Microencapsulation Characteristics Depending on Mixing Ratio of Wall Materials and Squid Liver Oil (피복물질과 오징어 간유의 혼합 비율에 따른 미세캡슐화 특성)

  • This study was carried out to investigate the microencapsulation characteristics of squid liver oil according to various ratios of wall materials and squid liver oil. The emulsion stability and the water binding capacity increased with an increase in wall materials contents. The microencapsulation efficiency was found to be in the order of 7:3>4:6>3:7>5:5>6:4 by mixing ratio of wall materials and squid liver oil; also, ratio of 7:3 was found to be inappropriate because the oil content in the powder was not sufficient. Regarding the fatty acid composition, the content of polyunsaturated fatty acid was found to be over 50% in all treatment groups. The ratio of polyunsaturated to saturated fatty acid of the powder was the highest (2.13) at the mixing ratio of 4:6.

Functional Characterization of the Squid Calexcitin-2, a Calcium and GTP-binding Protein

  • Calexcitin, a calcium-binding protein, was previously cloned and functionally characterized in the squid Loligo pealei. We now report the cloning of a second form of Calexcitin, Calexcitin-2, found in the squid Todarodes pacificus optic lobe. Calexcitin-2 has a significantly different carboxyl terminal region than Calexcitin-1. It lacks the CAAX motif, which is a farnesylation site. The amino acid sequence of Calexcitin-2 shows an 84% identity with Calexcitin-1 and also displays a strong cross immunoreactivity. Western blotting shows that Calexcitin-2 was expressed exclusively in the optic lobe region of squid, but not in other body organs. Regardless of its lack of conserved regions for GTP-binding, Calexcitin-2 shows moderately low affinity GTP-binding and also shows dramatic conformational change induced by GTP-binding. Three possible GTP-binding region mutations, K142A, D144A, and K157A, did not change the G TP binding affinity. This raises the possibility that Calexcitin-2 may have a novel GTP-binding motif.

  • The aim of this study is to confirm clinical usefulness of magnetocardiogram (MCG) by analyzing MCG data of health subjects and patients with Wolff-Parkinson-White (WPW) syndrome and dilated cardiomyopathy (DCM). Measurement of MCG signals was done with a home-made 40-channel SQUID system. MCG signals of 30 healthy subjects were measured as the reference of MCG signals. Among the DCM patients, 7 patients showed abnormal the direction of T wave vector. For a WPW syndrome patient, we measured the MCG signals before and after the surgery. and compared the difference. From the measured magnetic field distribution, current vector map was obtained to show the myocardium current activity. By comparing the MCG signals and current maps, we showed the differences in the analysis results between the healthy subjects and patients with heart diseases.

Proximate and Total Fatty Acid Compositions of the Reproductive Organs of Male and Female Common Squid Todarodes pacificus

  • We analyzed the compositions of proximate and total fatty acids in the reproductive organs of male (testis, vas deferens, seminal vesicle, and spermatophore sac) and female (ovary, oviduct, oviducal gland, and nidamental gland) common squid. The protein contents were 12.1-22.7 and 13.2-19.4% for males and females, respectively, and the lipid contents were 1.0-2.7 and 2.1-8.0%, respectively. The prominent fatty acids in all reproductive organs were 22:6n-3, 20:5n-3, 16:0, 20:1n-9, 18:0, 18:1n-9, and 20:4n-6. Females had 28.6-32.0% of 22:6n-3 and 11.3-22.6% of 20:5n-3, while males had 20.8-26.8% of 22:6n-3 and 14.4-28.7% of 20:5n-3. These results indicate that the reproductive organs of both male and female squid are potential sources of n-3 polyunsaturated fatty acid.

  • Squid catch in the East Sea has annual, 3-5 years and decadal periodicity. Position of main fishing ground depend on the pattern of the Tsushima Warm Current. Marine environment in spawning ground has close correlation with the variation of squid catch in the East Sea.

  • Heart consists of myocardium cells and the electrophysiological activity of the cells generate magnetic fields. By measuring this magnetic field, magnetocardiogram (MCG), functional diagnosis of the heart diseases is possible. Since the strength of the MCG signals is weak, typically in the range of 1-10 pT, we need sensitive magnetic sensors. Conventionally, superconducting quantum interference devices (SQUID)s were used for the detection of MCG signals due to its superior sensitivity to other magnetic sensors. However, drawback of the SQUID is the need for regular refill of a cryogenic liquid, typically liquid helium for cooling low-temperature SQUIDs. Efforts to eliminate the need for the refill in the SQUID system have been done by using cryocooler-based conduction cooling or use of non-cryogenic sensors, or room-temperature sensors. Each sensor has advantage and disadvantage, in terms of magnetic field sensitivity and complexity of the system, and we review the recent trend of MCG technology.

  • We have earlier developed a 40-channel SQUID system. An important figure of merit of a MEG system is the localization error, within which the underlying current source can be localized. With this system, we investigated the localization error in terms of the standard deviation of the coordinates of the ECDs and the systematic error due to inadequate modeling. To do this, we made localization of single current dipoles from tangential components of auditory evoked fields. Equivalent current dipoles (ECD) at N1m peak were estimated based on a locally fitted spherical conductor model. In addition, we made skull phantom and simulation measurements to investigate the contribution of various errors to the localization error. It was found that the background noise was the main source of the errors that could explain the observed standard deviation. Further, the amount of systematic error, when modeling the head with a spherical conductor, was much less than the standard deviation due to the background noise. We also demonstrated the performance of the system by measuring the evoked fields to grammatical violation in sentence comprehension.

Effects of Hydrolysates from Aquatic By-Product on Feeding Attraction and Growth of Sea Cultured Fish (수산 가공부산물 가수분해물에 의한 해산 양식어의 섭식 유인 및 성장에 미치는 효과)

  • This study was conducted to investigate the effects of different dietary enzymatic hydrolysates using squid and mackerel by-product on feed efficiency, growth and chemical composition in Korean rockfish (Sebastes schlegeli) after a eight weeks feeding experiment. Effects on the weight gain and feed efficiency in the enzymatic hydrolysates added groups were very effective. Weight gain were significantly higher in the squid hydrolysates whole added group than in the other groups. Moisture and crude ash contents of muscle were not significantly affected by hydrolysates supplementation in all groups. Crude protein and lipid contents of muscle were increased after eight weeks feeding trial. Amino acid contents of muscle were higher in squid hydrolysates whole added group than in the other groups. Amino acids were not affected by hydrolysates supplementation in all groups. Fatty acid contents of muscle were increased after eight weeks growth trial. Fatty acids were not significantly different in the dietary groups.

A Genetic Analysis of the Common Squid, Todarodes pacificus in the Korean Waters

  • In order to estimate the genetic variability and differentiation in common squid, eleven isozymic loci, coded for nine enzymes detected by starch gel electrophoresis, were scored from nine spawning cohorts in four localities. The expected average heterozygosity ranged from 0.00019 (between II-S$_2$ and Na-W) to 0.00814 (Between Bu-S and Na-W) in nine different spawning cohorts in four localities. A dendrogram, based on genetic distance mentioned, illustrated that nine different spawning cohorts were divided into three groups, similar to the result estimated by their ecological characterizations. From these results, we estimate that the common squid distributed throughout Korean waters will maintain this gene exchange. It is postulated that either the summer or the autumn spawning cohort has developed a local population that is isolated by hydrographic factors.

Pyrosequencing-Based Analysis of the Bacterial Community in Korean Traditional Seafood, Ojingeo Jeotgal

  • Jeotgal fermentation is dependent upon a diverse microbial community, although a detailed understanding of its microbial composition is limited to a relatively small number of jeotgal. Pyrosequencing-based bacterial community analysis was performed in fermented squid, ojingeo jeotgal. Leuconostoc was identified as the predominant bacterial genus, with Bacillus and Staphylococcus also accounting for a large proportion of the bacterial community. Phylogenetic analysis with 16S rRNA genes of Leuconostoc type species indicated that L. citreum- and L. holzapfelii-like strains could be the major Leuconostoc strains in jeotgal. High concentrations of NaCl were thought to be an important factor determining the makeup of the bacterial community in the fermented squid; however, a genomic survey with osmotic stress-related genes suggests the existence of more complex factors selecting the dominant bacterial species in fermented squid.
