Characterization and Bioactivities of a Novel Exopolysaccharide Produced from Lactose by Bacillus tequilensis PS21 Isolated from Thai Milk Kefir

  • The aims of this work were to characterize and determine bioactivities of crude exopolysaccharide (EPS) extract from Bacillus tequilensis PS21 isolated from milk kefir from Kampaeng Petch, Thailand. B. tequilensis PS21 produced 112.1 mg dried EPS/l from initial 80 g/l lactose in modified TSB media at 52 h, with EPS product yield of 8.9 mg EPS/g lactose and specific product yield of 0.3 mg EPS/mg biomass. The FTIR result confirmed EPS to be a protein-bound polysaccharide and SEM analysis showed the morphology to be a grainy appearance with an uneven surface, covered with pores. HPLC analysis determined EPS as a heteropolysaccharide consisting of five sugar units with the following molar ratios; xylose (17.65), glucose (2.54), ribose (1.83), rhamnose (1.23), and galactose (1). Chemical components of this EPS were predominantly carbohydrate at 697.8 mg/g EPS (65%), protein 361.4 mg/g EPS (34%), and nucleic acid 12.5 mg/g EPS (1%). The EPS demonstrated antioxidant activities at 57.5% DPPH scavenging activity, $37.2{\mu}M\;Fe(II)/mg$ EPS and $34.9{\mu}M\;TEAC\;{\mu}M/mg$ EPS using DPPH, FRAP and ABTS assays, respectively. EPS also exhibited anti-tyrosinase activity at 34.9% inhibition. This work represents the first finding of EPS produced by Bacillus sp. from Thai milk kefir which shows potential applications in the production of antioxidant functional foods and whitening cosmetics. However, optimization of EPS production for industrial exploitation requires further study to ascertain the economic potential.

Physicochemical and Rheological Properties of a Novel Emulsifier, EPS-R, Produced by the Marine Bacterium Hahella chejuensis

  • The rheological properties of an exopolysaccharide, EPS-R, produced by the marine bacterium Hahella chejuensis strain 96CJ 10356 were investigated. The $E_{24}$ of $0.5\%$ EPS-R was $89.2\%$, which was higher than that observed in commercial polysaccharides such as xanthan gum ($67.8\%$), gellan gum ($2.01\%$) or sodium alginate ($1.02\%$). Glucose and galactose are the main Sugars in EPS-R, with a molar ratio of ${\~}1:6.8$, xylose and ribose are minor sugar components. The average molecular mass, as determined by gel filtration chromatography, was $2.2{\times}10^3$ KDa, The intrinsic viscosities of EPS-R were calculated to be 16.5 and 15.9 dL/g using the Huggins and Kraemer equations, respectively, with a 2.3 dL/g overlap. In terms of rigidity, the conformation of EPS-R was similar to that of caboxymethyl cellulose ($5.0{\times}10^{-2}$). The rheological behavior of EPS-R dispersion indicated that the formation of a structure intermediate between that of a random-coil polysaccharide and a weak gel. The aqueous dispersion of EPS-R at concentrations ranging from 0.25 to $1.0\%$ (w/w) showed a marked shear-thinning property in accordance with Power-law behavior. In aqueous dispersions of $1.0\%$ EPS-R, the consistency index (K) and flow behavior index (n) were 1,410 and 0.73, respectively. EPS-R was Stable to pH and salts.

Metabolite profiling of fermented ginseng extracts by gas chromatography mass spectrometry

  • Background: Ginseng contains many small metabolites such as amino acids, fatty acids, carbohydrates, and ginsenosides. However, little is known about the relationships between microorganisms and metabolites during the entire ginseng fermentation process. We investigated metabolic changes during ginseng fermentation according to the inoculation of food-compatible microorganisms. Methods: Gas chromatography mass spectrometry (GC-MS) datasets coupled with the multivariate statistical method for the purpose of latent-information extraction and sample classification were used for the evaluation of ginseng fermentation. Four different starter cultures (Saccharomyces bayanus, Bacillus subtilis, Lactobacillus plantarum, and Leuconostoc mesenteroide) were used for the ginseng extract fermentation. Results: The principal component analysis score plot and heat map showed a clear separation between ginseng extracts fermented with S. bayanus and other strains. The highest levels of fructose, maltose, and galactose in the ginseng extracts were found in ginseng extracts fermented with B. subtilis. The levels of succinic acid and malic acid in the ginseng extract fermented with S. bayanus as well as the levels of lactic acid, malonic acid, and hydroxypruvic acid in the ginseng extract fermented with lactic acid bacteria (L. plantarum and L. mesenteroide) were the highest. In the results of taste features analysis using an electronic tongue, the ginseng extracts fermented with lactic acid bacteria were significantly distinguished from other groups by a high index of sour taste probably due to high lactic acid contents. Conclusion: These results suggest that a metabolomics approach based on GC-MS can be a useful tool to understand ginseng fermentation and evaluate the fermentative characteristics of starter cultures.

영지(Ganoderma lucidum) 균사체의 액체배양에 의한 세포외 생물고분자의 생산조건과 특성 (Production Conditions and Characterization of the Exo-biopolymer Produced by Submerged Cultivation of Ganoderma lucijum Mycelium)

  • For the screening and the development of the new bio-material, cultural conditions for the exo-biopolymer (EBP) production throught the submerged cultivation of Ganoderma lucidum mycelium were investigated. Also, the fractionations and the purifications of the exo-biopolymer were carried out and the chemical compositions of the exo-biopolymer were examined. The optimal culture conditions for the exo-biopolymer production were pH 5.0, 30$^{\circ}C$ and 100 rpm of agitation speed in the medium containing of 5% (w/v) glucose, 0.5%(w/v) yeast extract, 0.1% (w/v) ($(NH_4)_2HPO_4$, and 0.05% (w/v) $KH_2PO_4$. In the flask cultivation for 7 days under these conditions, the concentration of the maximum exo-biopolymer and the cell mass were 15.4g/l and 18.8g/l, respectively. The specific growth rate was 0.039 $hr^{-1}$. In addition, the substrate consumption rate, and the exo-biopolymer production rate were 0.043$gg^{-1}$$hr^{-1}$ and 0.025$gg^{-1}$$hr^{-1}$, respectively. The exo-biopolymer was fractionated into BWS (water soluble exo-biopolymer) and BWI (water insoluble exo-biopolymer) by the water extraction, and the sugar contents of two fractions were higher than 97% (based on dry basis). The components sugar of BWS and BWI fractions were glucose, galactose, mannose, xylose, and fucose. Their molar ratios were 3.6:1.5:2.1:0.5: trace and 2.9:3.1:2.0:1.6:0.3, respectively.

Opuntia ficus-indica 다당 A-1의 특성 및 알코올유도 간 산화스트레스의 보호 효과 (Characterization of polysaccharide A-1 from Opuntia ficus-indica and it's protection effect on alcoholic induced hepatic oxidative stress)

  • Reactive oxygen species(ROS) can induce hepatotoxicity and trigger apoptosis in the liver. In this study, we investigated the sulfated polysaccharide A-1 from Opuntia ficus-indica against alcoholic oxidative stress in human liver Hep G2 cell. An antioxidant substance A-1 obtained from the enzymatic extract of Opuntia ficus-indica fruit was purified by DEAE-cellulose ion exchange and sephadex G-100 gel permeation chromatography. The purification yield and molecular weight were 14.3% and 1.8 KDa, respectively. The A-1 predominately contained arabinose, galactose, rhamnose and also sulfate group. The structure of A-1 was investigated by periodate oxidation, FT-IR spectroscopy, $^1H$-NMR spectroscopy. The A-1 mainly composed of alternating unit of ${\rightarrow}4$)-$\alpha$-L- Rapp-2-$SO_3^-$-$\alpha$-L-Galp-($1{\rightarrow}$ and branched linkage of $\beta$-D-Arbp- ($5{\rightarrow}$. The antioxidative activity was measured using the SOD, CAT activity and GSH assay, respectively. The expression of Nrf2 protein was analyzed by western blotting. The viable cell count analyzed by autofluorescence. Oxidative stress induced by ethanol(1 M) were dramatically reduced by A-1 treatment. A-1 also prevented cell death induced by oxidative stress. It also increased expression Nrf2 protein level. We concluded that sulfated polysaccharide A-1 from Opuntia ficus-indica effectively protect Hep G2 liver cell from alcoholic oxidative stress.

Insight Into Genes Involved in the Production of Extracellular Chitinase in a Biocontrol Bacterium Lysobacter enzymogenes C-3

  • The chitinase producing Lysobacter enzymogenes C-3 has previously been shown to suppress plant pathogens in vitro and in the field, but little is known of the regulation of chitinase production, or its role in antimicrobial activity and biocontrol. In this study, we isolated and characterized chitinase-defective mutants by screening the transposon mutants of L. enzymogenes C-3. These mutations disrupted genes involved in diverse functions: glucose-galactose transpoter (gluP), disulfide bond formation protein B (dsbB), Clp protease (clp), and polyamine synthase (speD). The chitinase production of the SpeD mutant was restored by the addition of exogenous spermidine or spermine to the bacterial cultures. The speD and clp mutants lost in vitro antifungal activities against plant fungal pathogens. However, the gluP and dsbB mutants showed similar antifungal activities to that of the wild-type. The growth of the mutants in nutrient rich conditions containing chitin was similar with that of the wild-type. However, growth of the speD and gluP mutants was defective in chitin minimal medium, but was observed no growth retardation in the clp and dsbB mutant on chitin minimal medium. In this study, we identified the four genes might be involved and play different role in the production of extracellular chitinase and antifungal activity in L. enzymogenes C-3.

표고버섯 균사체 배양 및 그 추출물의 생리학적 특성

  • 한국산 표고버섯 균사체를 액체배양하여 천연항암물질로 알려진 단백다당체를 추출한 후 그 물질의 특성에 대하여 검토하였다. 균사체의 최적 재양조건을 TGY배지로 조사한 바 온도 $25^{\circ}C$, 배양초기 pH4.0, 교반속도 300rpm, 균사배지 접종량을 10.0%로 하고 산소 통기량을 1.0volume of ait/volume of medium/mimute으로 하였을때 가장 양호한 조건이였으며, 대량생산 하기 위한 SCM배지에서 최적의 C/N비는 13.1오써 7일간 배양하였을때 18.8g/L의 균사령을 얻었으며, 이때 생산수율은 0.46으로 나타났다. 발효가 끝난 배양액에서 균사, 여액 그리고 배양액의 전체에서 단백다당체를 분리한 결과 각 분획에서 단백다당체가 각각 0.55%, 0.12%, 0.69%가 회수되어 배양액 전체에서 단백다당체를 추출하는 것이 바람직하며, 추출방법으로 열수추출, glass bead추출 및 cellulasa처리를 하여 단백다당체의 수율을 비교한 결과 0.25-0.5mm glass nead로 30분간 균사체를 분쇄한 다음 열수추출을 1시간을 하였을때 990mg/100ml의 단백다당체를 얻을 수 있었다. 고단백다당체를 1차 단백질 가수분해 효소로 분해하고, EDAE cellulose 및 Sepadex G-100 column chromatography로 정제한 후, TLC/FLD, ultracentrifugation한 결과 순수한 물질임을 알 수 있었다. 단백다당체의 항암효과 조사중 in vitro배양에서 $P_{388}$$L_{1210}$에 대한 단백다당체의 활성단위 1 unit는 1mg정도였으며, 인체의 장암세포인 HCT-48, HRT-18, HT-29 밀 간암세포인 Hep G2 대한 생육저해 단위는 각각 4.4, 3.6, 6.6, 2.6mg이었다. HCT-48과 Hep G2 세포의 크기 분포도는 대조군에 비하여 시간이 경과함에 따라, 그리고 단백다당체의 농도가 증가함에 따라 peak가 작은 size 쪽으로 이동하였다. 또, 단백다당체를 첨가 배양한 HCT-48과 Hep G2세포의 현미경 관찰에서 본래의 암세포 형태가 변형되고 크기가 감소하며 세포사이의 경계막이 흐트러지면서 세포수가 감소하고 사멸하였다. In vivo실험에서는 대조군보다 단백다당체를 첨가한 군에서 항체 형성능력이 대조군에 비하여 형질세포가 2배로 증가하였다. 단백다당체의 화학적 성분 분석에서 다당함량은 46.1%이면 구성다당류는 glucose, galactose, mannose, xylose로 구성되었고 단백질의 함량은 7.28%이며, 구성아미노산은 15종의 아미노산으로 되었다. 또 무기물은 Na, K, Zn, Ca등의 순으로 이루어 짐을 알 수 있었다.

  • 한국산 고등균류의 성분중에서 항암성분을 탐색하고자, 경기도 수원에서 채집한 간버섯 Trametes sanguinea (L. ex Fr.) Lloyd.로 부터 무미 무취인 짙은 갈색 분말을 추출 분리하였다. 이 물질은 마우스에 이식된 sarcoma 180에 대해 높은 저지율을 보였고, 그 저지작용은 이 물질의 치료적인 투여에 의해서 나타났다. 이 물질의 성분은 다당류와 단백질이었으며, 다당류는 fucose, glucose와 galactose의 3종의 당을 함유하였다. 단백질 분획은 13종의 아미노산으로 구성되었으며, 특히 산성 아미노산이 많은 반면에 염기성 아미노산은 적었다.

생물전환으로 생리활성물질인 trifolin의 생합성 (Biosynthesis of trifolin, a bioactive flavonoid by biotransformation)

  • Trifolin (kaempferol 3-O-galactoside)는 플라보놀 그룹에 속하는 물질로 아토피, 항균, 폐암에 효과가 있는 것으로 알려져 있다. Trifolin은 다양한 식물에서 추출하여 사용하고 있지만 추출 과정이 복잡하고, 수율이 낮으며, 추출을 위한 바이오매스를 얻는데 계절적 어려움이 있다. 생물전환은 저렴한 화합물에서 고부가가치 화학물질을 생산할 수 있는 대체 수단으로 이용된다. 본 연구에서는 naringenin으로부터 trifolin을 생합성하기 위해 3개의 유전자(PeFLS 및 OsUGE-PhUGT)를 각각의 대장균에 도입한 BL-FLS균주와 BL-UGTE균주를 이용하여 공조배양시스템을 개발하였다. Naringenin으로부터 trifolin을 생합성하기 위해 세포의 밀도, 생물전환 온도, 재조합 단백질 유도의 적정 IPTG농도 및 시간, 기질 공급 농도 등의 최적화를 실시하였다. 최적화된 공동 배양 발효 시스템을 통해 67.3 mg/L의 trifolin을 성공적으로 생합성 하였다.

Constipation anti-aging effects by dairy-based lactic acid bacteria

  • Constipation, which refers to difficulties in defecation and infrequent bowel movement in emptying the gastrointestinal system that ultimately produces hardened fecal matters, is a health concern in livestock and aging animals. The present study aimed to evaluate the potential effects of dairy-isolated lactic acid bacteria (LAB) strains to alleviate constipation as an alternative therapeutic intervention for constipation treatment in the aging model. Rats were aged via daily subcutaneous injection of D-galactose (600 mg/body weight [kg]), prior to induction of constipation via oral administration of loperamide hydrochloride (5 mg/body weight [kg]). LAB strains (L. fermentum USM 4189 or L. plantarum USM 4187) were administered daily via oral gavage (1 × 10 Log CFU/day) while the control group received sterile saline. Aged rats as shown with shorter telomere lengths exhibited increased fecal bulk and soften fecal upon administration of LAB strains amid constipation as observed using the Bristol Stool Chart, accompanied by a higher fecal moisture content as compared to the control (p < 0.05). Fecal water-soluble metabolite profiles showed a reduced concentration of threonine upon administration of LAB strains compared to the control (p < 0.05). Histopathological analysis also showed that the administration of LAB strains contributed to a higher colonic goblet cell count as compared to the control (p < 0.05). The present study illustrates the potential of dairy-sourced LAB strains as probiotics to ameliorate the adverse effect of constipation amid aging, and as a potential dietary intervention strategy for dairy foods including yogurt and cheese.
