Effects of Seed Pre-treatment and Germination Environments on Germination Characteristics of Ligularia fischeri Seeds (종자 전처리 및 발아환경에 따른 곰취 종자의 발아특성)

  • This study was carried out in order to examine the germination characteristics of Ligularia fischeri seeds, and it was to develop the more efficient pre-treatment and production system of the seeds. It was performed by two ways - temperature control (10, 15, 20 and $25^{\circ}C$) and shading treatment (Full sunlight, 35%, 50%, 75% and 95% of full sunlight). Seed pre-treatment before the each experiment was carried out by temperature (with low temperature and wetting treatment (LTW) for 0, 15, 30, 45 and 60days) and shading treatment (with drying at room temperature (DRT), drying at low temperature (DLT) and water soaking (WS) for 2 days). Seeds of L. fischeri were, regardless of seed pre-treatment, germinated well at $10^{\circ}C$ and the more temperature went up, the more germination rate went down. As a result of surveying shading treatment, 75% shading with DRT was the highest germination rate (68.1%) and 95% shading with WS was the lowest germination rate (48.6%). It was showed over-growth under 95% shading treatment experiment and withered in the full sunlight. As a result of surveying the whole experiment, L. fischeri seeds pre-treated with LTW for 15 days germinated well at $10^{\circ}C$ and under 50~75% shading treatment.

Toxic Interactions of Perfluorinated Compounds (PFCs) with Heavy Metals Using Vibrio fischeri (발광박테리아 Vibrio fischeri를 이용한 과불화합물과 중금속의 복합독성평가)

  • The object of this study was to evaluate the combined toxic interactions of the perfluorooctanoic acid (PFOA) or perfluorooctane sulfonate (PFOS) with six heavy metals (Cu, Zn, Cr, Cd, Hg, and Pb). The individual and combined toxic effects were assessed using the Vibrio fischeri assay. In case of the individual toxicity, PFOA was higher toxic than PFOS and toxicity of PFOA and PFOS were lower than heavy metal. In the toxicity of heavy metals, the $Hg^{2+}$ was found to be most toxic followed by $Pb^{2+}$, $Cr^{6+}$, $Cu^{2+}$, $Zn^{2+}$, and $Cd^{2+}$. The combined toxicity of PFOA or PFOS with $Cr^{6+}$ were synergistic effect because the $EC_{50}$ mix values were less than 1 TU. PFOA + $Zn^{2+}$, PFOS + $Zn^{2+}$, PFOA + $Cd^{2+}$ and PFOS + $Cd^{2+}$ produced addictive effect. Except in these case, all of binary mixtures show antagonistic effect. This study proved potential risk of coexistent with perfluorinated compounds and heavy metals in water environment.

A Study on the Degradation and the Reduction of Acute Toxicity of Simazine Using Photolysis and Photocatalysis (광반응 및 광촉매 반응을 이용한 simazine의 분해 및 독서저감에 관한 연구)

  • The photocatalysis degradation of simazine, s-triazine type herbicide was carried out using circulating photo reactor systems. In order to search for the effective method to mineralize this compound into environmentally compatible products, this study compared the removal efficiencies of simazine by changing various parameters. First, under the photocatalytic condition, simazine was more effectively degraded than by photolysis and $TiO_2$ only condition. With photocatalysis, 5 mg/l simazine was degraded to approximately 90% within 30 min, and completely degraded after 150 min. Ionic byproducts such as ${NO_2}^-$, ${NO_3}^-$, and $Cl^-$ were detected from the photocatalysis of simazine, however, the recoveries were poor, indicating the presence of organic intermediates rather than the mineralization of simazine during photocatalysis. Two bioassays using V. fischeri and D. magna were employed to measure the toxicity reduction in the reaction solutions treated by both photocatalysis and photolysis. Simazine and its photocatalysis treated water did not exert any significant toxicity to V. fischeri, marine bacterium. However, the acute toxicity test using D. magna indicates that initial acute toxicity ($EC_{50}$ = 57.30%) was completely reduced ($EC_{50}$ = 100%) after 150 min under both photocatalysis and photoysis of simazine. This results indicates that photocatalysis and photolysis of simazine reduced the acute toxicity through mineralization.

Method for Validation of Caffeoylquinic Acid Derivatives in Ligularia fischeri Leaf Extract as Functional Ingredients (건강기능식품 기능성 원료로서 곰취잎 추출물의 Caffeoylquinic Acid계 성분 분석법 검증)

  • An HPLC analysis method was developed for standard determinations of chlorogenic acid, 3,4-di-O-caffeoylquinic acid, 3,5-di-O-caffeoylquinic acid, and 4,5-di-O-caffeoylquinic acid as functional health materials in Ligularia fischeri extract. HPLC was performed on a $C_{18}$ Kromasil column ($4.6{\times}250mm$, $5{\mu}m$ column) with a gradient elution of 0.1% (v/v) trifluoroacetic acid and acetonitrile at a flow rate of 1.0 mL/min at $30^{\circ}C$. The analytes were detected at 330 nm. The HPLC method was validated in accordance with the International Conference on Harmonization guideline of analytical procedures with respect to specificity, precision, accuracy, and linearity. The limits of detection and quantitation for the four compounds were 3.0~14.6 and $9.2{\sim}44.4{\mu}g/mL$, respectively. Calibration curves showed good linearity ($r^2$ > 0.999), and the precision of analysis was satisfied (less than 0.9%). Recoveries of quantified compounds ranged from 98.96 to 101.81%. This result indicates that the established HPLC method is very useful for the determination of marker compounds in Ligularia fischeri leaf extracts.

Effects of Ligularia fischeri Extracts on Oxidation of Low Density Lipoprotein (저밀도 지방단백질의 산화에 대한 곰취 추출물의 항산화 효과)

  • The study was carried out to investigate the antioxidant effects of ethanol, methanol, and water extracts or fractions of ethanol extract of Ligularia fischeri on low density lipoprotein(LDL) and ethanol extract was further fractionated. The methanol extract and ethyl acetate fraction showed strong antioxidant effect with 71.7% (13.36 nmol/mg) and 95% (1.35 nmol/mg) inhibition in the presence of $15\;{\mu}g/mL$, respectively. In the value of malondialdehyde(MDA) with oxidation time, ethanol, methanol and water extract in the presence of $25\;{\mu}g/mL$ inhibited the oxidation up to 4h incubation and ethyl acetate fraction showed strong antioxidant effect up to 8h incubation. Also, the ethanol, methanol and water extract showed antioxidant effects in the agarose gel electrophoresis test. The conjugated diene formation by lipid oxidation with addition of $Cu^{2+}$ in the Ligularia fischeri extracts and their fractions was decreased approximately 1.1 to 2.8 times and 2.2 to 3.2 times, respectively compared to control. In the degradation of apo B-100 by oxidation using SDS-PAGE, ethanol, methanol and water extract showed similar degradation band pattern compared to that of native LDL band. Apo B-100 contents using densitometer were 77.8, 92.5% and 82.3% in the ethanol, methanol and water extract, respectively, compared to 100% of native LDL. In the meanwhile, apo B-100 contents in hexane, ethyl acetate and water fraction were 38.8, 94.5 and 65.5% respectively. This results indicated that ethyl acetate fraction showed the strongest antioxidant effect on MDA value or apo B-100 contents of LDL.

Desmutagenic Activity of Heated Mountain Herb Juices (산채류(山菜類) 가열즙(加熱汁)의 돌연변이 억제 작용에 관(關)한 연구(硏究))

  • Potential mutagenicity of ten heated edible mountain herbs were examined with spore recassay, Ames test and DNA breaking test. Samples of edible mountain herbs were prepared with water extraction at $100^{\circ}C$ for 20 minutes. With the rec-assay, no significant mutagengic activity could be obtained from all of the samples, but among the eight of metal ions added to sample solution, $Pb^{2+}$ to R. crispus heated juice, $Zn^{2+}$ to L. fischeri and S. bracycarpa heated juice increased mutagenic activity of the samples. With the Ames test and DNA breaking test, all of the samples did not show mutagenicity. However, breaking action was activated on heated L. fischeri, P. japonicus. A. triphylla and A. tataricus juices in the presence of 25mM $Cu^{2+}$. But heated A. elata, H. aurantiaca, A. triphylla, S. bracycarpa and A. scaber juices were inactivated in the presence of 25mM $Fe^{2+}$. Desmutagenic activities against benzo$({\alpha})$pyrene significantly increased as increasing concentration of the heated edible mountain herb juices.

Antimicrobial Activity on Respiration Diseases Inducing Bacteria and Antioxidant Activity of Water Extracts from Wild Edible Vegetables (산채류 추출물의 항산화 활성 및 호흡기 질환을 유발하는 세균에 대한 항균활성)

  • To investigate the antimicrobial activity on bacteria causing a respiration disease and antioxidant effects of water extracts from 12 kinds of wild edible vegetables, we extracted the water extracts for 72 h in $7^{\circ}C$ using distilled water as solvent. The water extracts except Ixeris dentate and Allium monanthum had high concentrations of phenol compounds and flavonoids. Liguraia fischeri specially had the highest level on total phenol compounds and flavonoids with 205 ${\mu}g/mL$ and 98. 86 ${\mu}g/mL$, respectively. The each 0.05% extracts of Sedum sarmentosum and Liguraia fischeri had high effect on the DPPH radical scavenging activity among wild edible vegetables and the most extracts promoted antioxidant activity with increasing concentration of extract. The catalase activity of Erysimum aurantiacum and Aralia elata showed more than 150 units per g of fresh tissue. The effect of antimicrobial activity on water extracts showed characteristic activity. Only Staphylococcus aureus KCTC 1928 and Corynebacterium diptheriae KCTC 3075 were inhibited cell growth on the other hands, the remainder of bacteria was not inhibited cell growth. Nevertheless, the extracts of wild edible vegetables had specific concentration as MIC for antimicrobial activity respectively. In case of the extract of Aster scaber, Erysimum aurantiacum, and Allium monanthum had over 30% antimicrobial activity on the bacteria causing a respiration disease. In results, the wild edible vegetables include high concentrations of total phenol compounds and flavonoids that give a good antioxidant activity and antimicrobial activity. Therefore the wild edible vegetables are functional food for anti-aging and physiological activation.

Effects of Advanced Oxidation of Penicillin on Biotoxicity, Biodegradability and Subsequent Biological Treatment (고도산화공정 처리가 페니실린의 생독성, 생분해도 및 생물학적 분해에 미치는 영향)

  • Advanced oxidation processes (AOPs) composed of O3 and UV were applied to degrade penicillin (PEN). The degradation efficiency was evaluated in terms of changes in the absorbance (ABS) and total organic carbon (TOC). The combination of $O_3/H_2O_2/UV$ and $O_3/UV$ showed the best performance for the reduction of ABS (100% for 9 min) and TOC (70% for 60 min) values, although the mineralization was uncompleted under the experimental condition in this study. The change in biotoxicy was monitored with Escherichia coli susceptibility and Vibrio fischeri biofluorescence. The E. coli susceptibility was eliminated completely for 9 min by $O_3/UV$, and the toxicity to V. fischeri biofluorescence was 57% reduced by $O_3/H_2O_2/UV$. For the ultimate treatment of PEN, it is suggested that an AOP using $O_3/UV$ is followed by biological treatment, utilizing the enhanced biodegradability by the AOP. During 30 min of $O_3/UV$ treatment, the $BOD_5/COD$ ratio as an indication of biodegradability showed about 4-fold increment, compared to that of using a non-treated sample. TOC removal rate for AOP-pretreated PEN wastewater increased 55% compared to that of using the non-pretreated one through an aerobic biological treatment by Pseudomonas putida for artificial wastewater containing 20 mg/L of PEN. In conclusion, $O_3/UV$ process is recommended as a pretreatment step prior to an aerobic biological process to improve the ultimate degradation of penicillin.

  • This experiment was conducted to investigate the opimum media, media amount and planting density of nutri-culture for the high quality and yield of wild vegetable, Liguliaria fischeri Turez. The fried rice hull was recognized best media among perlite, sand and fried rice hull for nutri-culture of Ligularia fischeri Turcz. The yield on fried rice hull media at early growing stage was lower, but higher than in other media at late growing stage. Therefore, total yield in fried rice hull media was 15-29% higher than that of the existing perlite media. Growth and yield of Ligularia fischeri Turcz. were not significantly different between medium amounts, therefore considering economical efficiency and stability, the optimum medium amount was 105 $\iota/m^3$, Yield by planting density increased in the high density and 50 plants/$m^2$ was considered as proper planting density in nutri-culture of Liguliaria fishcheri Turcz.

The Quality Changes of Watery Kimchi made of Wild Vegetables by the Pre-Treatment Methods (전처리 방법에 따른 산채 물김치의 품질변화)

  • This study aimed to determine the physiochemical changes during maturation to examine the preparation of watery Kimchi with wild vegetables from Gangwon Province, and its use. The wild vegetables in this study included Ligularia fischeri, Aster scaber Thunb, and Cirsium setidens; a comparison was made between preservation in a saline solution and pre-treatment with blanching. As for treatment before preparation of watery Kimchi, the water content was the lowest at 81.45% for Aster scaber Thunb preserved in a saline solution and was found to be 87.39% for blanched Cirsium setidens. Blanching resulted in higher L, b, and a values for color value than treatment with a saline solution and Ligularia fischeri got the highest hardness, followed by Aster scaber Thunb and Cirsium setidens, from both types of treatment. As for the physiochemical properties during maturation, the total soluble solid content and salinity were kept higher by treatment with a saline solution than by blanching, whereas pH tended to get lower in both types of treatment. Turbidity became higher one week after treatment with a saline solution, but varied insignificantly afterward by any type of treatment. As for free sugar, both types of treatment increased the content of glucose and fructose and kept that of sucrose and maltose constant during maturation. As for overall acceptability, blanched watery Kimchi made of Cirsium setidens got the highest preference.
