29.78{\pm}0.31%$ at $80{\mu}g/mL$. Furthermore, mRNA expressions of tyrosinase and MITF were decreased after treatment with ethyl acetate fraction in a dose-dependent manner. As a result, the ethyl acetate fraction of A. semen could be considered as potential as whitening agents.

Antioxidant and Anticancer Properties of Methanolic Extracts from Different Parts of White, Yellow, and Red Onion

  • Antioxidant and anticancer effects of methanolic extracts from the flesh (WFME) and peel (WPME) of white onion, the flesh (YFME) and peel (YPME) of yellow onion, the flesh (RFME) and peel (RPME) of red onion were studied. The content of total phenolics in WFME, WPME, YPME, YFME, RPME, and RFME were $0.260{\pm}0.01$, $4.480{\pm}0.23$, $0.319{\pm}0.02$, $719.12{\pm}37.36$, $0.248{\pm}0.01$, and $806.21{\pm}26.38\;mg/g$, respectively. The quercetin content of WFME, WPME, YFME, YPME, RFME, and RPME were $12.56{\pm}0.19$, $3.57{\pm}0.14$, $15.24{\pm}0.65$, $755.29{\pm}22.24$, $5.70{\pm}0.23$, and $774.03{\pm}29.48\;mg$/100 g, respectively. Like total phenolics, the highest 2,2'-diphenyl-1-picrylhydrazyl (DPPH) scavenging activities were found in RPME. However, inhibitory effects on lipid oxidation of RPME were similar to those of WPME and YPME. In addition, inhibitory effect of WPME, YPME, and RPME for human breast cancer cell (MCF-7) growth were 78.43, 81.90, and 96.52% while those on human prostate cancer cell (LNcap) were 71.58, 77.93, and 98.47% at $100{\mu}g/mL$, respectively. Total phenolics, quercetin content, antioxidant, and anticancer activities exhibited significant variation among the 3 onion varieties in this experiment. Therefore, it is assumed that antioxidant and anticancer activities were affected by the total phenolics and quercetin level of onion.

Validating a Xylose Regulator to Increase Polyhydroxybutyrate Production for Utilizing Mixed Sugars from Lignocellulosic Biomass Using Escherichia coli

  • Polyhydroxybutyrate (PHB) production from lignocellulosic biomass is economically beneficial. Because lignocellulosic biomass is a mixture rich in glucose and xylose, Escherichia coli, which prefers glucose, needs to overcome glucose repression for efficient biosugar use. To avoid glucose repression, here, we overexpressed a xylose regulator (xylR) in an E. coli strain expressing bktB, phaB, and phaC from Cupriavidus necator and evaluated the effect of xylR on PHB production. XylR overexpression increased xylose consumption from 0% to 46.53% and produced 4.45-fold more PHB than the control strain without xylR in a 1% sugar mixture of glucose and xylose (1:1). When the xylR-overexpressed strain was applied to sugars from lignocellulosic biomass, cell growth and PHB production of the strain showed a 4.7-fold increase from the control strain, yielding 2.58 ± 0.02 g/l PHB and 4.43 ± 0.28 g/l dry cell weight in a 1% hydrolysate mixture. XylR overexpression increased the expression of xylose operon genes by up to 1.7-fold. Moreover, the effect of xylR was substantially different in various E. coli strains. Overall, the results showed the effect of xylR overexpression on PHB production in a non-native PHB producer and the possible application of xylR for xylose utilization in E. coli.

Screening of Effective Medium Composition for the Cultivation of Lactobacillus plantarum and Lactobacillus reuteri Using Statistical Methods (통계적 방법을 이용한 Lactobacillus plantarum과 Lactobacillus reuteri 의 유효 배지 성분의 탐색)

  • This study was conducted to develop an economical optimum medium composition for the mass production of $Lactobacillus$ $plantarum$ and $Lactobacillus$ $reuteri$, livestock probiotics. Medium ingredient factors were selected on the basis of MRS broth composition, and the 15 ingredient variables were as follows: sucrose, glucose, molasses, yeast extract, corn steep liquor, soy peptone, dipotassium phosphate, manganese chloride, magnesium chloride, tween 80, sodium chloride, sodium acetate, ammonium citrate, sodium sulphate, and ferrous sulphate. The Plackett Burman design, consisting of 20 runs, was employed for the analysis of ingredient effects on cell growth of $L.$ $plantarum$ and $L.$ $reuteri$. As a result, sucrose, glucose, molasses, yeast extract, corn steep liquor, soy peptone, sodium acetate, and ammonium citrate positively influenced the growth of $L.$ $plantarum$. Additionally, yeast extract, soy peptone, $K_2PHO_4$, and tween 80 positively influenced the growth of $L.$ $reuteri$. Positive effects were found from sucrose, yeast extract, and soy peptone in the integrated analysis of the effects of both $L.$ $plantarum$ and $L.$ $reuteri$. Finally, effective medium components for both strains were found as follows: sucrose (20.0 g/l), glucose (5.0 g/l), soy peptone (11.0 g/l), yeast extract (5.0 g/l), $K_2PHO_4$ (0.2 g/l), $CH_3COONa$ (2 g/l), and $MgCl_2$ (0.02 g/l).

Functionalities of Squid Liver Hydrolysates (오징어 간 가수분해물의 기능성)

  • The autolysate and hydrolysate of a common squid liver, Todarodes pacificus, were prepared. Autolysis (liver ratio, pH, temperature) and Protamex-treated hydrolysis (pH, temperature, ratio of protease to liver) conditions were optimized by response surface methodology using central composite design for under 1 hr of hydrolysis time. The desirability profile indicated that maximum DH could be achieved at a squid liver of 93.5%, pH 6.4, and $47^{\circ}C$ in autolysis, while that of Protamex-treated hydrolysis did at a Protamex-to-squid liver level of 0.33%, pH 6.0, and $55^{\circ}C$. Three amino acids, proline, cysteine, and methionine, were not detected in the total amino acid composition of the Protamex-treated hydrolysate, while they were detected in the free amino acid composition. Cadmium was $8.32{\pm}0.03$ mg/100 g-powder for raw, $3.56{\pm}0.02$ mg/100 g-powder for the autolysate, and $13.26{\pm}0.04$ mg/100 g powder for the Protamex-treated hydrolysate. The major molecular weight ranged from 1.0 to 1.5 kDa for the autolysate and from 210 to 470 Da for the Protamex-treated hydrolysate. Food functionalities of the autolysate, such as surface hydrolphobicity, emulsion activity index, emulsion stability, water, and fat adsorption, were similar to the Protamex-treated hydrolysate. Both the autolysate and Protamex-hydrolysate showed high inhibitory activities on the angiotensin-I converting enzyme. Cell toxicity against the HepG2 cell line was not detected in the autolysate or the Protamex-treated hydrolysate by 200 ${\mu}g/mL$.

Reductive acetogens isolated from ruminants and their effect on in vitro methane mitigation and milk performance in Holstein cows

  • This study was designed to evaluate the in vitro and in vivo effects of reductive acetogens isolated from ruminants on methane mitigation, and milk performance, respectively. Four acetogens, Proteiniphilum acetatigenes DA02, P. acetatigenes GA01, Alkaliphilus crotonatoxidans GA02, and P. acetatigenes GA03 strains were isolated from ruminants and used in in vitro experiment. A control (without acetogen) and a positive group (with Eubacterium limosum ATCC 8486) were also included in in vitro experiment. Based on higher acetate as well as lower methane producing ability in in vitro trial, P. acetatigenes GA03 was used as inoculum for in vivo experiment. Holstein dairy cows (n = 14) were divided into two groups viz. control (without) and GA03 group (diet supplied with P. acetatigenes GA03 at a feed rate of 1% supplementation). Milk performance and blood parameters were checked for both groups. In in vitro, the total volatile fatty acids and acetate production were higher (p < 0.05) in all 4 isolated acetogens than the control and positive treatment. Also, all acetogens significantly lowered (p < 0.05) methane production in comparison to positive and control groups however, GA03 had the lowest (p < 0.05) methane production among 4 isolates. In in vivo, the rate of milk yield reduction was higher (p < 0.05) in the control than GA03 treated group (5.07 vs 2.4 kg). Similarly, the decrease in milk fat was also higher in control (0.14% vs 0.09%) than treatment. The somatic cell counts (SCC; ×103/mL) was decreased from 128.43 to 107.00 in acetogen treated group however, increased in control from 138.14 to 395.71. In addition, GA03 increased blood glucose and decreased non-esterified fatty acids. Our results suggest that the isolated acetogens have the potential for in vitro methane reduction and P. acetatigenes GA03 strain could be a candidate probiotic strain for improving milk yield and milk fat in lactating cows with lowering SCCs.

Effect of Light Wavelengths on the Mycelial Browning of Lentinula edodes Strain Sanjo 701ho (광 파장이 표고 품종 산조 701호 균사의 갈변에 미치는 영향)

  • Mycelial browning, which protects the organism from contamination and moisture loss, is essential for sawdust cultivation of Lentinula edodes. The effects of light and light wavelengths on the mycelial browning of the L. edodes Sanjo 701ho strain, and the characteristics of its brown hyphae, were investigated. After the mycelia were cultured on potato dextrose agar medium under fluorescent lamps covered with colored cellophane filters (red, green, and blue) or under light emitted diodes (LED), with wavelengths ranging from 400 to 700 nm (far-red, red, green, and blue), for 14 h per day for 40 days, the mycelial browning rate was measured. The wavelength of fluorescent lamps, which range from 300 to 1,100 nm, was reduced to 360 to 1,022 nm with the use of three colored cellophane filters and the photosynthetic photon flux density was reduced by 42 to 71 % depending on the light wavelength. The browning rate by colony area of mycelia exposed to light was at an average of 64 %, whereas, that of unexposed mycelia was only 5 %. The browning rate was 0.02 % in far-red, 1.5 % in red, 53.8 % in green, 57.3 % in blue, and 64.0 % in fluorescent light. The white mycelia were resilient with actively growing hyphae, filled with cytoplasm, and thin cell walls less than $1{\mu}m$ thick. Conversely, the brown mycelia possessed dead, hard hyphal structures without cytoplasm, but with approximately $2-4{\mu}m-thick$-thick cell walls. In conclusion, lights of varying wavelengths, especially short-wavelength LEDs, are effective for forming dead, brown mycelia of L. edodes, thus, forming a protective functional layer for its living white mycelia.

Determination of selenium in grains by hydride generation-ICP/MS (수소화물발생-유도결합플라즈마 질량분석법(HG-ICP/MS)을 이용한 곡류 중 셀레늄의 정량분석)

  • The purpose of this study is to determine the quantity of selenium in grains by hydride generation-inductively coupled plasma mass spectrometry. Two sample preparation methods, beaker and microwave digestions, are compared and the former shows better result. The optimum condition for hydride generation is 4.0 M for HCl, 3% for $NaBH_4$ with the sample flow of 0.6 mL/min. The detection limit is 0.02 ${\mu}g/kg$($3{\sigma}$) and improved by 10 times. Isobaric interferences on Se is removed by Octopole Reactoin Cell and $H_2$ (3.8 mL/min) shows better performance over He. However, in case Br exists in a matrix, $H_2$ could induce interferences on m/z 80 and 82 ($^{80}[BrH]^+$ and $^{82}[BrH]^+$). The accuracy of this experiment is examined successfully by analyzing several reference materials. The results for several domestic grain analysis show that the concentrations are between 12.7 ${\mu}g/kg$ and 29.6 ${\mu}g/kg$.

Antioxidant and Inflammatory Mediators Regulation Effects of the Roots of Opuntia humifusa (천년초 뿌리의 항산화 및 염증 매게 물질 조절 효과)

  • The roots of Opuntia humifusa (OHR) were extracted with 80% aqueous MeOH and the concentrated extract was partitioned with EtOAc, n-butanol and $H_2O$, successively. The fractions were tested using DPPH and ABST radical scavenging method. The all fractions showed potent scavenging effects. The scavenging effect of the EtOAc fraction was higher than the other fractions, with $IC_{50}$ values as DPPH; $77.0{\pm}1.38{\mu}g/mL$, ABTS: $26.3{\pm}2.02{\mu}g/mL$. And, we investigated anti-inflammatory activities by examining the effects of the OHR fractions on pro-inflammatory cytokine release in the human mast cells (HMC-1). Treatment with OHR fractions clearly reduced the release of the proinflammatory cytokines tumor necrosis factor-alpha (TNF-${\alpha}$), interleukin (IL)-$1{\beta}$, interleukin (IL)-6 and interleukin (IL)-8) in PMACI-stimulated HMC-1 cells. The results showed the potential of OHR as an excellent antioxidant substance and inhibiting inflammatory mediators. Therefore, OHR may be used as a therapeutic approach to various inflammatory diseases.

Are the Clinical outcomes of Neonates and Infants Under 2 Months Old with Urinary Tract Infections Similar to those in Infants 2 to 12 Months Old?

  • Purpose: Although the American Academy of Pediatrics provides clinical guidelines for urinary tract infection (UTI) infants, guidelines are not appropriate for neonates and infants less than 2 months of age due to insufficient data. The aim of this study was to evaluate the characteristics of neonates and young infants less than 2 months old (group 1) with UTI compared to older infants from 2 to 12 months old (group 2). Methods: We reviewed UTI patients aged 0 to 12 months admitted to the pediatric department in the last 5 years. Clinical characteristics such as age, sex, fever duration, recurrence, progression to acute pyelonephritis (APN), malformations like hydronephrosis and vesicoureteral reflux (VUR), and laboratory results were compared between group 1 and group 2. Results: 615 patients were included in this study. Group 1 had 94 cases and group 2 had 521 cases. Escherichia coli was the most commonly isolated pathogen in urine cultures. Fever duration was shorter in group 1 (vs.) 2 ($1.91{\pm}1.43$ days vs. $3.42{\pm}2.40$ days, P<0.05). As compared to group 2, group 1 had a higher proportion of patients with antenatal hydronephrosis and hydronephrosis found after admission (10.6% vs. 3.6% and 75.5% vs. 55.9%, P<0.05). There were differences between two groups in white blood cell (WBC) count (Group 1: $13,694{\pm}5,315/{\mu}L$, Group 2: $15,271{\pm}6,130/{\mu}L$, P<0.05) and C-reactive protein (Group 1: $32.02{\pm}35.17mg/L$, Group 2: $46.51{\pm}46.63mg/L$, P<0.05). Conclusion: Compared to older infants, UTI in neonates and young infants shows milder clinical manifestations except higher rates of hydronephrosis but outcome is alike.
