Influences of Proteolytic Ability of Lactic Acid Bacteria on Acid Production and Precipitates Occurrence in Liquid Yogurt Preparation (액상발효유 제조시 유산균 Starter의 단백질 분해능이 산생성 및 침전발생에 미치는 영향)

  • In making liquid yoghurt, the influences of proteolytic ability of lactic acid bacteria on acid production and on protein stability were investigated. L. bulgaricus CH-2, L. helviticus IAM 1042 and L. jugurti 3048 showed a comparatively high proteolytic activity in milk, while L. casei YIT 9018 did not show any marked proteolysis. Starter organisms having high proteolytic ability showed more rapid growth and acid production than those having low ability in milk. The most active proteolysis occurred during logarithmic growth phase of yogurt organisms, and most of the proteolysis took place in the first 24-48 hrs of incubation. Highly proteolysed yogurts made by L. bulgaricus CH-2, L. jugurti 3048, L. helviticus IAM 1042, L. acidophilus L-54 and L. casei 3012 had low protein solubility at pH 3.5 and had much protein precipitates during storage of product, but those having little protein hydrolysates made by L. casei YIT 9018 or artificial acidification showed no precipitation during keeping.

The Effect of Concentration of Glucose and Salts on both the Growth and the production of Lipid and DHA of Thraustochytrium aureum ATCC 34304 (당농도 및 염농도가 해양미생물 Thraustochytrium aureum ATCC 34304의 성장 및 지질과 DHA 생성에 미치는 영향 규명)

  • The polyunsaturated fatty acids (PUFAs) are mostly synthesized by the marine original microorganisms. In this study, the effect of concentrations of glucose, NaCl, and $MgSO_4{\cdot}7H_2O$ on both the growth and the production of lipid and Docosahexaenoic acid (DHA) was investigated using Thraustochytrium aureum ATCC 34304. $Y_{Lipid/X}\;and\;Y_{DHA/Lipid}$ increased according to the increase of initial glucose concentration until 15 g/L of glucose. The maximum values of $Y_{Lipid/X}\;and\;Y_{DHA/Lipid}$ were 0.18 g/g and 0.41 g/g respectively at 15 g/L of glucose. The biomass yield, $Y_{x/s}$, however, was constant as 0.378 g/g regardless of initial glucose concentrations in the range of 5 g/L to 25 g/L. The specific growth rate of T. aureum increased also with initial glucose concentration from 5 g/L and 15 g/L. The maximum value was $0.79\;day^{-1}$ at 15 g/L of glucose, and the growth rate decreased to $0.57\;day^{-1}$ and remained constant according to initial sugar concentration in the range of higher than 15 g/L of glucose. The concentration of NaCl was changed from 0 g/L to 48 g/L, in order to study the characteristics of the growth and the production of lipid and DHA according to NaCl concentration. The growth and the production of lipid as well as DHA stopped all at 0 g/L of NaCl and the maximum values of all the three variables occurred at 24 g/L. The effect of $MgSO_4{\cdot}7H_2O$ concentration was also investigated not to find the growth and the production of lipid and DHA at 0 g/L. However, the growth and the Production with $MgSO_4{\cdot}7H_2O$ concentration increased to reach the maximum values at 18 g/L of $MgSO_4{\cdot}7H_2O$.

Determination of Optimum Coagulants (Ferric Chloride and Alum) for Arsenic and Turbidity Removal by Coagulation

  • The Raw water from Deer Creek (DC) reservoir and Little Cottonwood Creek (LCC) reservoir in the Utah, USA were collected for jar test experiments. This study examined the removal of arsenic and turbidity by means of coagulation and flocculation processes using of aluminum sulfate and ferric chloride as coagulants for 13 jar tests. The jar tests were performed to determine the optimal pH range, alum concentration, ferric chloride concentration and polymer concentration for arsenic and turbidity removal. The results showed that a comparison was made between alum and ferric chloride as coagulant. Removal efficiency of arsenic and turbidity for alum (16 mg/L) of up to 79.6% and 90.3% at pH 6.5 respectively were observed. Removal efficiency of arsenic and turbidity for ferric chloride (8 mg/L) of up to 59.5% at pH 8 and 90.6% at pH 8 respectively were observed. Optimum arsenic and turbidity removal for alum dosages were achieved with a 25 mg/L and 16 mg/L respectively. Optimum arsenic and turbidity removal for ferric chloride dosages were achieved with a 20 mg/Land 8 mg/L respectively. In terms of minimizing the arsenic and turbidity levels, the optimum pH ranges were 6.5 and 8for alum and ferric chloride respectively. When a dosage of 2 mg/L of potassium permanganate and 8 mg/L of ferric chloride were employed, potassium permanganate can improve arsenic removal, but not turbidity removal.

The Ultimate Load Capacity of the Parabolic Arches by Elasto-Plastic Model (탄소성 모델에 의한 포물선 아치의 극한 내하력 평가)

  • The advent or high-strength steel has enabled the arch structures to be relatively light, durable and long-spanned by reducing the cross sectional area. On the other hand, the possibility of collapse may be increased due to the slender members which may cause the stability problems. The limit analysis to estimate the ultimate load is based on the concept of collapse mechanism that forms the plastic zone through the full transverse sections. So, it is not appropriate to apply it directly to the instability analysis of arch structures that are composed with compressive members. The objective of this study is to evaluate the ultimate load carrying capacity of the parabolic arch by using the elasto-plastic finite element model. As the rise to span ratio (h/L) varies from 0.0 to 0.5 with the increment of 0.05, the ultimate load has been calculated fur arch structures subjected to uniformly distributed vertical loads. Also, the disco-elasto-plastic analysis has been carried out to find the duration time until the behavior of arch begins to show the stable state when the estimated ultimate load is applied. It may be noted that the maximum ultimate lead of the parabolic arch occurs at h/L=0.2, and the appropriate ratio can be recommended between 0.2 and 0.3. Moreover, it is shown that the circular arch may be more suitable when the h/L ratio is less than 0.2, however, the parabolic arch can be suggested when the h/L ratio is greater than 0.3. The ultimate load carrying capacity of parabolic arch can be estimated by the well-known formula of kEI/L$^3$where the values of k have been reported in this study. In addition, there is no general tendency to obtain the duration time of arch structures subjected to the ultimate load in order to reach the steady state. Merely, it is observed that the duration time is the shortest when the h/L ratio is 0.1, and the longest when the h/L ratio is 0.2.

  • Polyphenol oxidase (PPO) from Burdock (Arctium lappa L.) was purified and characterized. Purification of polyphenol oxidase was achieved by ammonium sulfate precipitation, Phenyl-sepharose CL-4B hydrophobic chromatography and Sephadex G-100 gel filtration chromatography. The molecular mass of the purified PPO was estimated to be 30 kDa by SDS polyacrylamide gel electrophoresis. In a substrate specificity, maximum activity was achieved with chlorogenic acid, followed by catechol and catechin. Whereas, there was low activity with hydroquinic acid, resorcinol or tyrosine. The optimum pH and temperature for enzyme activity were 7.0 and 35$\circC$ with catechol, respectively. The enzyme was most stable at pH 7.0 while unstable at acidic and alkaline pH. The enzyme was stable when heated to 40$\circC$. But heating at 50$\circC$ for more than 30 min caused 50% loss of activity. Ascorbic acid, L-cystein and $Cu^{2+}$ inhibited the activity of pholyphenol oxidase.

Experimental study of degradation and biodegradability of oxytetracycline antibiotic in aqueous solution using Fenton process

  • The degradation of aqueous oxytetracycline (OTC) from an aqueous solution antibiotic using H2O2/Fe2+ process was studied in one 1 L batch chemical reactor. The extent of OTC degradation (20 mg/L) was investigated from a known initial pH solution, temperature and the type of catalyst (Fe2+, Fe3+) and for various initial concentrations of OTC, H2O2 and Fe2+. The degradation efficiency achieved was found to be very important (90.82% and 90.63%) at initial pH solution of 3 and 4, respectively. However, the type of catalyst and the reaction temperature had a slight impact on the final degradation of OTC. The results showed that the OTC removal increased with increasing initial H2O2 concentration in the range of 70 to 150 mg/L and initial Fe2+ concentrations in the range of 2 to 5 mg/L. The highest degradation efficiency obtained at ambient temperature was 90.95% with initial concentration of OTC of 10 mg/L, H2O2 = 150 mg/L and Fe2+ = 5 mg/L. Moreover, biodegradability improved from 0.04 to 0.36 and chemical oxygen demand degradation was 78.35% after 60 min of treatment. This study proved that Fenton process can be used for pretreatment of wastewater contaminated by OTC before a biological treatment.

Culture characteristics of Leuconostoc mesenteroides WiKim32 in kimchi cabbage juice without supplements and sterilization (비살균 무첨가 배추즙에서 Leuconostoc mesenteroides WiKim32의 배양 특성)

  • This study was performed to develop an economical culture method of lactic acid bacteria (LAB) for kimchi fermentation. Leuconostoc mesenteroides WiKim32 was grown to $1{\times}10^9CFU/mL$ and maintained at 8.88 log CFU/mL for four days by culturing in kimchi cabbage juice (KCJ) without supplements and sterilization. Leuconostoc mesenteroides WiKim32 was cultured in 100 mL of KCJ by inoculating with 0.1% starter culture and adding 100 mL of KCJ everyday by adjusting pH to 5.5 using 1 M NaOH at $20^{\circ}C$ for four days. KCJ was prepared by extraction of kimchi cabbage leaves after washing them with citric acid and ethanol. Adjusting pH over 6.0 was favorable for the growth of LAB in the initial stage, but LAB growth was retarded in the later stage. In contrast, adjusting pH below 5.0 was not beneficial for the growth of LAB; therefore, pH was adjusted to 5.5.

$Hg^{2+}$-promoted Aquation and Chelation of cis-[Co(en)$_2$(L)Cl]$^{2+}$ (L = Amines) Complexes ($Hg^{2+}$에 의한 cis-[Co(en)$_2$(L)Cl]$^{2+}$ (L = 아민류) 착물의 아쿠아화 및 킬레이트화 반응)

  • It has been suggested that Hg$^{2+}$-promoted reaction of a series of cis-[Co(en)$_2$(L)Cl]$^{2+}$ (en = 1,2-diaminoethane) with L = NH$_3$, NH$_2$CH$_3$, glyOC$_2$H$_5$, glyOCH$_3$, dl-alaOC$_2$H$_5$, NH$_2$CH$_2$CONH$_2$, and NH$_2$CH$_2$CN proceeds by dissociative interchange(I$_d$) mechanism from kinetic data, circular dichroism spectra, analyses of products, and the values of m(Grunwald-Winstein plot) using Y (solvent ionizing power) in aqueous solution and in mixed aqueous-organic solvent. It has been found that chloride replacement by water (aquation) for the series with L = NH$_3$ and NH$_2$CH$_3$ and chelation of ligand L to Co(Ⅲ) for the series with L = glyOC$_2$H$_5$, glyOCH$_3$, dl-alaOC$_2$H$_5$, NH$_2$CH$_2$CONH$_2$, and NH$_2$CH$_2$CN occurs, respectively. The rate constants on Hg$^{2+}$-induced reaction of the series except cis-[Co(en)$_2$(NH$_2$CH$_2$CN)Cl]$^{2+}$ were increased with increasing the contents of ethanol in mixed water-ethanol solvents. In mixed water-30${\%}$ organic solvents, the rate constants of the series except cis-[Co(en)$_2$(NH$_2$CH$_2$CN)Cl]$^{2+}$ have also been measured in the order 30${\%}$ 2-propanol-water > 30${\%}$ ethanol-water > water. However, the rate constants of cis-[Co(en)$_2$(NH$_2$CH$_2$CN)Cl]$^{2+}$ were reversed. The rate constants of the series with L= NH$_3$ and NH$_2$CH$_3$ were related to ligand field parameter (${\Delta}$), but those of the series with L = glyOC$_2$H$_5$, glyOCH$_3$, dl-alaOC$_2$H$_5$, NH$_2$CH$_2$CONH$_2$, NH$_2$CH$_2$CN were not. The reaction between the series and Hg2+ in aqueous media containing NO$_3^-$ has been investigated. The results for the reaction do not alter the mechanism, but the rate only was altered.

Fermentation Properties of Rice Added Yogurt Made with Various Lactic Acid Bacteria (유산균주의 종류에 따른 쌀 첨가 요구르트의 발효 특성)

  • The objective of this experiment was to select the best strain of lactic acid bacteria for the manufacture of new type of yogurt with rice powders. Changes in pH, titratable acidity, viable cell counts, viscosity, organic acid contents, carbohydrates during fennentation were monitored and sensory evaluation were examined. The yogurt added with 4% rice or skim milk powders and L. salivarius ssp. salivarius culture did not reach pH 4.5, because the production of acids in this media for the culture was weak. The yogurt added with 4% rice or skim milk powder with L. casei, the pH was low and the titratable acidity was high, and therefore the quality of yogurt after 8 hours from fermentation was not high. The yogurt added with 4% rice or skim milk powders with a mixed culture of B. longum, L. acidophilus, Streptococcus salivarius ssp. thermophilus was considered best for achieving pH 4.5 and titratable acidity of 1.0 % from 8 to 14 hours. The yogurt with a mixed culture had more acetic acid. Galactose was accumulated when L. salivarius ssp. salivarius or the mixed culture were used for fermenting yogurt. In sensory evaluation, the yogurt with the mixed culture received high overall sensory score. From these results, a mixed culture of B. longum, L. acidophilus, Streptococcus salivarius ssp. thermophilus was identified as the best for the manufacture of yogurt added with rice powder.
