Cosmetical Properties of Polysccharides from the Root Bark of Ulmus davidiana var. Japonica (유근피로부터 추출한 다당류의 화장료적 특성)

  • The fruit or root of Ulmus davidiana var. Japonica and its extract has traditionally been used as anti inflammatation medicines as well as a remedy of the digestive ulcer for a long time. Ulmus davidiana var. Japonica has also been used to cure palliative and diuresis, because it contains lot of mucus and tannin. In this study, we examined the moisturizing effect and anti-inflammation effect of polysaccharides extracted from Ulmus davidiana var. Japonica to investigated whether it can be used as a cosmetic ingredient or not. The polysaccharides were extracted from Ulmus davidiana var. Japonica. Then the hydrolyzed was obtained through the hydrolysis. It has been ascertained that the polysaccharides we mainly composed of rhamnose, galactose, and glucose through HPLC experiments, and it has 20,000 of the molecular weight and 89.3 dL/g of intrinsic viscosity In a moisturizing test of measuring water loss in a desiccator and moisture content by Comeometer CM820, Ulmus davidiana root extract showed a similar moisturizing effect as hyaluronic acid. And its anti-inflammation effect on carrageenin-induced edema evaluated by measuring the thickness of swelling over paw induced by carrageenin (25 uL of $2\%$ carrageenin saline injection), the $\%$ swelling over paw treated with Ulmus davidiana root extract was much less than that of paw treated with carrageenin only. The anti-inflammatory effect of Ulmus davidiana root extract was almost similar to that of ketoprofen. These results can say that Ulmus davidiana root extract can be effectively used as a cosmetic ingredient.

Surface Display of Bacillus CGTase on the Cell of Saccharomyces cerevisiae (Saccharomyces cerevisiae에서 Bacillus CGTase의 표층발현)

  • For the expression in Saccharomyces cerevisiae, Bacillus stearothermophilus cyclodextrin glucano­transferase gene (cgtS) in pCGTS (4.8 kb) was subcloned into the surface expression vector, pYD1 (GALl promoter). The constructed plasmid, pYDCGT (7.2 kb) was introduced into S. cerevisiae EBY100 cells, and then yeast transformants were selected on the synthetic defined media lacking tryptophan. The formation of cyclodextrin (CD) was confirmed with active staining of culture broth of transformant grown on starch medium. Enzymatic reaction products with respect to the culture time and the reaction time were examined by TLC analysis. The results indicated that the enzyme activity was exhibited after 12 h cultivation and CD was produced after 10min of enzymatic reaction. When the surface-engineered yeast cells were cultured on galactose medium, maximum activities of CGTase were about 21.3 unit/l and 16.5 unit/l at $25^{\circ}C\;and\;30^{\circ}C$, respectively. The plasmids stability showed about $80\%\;even\;at\;25^{\circ}C\;and\;30^{\circ}C$.

Changes of Oligosaccharide and Free Amino Acid in Soy Yogurt Fermented with Different Mixed Culture (혼합균주를 이용한 대두유의 발효에 따른 당 및 유리아미노산의 변화)

  • This study was carried out to investigate the oligosaccharide and amino acid utilization by mixed cultures during soy yogurt fermentation. Three types soy yogurt were prepared by fermenting with Lactobacillus acidophilus and Streptococcus thermophilus, Streptococcus thermophilus and Saccharomyces uvarum, Lactobacillus acidophilus and Saccharomyces uvarum. The utilized amount of oligosaccharide and amino acid was determined by HPLC during the fermentation period. The oligosaccharide and amino acid utilization efficiency of S. thermophilus and Sac. uvarum was greater than the other mixed cultures. It was found that Sac. uvarum produced enzymes which can convert oligosaccharide and common sugars in soy milk into glucose, galactose and fructose which can be fermented by L. acidophilus and S. thermophilus, and in turn stimulated acid production and amino acid utilization of the latter.

Effects of Lactose and Yeast on the Growth of Lactic Acid Bacteria and Sensory Characteristics during the Fermentation of Soy Yogurts (Lactose와 효모의 첨가가 대두요구르트 발효 중 젖산균의 생육특성 및 관능적 특성에 미치는 영향)

  • Lactobacillus bulgaricus and Kluyveromyces lactic were inoculated to Jangyeob and Jinpum soymilks single or together after the addition of different amounts of lactose to increase the formation of lactic acid and sensory evaluation. The decrease of pH and the increase of acid production during the fermentation of soy yogurts were more effective when lactose was added. And the single culture method was more effective in decreasing pH and in increasing acid production than the mixed culture method. Without lactose, the growth of lactic acid bacteria in samples increased by mixed culture method than by single culture method. However, the growth of lactic acid bacteria increased more in the by single cultured samples than in the mixed cultured samples by the addition of lactose. Beany flavor decreased more in soy yogurts compared with Jangyeob and Jinpum soy milks, and Jinpum soy yogurts had less beany flavor than Jangyeob soy yogurts. In sour taste, Jangyeob soy yogurt prepared by mixed culture for 36 hr incubation with 4% lactose showed the poorest score, while soy yogurts containing 2% lactose showed significantly better scores and no significant difference compared with milk yogurt. Soy yogurts containing 2% lactose showed no significant difference in overall acceptability compared with milk yogurt.

Water-holding Capacity and Antimicrobial Activity and of 1, 2-Hexanediol Galactoside Synthesized by β-Galactosidase (베타-갈락토시데이즈를 이용하여 합성한 1, 2-Hexanediol Galactoside의 보습력과 항균력에 대한 연구)

  • We carried out the enzymatic synthesis of 1, 2-hexanediol galactoside (HD-gal) by transgalactosylation reaction using recombinant Escherichia coli ${\beta}-galactosidase$ (${\beta}-gal$). The amounts of ${\beta}-gal$ and 1, 2-hexanediol (HD), pH, and temperature, respectively, were first optimized (${\beta}-Gal$, 4.8 U/mL; HD, 75 mM; pH, 7.0; temperature, $37^{\circ}C$). Under these optimal conditions, about 96% HD was converted to HD-gal. When we investigated the water holding capacities (WHCs) of HD and HD-gal using pig epidermis in the concentrations of 84.4, 126.6, 168.8, 211.0 mM, WHC of HD-gal was superior to HD. In particular, at 168.8 mM HD and HD-gal, WHC of HD-gal showed about 20% greater than that of HD. However, it was observed that MIC values against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus of HD-gal were about three to ten times greater than those of HD, although MIC value of HD-gal against Enterococcus faecalis was almost the same as that of HD. Finally, it was concluded that the covalent bonding of a galactose molecule to HD (transgalactosylation) resulted in an increase in WHC of HD-gal and a decrease in anti-bacterial activity.

Bioconversion of Rare Sugars by Isomerases and Epimerases from Microorganisms (미생물 유래 당질관련 이성화효소 및 에피머효소를 이용한 희소당 생물전환)

  • The International Society of Rare Sugars (ISRS) defines rare sugars as monosaccharides and their derivatives that rarely occur in nature. Rare sugars have recently received much attention because of their many uses including low-calorie sweeteners, bulking agents, and antioxidants, and their various applications including as immunosuppressants in allogeneic rat liver transplantation, as potential inhibitors of various glycosidases and microbial growth, in ischemia-reperfusion injury repair in the rat liver, and in segmented neutrophil production without detrimental clinical effects. Because they rarely exist in nature, the production of rare sugars has been regarded as one of the most important research areas and, generally, they are produced by chemical synthesis. However, the production of rare sugars by bioconversion using enzymes from microorganisms has been receiving increased attention as an environmentally friendly alternative production method. In particular, D-allulose, D-allose, and D-tagatose are of interest as low-calorie sweeteners in various industries. To date, D-tagatose 3-epimerase, D-psicose 3-epimerase, and D-allulose 3-epimerase have been reported as D-allulose bioconversion enzymes, and L-rhamnose isomerase, Galactose 6-phosphate isomerase, and Ribose 5-phosphate isomerase have been identified as D-allose production enzymes. Elsewhere, D-tagatose has been produced by L-arabinose isomerase from various microorganisms. In this study, we report the production of D-allulose, D-allose, and D-tagatose by microorganism enzymes.

Physicochemical Characteristics of Galactomannan by Fractionation to Evaluate Heterogeneity (불균일 성질을 평가하기 위한 분획화된 galactomannan의 이화학적 특성)

  • Heterogenous samples of locust bean gum (galactomannan) were prepared into homogeneous substances. Locust bean gum was fractioned using ammonium sulfate (14.11-23.08%, w/w). The intrinsic viscosity was obtained by extrapolating reduced viscosity versus concentration by using an Ubbelohde viscometer. The ranges of intrinsic viscosity for fractions that not included protein (F3-F6) and fractions that included protein (F1-F2) were 9.89-8.10 and 8.44-4.59, respectively. Values for Huggins' coefficient (k'), which depends on physical interactions, were 0.46-0.78. Increasing ammonium sulfate concentration was associated with a weak trend towards lower molecular weight and intrinsic viscosity by size-exclusion chromatography (SEC): $M_w$ ranged from 674 to 617 kg/mol and [${\eta}$] from 9.80 to 8.10 dL/g between F3 and F6. The evaluations of those fractions by using SEC and the Ubbelohde viscometer produced very similar values, as predicted. We verified the application of a gradient of ammonium sulfate to precipitate locust bean gum into fractions of different molecular size and show structural variations.

Chemical Properties and Immuno-Stimulating Activities of Crude Polysaccharides from Enzyme Digests of Tea Leaves (녹차 효소 처리 다당의 화학적 특성 및 면역증진 활성)

  • In order to develop new immuno-stimulating ingredients from mature leaves of green tea, crude polysaccharides were isolated from pectinase digests of tea leaves (green tea enzyme digestion, GTE-0), after which their immuno-stimulating activities and chemical properties were examined. GTE-0 mainly contained neutral sugars (54.9%) such as glucose (14.2%), arabinose (12.2%), rhamnose (11.1%), and galacturonic acid (45.1%), which are characteristic of pectic polysaccharides. The anti-complementary activity of GTE-0 was similar to that of polysaccharide K (used as positive control). Number of morphologically activated macrophages was significantly increased in the GTE-0-treated group. GTE-0 significantly augmented $H_2O_2$ and reactive oxygen species production by murine peritoneal macrophage cells in a dose-dependent manner, whereas production of nitric oxide showed the highest activity at a dose of $100{\mu}g/mL$ among all tested concentrations. Murine peritoneal macrophages stimulated with GTE-0 showed enhanced production of various cytokines such as interleukin (IL)-6, IL-12, and tumor necrosis factors-${\alpha}$ in a dose-dependent manner. Further, GTE-0 induced higher phagocytic activity in a dose-dependent manner. In ex vivo assay for cytolytic activity of murine peritoneal macrophages, GTE-0-treated group showed significantly higher activity compared to the untreated group at an effector-to-target cell ratio of 20. The above results lead us to conclude that polysaccharides from leaves of green tea have a potent immuno-stimulating effect on murine peritoneal macrophage cells.

Analysis of Nutritional Components of Lepista nuda (민자주방망이버섯(L. nuda)의 영양성분 분석)

  • This study was carried out to analyze the nutritional component of Lepista nuda in order to estimate its nutritional and functional values. The content of moisture were $90.90{\pm}0.09%$ carbohydrate $4.34{\pm}0.07%$, crude protein $3.70{\pm}0.03%$, ash $0.55{\pm}0.04%$, and crude fat $0.47{\pm}0.01%$, respectively The reducing sugar was $636.17{\pm}7.50mg$. Total amount of free sugar was 97.32 mg, which were mainly composed of galactose (51.98 mg) and trehalose (22.48 mg). In the results of mined analysis, the content of K was the highest as $103.10{\pm}0.18mg$, followed by $56.01{\pm}0.20mg$ of Mg. The total content of hydrolyzed amino acid was 310.39 mg which consist of essential amino acids including valine (25.40 mg), leucine (22.52 mg), lysine (21.34 mg) and 198.89 mg of non-essential amino acids including glutamic acid (60.72 mg), aspartic acid (30.17 mg), and alanine (29.72 mg), as the major amino acids. free amino acids were 220.86 mg which were composed of essential amino acids (36.21 mg) inclusive of isoleucine (21.57 mg) and threonine (11.20 mg) and non-essential amino acids (184.65 mg) including glutamic acid (42.78 mg) and aspartic acid (32.44 mg) as major amino acids. Total content of amino acid derivatives was 187.85 me and that of hydroxyproline was highest as 121.50 mg.

Fermentation Properties of Yogurt Added with Rice (쌀 분말 첨가량에 따른 요구르트의 발효 특성)

  • Yogurt was prepared from skim milk added with 1, 2, 4 or 6% of rice- or skim milk powders and commercial lactic acid bacterial starters. Changes in pH, titratable acidity, viable cell counts, viscosity, organic acid contents and carbohydrates during fermentation were monitored and its sensory evaluation was also performed. The optimum level of additives such as rice- and skim milk powders for yogurt manufacture was selected. Fermentation properties of yogurt added with rice and skim milk powders were studied. The control yogurt reached pH 4.5 after 10 hours of fermentation, whereas the samples added with 4 or 6% rice powders reached pH 4.5 in 6 hours and those added with skim milk powder reached in 8 hours. After 4 hours of fermentation, the control yogurt reached a titratable acidity at 1.0 %, whereas other samples exceeded 1.0%. After 4 hours of fermentation, the viable cell counts in the samples added with 4% rice powders were the highest. At the level of 4% rice powders, the curd viscosity decreased. Except for the sample added with 6% skim milk powders, all the samples produced higher lactic acid during fermentation, and galactose level in yogurt was the highest when added with 4% rice powder or 6% skim milk powder. In overall sensory evaluation using 5 parameters, the sensory scores of all the yogurts were not significantly different. However, the sample added with 4% skim milk powders was the best for color and overall acceptability. The sample added with 4% rice powders was the best for the flavor and texture. From this experiment, the optimun level of additives such as rice or skim milk powders was selected to be 4%.
