Phellinus baumii으로부터 세포외 다당체 생산의 최적화 (Optimal Conditions of Mycelial Growth and Exopolysaccharide Production in Submerged Culture of Phellinus baumii)

  • 장수상황버섯(Phellinus baumii)의 균사체 성장 및 세포외 다당체(exopolysaccharides) 생산을 위한 액체배양의 최적 배지 및 배양조건에 관한 실험을 수행한 결과, 최적배양 온도와 초기 pH는 각각 3$0^{\circ}C$와 5.0으로 결정되었다. 탄소원의 경우, cellobiose와 maltose가 균사체 성장에는 양호하였으나 저조한 세포외 다당체 생산을 보였으며, 반면에 fructose 와 mannitol 은 상대적으로 높은 수율의 세포외 다당체 생산과 균사체 성장 또한 양호하였다. 최적 질소원으로는 yeast extract가 균사체 성장뿐만 아니라 세포외 다당체 생산에 효과적인 것으로 결정되었다. 최적 배지의 조성은 fructose 20 g/L, yeast extract 20 g/L 및 $CaCl_2$ 0.55 g/L이었으며, 최적 배양조건 하에서 5-L 교반 발효조를 운전한 결과, 최대 균사체 성장(17.43 g/L)과 최대 세포외 다당체 생산(3.6 g/L)을 얻을 수 있었다. 회수된 세포외 다당체는 glycoprotein이었으며 그 조성은 amino acid분석 결과 주로 arginine (14.1%)과 glycin (12.0%)이었으며, cabohydrate의 경우는 mannose (48.7%)와 arabinose (38.4%)이 주성분이었다.

귤피로부터 분리한 마우스의 장관면역 활성 다당류의 검색 (Isolation of Polysaccharides Modulating Mouse’s Intestinal Immune System from Peels of Citrus unshiu)

  • 90여종의 전통차 및 죽류용 식물로부터 조제된 다양한 용매추출물 중에서, 귤피(peels of C. unshiu)의 냉수추출물 (CUI-0)이 Peyer’s patch를 매개로 한 가장 높은 장관면역 활성을 나타내었으며 다시마(L. japonica), 둥글레(P. japonicum), 탱자(P. trifoliata)의 냉수추출물과 구기자(L. chinense) 및 치자(G. jasminoides)의 열수추출물을 제외한 나머지 식물의 용매추출물은 거의 활성을 가지지 못하였다. CUI-0는 MeOH-가용성 획분(CUI-1), MeOH-불가용성이면서 EtOH-가용성 획분(CUI-2)과 조다당 획분(CUI-3)으로 분획되었다. 이러한 획분들 중 CUI-3은 Peyer’s patch 세포를 매개로 하는 골수세포 증식의 자극활성이 가장 높았으며 arabinose, galacturonic acid, galactose, glucose, glucuronic acid와 rhmanose(molar ratio; 1.00:0.53:0.45:0.28:0.28:0.19) 등을 주요 구성당으로 함유하고 있었으며 소량의 단백질(9.4%)도 구성물질로 포함된 활성 획분임이 밝혀졌다. CUI-3의 장관면역 활성은 pronase 및 periodate 처리에 의해 감소되었으며 특히 periodate 산화는 CUI-3의 활성에 심각한 영향을 끼치는 것을 알 수 있었다. 활성 획분으로서 당 함량이 높은 CUI-3IIb-3-2는 DEAE-Sepharose FF, Sepharose CL-6B 및 Sephacryl S-200에 의해 귤피 냉수추출물의 조다당 획분으로부터 정제되었으며 HPLC에 의해 분자량 약 18,000 Da의 단일 peak임을 확인하였다. CUI-3IIb-3-2는 주로 arabinose, galactose, rhamnose, galacturonic acid와 glucuronic acid(molar ratio; 1.00:0.54:0.28:1.45:0.63) 등의 구성당으로 구성되어져 있었으며 소량의 단백질(3.2%)이 함유되어진 물질로 구성되어 있었다. 한편 CUI- 3IIb-3-2는 Peyer’s patch를 경유하였을 때만 골수세포 증식활성을 나타내었으며 활성물질 자체가 직접 골수세포 증식활성에 관여하지는 않는것으로 확인되었다. 이러한 결과로부터 귤피의 장관면역 활성은 측쇄에 arabinose와 galactose 등의 중성당이 결합된 polygalacturonan 구조를 갖는 펙틴계통의 다당류에 기인하고 있음을 보여주었다.

Penicillium sp. KJ 81에 의한 Erythritol 생산 최적 배양 조건 (Optimization of Cell Culture Condition for Erythritol Production by Penicillium sp. KJ8l)

  • Erythritol은 저 칼로리 건강 감미 물질로서 우리는 이미 자연계로부터 폭넓은 탐색을 통하여 Penicillium sp. KJ8l을 분리, 동정하였다. 본 연구에서는 Penicillium sp. KJ8l의 erythritol 생산을 위한 최적 배양 조건을 검토하였다. 이 균주는 기질로서 glucose, sucrose, fructose, mannose, lactose, maltose 그리고 galactose를 이용하여 erythritol을 생산할수 있었으나 mannitol, arabinose, sorbitol 그리고 xylose를 이용하여서는 erythritol을 생산할 수 없었다. 또한 sucrose를 기질로 사용하였을 때 가장 높은 erythritol 생산성을 보였다. Penicillium sp. KJ8l은 배지의 sucrose 농도가 60%까지 erythritol을 생산할 수 있었으나, 30% sucrose농도에서 erythritol 생산성이 가장 좋았다. 또한 배지내 yeast extract의 농도가 0.5%일 때 erythritol 생산성 이 가장 좋았다. 본 균주는 30% sucrose와 0.5% yeast extract 배지에 0.5% ammonium sulfate를 첨가해 주었을 때 erythritol의 생산성이 향상되었다. Penicillium sp. KJ8l은 5ι jar. fermentor에서 30% sucrose, 0.5% yeast extract, 0.5% ($(NH_{4})_{2}SO_{4}$, 0.1% $KH_{2}PO_{4}$, 0.01% $MgCl_{2}$ 조성의 배지에서 1 vvm으로 산소를 공급해 주면서 200 rpm으로$37^{\circ}C$에서 11 일간 배양하였을 때 최고의 erythritol 생산을 보였다. 이 때 erythritol 생산 수율은 28.2g/l이었다.

  • Antitumor and antimicrobial activities of water soluble fractions of Duchesnea indica (Andr.) and Focke (Rosaceae), have been studied. The contents of total polysaccharide were 22% and its composition was identified by G.C.: L-arabinose(Rt: 7.71), D-xylose (Rt. 9.91), D-glucuronic acid (Rt. 12.77), D-fructose (Rt. 13.63), D-galactose (Rt. 14.68), D-glucose (Rt. 15.42). The antimicrobial activity of Duchesnea indica against four bacterial species, S. aureus, S. paratyphi, S. dysenteriae and Ps. aeruginosa was observed.

Microbiological Purification of L-Arabitol from Xylitol Mother Liquor

  • As a rare sugar alcohol, L-arabitol can be used in food and can prevent extra fat deposits in the intestinal tract. Commercially, L-arabitol is prepared from pure L-arabinose by hydrogenation, which needs a high temperature and high pressure, leading to a high production cost for Larabitol. Therefore, this study describes a novel L-arabitol production method based on biological purification from the xylitol mother liquor, a cheap and readily available raw material that contains a high concentration of Larabitol. First, a novel Bacillus megaterium strain was screened that can utilize xylitol, sorbitol, and mannitol, yet not L-arabitol. The isolated strain was inoculated into a medium containing the xylitol mother liquor under formulated culture conditions, where a high L-arabitol yield (95%) and high purity (80%) were obtained when the medium was supplemented with 50 g/l of xylitol mother liquor. Upon further purification of the fermentation broth by ion exchange and decolorization, L-arabitol was crystallized with a purity of 98.5%.

도축돈에서 분리된 Strptococcus suis에 대한 연구 (Studies on the Streptococcus suis isolated from slaughtered pigs)

  • Streptococcus suis has been identified as a major cause of contagious disease in pigs Ithas been isolated worldwide from pigs suffering from meningitis, bronchopneumonia, polyserositis, polyarthritis and septicemia. Two hundred and fifty-five lung samples of slaughter pigs with gross lung lesions were collected from Jan. to Dec. 1995 in southern Chonnam area. Isolation of S suis were tested by biochemical, serological methods and antimicrobial susceptibility. The results were summerrized as follows ; 1. S suis was Isolated from 30 of 254 pneumonic lungs, 14 Isolates were isolated only, 12 were isolated with p. multocida and 4 were isolated with unidentified Gram positive cocci. 2. In biochemical characteristics studies, all isolates were not grown in 6.5% NaCl medium, and most isolates utilized L-leucine-2-naphtylamide and trehalose, didn't utilize sorbitol, ribose and L-arabinose. 3. In slide agglutination test, 6(20%) Isolates were classified as serotype 2, 4(13.3%) isolates were as serotype l/2, 16, 2 and 2(6.6%) were as serotype 1, 4, 13. 4. S suis isolates showed marked susceptibility to amoxicillin, oxacillin, cephalothin and cepoferazone and high resistance to kanamycin, streptomycin, tetracycline and erythromycin.

Isolation and Characterization of Lactobacillus buchneri Strains with High ${\gamma}$-Aminobutyric Acid Producing Capacity from Naturally Aged Cheese

  • Two lactic acid bacteria (LAB) with high ${\gamma}$-aminobutyric acid (GABA)-producing capacity were isolated from naturally aged cheese. Examination of the biochemical features using an API kit indicated that the two strains belonged to Lactobacillus. They were gram positive, rod-type bacteria, and fermented arabinose, melezitose, melibiose and xylose, but did not utilize cellobiose or trehalose. 16S rDNA sequencing analysis confirmed that they were Lactobacillus buchneri and Lactobacillus sp. They were accordingly named as Lactobacillus buchneri OPM-1 and Lactobacillus sp. OPM-2, and could produce GABA from MRS broth supplemented with 10 g/L of monosodium glutamate (MSG) at a productivity of 91.7 and 116.7 mg/L/hr, respectively. Cell extracts of L. buchneri OPM-1 and Lactobacillus sp. OPM-2 showed glutamate decarboxylase (GAD) activity, for which the optimum pH and temperature were 5.5 and $30^{\circ}C$, respectively.

Production of Soluble Human Granulocyte Colony Stimulating Factor in E. coli by Molecular Chaperones

  • The effects of coexpression of GroEL/ES and DnaK/DnaJ/GrpE chaperones on the productivity of the soluble form of human granulocyte colony stimulating factor (hG-CSF) in E. coli were examined. Recombinant hG-CSF protein was coexpressed with DnaK/DnaJ/GrpE or GroEL/ES chaperones under the control of the araB or Pzt-1 promoter, respectively. The optimal concentration of L-arabinose for the expression of DnaK/DnaJ/GrpE was found to be 1 mg/ml. When L-arabinose was added at $OD_{600}$=0.2 (early-exponential phase), soluble hG-CSF production was greatly increased. In addition, it was observed that the DnaK/DnaJ/GrpE and GroEL/ES chaperones had no synergistic effects on preventing aggregation of hG-CSF protein. Consequently, by coexpression of the DnaK/DnaJ/GrpE chaperone, the signal intensity of the hG-CSF protein band in the soluble fraction of cell lysate was increased from $3.5\%\;to\;13.9\%$, and Western blot analysis also revealed about a 4-5-fold increase of production of soluble hG-CSF over the non-induction case of DnaK/DnaJ/GrpE.

Xylanase를 생산하는 Streptomyces sp. YB914의 특성과 효소 생산성 (Characterization and Xylanase Productivity of Streptomyces sp. YB914)

  • 토양으로부터 세포외로 xylanase를 분비 생산하는 방선균 YB-914가 분리되었으며, 형태, 배양, 생화학적 특성을 조사한 결과 Streptomyces 속 균주로 확인되었다. 분리균의 배양상등액에 존재하는 xylanase는 pH 5.5과 $55^{\circ}C$의 반응조건에서 반응성이 가장 높았으며, pH 4.5~7.0 범위에서 최대활성의 80% 이상을 나타냈다. Xylanase의 생산을 위한 배지를 최적화하기 위해서 G.S.S 배지성분을 여러 종류의 탄수화물로 대체하였다. Oat spelt xylan, corn cob xylan, 밀기울 및 유당과 같은 탄수화물은 Streptomyces sp. YB914의 xylanase 생산성을 증가시키는 것으로 확인되었으며, galactose와 arabinose는 효소 생산을 크게 억제하였다. Oat spelt xylan(1%)와 유당(1.5%)을 함유한 변형배지에서 xylanase의 최대생산성이 48 U/mL로 확인되었다.

  • This experiment was carried out to elucidate the sugar composition of polysaccharides and the structural features of water-soluble polysaccharides(WSP) isolated from Korean oak mistletoe, Loranthus yadoriki Sieb. The 48-hours ball-milled meals of extractive-free dried mistletoe sawdusts were extracted with distilled water for $24hrs{\times}2$ at room temperature. The extracts poured into 95% ethyl alcohol to precipitate. The separated precipitate of WSP, in form of yellowish white powder by lyophilization, was fractionated into four subfractions of WSP-1, WSP-2, WSP-3 and WSP-4 by anion exchange chromatography on DEAE-cellulose column. The sugar composition of WSPs was analyzed by GLC in form of their glycitol acetates, and the structure of polysaccharides in Fractions WSP-1 and WSP-2 was determined by FT-IR and GC-MS after methylation through and acetylation. The sugars of WSPs from Korean oak mistletoe, Loranthus yadoriki, are majorly arabinose and galactose in stem, galactose in leaves very high in content and showed difference in composition and monomeric units between stems and leaves. D-galactose, D-glucose and L-arabinose are the simple sugars consisting of polysaccharides in WSP-1. ($1{\rightarrow}3$)-Linked galactan is the bakcbone with side chain of ($1{\rightarrow}5$)- -L-arabinofuranosyl residues and ($1{\rightarrow}6$)- -D-galactopyranosyl residues, and ($1{\rightarrow}4$)-linked glucan also presents. ($1{\rightarrow}4$)-Linked rhamnogalacturonan and ($1{\rightarrow}4$)- and ($1{\rightarrow}3$)-linked galactan present in WSP-2.
