Development of Phage-resistant Mutants from Lactobacillus casei (Lactobacillus casei의 Bacteriophage내성돌연맥리균분리)

  • A lactic starter organism, Lactobaciilus casei YIT 9018 was treated with N-methyl-N'-nitro-N-nitrosoguanidine (NTG) to obtain phage-resistant mutants. Freshly grown cells suspended in citrate buffer were exposed to NTG of 50 g/$m\ell$ for 40 min. Among 88 colonies isolated eight colonies showed distinct resistance to phages isolated previously from milk plants. The eight new colonies showed character similar to the original L. casei except that they responded differently to phage of different sources and thus were designated as eight different mutants of L casei. From the phage resisting toaether with the fermentative ability equivalent to the mother organism the mutants may be considered to be used as starter cultures for fermented milk.

  • The broad-host plasmid PAM $\beta_1$ of Streptococcus faecalis DS 5 which codes for erythromycin resistance and lactose utilization was transferred into L. casei M-3 (lac-mutant) by conjugation, but was not transferred by protoplast fusion and protoplast transformation. For conjugal transfer of plasmid PAM $\beta_1$ the method of membrane filter mating was more efficient than that of agar surface mating. The rate of acid production of transconjugant C-1, C-3 was similar to L. casei YIT 9018. The proteolytic activity of transconjugant C-3 was increased 20% higher than that of wild type. Plasmid PAM $\beta_1$ was detected by a11 of the transconjugants. The transconjugants expressed lactose ulitization and erythromycin resistance.

Selection and Physico-Chemical Characteristics of Lactic Acid Bacteria which had Cholesterol Lowering Activities (콜레스테롤 저하 유산균의 분리 및 이들 균주의 이화학적 특성)

  • Four strains of lactic acid bacteria which had cholesterol lowering activities were selected from foreign fermented milk. The strains were identified as Lactobacillus(L.) rhamnosus 2084, L. casei 0781, Lactococcus (Lacto.) lactis spp. 204, and Enterococcus(E.) faecium 402. We observed that the L. rhamnosus 2084 was the most tolerant against pH 1.5, L. casei 0781 against pH 2.0, but not significantly different in the tolerance against pH 3.0. The L. rhamnosus 2084 was the most tolerant against bile acid and prominent in the degree of lowering cholesterol level. All four strains were used as starters in producing yogurt, and then investigated physico-chemical characteristics, such as pH, titratable acidity, and viable cell counts of yogurt base. L. casei 0781, L. rhamnosus 2084, Lacto. lactis 204, and E. faecium 402 were incubated for 6 hours at $40^{\circ}C$, 4 hours at $40^{\circ}C$, 6 hours at $37^{\circ}C$, and 12 hours at $37^{\circ}C$ and $40^{\circ}C$, respectively, for the optimum conditions of fermented milk.

Characterization of Isolated Lactobacillus spp. And classification by RAPD-PCR Analysis

  • The genetic relationship of six Lactobacillus strains and five laboratory isolated form fermented milk were determined by a random amplified polymorphic DNA(RAPD)-Polymease chan reaction (PCR) method. With 42 random primers. the result were analyzed by using the NTSYS-PC software for phenetic analysis. it revealed that all tested bacteria were divided into three distinct clusters. The clusters implied three subgenuses existed for the genus Lactobacillus, which were previously proposed by Rogosa and Sharpe. From the results, it was also possible to determine that the isolated Lactobacillus strains from fermented milk were grouped into L. acidophilus or L. bulgaricus. Interestingly. the three tested L. casei strains were divided into different clusters implying different subgenuses, i.e., Thermobacterium (L. casei YIT 9018) and Streptobacterium(L. casei CHR. Hansen and L.casei ATCC 4646). According to the distance matrix generated by an UPGMA program, the isolated bacteria LT01 and LT02 were determined as a subspecies of L. bulgaricus. The HK01, HK02 and HK03 were very closely related to either L. acidophilus or L. case YIT 9018. Hence, RAPD-PCR appears to be a very practical method to determine the genetic relationships of the Lactobacillus species and to characterize the unknown Lactobacillus strains at the subspecies level.

  • The best conditions for the protoplast fusion of Lactobacillus casei have been searched for in this study. Antibiotic resistance was used as the selective marker for enumerating and selecting the recombinants. Antibiotic resistant mutants were isolated after treating cells with N-methyl-N'-nitro-N'-nitrosoguanidine. High frequency fusion of protoplasts of L. casei strains were obtained in the presence of 40% (wt/vol) polyethylene glycol 4,000 after 1 min at 3$0^{\circ}C$ at around neutral pH. Spontaneous mutations of drug-resistance of L. casei were two or three orders lower than the recombination frequency. Recombination frequencies were about 10$^{-4}$ per parent cells employed.

  • The anti-inflammatory effect of ethanol extracts from Hizikia fusiformis fermented with and without lactic acid bacteria was compared in lipopolysaccharide (LPS)-stimulated RAW 264.7 mouse macrophages. The fermentation was done using Weissella sp. SH-1 and Lactobacillus casei in a mixture of glucose and lactate source at $30^{\circ}C$ for 30 days. As a result, we confirmed that the fermentation of H. fusiformis with lactic acid bacteria inhibited LPS-stimulated nitric oxide (NO) production and the expression of inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2, interleukin (IL)-6, tumor necrosis factor ${\alpha}$, and IL-$1{\beta}$ as important inflammatory factors. During a comparison analysis, we found that L. casei fermented groups significantly suppressed NO production by regulating iNOS and COX-2 expression. Also, the effective suppression of pro-inflammatory cytokine and LPS-induced activation of mitogen- activated protein kinase indicated that the fermentation using Weissella sp. SH-1 and L. casei may provide an increment towards the extraction of active components, which are effective anti-inflammatory agents.

Influence of Culture Comditions on Acid Tolerance of Lactobacillus casei YIT 9018 (배양조건에 따른 Lactobacillus casei YIT 9018의 니산성 변화)

  • We studied the influence of culture conditions on the acid tolerance of Lactobacillus casei YIT 9018 in artificial gastric juice with respect to relative amount of membrane bound ATPase and their biochemical characteristics. With raising incubation temperature from 30.5$\circ$C to 40.5$\circ$C and lengthening incubation time from exponential phase to late stationary phase, the acid tolerance of L. casei YIT 9018 was increased. As acid tolerance enhanced, C$_{18:1}$ content of membrane fatty acid was reduced and C$_{19:0 cyclo}$ was enriched but the others were not changed greatly. At high ATPase activity, proton permeability was relatively low but this phenomenon did not correspond to acid tolerance. In conclusion, it was considered that changes of C$_{18:1}$ and C$_{19:0 cyclo}$ were closely related to the acid tolerance of L. casei YIT 9018.

A Novel Lactobacillus casei LP1 Producing 1,4-Dihydroxy-2-Naphthoic Acid, a Bifidogenic Growth Stimulator

  • 1,4-Dihydroxy-2-naphthoic acid (DHNA) is a bifidogenic growth stimulator (BGS) and could be a functional food ingredient since bifidobacteria are beneficial for human health. For that reason, lactic acid bacteria producing DHNA have been screened. A lactic acid bacterium LP1 strain isolated from a natural cheese was confirmed to produce DHNA, analyzed by a HPLC method. The strain was identified as Lactobacillus casei by 16S rRNA gene sequence analysis. The cell-free supernatant of fermented whey produced by L. casei LP1 presented the BGS activity for three bifidobacterial strains such as Bifidobacterium longum subsp. infantis KCTC 3127, Bifidobacterium bifidum KCTC 3202, and Bifidobacterium breve KCTC 3220 which were human-originated. To the best of our knowledge, a L. casei strain which can produce DHNA was firstly identified in this study.

Microbiological, Physicochemical, and Antioxidant Properties of Plain Yogurt and Soy Yogurt (플레인 요구르트와 소이 요구르트의 미생물학적, 물리화학적 및 항산화 활성)

  • This study evaluated the physicochemical and microbiological characteristics and antioxidant properties of yogurt samples fermented with lactic acid bacteria (LAB) obtained from pickled cabbage. API 50 CHL systems and 16S rRNA nucleotide sequence analyses revealed that the isolates were Lactobacillus casei PC05 and L. acidophilus PC16. Cell counts, titratable acidity, and viscosity of the yogurt samples fermented with L. acidophilus PC16 were significantly higher than those of the samples fermented with L. casei PC05 (P<0.05). The detected cell counts and physicochemical properties were significantly lower in plain yogurt than in soy yogurt (P<0.05). Yogurt samples fermented with L. acidophilus PC16 exhibited higher antioxidant activity, measured as ability to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and chelate ferrous ions, than those fermented with L. casei PC05. However, the ability to scavenge superoxide anions and superoxide dismutase (SOD) activity were significantly (P<0.05) higher in yogurt samples fermented with L. casei PC05 compared to those in samples fermented with L. acidophilus PC16. The antioxidant activity of soy yogurt was significantly (P<0.05) higher than that of plain yogurt. The antioxidant activity of the tested strains resulted in lipid peroxidation inhibition (in vitro), which may be related to the elimination of free radicals, chelating ability, and reducing power. There were no significant differences in the physicochemical properties and antioxidant activities of the yogurt samples during cold storage.

Comparative Tests on the Acid Tolerance of Some Lactic-Acid-Bacteria Species Isolated from Lactic Fermented Products (젖산 발효제품에서 분리한 유산균의 내산성 비교)

  • We isolated sixty lactic acid bacteria(LAB) from lactic fermented products. Among 60 isolates of LAB, 30 isolates were identified as Lactobacillus casei ssp.(5 strains), Lactobacillus acidophilus(2 strains), Lactobacillus delbrueckii ssp. bulgaricus(6 strains), Lactobacillus plantarum(4 strains), Streptococcus salivarius ssp. thermophilus(11 strains), and Streptococcus faecalis(2 strains). The acid tolerance and bile resistance of 30 LAB were determined. Because the acid tolerance was affected by the initial cell concentrations, the analysis of covariance could be used to remove the effect of initial cells on acid tolerance when testing for differences in acid tolerance among six species. Viability of LAB under acidic condition, pH 3 for 2 hours at $37^{\circ}C$, was significantly different among the species. L. casei and L. acidophilus strains showed great viability, but L. bulgaricus and S. thermophilus strains were very weak in acid tolerance.
