식중독 미생물에 대한 유기산 및 에탄올의 항균활성 비교연구 (Antimicrobial Effects of Organic Acids and Ethanol on Several Foodborne Microorganisms)

  • Automated turbidometer, Bioscreen(Labsystem)을 이용하여 tryptic soy broth에 에탄올(3%, 5%. 7%(v/v)) 및 유기산( 초산, 구연산 .젖산, 피로피온산 및 주석만(W/V))을 단독으로 첨가하거나, 혼합하여 Staphylococcus aureus, Salmonella typimurium, Escherichia coil O157 : H7, Listeria monocytogenes의 증식 곡선의 면적(AREA)과, 최대성장율(MGR), MGT(minimum generation time), DT(detection time)를 조사하였다. 모든 균주는 7% 에탄올에서 24 시간 동안 전혀 증식하지 못했으나 0.1% 유기산에서는 미생물의 종류와 유기산의 종류에 따라 다양한 양상을 보였다. S. aureus의 경우 프로피온산이 뚜렷한 증식 저해 효과를 나타내었으며, S. typhimurium의 경우 초산이 비교적 높은 효과를 보였다 L. monocytogenes에 대해서는 프로피온산, 주석산 및 젖산이 뚜렷한 증식 저해 효과를 보인 반면, E. coil O157 : H7의 경우에는 초산과 프로피온산이 효과가 있었다. 1%· 에탄올과 0.01% 유기산을 병용했을 때 S. typhimurium은 AREA에서 2% 에탄올 및 0.05% 초산의 증식 저해율과 비슷한 효과를 보였으며, L. monocytogenes는 AREA에서 0.05%프로피온산 및 MGR, MGT, DT에서 2% 에탄올과 비슷한 저해율을 보였다. S. aureus은 0.01% 프로피온산을 단독으로 사용했을 때가 오히려 0.01% 프로온피산과 1% 에탄올을 동시에 처리한 경우보다도 월등히 증식 저해 효과를 나타내었으며, E. coil O157 : H7은 1% 에탄올과 0.01% 유기산을 동시에 사용해도 AREA를 제외하고는 증식 저해를 받지 않았다.

식품 부패미생물에 대한 천연 항균성물질의 농도별 및 분획별 항균 특성 (Antimicrobial Effect of Some Plant Extracts and Their Fractionates for Food Spoilage Microorganisms)

  • 천연물에 존재하는 항균성물질을 정제, 분리할 목적으로 대량생산이 가능하며 식용하고 있는 식물로서 비교적 항균성이 높은 황백, 느릅뿌리, 질경이 및 민들레의 에탄올 추출물로 농도별 항균성을 시험하였고, 클로르포름, 에틸 아세테이트, 부탄올 순으로 분획하여 얻은 분획물의 항균성 검색과 항균성이 우수한 황백과 질경이 분획물은 농도별로 항균효과를 검토하였다. 황백, 느릅뿌리, 질경이 및 민들레의 에탄올 추출물로 농도별 항균성을 시험한 결과 균종에 따라 차이가 있지만 일반적으로 $500{\sim}2000\;ppm$에서 완전 혹은 상당한 증식 저지현상을 보였다. B. cereus는 황백 1000 ppm, 느릅뿌리 500 ppm에서 증식 저지를 보였고, L. plantarum은 황백 1000 ppm, L. mesenteroides는 500 ppm에서 완전 저지 되었으며 P. fluorescens는 황백, 느릅뿌리 및 질경이 추출물 2000 ppm에서 상당한 저지효과를 보였다. 항균성이 확인된 식물 추출물의 유효성분을 확인하기 위하여 클로르포름, 에틸 아세테이트, 부탄올 및 물로써 계속 분획하여 농축물을 얻고 이에 대하여 항균성을 시험한 결과 일반적으로 부탄올과 클로르포름 분획에서 얻어진 물질이 높은 항균성을 보였다. 항균성이 가장 높은 황백을 분획하여 농도별로 각 대상균에 대하여 항균성을 비교한 결과 B. cereus는 클로르포름 분획물 500 ppm, 에틸 아세테이트 및 부탄올 분획물 1000 ppm에서 완전 증식저지를 보였고, L. mesenteroides는 부탄올 분획물 500 ppm에서 완전 증식저지를 보였으나, P. fluorescens는 부탄올과 에틸 아세테이트 분획물 500 ppm에서 저지효과를 나타냈다.

  • Disease associated with microorganisms are far from resolved by current therapeutics. One of effective approach to health maintenance and disease control is the use of dietary bacterial and carbohydrate supplements. This comprises use of probiotics and prebiotics. Probiotics mean the live microorganisms, which when administered in adequate amounts confer a health benefit on the host. Prebiotics mean a nondigestible food ingredient that beneficially affects the host by selectively stimulating the growth and/or activity of one or a limited number of bacteria that can Improve the host health. Especially, probiotics has the relation which is close with innate immunity and adaptive immunity. And probiotics has the clinical value with many disease like lactose intolerance, constipation, acute gastroenteritis, food hypersensitivity and allergy, atopic dermatitis, crohn's disease, rheumatoid arthritis, pelvic radiotherapy, intestinal inflammation and chemical exposure, colon cancer, inhibitory effect of Helicobacter pylori and lowering the level of cholesterol. We use jointly korean medicine and probiotics and it has the more therapeutic effect in the many disease.

배양검사를 통해 확인한 중환자실 직원의 손씻기 효과 (Hand Hygiene Effects Measured by Hand Culture in Intensive Care Unit)

  • The effect of hand hygiene was measured by hand culture before and after hand hygiene for 86 nurses, doctors, and nurses aide/housekeepers in Surgical Intensive Care Unit. The subjects were asked to press their dominant hand in hand-shaped Mannitol salt agar immediately after patient contact and then washed their hand by preferred hand hygiene agents [soap and water, waterless alcohol gel, or 4% chlorhexidine gluconate detergent (CHG)], and cultured one hand again Amount of isolated microorganism was calculated by counting the number of divided areas ($1{\times}1cm$) which is culture positive in hand culture plate. The amount of microorganisms were significantly reduced from 58.1(${\pm}38.59$) to 27.4(${\pm}30.4$) cells after hand hygiene. The staff nurse's hand hygiene was more effective compared to medical doctors and nurses aide/housekeepers. Methicillin-resistant Staphylococcus aureus(MRSA) was isolated in 41(47.1%) subjects ; but only removed 100% in 28(32.2%) subjects. When the amount of hand microorganisms was compared by subject's preferred hand hygiene agents, it was decreased in order of 4% CHG, waterless alcohol solution, soap and water, and water. The hand hygiene practice was inadequate to reduce hand microorganisms and significantly different by occupations. Further research and development of hand hygiene improvement program which emphasize the quality of hand hygiene is recommended.

Bacteriological profiles of dressed broilers at different conditions and frozen storage periods

  • This study was conducted to determine the incidence of microorganisms associated with dressed broiler with intact skin and without skin at different frozen storage periods such as 0, 10, 20, 30 days and to demonstrate the role of packaging and pretreatment chilling on the changes of carcass quality. The values of total viable count (TVC), total coliform count (TCC), total streptococcal count (TStC) and total staphylococcal count (TSC) were determined for meat samples of thigh and breast and swab samples of visceral surfaces of the broilers with intact skin and without skin. It was observed that the values of TVC, TCC, TStC and TSC in both cases of dressed broiler with intact skin and without skin exceeded the International Commission on Microbiological Specification for Foods. However, numbers of microorganisms were considerably decreased during the frozen storage. Packing and prechilled conditions were generally better effective in decrease of the loads of microorganisms than without packing and prechilled conditions, and lower bacterial numbers were also found in dressed broiler with intact skin than that without skin. The highest sensory panel score was obtained at 10 days of frozen storage. These results, thus, indicate that usages of appropriate periods and conditions of frozen storage and packaging systems can minimize the potential health hazards associated with contaminants gaining access to the dressed or processed broilers and improve the quality and shelf life of dressed broilers.

유기물 생흡착 현상에 관한 기초연구 (Characteristics of Carbon Source Biosorption)

  • Biosorption technology was used to remove hazardous materials from wastewater, herbicide, heavy metals, and radioactive compounds, based on binding capacities of various biological materials. Biosorption process can be explained by two steps; the first step is that target contaminants is in contact with microorganisms and the second is that the adsorbed target contaminants is infiltrated with inner cell through metabolically mediated or physico-chemical pathways of uptake. Until recently, no information is available to explain the definitive mechanism of biosorption. The purpose of this study is to evaluate biosorption capabilities of organic matters using activated sludge and to investigate affecting factors upon biosorption. Over 49% of organic matter could be removed by positive biosorption reaction under anoxic condition within 10 minutes. The biosorption capacities were constant at around 50 mg-COD/mg-MLSS for all batch experiments. As starvation time increased under aerobic or anaerobic conditions, biosorption capacity increased since higher stressed microorganisms by starvation was more brisk. Starvation stress of microorganisms was higher at aerobic condition than anaerobic one. As temperature increased or easily biodegradable carbon sources were used, biosorption capacities increased. Consequently, biosorption can be estimated by biological -adsorbed capability of the bacterial cell-wall and we can achieve the cost-effective and non -residual denitrification with applying biosorption to the bio-reduction of nitrate.

철산화 박테리아의 생장 및 활성 억제를 통한 산성광산배수의 발생 저감 (Inhibition of Growth and Activity of Iron Oxidizing Bacteria for the Prevention of Acid Mine Drainage Production)

  • 폐광산의 산성배수(AMD)는 황철석을 비롯한 다른 금속 황화물의 산화를 통해 발생한 폐광산의 산성배수는 환경오염의 원인 중 하나이다. 본 연구에서는 이러한 폐광산의 산성배수가 생성되는 과정에서 산화미생물의 관여 정도를 알아보고, 이를 억제할 수 있는 방법에 대해여 살펴보았다. 산성배수 발생에 영향을 미치는 산화미생물로 Acidithiobacillus ferrooxidans, Acidithiobacillus thiooxidans을 선정하였으며, 이 산화미생물의 활성 및 생장 속도를 측정하였으며, 이산화염소$(ClO_2)$, NaCl, 그리고 계면활성제(ASOR-770) 를 산발생 억제제로 이용하여 실험을 진행하였다. 실험 결과 10ppm 이산화염소가 가장 효과적인 억제제였으며, 산화미생물의 활성도와 생장도를 20% 까지 감소시켜주었다.

Plant Growth Promoting Activities of Some Rhizosphere Bacteria and their Effect on Brassica rapa Growth

  • The necessity to develop economical and eco-friendly technologies is steadily increasing. Plant growth promoting rhizomicrobial strains PGPR are a group of microorganisms that actively colonize plant roots and increase plant growth and yield. Pot experiments were used to investigate the potential of some rhizobacterial strains to enhance the Brassica rapa growth. Microbial strains were successfully isolated from the rhizosphere of Panax ginseng and characterized based on its morphological and plant growth promotion characters. Surface disinfected seeds of Wisconsin Fast B. rapa were inoculated with the selected PGPR microorganisms. The different pots treatments were inoculated by its corresponding PGPR ($10^7cfu\;mL^{-1}$) and incubated in the growth chamber at $25^{\circ}C$ and 65% RH, the light period was adjusted to 24 hours (day). NPK chemical fertilizer and trade product (EMRO, USA) of effective microorganisms as well as un-inoculated control were used for comparison. Plants harvested in 40 days were found to have significant increase in leaf chlorophyll units and plant height and also in dry weight of root and shoot in the inoculated seedlings. Root and shoot length and also leaf surface area significantly were increased by bacterial inoculation in sterile soil. The study suggests that Rhodobacter capsulatus and Azotobacter chroococcum are beneficial for B. rapa growth as they enhance growth and induced IAA production and phosphorus solubilization. This study presents some rhizomicrobial strains that significantly promoted growth of Wisconsin Fast Plant B. rapa in pot experiment under different soil conditions.

Antimicrobial Activity of Essential Oil against Oral Strain

  • Objective: In this study, we tried to investigate the antimicrobial activity of natural herbaceous plant-derived essential oils against oral disease-causing bacterial strains and establish the scientific evidences to apply them to the dentifrice, mouthwash, and other uses. Methods: We used the disk diffusion method with 23 types of 100% natural essential oils to verify the antimicrobial effect against Streptococcus mutans, Porphyromonas gingivalis, and Lactobacillus rhamnosus. After adhering to the surface of the plate culture medium, $20{\mu}l$ of the essential oil was prepared, and the diameter of the transparent ring was measured after incubation for 24 hours. All the experiments were repeated three times. Results: Among 23 types of oils, 13 were effective against all three strains: myrrh, ginger, basil, carrot seed, tea tree, patchouli, ylang ylang, cypress, lemongrass, cinnamon, peppermint, lavender, and eucalryptus. Seventeen oils were effective against S. mutans, and myrrh, basil, and carrot seed showed high antimicrobial activity. Eighteen oils were effective against P. gingivalis, and tea tree, carrot seed, and cinnamons showed high antimicrobial activity. Sixteen oils were effective against L. rhamnosus, and carrot seed and peppermint cinnamon showed high antimicrobial activity. Conclusion: Consequently, 13 essential oils showed the antimicrobial activity against three bacterial strains, which indicates these essential oils might be used as the effective materials to suppress the growth of oral-disease inducing microorganisms. Moreover, essential oils that have been analyzed activities in this study will be mixed up within the constant range and analyzed their antimicrobial effects to examine the synergistic activity among them.

Discrimination of Bacillus anthracis Spores by Direct in-situ Analysis of Matrix-Assisted Laser Desorption/Ionization Time-Of-Flight Mass Spectrometry

  • The rapid and accurate identification of biological agents is a critical step in the case of bio-terror and biological warfare attacks. Recently, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry has been widely used for the identification of microorganisms. In this study, we describe a method for the rapid and accurate discrimination of Bacillus anthracis spores using MALDI-TOF MS. Our direct in-situ analysis of MALDI-TOF MS does not involve subsequent high-resolution mass analyses and sample preparation steps. This method allowed the detection of species-specific biomarkers from each Bacillus spores. Especially, B. anthracis spores had specific biomarker peaks at 2503, 3089, 3376, 6684, 6698, 6753, and 6840 m/z. Cluster and PCA analyses of the mass spectra of Bacillus spores revealed distinctively separated clusters and within-groups similarity. Therefore, we believe that this method is effective in the real-time identification of biological warfare agents such as B. anthracis as well as other microorganisms in the field.
