The conditions for the extraction of effective substances from the stem of Opuntia ficus-indica were optimized by a response surface methodology. The total extract yield was maximized under the temperature of $97.77^{\circ}C$, at a time duration of 145.82 min and a water to sample ratio 16.59 mL/g. Moreover, the optimum conditions for the extraction of effective substances were as follows: $84.95^{\circ}C$, 156.50 min and a water/sample ratio of 7.46 mL/g for the phenolics content; and $97.11^{\circ}C$, 139.03 min and a water/sample ratio of 10.91 mL/g for the pectin content. The range of optimum extraction conditions in consideration of the physicochemical properties of the extracts were shown to be as $95-100^{\circ}C$ as the extraction temperature, 120-180 min as extraction time and a water to sample ratio of 5-18 mL/g.
교대로 간헐적으로 포기되는 2개의 부직포 여과막 생물반응조로 폐수를 처리할 때 반응조의 MLSS 농도와 유입수의 C/N 비가 질소 제거효율에 미치는 영향을 파악하기 위하여 MLSS 농도를 약 5,500 mg/L, 10,000 mg/L 및 15,000 mg/L로 유지하면서 $NH_4Cl$을 첨가하여 유입수의 TCOD/TKN 비를 5, 4, 3 및 2로 감소시켰다. 유입수는 음식물 쓰레기 침출수를 COD농도가 약 300 mg/L되도록 희석시킨 것이었다. 실험 결과, 반응조의 F/M 비는 0.112 g COD/g MLSS-day 이하, COD 제거효율은 95% 이상, 그리고 미생물 성장계수($Y_{obs}$) 값은 평균 0.283 g MLSS/g COD로 나타났다. 질산화 효율은 MLSS 농도가 5,500 mg/L이고 유입수의 TCOD/TKN 비가 2인 경우의 90.5%를 제외하고 모두 96% 이상이었다. 탈질효율은 유입수의 TCOD/TKN 비가 감소할수록 악화되었다. MLSS 농도가 5,500 mg/L인 경우에 비하여 10,000 mg/L인 경우에 탈질효율이 평균 10.7% 더 높아, MLSS 농도가 1,000 mg/L 증가함에 따라 평균 2.66 mg N/L의 율로 탈질율이 증가하였다. 그러나, MLSS 농도가 15,000 mg/L로 유지된 경우에는 5,500 mg/L인 경우에 비하여 탈질효율이 평균 4.6%만 더 높아 MLSS 농도가 1,000 mg/L 증가함에 따라 평균 0.75 mg N/L의 율로 탈질율이 증가하였다. 따라서 MLSS 농도와 내생 탈질율 간에 비례관계가 성립되지 않았다. 알칼리도 소모량은 유입수의 TCOD/TKN 비가 5인 경우에 제거된 T-N 1 mg당 평균 3.36 mg으로서 이론값인 3.57 mg에 가까웠으나 유입수의 TCOD/TKN 비가 감소함에 따라 증가하는 경향을 나타내었다.
하악골 성장양상이 진단과 치료, 치료후 예후에 특히 중요한 III급 부정교합자에 있어 그 성장양상을 예측하는데 도움을 주고자 사춘기 전으로 생각되는 남자 10-12세군(G1군)과 악골성장이 거의 끝난 것으로 생각되는 성인으로 남자 20세 이상군(G2군)에 대하여 각각 이부비율에 따라 다시 3군으로 분류하고 이부형태와 두개안면형태의 연관성을 연구 한 결과 다음과 같은 결론을 얻었다. 1. 평균 이부비율에 있어 G2군이 G1군에 비해 통계학적으로 유의성 있게 크게 나타났다(p<0.05). 2. G1군과 G2군 모두 이부비율과 두개저 계측항목과는 상관성이 없었다(P>0.05). 3. 상하악골의 수평적 관계 계측항목에서 G1군과 G2군 모두 이부비율과 상악골 계측항목과는 상관성이 없었다(p>0.05). 또한 G1군과 G2군 모두 L, A, S군간 하악의 전후적 위치는 뚜렷한 차이가 없고, 단지 G1군의 Y-axis angle에서 L군이 S군에 비해 크고 G2군은 Pog to Na perp.에서 S군이 L군에 비해 통계학적으로 유의성 있게 크게 나타났다(p<0.05). 4. 하악골의 형태 계측항목에서 gonial angle은 G1군과 G2군 모두 이부비율이 클 수록 수치적으로 증가하나 통계학적인 유의성은 없었다(p>0.05). Lower genial angle과 chin angle은 G1군에서 L군과 A군이 S군보다 통계학적으로 유의성 있게 크게 나타났다(p<0.05). 5. 안면의 수직적 관계 계측항목에서 G1군은 L군이 S군에 비해 전안면고경, 전하안면고경이 통계학적으로 유의성 있게 크며(p<0.05) 하악골이 하방성장한 경향을 보여주었다. 6. 치아의 위치 및 경사도 계측항목에서 G1군은 하방성장을 많이 한 L군이 S군에 비해 상하악 치아들이 정출되어 있는 경향을 보여주었지만 G2군은 단지 하악치아 만이 그러한 경향이 있었다. 7. 이부비율과 다른 계측항목과의 상관분석에서 G1군은 chin angle, PP/MP angle, ANS-Me등 많은 계측항목에서 G2군은 단지 MP-LIT와 MP-LMMC에서 통계학적으로 유의성 있는 상관성을 보였다(p<0.05, p<0.01). 이상을 종합하면 어린 남자군인 G1군에서는 이부비율이 큰 군(L군)과 작은 군(S군)사이에 수평적 골격관계는 차이가 없으나 수직적 골격관계는 L군이 S군에 비해 하악골이 하방으로 많이 성장한 경향을 보여주었다. 하지만 성인 남자군인 G2군에서는 이부비율에 따른 안면골격의 뚜렷한 형태적 차이를 보여주지 않았다.
무화과를 이용한 기능성 음료의 제조를 위하여 반응표면 분석법에 의하여 무화과의 추출조건에 따른 무화과 유용성분의 추출특성을 모니터링하였다. 수율의 최대값은 66.46%이었으며, 이때의 추출조건은 시료에 대한 용매비 22.08 mL/g, 추출온도 $90.59^{\circ}C$ 및 추출시간 148.04 min이었다. 총페놀성 화합물 함량은 시료에 대한 용매비 17.87 mL/g, 추출온도 $98.82^{\circ}C$ 및 추출시간 130.80 min에서 최대 값 $121.31{\mu}g/mL$을 나타내었으며, 전자공여능은 시료에 대한 용매비 18.13 mL/g, 추출온도 $98.81^{\circ}C$ 및 추출시간 127.95 min일때 최대값을 나타내었다. 무화과 추출물의 단백분해효소의 활성은 시료에 대한 용매비 17.45mL/g, 추출온도 $99.01^{\circ}C$ 및 추출시간 131.43 min일 대 최대값을 나타내었으며, 환원당 함량은 시료에 대한 용매비 22.66 mL/g, 추출온도 $86.30^{\circ}C$ 및 추출시간 153.59 min에서 최대값이 19.14 mg/mL이었다. 무화과 유용성분의 추출을 극대화할 수 있는 최적 추출조건의 범위는 시료에 대한 용매비 $17{\sim}25$ mL/g, 추출온도 $80{\sim}100^{\circ}C$ 및 추출시간 $100{\sim}170$ min으로 예측되었으며, 실제 실험을 통하여 그 신뢰성을 검증하였다.
Objective: Purpose was to determine the effects of grazing system and floor type on concentrations of blood metabolites, activity of creatine kinase, body weight and carcass characteristics of castrated Nguni goats. Methods: Forty eight, 7 month old goats were randomly allocated to herding and tethering treatments from 0800 to 1300 hours and accommodated on slatted and earth floors daily. Blood samples were collected by jugular venipuncture every fifteenth day for metabolite analysis. Slaughter was done at a commercial abattoir following 5 months of monitoring. Results: Tethered goats had significantly higher concentrations of urea (5.19 mmol/L) (p<0.001), creatinine ($55.87{\mu}mol/L$) (p<0.05), total protein (64.60 g/L) (p<0.01), and globulin (49.79 g/L) (p<0.001), whereas herded goats had higher glucose (3.38 mmol/L) (p<0.001), albumin (15.33 g/L) (p<0.05), albumin/globulin ratio (0.34) (p<0.01), and body weight (24.87 kg) (p<0.001). Slatted floors caused higher (p<0.01) albumin at 15.37 g/L. The interaction of grazing system and floor type affected creatinine, total protein, globulin at (p<0.01) and albumen/globulin ratio at (p<0.01). The least creatinine concentration and albumin/globulin ratio was in herded and tethered goats that were accommodated on earth floors, respectively. The highest total protein and globulin concentrations were in serum of tethered goats that were accommodated on earth floors. The highest (p<0.05) dressing percentage (45.26%) was in herded goats accommodated on slatted floors. Conclusion: Herding of goats lowered globulin concentration, improved estimated feed intake, blood glucose and albumin concentrations, albumin globulin ratio, increased body weights and weight related carcass characteristics. Floor type had very little effects on metabolites where earth floors only reduced albumin concentration. Tethering and housing goats on earth floors resulted in double stress that increased chronic infections.
Pinosylvin, a stilbenoid phytoalexin, is a health ingredient to be extracted from pine leaves. In this study, ultrasonication-assisted extraction process for manufacturing extracts with high content of pinosylvin from pine leaves was investigated. As process and system variables, ultrasonic power, sonication time and solvent ratio were selected. According to the experimental results, the effective yield of pinosylvin increased with the increase of ultrasonic power and sonication time and the decrease of solvent ratio. When the ultrasonic power of 2400 W/L was added to the solution of pulverized pine leaves of 8 g per 1 L of a solvent for 10 minutes, yield of extracts and purity, effective yield and concentration ratio of pinosylvin were 0.3166 g/g, 0.7247 mg/g, 0.2294 mg/g and 23.0, respectively.
본 연구는 Lentinus edodes의 액체배양을 통하여 물리 ${\cdot}$ 화학적인 방법을 이용, 고농도의 탄수화물 복합효소를 얻어 hemicellulose와의 반응을 거쳐 고수율의 아라비 녹실란을 얻는데 그 목적이 있다. 온도 $24^{\circ}C$, pH 5.5. working volume 100mL, 접종비 5%. 200rpm이며 가장 적합한 C-source 로서는 glucose가 선택되었으며 N - source로서는 yeast extract가 선택되었다.
수산물 수입이 전면 개방됨에 따라 앞으로 외국으로부터의 수산물 수입이 급증할 것으로 여겨지는 시기에 외국산 복어 가운데 멕시코산 황소눈복어에 대한 독성 및 정미성분에 관한 자료를 얻기 위하여 mouse assay법에 의하여 독성을 조사하였고, 아울러 정미성분인 ATP관련물질과 유리아미노산도 분석하였다. 생식선은 약한 독성 (무독 내지 $25{\mu}/g$)을 나타내었으나, 그 이외의 부위 및 가식부인 근육 및 껍질의 독성검사 결과는 모두 $10{\mu}/g$이하로서 무독한 것으로 나타났다. 핵산관련물질 가운데 IMP 및 ADP가 $5.61{\mu}mol/g$ 및 $2.7{\mu}mol/g$으로 많았고, 전체 ATP 관련물질의 $41.4{\%}$를 차지하였다. 유리아미노산은 모두 19종이 검출되었으며, 총량은 1,648.2mg/100g이었다. 유리아미노산 중 함량이 많은 아미노산은 L-glycine, L-alanine, L-anserine, L-threonine 및 L-valine으로 각각 233.5 mg/100 g, 169.0 mg/100 g, 149.1 mg/100 g, 135.7mg/100 g 및 132.3 mg/100g이었고, 또한 이들 아미노산의 전체 유리아미노산에 대한 중량 비율은 각각 $14.28{\%},\;10.33{\%],\;9.12{\%},\;8.30{\%}$ 및 $8.09{\%}$로서 이들 5종류의 아미노산이 전체 유리아미노산의 $50.12{\%}$를 차지하였다. 또한 taurine의 경우는 119.3mg/100 g, L-histidine의 경우는 14,7mg/100 g이었다.
The goal of this study was to evaluate micro-bubble concentration ($C_{air}$) in water by air/water ratio (A/W ratio) with a micro-bubble generating pump. The estimation of micro-bubble concentration is based on the balance of inlet/outlet air and water flow rate. On net A/W ratio to be generated micro-bubble, we found that the obtained the $C_{air}$ are shown as a function of discharge pressure ($P_g$) of the micro-bubble generating pump. The correlation of the $C_{air}$ and the $P_g$ ($C_{air}=3.261P_g-1.754$) was adequately described by the least square methods with a high correlation coefficient (r = 0.9459) and calculated values fit the experimental data quite well. The $C_{air}$ was lower than theoretical dissolved air concentration ($C_{aq}$) calculated by Henry's law. The $C_{air}$ for being operated the micro-bubble generating pump was 6.75 - 39.53 mL/L, however, we found that the optimum of the $C_{air}$ to generate micro-bubble was the range from 10 to 12 mL/L.
This study was investigated the effects of exercise and supplementation of L-carnitine and antioxidants on hepatic mitochondrial function, especially oxidative phosphorylation (OXPHOS). Isolated hepatic mitochondria from 4 rat groups were functionally tested by an analysis of respiration and the coupling of this process to ATP synthesis in the presence of ADP. Four groups were non-trained, non-supplemented group (NTNS), non-trained, supplemented group (NTS), long term-trained, non-supplemented group (LTNS) , and long term-trained, supplemented group (LTS). The trained rats run on a treadmill (grade 10°,20 m/min) for 60min/day for 8 weeks. The supplemented rats were treated with L-carnitine (0.5% diet), vitamin E(0.5mg/g BW), vitamin C (0.5mg/g BW) and melatonin (1 $\mu$ g/g BW) for 8 weeks. There were exercise effects on improving mitochondrial OXPHOS. Within non-supplemented groups, exercised rats resulted in a significant decrease in state 4 oxygen consumption, which increased the respiratory control (RC) ratio and ADP : O (P/O) ratio. There were supplementation effects on improving mitochondrial OXPHOS, too. Within non-exercised rats, supplemented rats resulted in a significant decrease in state 4 oxygon consumption. which increased the RC ratio and P/O ratio. There were additive effects of exercise and supplementation on OXPHOS. Within supplemented rats, exercise resulted in an increase in RC ratio. Significant effects of exercise-supplement interaction on improving OXPHOS were identified. It suggests that exercise and supplementation of L-carnitine and antioxidants might improve more efficiently the impaired OXPHOS efficiency in mitochondrial dysfunction that recognized as is an important cause of degenerative diseases.
