A Study on the Perception and Awareness of Native Local Foods - Centering around Gimhae, Gyeongnam Province - (향토음식에 대한 의식 및 인지도에 관한 연구 - 경남 김해지역을 중심으로 -)

  • This study aims to furnish the basic data in securing momentum for taking interest in inheriting and developing the native local foods in Gimhae by examining the perception and awareness of the native local foods among the local residents in Gimhae, Gyeongnam Province. As the results of this study, the definition of native local foods was surveyed as follows. Foods that have been handed down for generations by using local agricultural products as their ingredients; a local food restaurant was the most preferred place to eat native local foods; the most chosen reason why they should eat native local foods was 'good for health'; as for the necessity of inheriting and developing the native local foods, most of them agreed, and it was because they felt they should protect their food culture; and 'native local foods contests' were the best way to promote the native local foods. As the results of analyzing the awareness of 12 kinds of native local foods in Gimhae, there were significant differences in Kongnamul-bab, Minmul Jangeo-gui, Gajuk-jaban, Kongip-mulkimchi by gender, in Hobakogari-namul, Moomallaengi-jangachi, Kongip-mulgimchi by ages, and in Kongnamul-bab, Gajuk-jaban, Kongip-mulgimchi by the residential districts. Accordingly, in order to inherit and develop the native local foods which are the precious cultural assets in Gimhae, there should be continued opportunities and interest to promote them through the local festivals and events along with publicity activities for the native local foods.

Comparative liver drug metabolizing enzymes activities between Korean native cattle and swine (한우와 돼지에서 간의 약물 대사효소의 활성 비교)

  • Drug-metabolizing activities of Korean native cattle and swine were investigated from viewpoints of the cytochrome P-450's level, their dependent mixed function oxidase activities, the reactive oxygen species formation and cytosolic enzyme acitivities from each liver homogenates. Level of cytochrome P-450 in the liver microsome of Korean native cattle was $0.28{\pm}0.05nmole/mg$ and that in pigs $0.35{\pm}0.03nmole/mg$. Level of cytochrome $b_5$ of Korean native cattle was $0.24{\pm}0.06nmole/mg$, and that of pigs $0.2{\pm}0.05nmole/mg$, showing no difference between two species. NADPH P-450 reductase were higher in Korean native cattle ($58.3{\pm}5.3nmole/mg/min$) than in pigs ($29.9{\pm}3.8nmole/mg/min$)(p<0.01). The activities of cytochrome P-450 dependent monooxygenases such as ethoxyresorufin O-deethylase (cattle, $96.5{\pm}12.5nmole/mg/min$ ; pigs, $13.6{\pm}2.1nmole/mg/min$), N-benzphetamine N-demethylase (cattle, $5.23{\pm}0.82nmole/mg/min$ ; pigs, $0.76{\pm}0.3nmole/mg/min$) and aniline hydroxylase (cattle, $0.95{\pm}0.1nmole/mg/min$ ; pigs, $0.33{\pm}0.08nmole/mg/min$) were much higher in Korean native cattle than in swine(p<0.01). However, the activity of testosterone $7{\alpha}$-hydroxylase was higher in swine ($90.4{\pm}1.2nmole/mg/min$) than cattle (cattle, $32.8{\pm}1.2nmole/mg/min$). Interestingly, testosterone $16{\alpha}$-hydroxylase, a marker enzyme for P-450 IIA was not detected in both animal species. These results suggest that Korean native cattle and pigs have high contents of P-450 IA1 and P-450 IIIA. Total sulfhydryl compound (cattle, $10.3{\pm}1.1nmole/mg$ ; Pigs, $14.5{\pm}1.8nmole/mg$) and glutathione related enzymes except glutathione reductase (cattle, $38.1{\pm}7.9nmole/mg/min$; swine, $22{\pm}3.6nmole/mg/min$) showed higher levels in swine than in Korean native cattle. Superoxide dismutase (cattle, $7.64{\pm}0.84nmole/mg/min$ ; pigs, $4.47{\pm}0.94nmole/mg/min$) and catalase (cattle, $30.4{\pm}3.7nmole/mg/min$ ; pigs, $17.2{\pm}1.8nmole/mg/min$) were remarkably higher in Korean native cattle than in swine (p<0.05).

Characteristics of Seed Germination and Bulb Dormancy in Korean- Native Allium victorialis L. (韓國産 산마늘 種子의 發芽와 種球의 休眠 特性)

  • The experiments were conducted to investigate the characteristics of the seed germination and the bulb dormancy in Korean-native Allium victorialis L. by the physicochemical treatments. The characteristics of two Allium victorialis seeds was similar in shape, color and threshing but different in other characters. Weight of 1,000 grains in Ullrung-native was 14.8g corresponding to 2.1 times as compared with Odae-native. Seeding immediatly after seed collection of Ullrung-native only germinated and chemicals didn't affect in germination improvement. The germination rate, average days to germination and coefficient of germination of Ullrung-native was 86%, 42 days and 2.05 at the dark condition of control, respectively. The germination rate, average days to germination and coefficient of germination of Odae-native was 70%, 47 days and 1.5 at the dark condition of control and 82%, 47 days, 1.75 at the light condition after 5$^{\circ}C$ prechilling for 30 days, respectively. The bedding days to reduce the physiological dormancy in the bulb of Odae-native assumed to be appropriate up to 45 days after the first prechilling.

Identification of Chromosomal Band Markers of the Korean Native Chicken (한국재래계의 염색체 분염 표지 분석)

  • The present study was carried out to establish the standard karyotype of the Korean Native Chicken and to find their chromosomal band markers using high-resolution banding technique. Chromosome analysis was performed on early chick embryos following in vitro culture of fertilized eggs of the yellow-brown and the red-brown lines of the Korean Native Chicken which had been established at National Livestock Research Institute. The high-resolution banding of the chromosome was achieved by treating the embryos with ethidium bromide and colchicine during culture. On GTG-banding, the Korean Native Chicken exhibited a typical chick banding pattern in all the macrochromosomes. Overall chromosomal morphology and positions of typical landmarks of the Korean Native Chicken were virtually identical to those of White Leghorn and International System for Standardized Avian Karyotypes(ISSAK). However, the lengths and G-band numbers of the Korean Native Chicken macrochromosomes were greater than those of White Leghorn and ISSAK. Especially in chromosomes 1 and Z, the Korean Native Chicken exhibited more separated bands in compared with ISSAK. In C-banding patterns, although a lot of observed cells had C-band polymorphic patterns, almost the Korean Native Chicken macrochromosomes had heterochromatic C-band on centromeres and/or near terminal part. However, the heterochromatic C-band was constantly observed at the end of q-arm of Z chromosomes and on the whole W chromosome. In addition, the Korean Native Chicken exhibited distinctive heteromorphic patterns of C-bands on the centromere of chromosome 3 and at the end of q-arm of Z chromosome between homologous chromosomes.

Quality Comparison between Korean Native Black Ground Pork and Modern Genotype Ground Pork during Refrigerated Storage (재래종과 개량종 세절 돼지고기의 저장 중 품질 특성 비교)

  • This study was carry out to investigate the quality characteristics of Korean native black ground pork compared with modern genotype ground pork during refrigerated storage. Korean native black pig and modern genotype pig were slaughtered at 75 kg and 105 kg of live weight, and for 240 days and 210 days of feeding periods, respectively. The ground lean pork (M. semimembranosus) was stored for 9 days at 4℃. The crude fat and crude protein contents were significantly (p<0.05) higher in Korean native black pork. The pH value after 5 days of storage was significantly (p<0.05) lower in Korean native black pork than in modern genotype pork. WHC of Korean native black pork was significantly (p<0.05) higher than that of modern genotype pork over time. The Korean native black pork maintained black reddish color because it had lower CIE L/sup */ value and higher CIE a/sup */ value than the modern genotype pork. CIE L/sup */, b/sup */, C/sup */ and h/sup O/ values decreased as storage time increased. TBARS (thiobarbituric acid reactive substance), POV (peroxide value) and FOX (ferrous oxidation xylenol orange) tended to increase as storage time increased in all of the groups, in particular, those values increased more rapidly in Korean native black pork. Total saturated fatty acid and stearic acid contents had significantly higher in Korean native black pork (p<0.05).

  • This study was carried out to analyze the genetic polymorphisms of genomic DNA of blood and meat for conservation of the genetic resources and genetic improvement of Korean Native goat. The genetic identification between Korean Native goat and imported goat was examined using RAPD(random amplified polymorphisms DNAs) analysis with 30 Korean Native goat, 10 hybrid, 10 imported goat. 10 Korean native goat meat and 10 imported goat meat. The results obtained from this study were summarized as follows: 1. Genomic DNA from Korean native goat, hybrid and imported goat could be obtained above about 23kb size using 0.5% agarose gel electrophoresis and the ratio of optical density at 260nm to that at 280nm was between 1.7 and 2.0 using UV spectrophtometer instrument. 2. In the results of the gene identification between Korean Native goat and hybrid, and imported goat using RAPD methods with random primer of 110 kinds, only Korean native goat showed a specific band at about 369bp using a random primer OPO-19 (5'-CAA ACG TCG G-3'), but imported goat and hybrid not showed. 3. Also, in the results of the gene identification between Korean Native goat meat and imported goat meat using RAPD methods with random primer, Korean native goat only showed a specific band at about 369bp using a random primer No. 19(5'-CAA ACG TCG G-3'), but imported goat not showed.

Morphological Observations of Ovaries in Relation to Infertility in Slaughtered Cows in Kyungnam Province 2. Incidences and Morphological Findings of Ovarian Cysts (경남지방의 도태우에 불임과 관련된 난소의 형태학적 관찰 2. 난소낭종의 발생과 낭종형태에 대하여)

  • Ovaries from total 192 slaughtered cows(154 Korean native cows and 38 Holstein cows) were collected during the slaughtering process in Kimhae, Changyoung and Yangsan abattoirs in Kyungnam province from January 2001 to January 2002. In order to investigate incidence of the ovarian cysts, anatomical, histological observations were performed and also TUNEL methods and PCNA antibody by immunogistochemical methods for diagnostic accuracy of cysts in a few ovaries were applied. Apoptotic positive cells by TUNEL method appeared not or a few in cystic walls but appeared more number in normal large follicular walls and the proliferative positive cells by PCNA antibody appeared numerous in normal large follicular walls but not or a few in cystic walls. The incident rates of ovarian cysts were 19.5% in Korean native cows and 18.4% in Holstein cows. The incident rates of ovarian cysts in Holstein cows were lower than that of Koran native cows. The incident rates of follicular cysts and luteal cysts in Korean native cows were 11.7% and 7.8% respectively. The incident rates of follicular cysts and luteal cysts in Holstein cows were 10.5% and 7.9%, respectively. Higher incidence proportions of ovarian cysts according to seasons in Korean native cows were ordered as spring (29.8%), autumn (21.4%) winter (14.3%) and summer (6.7%). Rates of cows with single cyst and multiple cysts were 63.3%(19 heads /30 heads) and 36.7%(11 heads/30 heads) in 30 cystic Korean native cows, respectively. Cystic cows with corpus luteums were 50.0%(15 heads) in 30 Korean native cows and 42.9%(3 heads) in 7 dairy cows, respectively. Among 15 cystic Korean native cows with corpus luteums, rates of cows with single corpus luteum were 66.7%(10 heads) and rates of multiple corpus luteum were 33.3%(5 heads ), respectively. The average diameter of cysts and corpus luteums in cystic ovaries were 21.0$\times$17.1 mm and 18.1$\times$13.8 mm in 30 Korean native cows and 20.6$\times$17.7 mm and 19.3 $\times$ 14.9 mm in 7 Holstein cows, respectively. So the average sizes of cysts in cystic ovaries were larger than those of corpus luteums.

Comparison and Validation of Genetic Diversity and Population Structure Using Monomorphic SNP Data of the Korean Native Black Goat and Crossbred Goat (재래흑염소와 교잡종 염소의 Monomorphic SNP 분석을 통한 유전적 다양성과 집단구조의 비교 및 검증)

  • This study was conducted to analyze the genetic diversity and relationships that discriminate between Korean native black goat populations (Dangjin, Jangsu, Tongyoung, and Gyeongsang National University strains) and crossbred goats. Monomorphic single nucleotide polymorphisms (SNPs) in each strain were collected, and 133 common SNPs were selected for analysis. These 133 monomorphic SNPs showed differences in the genetic structure of the Korean native black goat and crossbred goats, and results from the principal component analysis (PCA) showed that the two can be clearly separated. Furthermore, analysis of the validation population comprising 70 individuals (Korean native black goats, n = 24; crossbred goats, n = 46) with the reference population showed that Korean native black goat strains and the reference population have the same genetic structure, and the crossbred goats shared only part of the genetic structure with the reference population. The result of the PCA analysis showed that the Korean native black goat strains form one population, whereas the foreign strains form another population which is more widely dispersed than the Korean native black goat strains. Thus, the results from this study can be used as baseline data for the conservation of genetic resources of Korean native black goat communities through utilization of monomorphic SNPs and for the introduction of exotic species for further improvement in genetic diversity. This study can also help reduce unnecessary inbreeding and gene flow between native strains.

Growth of Two Native Zoysiagrasses Collected from Sea Side and Mountain Area in Indonesia on Growing Media Composed of Sand and Clay

  • Zoysiagrass (Zoysia spp.) exists spotly in Indonesia and it has potential to be used in parks, golf courses, and football fields. Many football fields and golf course fairways use sand as top soil over native soil. This study aims to analyze growth and quality of two native zoysiagrasses Zis and Zim. Zis is a native zoysiagrass collected in sea-side and Zim is a native zoysiagrass collected in mountain area. Both types of zoysiagrasses were planted at field with altitude of 300 m with various growing media mixes of sand and reservoir's sediment. Thickness of the growing medium was 10 cm over an alfisol clay soil. Experimental plots were constructed using factorial completely randomized design with two native zoysiagrasses and 5 types of growing media. Two ecotypes of native zoysiagrasses showed different in growth habits combined with mixtures of growth media. Zim showed higher growing speed including more vigor with uniformity and texture than Zis. There were not significanthly differences on leaf color and root length between two ecotypes of native zoysiagrasses collected in Indonesia.

Comparison of Functional Compounds and Micronutrients of Chicken Breast Meat by Breeds

  • The concentrations of functional compounds and micronutrients of chicken breast from native chickens were compared with those from broiler. Totally 200 male chicks from a commercial native chicken (HH) and three newly bred native chicken strains (2A, 2C, and 2D) were reared for about 2 kg of final live weight up to 12 wk. After slaughtered, antioxidant dipeptides, reducing sugar, free amino acids, vitamins, and minerals of the breast muscles were analyzed with those from broilers with similar live weight. Mostly native chicken strains had higher contents of carnosine, anserine, and reducing sugar than the broiler. Especially HH implied the highest values of carnosine and anserine, and 2C did the highest of reducing sugar (p<0.05). Vitamin A contents between native chickens and broiler were not significantly different (p>0.05). The contents of ${\alpha}-tocopherol$ were significantly higher in 2C than those of HH or broiler (p<0.05). Native chicken strains contained lower cholesterol levels than the broiler. Broiler had higher contents of P, Mg, and Na than native chickens (p<0.05), but it had lower content of Cu than HH or 2A. The savory free amino acids including glutamic acid was highest in 2A than the other native chickens and broiler (p<0.05). This study confirms that certain new strains of native chickens be a good source in terms of functional compounds and micronutrients which can be attractive health promoting nutritional quality factors.
