겨우살이(Viscum album var. coloratum) 추출물의 항산화, 항응고 및 α-glucosidase 저해 효과 (Antioxidant, anticoagulant, and α-glucosidase inhibitory effects of mistletoe (Viscum album var. coloratum) extract)

  • 겨우살이(Viscum album var. coloratum)의 80% 에탄올 추출물의 항산화, α-glucosidase 저해 및 항응고 효과를 확인하였다. 겨우살이 추출물의 수율은 30.9±0.4%이었고, 폴리페놀 함량은 57.6±1.5 mg GAE/g으로 나타났다. 겨우살이 추출물의 농도에 비례하여 유리라디칼 및 양이온라디칼 소거능, 아질산염 소거능, 환원력의 항산화 효과가 증가하였다. 또한 겨우살이 추출물은 α-glucosidase의 활성을 저해하였고, 혈액응고 체계의 외인성 경로와 공통 경로를 주로 저해함으로써 혈장 응고를 억제하였다.

  • The mistletoe lectins are major active components in the extract of European mistletoe (Viscum album L) that have been widely used in adjuvant chemotherapy of cancer. This study was performed to investigate the antitumor activity of extract of Korean mistletoe (Viscum album var. coloratum) modified with Korean mistletoe lectin (Viscum album var. coloratum agglutinin, VCA). Compared with the results of VCA, survival rate was increased and experimental lung metastasis was reduced by treatment of modified extract (VCM). In addition, the treatment of VCM reduced angiogenesis and VCA-induced toxicity measured by a CAM assay. And VCM inhibited proliferation and induced apoptosis in vitro in tumor cells originated from tissues which are possible to apply topically without surgery. Taken together, the antitumor activities of VCM-treated group outperformed the activities of the VCA-treated group.

Bleeding Time Prolongation Effect of Methanol Extract of Viscum album var. coloratum

  • The methanol extract of Viscum album var. coloratum, Korean mistletoe, showed potent prolongation effects on the bleeding time in rats in vivo, and whole blood clotting time and plasma recalcification time in rats ex vivo. The prolongation effect on the bleeding time of Korean mistletoe is comparable to that of Viscum album L., European mistletoe, 185.6% and 176.5%, respectively. However, the water extracts of the both plants did not show any prolongation effects. Platelet activating factor (PAF) receptor binding assay was carried out to elucidate the action mechanisms of the extracts, and both of the methanol extracts did not show any inhibitory activity. The $LD_{50}$ of the methanol extracts of both mistletoes are more than 2 g/kg. These results suggest that the mehtanol extract of Korean mistletoe might be a potential candidate to develop new drug to improve microcirculation.

Preparation of Alginate/Chitosan Microcapsules and Enteric Coated Granules of Mistletoe Lectin

  • The aqueous extract of European mistletoe (Viscum album, L.) has been used in cancer therapy. The purified mistletoe lectins, main components of mistletoe, have demonstrated cytotoxic and immune-system-stimulating activities. Korean mistletoe (Viscum album L. coloratum), a subspecies of European mistletoe, has also been reported to possess anticancer and immunological activities. A galactose- and N-acetyl-D-galactosamine-specific lectin (Viscum album L. coloratum agglutinin, VCA) with Mr 60 kDa was isolated from Korean mistletoe. Mistletoe preparations have been given subcutaneously due to the low stability of lectin in the gastrointestinal (GI) tract. In the present study, we investigated the possibility of alginate/chitosan microcapsules as a tool for oral delivery of mistletoe lectin. In addition, our strategy has been to develop a system composed of stabilizing cores (granules), which contain mistletoe lectin, extract or powder, coated by a biodegradable polymer wall. Our results indicated that successful incorporation of VCA into alginate/chitosan microcapsules has been achieved and that the alginate/chitosan microcapsule protected the VCA from degradation at acidic pH values. And coating the VCA with polyacrylic polymers, Eudragit, produced outstanding results with ideal release profiles and only minimal losses of cytotoxicity after manufacturing step. The granules prepared with extract or whole plant produced the best results due to the stability in the extract or whole plant during manufacturing process.

Korean Mistletoe (Viscum album Coloratum) Extract Induces Eel (Anguilla japonica) Non-specific Immunity

  • Background: The immunomodulatory effects of Korean mistletoe (Viscum album Coloratum) on the innate immune responses of eel (Anguilla japonica) were studied. Methods: Mistletoe, Freund’s complete adjuvant (FCA), or phosphate-buffered saline (PBS) as a control was injected into eel peritoneal cavities. Results: Nitroblue tetrazolium (NBT)-positive cells in the head kidney of eel were significantly augmented by the second day post-injection of mistletoe. Reactive oxygen intermediates (ROI) were more produced in mistletoe-injected fish kidney leucocytes than in FCA-injected ones. The level of lysozyme activity in the serum of fish 2 days after injection with mistletoe was also significantly higher than that in the serum of the control fish. The optimal concentration of mistletoe in inducing the highest serum lysozyme activity was revealed to 500${\mu}$g/200 g of fish. In phagocytic activity assay, mistletoe-sensitized eel kidney phagocytes captured more zymosan than did the control fish. Conclusion: Korean mistletoe appeared to be a good activator of the non-specific immune responses of eel.

  • A lectin (agglutinin, VCA) from Korean mistletoe (Viscum album L. coloratum) was isolated by affinity chromatograpy on a asialofetuin-Sepharose 4B. The molecular weights of A- and B-chains of VCA were different from those of VAAS. The VCA recognized the antibody of VAAs in the Western blot analysis and ELLA system. We also investigated the synergistic effects of the components in mistletoe by dividing the extract into different molecular weight fractions.

  • The mistletoe lectins are major active components in the extrct of Viscum album var. coloratum. This study was performed to investigate the anti- skin cancer effect of Korean mistletoe extract intensified with mistletoe lectin (KI 2103S). B16F10 melanoma cells were allografted in C57BL/6 mice and F344 rats. The effect of KI 2103S on melanoma was measured by monitoring tumor index. The KI 2103S was injected intra-tumorally and tumor index was decreased in dose dependent manner.

  • Viscum album coloratum (Korean mistletoe; KM) is a semi-parasitic plant growing on various trees. This plant hasbeen shown to possess a variety of biological functions such as immunomodulation, apoptosis-induction and antitumor activity. However, there is no information about how the extract of KM affects life span of the host. In this study, in order to examine the effect of KM on the longevity of the host, we investigated whether a Korean mistletoe extract (KME) was able to extend the life length in an experimental model using the spring silkworm moth of Bombyx mori (Lepidoptera bombycidae). We established a model test for survival in which all of the male and female moths died within 18 days after the beginning of the experiment. Mean survival time of female moths was longer than that of male moths. Both male and female moths fed with KME containing diet (200 or 400 ${\mu}g$/head/day) showed significantly higher mean survival times than those of the control moths. Studying the effect of KME on prolongation of mean survival time showed that male moths were more susceptible than female moths. The moths fed with KME-containing diet showed a slight decrease in body weight. Interestingly, however, no difference in food intake was observed between moths fed with KME-containing diet and those with control diet. In analysis for mRNA increase in the SIRT2 gene, a member of the Sir2 gene family playing important roles in regulation of cell death and prolongation of life span, moths fed with KME-containing diet showed a significant increase in SIRT2 gene expression. These results suggest that KME has a potential to extend the life span in Bombyx mori, and its effect is partly associated with increase in SIRT2 gene expression.

Mistletoe Lectin (Viscum album coloratum) Modulates Proliferation and Cytokine Expressions in Murine Splenocytes

  • It is well documented that an extract of European mistletoe has a variety of biological effects, such as the stimulation of cytokine production from immune cells, and additional immunoadjuvant activities. While the European mistletoe has been studied intensively, we know less about Korean mistletoe as a therapeutic plant, especially as a possible immunomodulating drug. This study will investigated the effects of Korean mistletoe lectin (Viscum album L. var. coloratum agglutinin, VCA) on murine splenocytes to investigate whether VCA acts as an immunomodulator, which could lead to improved immune responses in these cells. The results showed that VCA inhibited cell proliferation at higher concentrations (at 1-8 ng/ml) and enhanced cell proliferation at lower concentrations (at 4-32 pg/ml). Further studies were carried out to determine if the pro-proliferative or anti-proliferative activity exhibited by VCA was correlated with cytokine secretion. Consequently, interferon (IFN)-$\gamma$ secretion was decreased in concanavalin A (ConA)-stimulated murine splenocytes by VCA (4-64 ng/ml), but there was no change in IL-4 levels. This suggests that VCA has the ability to modulate murine splenocyte proliferation and can possibly act on the balance of Th1/Th2 cellular immune responses.
