Mineral Contents of Brown and Milled Rice (현미와 백미의 품종별 무기질 함량)

  • The objectives of this study were to analyze mineral contents of brown and milled rice of five varieties (Kwangan, Daean, Daejin, Sura, Hwaseong) and to compare those with Korean recommended dietary allowance. The ash contents of brown rice ranged from 1.28 to 1.45% and those of milled rice ranged from 0.51 to 0.62% The mineral contents of brown rices were P, 270.8∼327.2 mg/100 g; K, 216.0∼ 249.0 mg/100 g: Mg, 102.0∼111.0 mg/100 g, Ca, 11.8∼13.2 mg/100 g. The major minerals of milled rices were P, 125.3∼153.2 mg/100 g; K, 96.5∼118.6 mg/100g; Mg, 34.7∼40.0 mg/100 g; Ca, 6.0∼9.4 mg/100 g. Contents of phosphorus from average daily rice consumption (215.9 g) correspond to 90.7% of Korean RDA of phosphorus in brown rice while 42.3% in milled rice.

Characteristics of Vibrio mimicus K-1 Isolated from Coastal Sea Water (연안 해수에서 분리된 Vibrio mimicus K-1의 특성)

  • An environmental study was done to examine the distribution of Vibrio mimicus in aquatic environments of Kwangan and Minrak beach, Pusan, Korea. Moreover, both bacteriological characteristics and lethal effects of isolated V. mimicus were observed. Sea water samples were collected monthly from January to September, 1993, and quantitatively analyzed for V. mimicus. This organism was isolated from April(water temperature was $16.3^{\circ}C$), whereas it was not isolated when the water temperature fell below $15^{\circ}C$. V. mimicus counts were not remarkably high, however this study at least describes the distribution and occurrence of the possible highest density in aquatic environments of this region. Among the confirmed V. mimicus strains, the author chose the strongest antibiotic resistant bacterium and named it V. mimicus K-1. This strain has antibiotic resistance to colistin, erythromycin, tetracycline, and penicillin, and most isolates had a higher level of antibiotic resistance than V. mimicus ATCC 33653. The optimum growth for V. mimicus K-1 was observed at $37^{\circ}C$, pH 7.5, and $1\%$ NaCl, respectively. This organism was mostly inactivated by Ultra Violet irradiation (30W, $50^{\circ}C$) for 70 seconds and death lethality increased in proportion to treatment temperature ($D_{50}=5.7min,\;D_{60}=\;2.1min,\;and\;D_{70}=0.7min$).

Increase of isoflavones in soybean callus by Agrobacterium-mediated transformation

  • Plant secondary metabolites have always been a focus of study due to their important roles in human medicine and nutrition. We transferred the isoflavone synthase (IFS) gene into soybean [Glycine max (L.) Merr.] using the Agrobacterium-mediated transformation method in an attempt to produce transformed soybean plants which produced increased levels of the secondary metabolite, isoflavone. Although the trial to produce transgenic plant failed due to unestablished hygromycin selection, transformed callus cell lines were obtained. The induction rate and degree of callus were similar among the three cultivars tested, but light illumination positively influenced the frequency of callus formation, resulting in a callus induction rate of 74% for Kwangan, 67% for Sojin, and 73% for Duyou. Following seven to eight subcultures on selection media, the isoflavone content of the transformed callus lines were analyzed by high-performance liquid chromatography. The total amount of isoflavone in the transformed callus cell lines was three- to sixfold higher than that in control callus or seeds. Given the many positive effects of isoflavone on human health, it may be possible to adapt our transformed callus lines for industrialization through an alternative cell culture system to produce high concentrations of isoflavones.

Maturity Grouping of Korean Soybean Cultivars and Character Relationships According to the Planting Date

  • This study was carried out to classify Korean soybean varieties base on maturity group (MG) and to find character relationships according to planting date for high quality soybean seed production adapted to early season cultivation environment of Miryang. Results of maturity grouping of Korean soybean varieties showed that Keunol (3 cultivars), belonged to Group 0; Seonnok and Danmi in Group II, Shinrok in Group III, Seonyu (17 cultivars), in Group IV, Taekwang (44 cultivars) in Group V, Daewon (25 cultivars) in Group VI, and Kwangdu and Keumdu in Group VII. Agronomic characteristics of 100 soybean varieties were compared based on MG, cultivation year and seeding date. Soybean varieties belonging to the MG $VI{\sim}VII$ showed longer days to flowering and growth period, high lodging density and higher yield. Seed quality analysis revealed that as maturity was delayed, seed weight becomes heavier while seed cracks become abundant. In addition, occurrence of purple seed and phomopsis were higher in MG $0{\sim}III$. Protein content was higher in MG $0{\sim}III$, and isoflavone content was higher as maturity was delayed. On the other hand, lipid content was generally similar across MGs. Correlation analysis of major agronomic characters showed positive relationships between days to flowering and growth days, seed weight and lodging in MG $IV{\sim}V$, seed crack and growth days in MG $0{\sim}III$, seed crack and days to flowering in MG $IV{\sim}V$ and MG $VI{\sim}VII$, seed crack and lodging in MG $IV{\sim}V$ and MG $VI{\sim}VII$, seed crack and seed weight in MG $IV{\sim}V$ and MG $VI{\sim}VII$, purple seed and growth days in MG $IV{\sim}V$, purple seed and seed weight in MG $VI{\sim}VII$, phomopsis and growth days in MG $IV{\sim}V$ and MG $VI{\sim}VII$, and phomopsis and purple seed in MG $IV{\sim}V$ and MG $VI{\sim}VII$. In contrast, a negative relationship was observed between seed weight and lodging in MG $0{\sim}III$. Correlating yield and major characters revealed negative relationships between days to flowering and growth days in MG $0{\sim}III$ and MG $IV{\sim}V$, whereas positive relationships were obtained on MG $VI{\sim}VII$ seeded on April 30. Lodging, seed weight and seed crack were all negatively correlated with yield in the MG $IV{\sim}V$ and MG $VI{\sim}VII$. Soybean cultivars identified as adaptable to early season planting for production of high quality soy curd and fermented soybean paste were Seonyu, Kwangdu, and Soho while those suited for the manufacture of soybean sprouts were Sobaeknanul, Kwangan, Sowon, and Bosuk. Geomjeong 2 chosen as best for mixing with rice.

Effect of γ-Ray Mutagen Treatment on Botanical Characteristics of Local Small Grain Soybean (Glycine max. L) Lines (돌연변이원(突然變異源) γ-선(線) 처리(處理)가 재래종(在來種) 소립(小粒)콩의 식물학적(植物學的) 특성(特性)에 미치는 영향(影響))

  • This experiment was carried out to obtain a basic information for new variety development. The local soybean (Glycine max. L) lines used were endowed from Chungnam Agricultural Research and Extension Services. These lines were planted at upland farm of Chungnam Nat'l Univ., College of Agri. after treatment with mutagen $\gamma$-ray. All characteristics measured were decreased due to the $\gamma$-ray treatment than check except the branches of main stem of the Hannam and plant height of the Sobak. Especially the Sobak and the Eunha among these lines were shown very sensitivly than others treated with $\gamma$-ray of 15Krad. Of characteristics surveyed the nodes per plant was very sensitive to $\gamma$-ray, while the grain weight per plant, the pods per plant and the branches per plant were gradually decreased to further radioactivity about $\gamma$-ray treatment. In the Pungsan and the Sobak lines there were not shown significant between major characteristics at treatments including check. Other lines were also not shown positive correlation among characteristics except between stem diameter and branches(0.64*), grain weight and plant height(0.77**) at check of the Eunha, between stem nodes and stem diameter(0.65*) at check of the Kwangan and between number of pods and stem diameter(0.70*) at check of the Danyeop. From result this experiment, some of lines used were to be appeared characters of interest, for example, such as the short stem and the early flowering. Therefore these lines were needed more advanced generations for stability and performance.

DISTRIBUTION OF SOME CHEMICAL POLLUTANTS IN SUYEONG BAY (수영만 인근해수의 오탁분포에 대하여)

  • In order to study the water pollution in Suyeong Bay, Busan, some chemical constituents were determined at 25 stations in the neap tides on 9 Aug. 1977 and spring tides on 30 Aug. 1977. Range and mean values of the constituents in the spring tides are as follows: $pH\;6.54\~8.06,\;7.54;$ electrical conductivity $0.413\~0.481\times10^5\;\mu\mho/cm,\;0.467\times10^5\;\mu\mho/cm;\;transparency\;0.2\~5.5m,\;2.2m;$ turbidity $1\~60ppm$, 14ppm, chlorosity $15.20\~18.11g/\ell,\;17.67g/\ell;$ fluoride ion $0.94\~1.03ppm$, 0.99ppm; dissolved oxygen $0.17\~7.60ppm$, 4.77ppm; sulfide $0\~0.46ppm$, 0.07ppm; chemical oxygen demand $1.20\~40.74ppm$, 6.11ppm; ammonia-nitrogen $0.060\~0.520ppm$, 0.180ppm; nitrite-nitrogen $0.001\~0.026ppm$, 0.009ppm; nitrate-nitrogen $0\~0.037ppm$, 0.014ppm; phosphate-phosphorus $0.002\~0.261ppm$, 0.050ppm; n-Hexane soluble $0.5\~5.4ppm$, 2.1ppm ; iron $1.0\~104.11\;ppb$, 24.15ppb ; copper $0\~27.45ppb$, 4.19ppb; lead $0\~2.50ppb$, 0.92ppb; zinc $0\~5.15ppb$, 1.47ppb ; cadmium $0\~0.26ppb$, 0.04ppb; and mercury $0.05\~0.37ppb$, 0.11ppb respectively. The variations of the contents of the chemical constituents in the spring tides were larger than in the neap tides. The contents of COD, sulfide, nutrient salts and heavy metals were the highest in the estuary of Suyeong River, and decreased in order of off Kwangan-Ri region, outer Bay and off Haeun-Dae region. The water quality in Suyeong Bay was particularly shown that the concentrations of COO, iron, copper and mercury were higher than those of other coastal aseas and deficiency in dissolved oxygen was observed in some parte of Suyeong Bay. In consideration of the relationship between the chlorosity and the concentrations of nutrient salts, COD and total heavy metals, water pollution of this area is considered due to the inflow of Suyeong River which was extremely polluted by sewage and industrial wastewaters.
