배지에 따른 Genus Monascus의 증식도와 색소함량 (Growth and Pigment Formation of Genus Monascus on Medium Compositions)

  • Genus Monascus(M. pilosus and M purpureus)의 배지(Lin, SP, YM YE, GMIN, SMO, MY, GY, Nishikawa, sucrose, stock culture, Mizutani, modified Lit Toya 및 쌀배지)에 따른 균의 증식도와 색소생성량을 조사하였다. M. pilosus 및 M purpureus의 증식도가 높은 배지는 Mizutani 배지와 Lin 배지 였으며 3$0^{\circ}C$에서 10일간 배양하였을 때 생균체 무게는 각각 24.5~26.9 g/100 mL 및 15.9~17.2 g/100 mL이였고 M. pilosus의 성장률이 높았다. 색소생성량이 높은 배지는 두 균주 다같이 Lin 및 Mizutani 배지였으며 각각 흡광도는 1.2~1.6 및 0.8~l.0이었다. 균주 중에서는 M. pilosus에서 높았다.

  • Eubacterium is one of the predominant bacteria in the human large intestine. currently ES (Eubacterium Selective) medium developed by T. Mitsuoka is commonly used as a selective medium. neomycin sulfate which is one of the selective agents of ES medium inhibited about 50% of the growth of Eubacterium isolated, whereas malidixic acid inhibited only 5% while inhibiting other intestinal bacteria. NES medium which replaced neomycin with nalidixic acid in the ES medium was designed and shown to be better for the isolation and enumeration of Eubacterium sp. than ES medium.

돼지 난관액과 Oviductal Conditioned Medium 이 다정자침입과 체외배발달에 미치는 영향 (Effect of Oviductal Fluid and Oviductal Conditioned Medium on Polyspermy and In Vitro Development of Porcine Oocytes)

  • 본 연구는 난관액과 oviductal conditioned medium 이 다정자 침입과 체외배발달에 미치는 효과를 규명하기 위하여 수행하였다. 배양액내 난관액과 oviductal conditioned medium 의 첨가는 다정자침입율과 난자내 침입한 평균정자수를 감소시켰다 (P<0.05). 정자와 난관액 그리고 oviductal conditioned medium 과 l.5, 3, 4.5 시간 공배양후 첨체반응의 성적은 대조구에 비하여 증가하였다. 체외수정후 체외발달 배양액에 난관액이나 oviductal conditioned medium 을 첨가하여 192 시간동안 배양했을 때 상실배와 배반포배발달율이 난관액과 oviductal conditioned medium 첨가구에서 유의적으로 높았다 (P<0.05). 이러한 연구결과는 난관액과 OVCM 등 난관 유래물질은 다정자침입율과 난자내 침입한 평균정자수를 감소시키며 체외발달율을 높인다고 사료된다.

CT 조영제가 MR 조영제의 이완율에 미치는 영향 (Effects of CT Contrast Medium on the Relaxation Rate of MR Contrast Medium)

  • In MR, the iodine CT contrast medium reduces the T1 and T2 relaxation times of the substance, resulting in a change in signal intensity. This study aimed to measure the relaxation rate of MR contrast medium with or without diluting CT contrast medium and analyzed the effect of CT contrast medium. Undiluted Gadoteridol solution was diluted with saline to prepare MR contrast medium phantoms with various levels of Gadoteridol concentrations. Moreover, undiluted Iomeprol was mixed with the prepared MR contrast medium phantoms at 1:1 ratio to make MR contrast medium phantoms with containing CT contrast medium for the experiment. T1 and T2 mappings were conducted to quantitatively evaluate the relaxation time and relaxation rate of these phantoms. The results showed that the T1 and T2 relaxation time and relaxation rate of MR contrast medium diluted with CT contrast medium were significantly (p<0.05) shorter than those of MR contrast medium not diluted with CT contrast medium. The results of this study imply that, when MR contrast medium shall be used after injecting CT contrast medium, CT contrast medium should be discharged enough. Moreover, it would be desirable to conduct CT test after taking MRI test in order to reduce the effects of CT contrast medium on MR contrast medium.

몇가지 물질(物質)이 Agaricus campestris균사생장(菌絲生長)에 미치는 영향(影響) (제(第) II 보(報)) (Effects of the special media on the mycelium growth in Agaricus campestris(II))

  • 상기(上記) 실험(實驗)에서는 Agaricus campestris의 basal medium에 몇종(種)의 생즙(生汁)을 첨가(添加)하여 균사생장(菌絲生長)에 미치는 영향(影響)을 조사(調査)하였다. 이때 실험실(實驗室) 온도(溫度)는 $23{\pm}1^{\circ}C$, 습도(濕度)는 $70{\pm}5%$, medium pH $5.4{\sim}5.5$로 하였다. 위 환경(環境) 조건(條件)에서 자란 결과(結果)는 다음과 같다. (이상(以上)은 144hrs후(後) 성적(成積)이다) 1. Carrot basal medium에서는 100ml의 basal medium에 4ml 의 carrot extract를 첨가(添加)할때가 control에 비(比)해 가장 좋은 유의적(有意的)인 성적(成積)을 나타냈고 Cucumber basal medium에서는 100ml basal medium에 3ml의 cucumber extract를 첨가(添加)했을 때가 우세(優勢)하였으며 radish basal medium에서는 100ml의 basal medium에 radish extract 3ml 첨가시(添加時) 성적(成積)이 양호(良好)하게 나타남으로써 유의적(有意的)인 차이(差異)를 보였다. 2. cabbage basal medium에서는 100ml의 basal medium에 2ml의 cabbage extract를 첨가(添加)했을 경우(境遇)에 control에 비(比)해 약간(若干) 우위(優位)였으나 대차(大差)없었고 Lettuce basal medium 역시(亦是) 100ml의 basal medium에 2ml의 lettuce extract를 첨가(添加)했을 경우(境遇)만 조금 성적(成積)이 좋았을 뿐이다.

Morphological Variations in Conidia of Arthrobotrys oligospora on Different Media

  • Most commonly occurring predacious fungus Arthrobotrys oligospora showed great variation in size and shape of conidia on some media. The formation of larger conidia was recorded on beef extract and nutrient agar media. The length of conidia in Richard's YPSS, Sabouraud's, PDA and com meal agar media was of medium size while smaller conidia were produced on Czapek's, Jensen's, Martin's medium. Maximum width of conidia was recorded on YPSS medium followed by Sabouraud's medium. The average size of spores on nematode infested com meal agar medium was slightly increased than those on com meal agar medium.

Effects of culture media conditions on production of eggs fertilized in vitro of embryos derived from ovary of high grade Hanwoo

  • Background: This study was investigated the effects of culture media conditions on production of eggs fertilized in vitro of embryos from ovaries of high grade Korean native cow, Hanwoo. Methods: The IVMD 101 and IVF 100 were used for in vitro maturation of selected Hanwoo oocytes and In vitro embryo culture after in vitro fertilization, respectively. The IVMD 101 and IVD 101 were used for in vitro culture and completely free of serum. Results: The cleavage rates of 2-cell embryos in reference to Hanwoo oocytes were 86.7, 92.9, and 90.1 % in the control group, IVDM101 medium and IVD101 medium, respectively which indicates that the IVDM101 medium and IVD101 medium may result favorable outcomes. The in vitro development rates of blastocysts were 12.4, 38.4 and 32.4 % in the control group, serum free IVMD101 medium and IVD101 medium, respectively. For hatched blastocysts, it was 5.3, 33.9, and 28.6 % in the control group, serum free IVMD101 medium and IVD101 medium, respectively. Hence, more favorable results were expected for the hatched blastocysts in which the IVMD101 medium and IVD101 medium were used than the control group. Average cell numbers of blastocysts were 128.3, 165.7, and 163.6 in the groups of TCM-199 + 10 % FBS medium, IVMD 101 medium, and IVD 101 medium, respectively which clearly show that the IVMD 101 and IVD 101 medium consequence significantly higher cell numbers compared to the control group (i.e., TCM-199 +10 % FBS medium). Pregnancy rate after embryo transfer was 39.6 % when the serum free medium was used which is higher than that of the medium supplemented with serum (32.8 %). In addition, stillbirth rates were 4.9 % in the group of serum free medium whereas it was 13.6 % in the serum supplemented medium (13.6 %). Conclusions: Taken altogether, serum free media, the IVMD 101 and IVD 101 represented more favorable results in the embryo development rate of embryos, cell numbers of blastocyst, and pregnancy rate. Of note, the IVMD 101 medium showed better outcomes hence, it might be a better option for future applications for in vitro culture of bovine embryos.

Exhausted Medium에 의한 각막상피 세포의 세포고사 유도 (Apoptosis in Human Corneal Epithelial cells induced by Exhausted Medium)

  • 본 연구는 각막상피가 저절로 탈락해 나가는 과정을 이해하기 위해 사람의 각막상피 세포를 배양하여 배지의 영양이 고갈될 때까지 약 7일 정도 배지를 교환하지 않고 계속 배양하여 세포고사를 조사하였다. 영양이 고갈된 배지인 Exhausted Medium을 여과하여 새로운 각막 상피세포에 넣어 2일 정도 배양을 계속하였다. Exhausted Medium에서 배양된 세포를 수확하여 세포고사를 검정하기 위해 Agarose gel electrophoresis로 DNA 단편을 확인하고 세포고사를 초기에 감지할 수 있는 방법인 M30 $CytoDEATH^*$. Fluorescein으로 확인하였다. 또한 Exhausted Medium에 의한 세포고사의 유발 경로를 조사하기 위해 사이토카인에 의한 유도인자 중 가장 많이 알려진 FAS나 FAS Ligand에 의한 유발여부를 soluble FAS and FAS Ligand에 대한 sandwich ELISA로 조사하였다. 그 결과 전형적인 DNA Ladder패턴을 보였으며 M30 $CytoDEATH^*$. Fluorescein에도 선명하게 염색되어 세포고사를 확인할 수 있었다. 또한 soluble FAS and FAS Ligand ELISA Kit로 Exhausted Medium이 FAS와 관련이 있는지를 조사한 결과 거의 무관한 것으로 나타났다. 본 연구의 결과는 영양 부족상태의 배지는 각막상피 세포가 세포고사를 거쳐 소모되는데 FAS and FAS Ligand system과는 무관한 것으로 나타났다.

  • In Korea small-medium companies constitute 96.7% of all of the mining and manufacturing industries, about 34.2% of the total production of value added business, an average of 34.8% of #he actual export of industrial goods, and 49.4% of employment at the end of 1980. Now then small-medium industry companies organization style constitutes 82.1% of private companies and 82.7% of the total on an small scale under 49 persons in regular empolyees. Small-medium industry Companies have growing problems and bottle-necks in their physical conditions. Total Quality Control is contributed by small-medium industry company for more growth than present conditions. But now, Small-medium industry company do not use systematic application of Quality Control Method, the small-medium business company products quality is not better as compared with big business company. Specially, TQC is needed for Small-medium company for imperishable enlargement . In order to promote good will and the major stress of this paper is first of all on the problems of Quality Control, which covers the present states and derived problems of Quality Control, ill Korea Small-medium industry Companies. These are investigated and analyzed to insure the rational application of Quality Control to the Small-medium companies with a view of the establishment of the direction and systematic improving means of Quality Control application Model building for Korea Small-medium industry.

벼 종자에서 Xanthomonas campestris pv. oryzae의 분리를 위한 선택배지 (A Semiselective Medium for the Isolation of Xanthomonas campestris pv. oryzae from Rice Seed)

  • A semiselective agar medium (XCO) was developed for the isolation of bacterial blight pathogen, Xanthomonas campestris pv. oryzae, from rice seed. The medium contained yeast extract 1.0 g, peptone 2.0 g, sucrose 5.0 g, sodium glutamate 1.0 g, FeSO4.7H2O 0.05 g, Fe.EDTA 1 mg, cephalexin 20 mg, Evan blue (0.1%) 1.5 ml, bromcresol purple (0.1%) 2.5ml, cycloheximide 100 mg and agar 15.0 g per liter. Colonies of X. c. pv. oryzae were 4~5 mm in diameter, smooth, round, blue (darker center) and convex after 6 days incubation at 28$^{\circ}C$. The recovery of 6 isolates of X. c. pv. oryzae on the XOC medium ranged from 81% to 120% (mean 98.2%) in comparison to Wakimoto's medium. The number of saprophytic bacteria from 10 rice seed lots on XCO medium was reduced to 70.4% of that on Wakimoto's medium. The recovery of X. c. pv. oryzae added to rice seed on XOC medium ranged from 67% to 87% (mean 75.6%) of that on Wakimoto's medium.
