Effects of Eicosapentaenoic Acid and Docosahexaenoic Acid on Mitochondrial DNA Replication and PGC-1α Gene Expression in C2C12 Muscle Cells

  • Mitochondrial biogenesis is a complex process requiring coordinated expression of nuclear and mitochondrial genomes. The peroxisome proliferator-activated receptor gamma co-activator 1-alpha (PGC-$1{\alpha}$) is a key regulator of mitochondrial biogenesis, and it controls mitochondrial DNA (mtDNA) replication within diverse tissues, including muscle tissue. The aim of this study was to investigate the effects of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) on mtDNA copy number and PGC-$1{\alpha}$ promoter activity in $C_2C_{12}$ muscle cells. mtDNA copy number and mRNA levels of genes related to mitochondrial biogenesis such as PGC-$1{\alpha}$, nuclear respiratory factor 1 (NRF1) and mitochondrial transcription factor A (Tfam) were assayed by quantitative real-time PCR. The PGC-$1{\alpha}$ promoter from -970 to +412 bp was subcloned into the pGL3-basic vector, which includes a luciferase reporter gene. Both EPA and DHA significantly increased mtDNA copy number, dose and time dependently, and up-regulated mRNA levels of PGC-$1{\alpha}$, NRF1, and Tfam. Furthermore, EPA and DHA stimulated PGC-$1{\alpha}$ promoter activity in a dose-dependent manner. These results suggest that EPA and DHA may modulate mitochondrial biogenesis, which was partially associated with increased mtDNA replication and PGC-$1{\alpha}$ gene expression in $C_2C_{12}$ muscle cells.

  • Korean native goats have lived on the Korean peninsula for more than 2,000 years and are regarded as a valuable genetic resource for the world. As an initial step to investigate the genetic structures of this breed, phylogenetic analysis and calculation of genetic diversities have been performed using mitochondrial DNA (mtDNA) sequence variations. A total of 19 Korean native goats were grouped into six haplotypes and the large majority of haplotypes were present in 13 animals. All mtDNA of these Korean goats belonged to the mitochondrial (mt) lineage A and revealed remarkably small genetic distances within the population when compared with other Asian goat populations, indicating less genetic variation in the Korean native goats. These results indicate high-inbred status of the Korean native goats and will influence breeding and conservation strategies adopted for this breed.

DNA Heteropolymorphism of Chum Salmon Detected by Denaturing Gradient Gel Electrophoresis and Real Time PCR (Denaturing gradient gel electrophoresis와 real time PCR 방법을 이용한 연어 유전자들의 DNA 이형 다양성 검색)

  • In order to detect the DNA heteropolymorphism of chum salmon, selected essential genes were examined in different regional chum salmons, i.e., Korean, Japanese and American by denaturing gradient gel electrophoresis (DGGE) and real time PCR methods. From the promoter regions and introns of growth hormone, mtDNA NDI region, D-loop region, IGF-I, histone H3 and MCH2 several representative primer pairs were obtained and employed for the DGGE with the PCR products from the genomic DNAs of the different regional chum salmons. mtDNA NDI, D-loop region and IGE-I genes showed marked heteropolymorphism between Korean and American chum salmons. Intron C of growth hormone also showed a heteropolymorphism between Korean and Japanese chum salmons. Whereas heteropolnnorphism of histone liH and MCH2 genes was detected among in Korean, Japanese and Asnerican chum salmons in the examined region. The real time PCR disclosed the characteristic incremental production of target DNAs dependent on the heteropolymorphic conditions of genomic DNAa of chum salmons, thus the different regional chum salmons could be grouped by the variable incremental curies. Although the DGGE and real time PCR did not produce the identical results in this study, we suggest that the DGGE and real time PCR could be used for the primary screening of the DNA heteropolymorphism of different animal genome.

뱀장어속 어류 2종의 동위효소 및 mtDNA 분석

  • 뱀장어속(Anguilla)의 뱀장어(A. ioponica)와 무태장어(A. marmoruta) 및 꾀붕장어(Anago anogo)의 유전적 특징과 종간 유연관계를 밝히기 위하여 isozyme 분석과 mtDNA 분석을 실시하였다. Isozyme 분석결과 20개의 효소 및 비효소단백질에서 총 39개의 유전자를 검출하였고 각종 특유의 genetic marker를 확인하였다. 3종의 평균 유전적 변이는 뱀장어가 HD=0.057. HG=0.065, 무태장어는 HD=0.067 HG=0.053, 체불장어가 HD=0.018. HG=0.020으로 각각 나타났다. 뱀장어와 무태장어의 평균 종간유전적 근인관계는 S=0.420 (D=0.869)로 멀게 나타났다. 6 base를 인지하는 10종류의 제한효소를 처리한 결과 mtDNA 절편양상은 각 종내에서 동일하게 나타났으나 각종간에 차이를 보였고, 종간 평균염기 치 환율은 뱀장어와 무태장어가 p=3.4%, 뱀장어속 2종과 꾀붕장어는 p=9.6%로 나타났다.

  • The venus clam, Ruditapes philippinarum, is an aquaculture shellfish mainly distributed in an intertidal zone of East Asia including Korea, China and Japan. The morphological variation of this species is great. In fact, two of the most popular markers used in molecular evolution, mitochondrial DNA (mtDNA) and nuclear ribosomal DNA (rDNA), have quite different properties, which could translate into different consequences of mutation, drift, migration and selection on patterns of geographical variation and molecular divergence.   • The mitochondrial DNA (mtDNA) restriction fragment length polymorphi는 (RFLPs) of five populations were analyzed to delineate the stocks of Penaeus chinensis (Osbeck) in the Yellow Sea. Comparison of P. chinensis with P. japonicus to clarify the nucleotide divergence between two species was also carried out. Based on the fragment patterns, three composite haplotypes were analyzed in P. chinensis mtDNA as four naplotypes were in P. japonicus. Most individuals of each P. chinensis population are shared by one dominant haplotype. Another two haplotypes haying variations at the C/a I and hull sites were also distributed evenly in the Korean and Chinese populations. It is suggested that the gene exchange occurring between populations in the Yellow Sea is frequent. Average length of the mtDNA molecule was estimated to be about 16.44 kb in P. chinensis and 16.31 kb in P. japonicus, Sequence divergence (p) of mtDNA between two species estimated by using Upholt's (1977) fomula was $13.7\%$.

오골계의 기원과 유전적 다양성

  • Korean Ogol Chicken is a natural treasure in Korea and expected to be a valuable genetics resource in the world. As an initial step to investigate the genetic structures of this breed, phylogenetic analysis has been performed using mitochondrial DNA (mtDNA) sequence variations. Total 30 Korean Ogol Chickens were investigated in this study and they were grouped into 4 haplotypes, consisting 11 birds in the largest haplotype. Based on the phylogenetic analysis, chicken breeds were divided into three major groups and Korean Ogol Chicken were appeared all of these three groups indicating their large genetic mtDNA diversity. These results will be used for making breeding and conservation strategies for this breed.

Effect of Sequence Variation in Bovine Mitochondrial DNA D-loop Region on Economic Traits for Hanwoo (한우 경제형질에 미치는 Mitochondrial DNA D-loop 영역의 염기서열 변이효과)

  • This study was performed to analyse the sequences of variations of mtDNA D-loop and their effects on carcass traits in Hnawoo(Korean cattle). The resulting sequences were compared with previously published sequences for other cattle breeds(GenBank J01394). The PCR was used to amplify a total of 964 bp between nucleotide 15758 and 383 within D-loop region of mtDNA using specific primers. Twenty five polymorphic sites by nucleotide substitution were found in mtDNA of Hanwoo. The frequencies of positions at 169, 16042, 16093, 16119, 16255 and 16302 nt with high levels of sequence polymorphism were 0.891, 0.117, 0.109, 0.182, 0.197 and 0.117, respectively. The substitution effect at 169 and 16119 nt was found significant on marbling score. Also substitution effect at 169 and 16042 nt was highly significant(p〈0.01) on backfat. thickness. Polymorphism of mtDNA sequence in D-loop region could be useful for the analysis of cytoplasmic genetic variation and associations with the other economically important traits and maternal lineage analysis in Hanwoo.

The frequencies of positions at 8, 169, 16042, 16051, 16057, 16093, 16119, 16122, 16209, 16255 and 16302 nt with high levels of sequence polymorphism were 0.150, 0.950, 0.085, 0.138, 0.106, 0.085, 0.138, 0.212, 0.085, 0.148 and 0.180, respectively. The substitution effect at 16119(p<0.1) and 16185(p< 0.05) nt was found significant on milk production. Polymorphism of mtDNA sequence in D-Ioop region could be useful for the analysis of cytoplasmic genetic variation and associations with the other economically important traits and maternal lineage analysis in Hanwoo.

Phylogenetic relationship of ribosomal ITS2 and mitochondrial COI among diploid and triploid Paragonimus westermani isolates

  • We compared patterns of intraspecific polymorphism of two markers with contrasting modes of evolution, nuclear ribosomal DNA (rDNA) and mitochondrial DNA (mtDNA), in the lung fluke, diploid and triploid Paragonimus westermani from three geographical regions of Korea. The genetic distances between three populations of Korean diploid and triploid P. westermani showed no significant difference in the nucleotide sequences of the mitochondrial cytochrome c oxidase subunit 1 (mtCOI) and ribosomaal second internal transcribed spacer (ITS2) genes. A highly resolved strict-consensus tree was obtained that illustrated phylogenetically useful information of the ITS2 and mtCOI sequences from diploid and triploid P. westermani.
