The reaction steps involved in the bioconversion of a chemically synthesized precursor, $D,L-2-amino-{\Delta}^2-thiazoline-4-carboxylic$ acid (D,L-ATC), to L-cysteine and the properties of the involved enzymes were investigated. It was found that the conversion consisted of two steps, i. e., D,L-ATC to S-carbamyl-L-cysteine (S-C-L-cysteine) and S-C-L-cysteine to L-cysteine, and the S-C-L-cysteine was an intermediate between them. While the enzymes involved in the reactions were induced by the addition of D,L-ATC as an inducer, S-C-L-cysteine induced only the enzyme involved in the latter step. The conversion of S-C-L-cysteine to L-cysteine could be also carried out in the presence of hydroxylamine and its rate was much faster than that by the corresponding enzyme. On the other hand, L-cysteine (or L-cystine) was decomposed to evolve $H_2S$ by the enzyme considered to be a kind of desulfhydrase. However, hydroxylamine was a perfect inhibitor for this enzyme.
This study was conducted to investigate the production of lipid, and removal of nitrogen and phosphorus from swine wastewater by Botryococcus braunii UTEX 572 in a tubular bioreactor. The rate of dry cell weight increase of B. braunii was highest at 20.1mg/l/din a modified Chu 13 medium at $25^{\circ}C$. Under the above conditions, the rate of lipid content increase was also highest at 6.1mg/l/d. The lipid content of B. braunii on a dry weight basis ranged from 30.5 to 34.1% with an average value of 32.3%. When B. braunii was cultured in a secondary-treated swine wastewater diluted to 50% with tap water, the rate of dry cell weight increase was 18.6mg/l/d and the rate of lipid content increase was 6.0mg/l/d. The lipid content ranged from 30.3 to 34.2%. No significant difference was observed between lipid content and growth conditions. The removal rates of nitrogen and phosphorus in swine wastewater were 43.9% and 41.7%, respectively, after 14 days of incubation.
This study investigates the effect of sodium bicarbonate ($NaHCO_3$) on growth of S.dimorphus. $NaHCO_3$ concentration was varied from 0 to 2 g-C/L. As a result, the increase in concentration of $NaHCO_3$ up to 1.5 g-C/L increased dry weight of algae. The highest specific growth rate of S. dimorphus was $0.36day^{-1}$ which was obtained at concentration of 0.5 g-C/L $NaHCO_3$. pH showed a large variation range at the concentrations lower than 0.5 g-C/L $NaHCO_3$ whereas inorganic carbon, nitrate and phosphorus removal rates were almost same at the concentrations higher than 0.5 g-C/L $NaHCO_3$ (0.75, 1, 1.25, 1.5, 2 g-C/L $NaHCO_3$). Their average inorganic carbon, nitrate and phosphorus removal rate were 70 mg-C/L/d, 11.3 mg-N/L/d, and 1.6 mg-P/L/d, respectively. Thus, $NaHCO_3$ didn't effect on inorganic carbon, nitrate and phosphorus removal rate of S. dimorphus.
본 연구는 미나리와 울금의 방사선 피폭이 어떠한 영향이 미치는지 알아 보고자 하였다. 실험동물은 ICR계 5주령 25~30g 수컷 50 마리를 마우스를 사용하여, 5 마리를 하나의 군으로 3 Gy 대조군(A), 5 Gy대조군(B), 3 Gy 미나리 전처치(C), 5 Gy 미나리 전처치(D), 3 Gy 미나리 후처치(E), 5 Gy 미나리 후처치(F), 3 Gy 울금 전처치(G), 5 Gy 울금 전처치(H), 3 Gy 울금 후처치(I), 5 Gy 울금 후처치(J)으로 설정하였고, 대조군(A, B)은 방사선 조사 후 7 일 동안 생리식염수 500mg/kg/day)를 마우스에 경구투여 하였고, 후처치 대조군(E, F, I, J)도 방사선조사 후 7일 동안 미나리(500mg/kg/day)와 울금(500mg/kg/day)을 각각 마우스에 경구투여 하였다. 또 이와 같은 방법으로 전처치 대조군(C, D, G, H)은 미나리(500mg/kg/day)와 울금 (500mg/kg/day)을 각각 마우스에 7 일 동안 경구투여 후 방사선 조사를 하였다. 그 결과 3 Gy 대조군에서 백혈구 평균수치(마우스 5 마리)는 평균 200K/${\mu}L$, 3 Gy 미나리 전처치군에서는 평균 6,46K/${\mu}L$로 정상수치(1.8-10.7 K/${\mu}L$)로 되었다. 3 Gy 울금 전처치의 백혈구 평균수치는 23.33K/${\mu}L$로 정상수치는 아니지만, 200K/${\mu}L$에 비하여 정상수치에 가까워졌음을 알 수가 있다. 3 Gy 대조군에서 림프구의 평균 수치는 118.87K/${\mu}L$로, 3 Gy 울금 전처치군은 1.51 K/${\mu}L$로 림프구 정상수치(0.9-9.3K/${\mu}L$)로 나타내었다. 혈소판에서는 3 Gy 대조군의 평균 수치는 4000K/${\mu}L$, 3 Gy 미나리전처치군은 1107 K/${\mu}L$, 3 Gy 울금 전처치군은 2759.6 K/${\mu}L$로 혈소판정상수치(592-2972 K/${\mu}L$)로 나타내었다. 위의 결과로 미나리와 울금이 방사선 피폭에 대한 예방적 차원에서 방어효과가 있는 것으로 사료되었고, 동물실험에서는 혈구수치가 정상수치로 돌아오는 것을 알 수가 있었다. 이 실험을 바탕으로 향 후 다양한 연구가 선행되어야 할 것으로 사료되는 바이다.
Poly(D,L-lacic acid)(PLA) microshperes containing loazepam were prepared by a solvent-emulsion evaporation method and their release patterns were investigated in vitro. Various batches of microspheres with different size and drug content were obtained by changing the ratio of lorazepam to PLA, PLA concentration in the dispersed phase and stirring rate. Rod-like lorazepam crystals on microsphere surface, which were released rapidly and could act as a loading dose, were observed with increasing drug content. The release rate was increased with increase in drug contents and decrease in the molecular weight of PLA. The release rate of lorazepam for long-acting injectable delivery system in vitro, which would aid in Predicting in vivo release Profile, could be controlled by properly optimizing various factors affecting characteristics of microspheres.
This study was aimed to decrease AOX(Adsorbed Organic Halide Compounds) by applying bleaching methods of A(HNO$_3$ + NaNO$_2$) and $D^{(H/L)}$(dual pH chlorine dioxide bleaching) to conventional OCEDED bleaching stages. And so we investigate the effects of NSA treatment and the influence of production rate of ion species in chlorine dioxide and pulp mixtures according to various pH as well as dual pH on pulp bleaching. Finally the effects of AOC_DED^{(H/L)}ED^{(H/L)}$ bleaching stage were investigated by measuring AOX, brightness, kappa number and viscosity of pulps. A stage was treated by using 4% $HNO_3$ and 0.05~2.0% $NaNO_2$ on pulp and $D^{(H/L)}$ stage was like that pulp mixed with chlorine dioxide solution was adjusted at pH 7.0 and reacted in pulp cosistency 3%, S~20min, $70^{\circ}C$, and then successively for 160~175min at pH 4.0. It was found that suitable $NaNO_2$ addition rate was at 0.6% where the brigtness of pulp was most higher and pulp viscosity was not decreased much. Chlorate ($CIO_3$) was decreased according to pH increase but chlorite($CIO_2$) was highly decreased according to pH increase. And chloride was slightly increased with pH increase. The applying of A and D^{(H/L)}$ bleaching stages to some various multiple bleaching stages ($AOC_DED^{(H/L)}/EopD^{(H/L)} etc.$) had good results that brightness was increased more 2~3% ISO than conventional method(OCEDEopD), but viscosity was dropped by only 1~2cps.
Ettlia sp. YC001, a highly settleable and productive microalga, was shown to be effective in removing nutrients and capturing suspended solids from eutrophic pond water. The optimum conditions for the Ettlia sp. YC001 cultivation were investigated using water from a landscape pond. The pond water was supplemented with different N : P ratios by weight, and the biomass production and nutrient removal compared in batch cultures. The maximum removal rate of N and P was with an N : P ratio of 16 : 1. Plus, the turbidity dropped to near zero within 4 days. Meanwhile, chemostat cultivation showed that the biomass productivity and nutrient removal rate increased when increasing the dilution rate, where a dilution rate of $0.9d^{-1}$ showed the highest N and P removal rate at $32.4mg\;L^{-1}\;d^{-1}$ and $1.83mg\;L^{-1}\;d^{-1}$, respectively, and highest biomass and lipid productivity at $0.432g\;L^{-1}\;d^{-1}$ and $67.8mg\;L^{-1}\;d^{-1}$, respectively. The turbidity was also reduced by 98% in the chemostat cultivation. Moreover, auto-flocculation and pH were closely connected to the turbidity removal. As a result, this study identified the optimal N : P ratio for small pond water treatment using an Ettlia sp. YC001, while also establishing the optimal conditions for nutrient removal, turbidity reduction, and biomass production.

For the first time, the characteristics of solid circulation rate ($G_S$) were investigated in a three-phase (gas-liquid-solid) viscous circulating fluidized bed (TPCFB). The solid circulation rate was controlled separately by adjusting the experimental apparatus as well as operating variables. Effects of primary and secondary liquid velocities ($U_{L1}$ and $U_{L2}$), gas velocity ($U_G$), particle size ($d_p$), height of particles piled up in the solid recycle device (h), and viscosity of continuous liquid media (${\mu}_L$) on the value of $G_S$ were determined. The experimental results showed that the value of $G_S$ increased with increases in the values of $U_{L1}$, $U_{L2}$, h and ${\mu}_L$, while it decreased with increasing $U_G$ and $d_p$ in TPCFBs with viscous liquid media. The values of $G_S$ were well correlated in terms of dimensionless groups within this experimental conditions.
Rhodobacter sphaeroides 균주가 생산하는 ${\delta}-aminolevulinic\;acid(ALA)$의 생산성에 관하여 in vivo, in vitro 상에서 기질 및 관련 화합물을 이용하여 검토하였다. $C_5\;ALA$ 생합성계에 의한 ALA yield 대비의 $C_4$ 생합성계에 의한 비율은 in vive 상에서 0.78인 반면, in vitro 상에서의 비율은 1.37이었다.$C_4\;C_5$ 각 계의 기질 첨가 배양에 의한 cell-free system의 $C_4,\;C_5$ 계의 발현도는 미첨가 배양에 의한 system과 비교하여 각각 1.35, 1.52로 증가하였으나, 증가한 계에 대한 상대계의 발현도가 억제되어, $C_4$ 와 $C_5$ 계가 각각 0.91, 0.83으로 나타났다. ${\gamma}-Glutamyl\;derivatives$의 세포내 uptake rates는 L-glutamic acid를 기준으로 비교해서 D-glutamic acid, 0.55: D-glutamine, 0.5: L-glutamine, 0.4: ${\gamma}-L-glutamyl\;ethylester$, 0.3: GSH 및 Glu-pNA, 0의 순서를 보였다. Uptake rate와 관계없이 in vivo 상에서 L-과 D-glutamine이 L-, D-glutamic acid보다 균체 외 ALA의 생산에 있어서 각각 높은 yield의 효과를 보였다.
We investigated the effects of dissolved oxygen (D.O.) and fructose (C-source) on cell growth and biosynthesis of cyclosporin A (CyA) produced as a secondary metabolite by a wild-type filamentous fungus, Tolypocladium inflatum. This was performed by controlling the level of D.O. and the residual C-source, as required, through adjustment of medium flow rate, medium concentration and agitation rate in fed-batch cultures. CyA production was furned out to be maximal, when D.O. level was controlled around 10% saturated D.O. and concentration of the C-source was maintained sufficiently low (below 2 g/L) not to cause carbon catabolite repression. Under this culture condition, we obtained the highest values of CyA concentration (507.14 mg/L), Qp (2.11 mg CyA/L/hr), $Y_x/s$ (0.49 g DCW/g fructose), $Y_p/s$<(22.56 mg CyA/g fructose), and YTEX>$_p/x$ (48.31 mg CyA/g DCW), but relatively lower values of cell concentration (11.98 g DCW/L) and cell productivity (0.043 g DCW/L/hr), in comparison with other parallel fed-batch fermentation conditions. These results implied that, in the carbon-limited culture with 10% saturated D.O. level, the producer microorganism utilized the C-source more efficiently for secondary metabolism.
