The Characteristics of Tetrachloroethylene (PCE) Degradation by Pseudomonas putida BJ10 (Pseudomonas putida BJ10의 Tetrachloroethylene (PCE) 분해 특성)

  • In this study, biological PCE degradation by using a BTEX degrading bacterium, named BJ10, under aerobic conditions in the presence of toluene was examined. According to morphological, physiological characteristics, 16S rDNA sequencing and fatty acid analysis, BJ10 was classified as Pseudomonas putida. As a result of biological PCE degradation at low PCE concentrations (5 mg/L), PCE removal efficiency by P. putida BJ10 was 52.8% for 10 days, and PCE removal rate was 5.9 nmol/hr (toluene concentration 50 mg/L, initial cell density 1.0 g (wet weight)/L, temperature 30, pH 7 and DO $3.0{\sim}4.2\;mg/L$. At high PCE concentration (100 mg/L), PCE removal efficiency by P. putida BJ10 was 20.3% for 10 days, and PCE removal rate was 46.0 nmol/hr under the same conditions. The effects of various toluene concentration (5, 25, 50, 100, 200 mg/L) on PCE degradation were examined under the same incubation conditions. The highest PCE removal efficiency of PCE was 57.0% in the initial PCE concentration of 10 mg/L in the presence of 200 mg/L toluene for 10 days. Furthermore, the additional injection of 5.5 mg/L PCE (total 7.6 mg/L) made 63.0% degradation for 8 days in the presence of 50 mg/L toluene under the same conditions. Its removal rate was 13.5 nmol/hr, which was better than the initial removal rate (8.1 nmol/hr).

Effect of Triazole Compounds Treatment on Quality of Cut Flower of Dendranthema grandiflorum 'Baegkwang' (트리아졸계 화합물 처리가 절화국화의 품질에 미치는 영향)

  • The objective of this study was to investigate the effect of triazole compounds [hexaconazole ($50mg{\cdot}L^{-1}$), diniconazole ($50mg{\cdot}L^{-1}$), myclobutanil ($150mg{\cdot}L^{-1}$), difenoconazole ($50mg{\cdot}L^{-1}$), tebuconazole ($500mg{\cdot}L^{-1}$), and bitertanol ($50mg{\cdot}L^{-1}$)] on commercial quality improvement of cut flowers of Dendranthema grandiflorum 'Baegkwang' Plant height, cut flower length and internode length decreased by all triazole compounds. Plant height, cut flower length, internode length and flower neck length were shortened by treatment of $50mg{\cdot}L^{-1}$ diniconazole and $500mg{\cdot}L^{-1}$ tebuconazole. Flower neck was thickened by hexaconazole and myclobutanil. Flower diameter increased with myclobutanil and bitertanol, while it decreased with tebuconazole. Among physiological injury, open center increased with bitertanol treatment. Overall $50mg{\cdot}L^{-1}$ diniconazole and $500mg{\cdot}L^{-1}$ tebuconazole showed a possitive effect on length flower neck and increasing chlorophyll content, while decreasing plant height, cut flower length and diameter of flowers. Thus it is necessary to consider kind, concentration, and treatment times for effective control of cut flower quality.

Effect of Pretreatment and Holding Solution on Vase Life and Quality of Cut Flowers of Aster koraiensis Nakai (벌개미취 절화의 품질향상과 수명연장을 위한 전·후처리제의 효과)

  • This experiment was conducted to improve postharvest quality and extended vase life of Korean native Aster koraiensis Nakai for use as cut flowers. Aster koraiensis Nakai cut flowers were treated with various pretreatment and holding solution. Postharvest pretreatment with 3% sucrose + $200mg{\cdot}L^{-1}$ HQC + $50mg{\cdot}L^{-1}\;AgNO_3$ + $50mg{\cdot}L^{-1}$ BA and 3% sucrose + $200mg{\cdot}L^{-1}$ HQC + $50mg{\cdot}L^{-1}\;AgNO_3$ + $150mg{\cdot}L^{-1}$ citric acid for 16 hours extended vase life of cut Aster koraiensis Nakai flowers by 1.4 times as compared with the control (distilled water). Holding solution of 3% sucrose + $200mg{\cdot}L^{-1}$ HQC + $50mg{\cdot}L^{-1}\;AgNO_3$ + $50mg{\cdot}L^{-1}$ BA and 3% sucrose + $200mg{\cdot}L^{-1}$ HQC + $50mg{\cdot}L^{-1}\;AgNO_3$ + $150mg{\cdot}L^{-1}\;GA_3$ extended vase life of cut Aster koraiensis Nakai flowers by1.6 and 1.7 times as compared with control (distilled water). Aster koraiensis Nakai.flowers held in this preservative solution increased fresh weight and were maintained positive water balance for a long vase period. It was suggested that the vase life of cut Aster koraiensis Nakai flowers was closely related to fresh weight and water balance of the cut flower.

Evaluation of γ-Aminobutyric Acid (GABA) Production by Lactic Acid Bacteria Using 5-L Fermentor (Lactic Acid Bacteria (LAB)와 5-L 발효기를 이용한 γ-Aminobutyric Acid 생산기술 개발)

  • This study aimed to optimize gamma-aminobutyric acid (GABA) production by employing five strains of lactic acid bacteria (LAB) that were capable of high cell growth and GABA production using a modified synthetic medium. GABA production in the strains was qualitatively confirmed via detection of colored spots using thin layer chromatography. Lactobacillus plantarum SGL058 and Lactococcus lactis SGL027 were selected as the suitable strains for GABA production. The conditions of the carbon and nitrogen sources were determined as 5 g/l glucose (L. plantarum SGL058), 5 g/l lactose (L. lactis SGL027), 10 g/l yeast extract (L. plantarum SGL058), and 20 g/l yeast extract (L. lactis SGL027) for GABA production. The cell growth, monitored by optical density at 600 nm, was 5.93 for L. plantarum SGL058. This value was higher than the 3.04 produced by L. lactis SGL027 at 36 h using a 5-L fermenter. The highest concentration of GABA produced was 546.7 ㎍/ml by L. plantarum SGL058 and 404.6 ㎍/ml by L. lactis SGL027, representing a GABA conversion efficiency of (%, w/w) of 4.0% and 3.4%, respectively. The fermentation profiles of L. plantarum SGL058 and L. lactis SGL027 provide a basis for the utilization of LAB in GABA production using a basal synthetic medium.

A Study on Serum Lipid Levels of Elderly People in Wando Area -Based on Dietary Behaviors- (완도지역 중·장년층의 혈중지질 수준에 관한 연구 -식행동을 중심으로-)

  • This study was performed to document the association between eating behaviors and major risk factors for cardiovascular diseases. Those who stated that they ate a daily breakfast for male and female were 88.6% and 96.0%, $1{\sim}2$ times a week were 9.1% and 2.0%, $3{\sim}4$ times a week were 2.3% and 2.0%, respectively. Those who stated that they were overeating of $0{\sim}1$ time a week for man and female were 80.7% and 89.9%, overeating of $2{\sim}3$ times a week were 19.3% and 10.1%, respectively. Those who said that they were regular of meal time for man and female were 38.6%, and 37.4%, sometimes irregular of meal time were 14.8% and 19.2%, irregular of meal time were 46.6% and 43.4%, respectively. Those who said that they were light of eating volume for man and female were 20.5% and 25.3%, moderate of eating volume were 69.3% and 61.6%, heavy of eating volume were 10.2% and 13.1%, respectively. Those who said that they were very fast of eating speed for man and female were 15.9% and 8.1%, fast of eating speed were 51.1% and 34.3%, moderate of eating speed were 4.5% and 20.2%, slow of eating speed were 17.0% and 14.1%, and very slow of eating speed were 11.4% and 23.2%, respectively. Higher frequency of breakfast a week resulted in higher serum total cholesterol and blood sugar for the daily eating group for both genders with women having high LDL-cholesterol and HDL-cholesterol. Both group had high HDL-cholesterol and low blood sugar with less number of overeating, with men having low triglyceride and LDL-cholesterol. With regular meal, both group had low triglyceride, total-cholesterol, atherogenic index, and blood sugar with women having low LDL-cholesterol. For both groups, the triglyceride, total-cholesterol, LDL-cholesterol, atherogenic index, and blood sugar had higher figures for overeating, with men having low HDL-cholesterol and women having high HDL-cholesterol. This study revealed that less number of overeating, regular mealtime, and less volume of food intake are effective in preventing and treating for the cardiovascular diseases.

  • The polysaccharide C prepared from gum tragacanth powder (U. S. P. grade) by the precipitation method with 85% ethanol was a neutral polysaccharide, $[{\alpha}]^{30}_D-72.2$. The polysaccharide C consisted of L-rhamnose, D-xylose, L-arabinose and D-galactose in the molar ratio 2:1:17:9 (Table 1, 2, 3, ). The polysaccharide C was methylated with dimethylsulphate and 40% NaOH, and Purdies regent. The hydrolyzate of fully methlated product ($[{\alpha}]^{22}_D-102$ in chloroform, the methoxy content 40.6%) was composed of 2, 3, 5-tri-O-methyl-L-arabofuranose (I), 3,4-di-O-methyl-L-rhamnopyranose (II), 2,3-di-O-methyl-D-xylose (III), 2,3,4-tri-O-methyl-D-galactopyranose (IV), 2,4-di-O-methyl-L-arabopyranose (?), 2,4-di-O-methyl-D-galactose(VI), 2-O-methyl-D-arabinose (VII), and L-arabopyranose(VIII) (Table 4, 5, and Fig. 4). The first partial hydrolysis (A) of the polysaccharide C with 0.05N-HCl for 4.5 hours at $80-85^{\circ}C$ released only L-arabinose: the second hydrolysis (B) with 0.1N-HCl for 5 hours at $80-85^{\circ}C$, L-arabinose and D-galactose; and the third hydrolysis (C) with 0.3N-HCl at $90-95^{\circ}C$ in sealed tube, L-rhamnose, D-xylose, L-arabinose and D-galactose. From the unhydrolyzate A' were found L-rhamnose, D-xylose, L-arabinose, and D-galactose; from B' L-rhamnose, d-xylose, L-arabinose and D-galactose; and from C' D-xylose and D-galactose respectively (Table 6). The periodate consumption and formic acid production of the polysaccharide C were measured at various time intervals. After 120 hours periodat was consumed by 1.23 mole per $C_5H_8O_4$ and formic acid was produced 0.78 mole per $C_5H_8O_4$ (Table 7). Although a definite chemical structure for this polysaccharide C may not be formulated, experimental data, especially, from methylation, partial hydrolysie and determination of its molar ratio, and periodate analysis showed that the polysaccharide C is a highly branched polysaccharide and would be constructed of galactoaraban as a main chain residue and L-arabofuranose, D-galactopyranosyl $(1{\rightarrow}1)$-L-arabofuranose, D-xylopyranosyl $(1{\rightarrow}2)$-L-rhamnopyranosyl $(1{\rightarrow}1)$-L-arabofuranose, and L-rhamnopyranosyl $(1{\rightarrow}1)$-arabofuranose, and D-galactopyranosyl-$(1{\rightarrow}2)$-L-arabopyranosyl-$(1{\rightarrow}1)$-I-arabofuranose as a branch chain or end group (page 21).

  • This paper presents characterizations of the power distribution with the parameter $\beta=1$ by the independence of the lower record values. We prove $X\;{\in}\;POW({\alpha},\;1)$ for ${\alpha}\;>\;0$, if and only if $\frac{X_{L(n)}}{X_{L(m)}}$ and $X_{L(m)}$ for $1\;{\leq}\;m\;<\;n$ are independent. And we prove that $X\;{\in}\;POW({\alpha},\;1)$ for ${\alpha}\;>\;0$, if and only if $\frac{X_{L(m)}-X_{L(m+1)}}{X_{L(m)}}$ and $X_{L(m)$ for $m\;{\geq}\;1$ are independent or $\frac{X_{L(m)}-X_{L(m+1)}}{X_{L(m+1)}}$ and $X_{L(m)}$ for $m\;{\geq}\;1$ are independent.

Oxidative Modification of Neurofilament-L by Copper-catalyzed Reaction

  • Neurofilament-L (NF-L) is a major element of neuronal cytoskeletons and known to be important for neuronal survival in vivo. Since oxidative stress might play a critical role in the pathogenesis of neurodegenerative diseases, we investigated the role of copper and peroxide in the modification of NF-L. When disassembled NF-L was incubated with copper ion and hydrogen peroxide, then the aggregation of protein was proportional to copper and hydrogen peroxide concentrations. Dityrosine crosslink formation was obtained in copper-mediated NF-L aggregates. The copper-mediated modification of NF-L was significantly inhibited by thiol antioxidants, N-acetylcysteine, glutathione, and thiourea. A thioflavin-T binding assay was performed to determine whether the copper/$H_2O_2$ system-induced in vitro aggregation of NF-L displays amyloid-like characteristics. The aggregate of NF-L displayed thioflavin T reactivity, which was reminiscent of amyloid. This study suggests that copper-mediated NF-L modification might be closely related to oxidative reactions which may play a critical role in neurodegenerative diseases.

  • The fused lumbar vertebrae (L) of the female Cheju native horse were observed macroscopically. The 5th L and 6th L were partially fused. They were composed of four parts between the spinous process of the 5th L and 6th L, left and right transverse process of the 5th L and 6th L, caudal articular process of the 5th L and cranial articular process of the 6th L, and fossa of vertebra of the 5th L and head of vertebra of the 6th L. The dorsal lumbar foramen and ventral lumbar foramen were formed each at left and right of medial parts in the fused transverse processes of the 5th and 6th lumbar vertebrae.

A Study on the Water Quality of Domestic Mineral Water (Saeng Su) (국내 보존음료수(생수)의 성분에 관한 연구)

  • This study was performed to analyze for the mineral water (Saeng Su) from 12 pure beverage Co. products, which was obtained during March to May in 1988. The results were as follows: 1. The analytical results of contaminant materials in all the samples were suitable for drinking water standard. The mean value of NH$_{3}$-N, NO$_{3}$-N, Cl-and KMnO$_{4}$ consumption were ND, 49mg/l, 1.08mg/l and 8.14mg/l, respectively. 2. Total bacterial counts were 3/cc in average and Coliforms was negative in all samples. 3. The analytical results of Heavy metals were not detected or detected in a small amount, and the contents were different from each company. The CN, Hg, P were not detected in all the samples and the mean value of Cu, Fe, Mn were 0.0087mg/l, 0.0303mg/l, 0.018mg/l, respectively and it's contents were low. 4. The mean value of Pb, Zn, Cr$^{+6}$, As and Cd were 0.016mg/l, 0.015mg/l, 0.0025mg/l. 0.0059mg/l and 0.0011mg/l respectively.
