First Report of Charcoal Rot Caused by Macrophomina phaseolina on Glycine max in Korea (Macrophomina phaseolina에 의한 콩 균핵마름병(가칭) 발생)

  • Stem blight symptom of soybean was severely developed in 2016 in Hwaseong and Yeoncheon. During the seedling period, the damping-off of seedlings and the brown or black spots of cotyledons were observed. After August, the leaves began to be yellowed, and partially browned areas on leaves began to develop. After September, microsclerotia began to form even on the surface of the stems that had exhibited water-soaking symptom. After mid-October of the harvest season, the epidermis of the stem was peeled off, resulting in the formation of a large number of microsclerotia in the cortex. The pathogens isolated from these symptoms were the best in mycelial growth at 32-35℃, and the formation of microsclerotia was the most at 20-28℃. The pathogen was identified as Macrophomina phaseolina through the morphological characteristics of the pathogen and the sequencing of the internal transcribed spacer region gene. In addition, when inoculated with a soybean stem using toothpicks cultured with the pathogen, the same symptoms as seen on the soybean field occurred. When the pathogen was re-isolated at the lesion site, the same pathogen was isolated and identified as Macrophomina phaseolina. Based on the results, the disease is reported as soybean charcoal rot.

In vitro shoot propagation of Ranunculus kazusensis Makino, an endangered aquatic plant (멸종위기 수생식물인 매화마름(Ranunculus kazusensis Makino)의 기내 신초 증식)

  • A micropropagation method via callus for Ranunculus kazusensis Makino, an endangered species, was established. When stem segments were cultured on MS media supplemented with 1.0 mg/L IAA, NAA, IBA and 2,4-D, the highest frequency of callus induction was achieved on MS medium supplemented with 1.0 mg/L NAA. Multiple shoot per explant was obtained, the MS medium containing 1.0 mg/L BA and 0.5 mg/L NAA. Additionally, effect of activated charcoal (AC) and sucrose on shoot growth in in vitro culture were examined. The most suitable conditions for shoot growth after 4 weeks of culture were the MS medium with AC and sucrose. This in vitro propagation protocol will be valuable for conservation and mass propagation of this endangered plant.

Effects of Activated Charcoal on in vitro Ruminal Fermentation Characteristics and Nutrient Disappearances (사료에 대한 활성탄의 첨가가 in vitro 시험시의 발효성상 및 영양소 소실율에 미치는 영향)

  • An in vitro study was conducted to examine the effects of the addition of activated charcoal (AC) on the ruminal fermentation characteristics, nutrient disappearance, and ruminal gas production. AC was added at the levels of 0.00, 0.25, and 0.50 % to each of the four types of diets (roughage/concentrate ratio : 8/2, 6/4, 4/6 and 2/8), respectively. Although not significant, ruminal pH tended to increase by adding AC, and as the concentrate level increased, ruminal pH decreased (P<0.05). Acetate concentration and acetate/propionate molar ratio tended to decrease in AC diets. but molar % of propionate tended to increase by the addition of AC. Ruminal degradation of dry matter, crude protein, NDF, ADF, and hemicellulose in AC diets tended to increase than in non-AC diet, however, no tendency in ruminal degradation of crude fat was observed. As the concentrate level increased, rumunal degradation of dry matter and nutrients in AC diets increased significantly(P<0.05). Ruminal gas production tended to decrease in the 0.50 % AC diets, however, it tended to increase in high roughage diets. Although there appeared some beneficial effects in adding AC to ruminant diets in this study, more works should be done with AC before we can make clear conclusion on the use of AC in the ruminant diets.

Establishment of proliferation and regeneration system of PLBs in Phalaenopsis by treatments of a variety of types of medium, sucrose concentrations and anti-browning agents (다양한 배지종류, sucrose 농도 및 갈변억제물질 처리에 의한 팔레놉시스 PLB 증식 및 재분화 체계확립)

  • To establish an efficient proliferation and regeneration of PLBs (protocorm-like bodies) of Phalaenopsis plants, a variety of propagation medium types, various concentraions of sucrose as well as liquid and solid type were tested in this study. Further, activated charcoal, citric acid and ascorbic acid were compared whether these agents are suppose to reduce the browning in culture process using PLBs of Phalanopsis plants. With regard to the proper propagation medium, VW medium showed 1.3 ~ 2 times highr than those of other medium in an index of increasing for fresh weight and 50% higher than those of other medium in the frequency of shoot regeneration. However, regarding liquid and solid types of culture, there were no significant differences in the proliferation of PLBs and regeneration of shoots from PLBs. In the experiment for a variety of sucrose concentrations (0 ~ 50 g/l), 10 g of sucrose showed 30 ~ 50% higher than other concentrations in increasing index and 10 ~ 50% higher in the regeneration of shoots from PLBs. Regarding the reduction of browning in tissue culture via PLBs of Phalaenopsis plants, 1 g of activated charcoal showed only 1.5% browning of PLBs cultured. Whereas, other treatments including citric acid and ascorbic acid showed 6 ~ 16% of browning of PLBs. Therefore, activated charcoal was selected as an efficient anti-browning agents for the culture of PLBs in Phalaenopsis plants. Using above-described results can be contibuted to the establishment of mass propagation system using PLBs of Phalaenopsis plants in the future.

Culture condition for gametophyte and sporophyte masspropagation of bamboo fern (Coniogramme japonica) using tissue culture (조직배양을 이용한 가지고비고사리의 전엽체와 포자체 대량번식을 위한 배양조건)

  • This study was conducted to investigate the optimal culture method for gametophyte and sporophyte propagation in Coniogramme japonica (Thunb.) Diels, which can be used in various fields. The propagation of prothallus were cultured in 1/4 - 1 Murashige and Skoog medium and Knop medium for 10 weeks. The results indicated that the fresh weight of prothallus was the highest (14.5 g) in 1MS medium. Subsequently, various concentrations of sucrose, activated charcoal and nitrogen source were also added to 1MS medium and cultured for 8 weeks. The results provided that the sucrose concentration was 3% and the fresh weight of prothallus was the highest 10.8 g. According to the concentration in the range of 8.8 ~ 10.8 g, in the case of activated charcoal, the four treatments showed no significant difference. The nitrogen source was added at a concentration of 30, 60 and 120 mM with the ratio of ${NH_4}^+:{NO_3}^-$ being 1 : 2. As a result, the fresh weight of all treatments increased to similar level but there was no significant difference. We investigated sporophyte formation according to soil type and the highest number of sporophytes at 228.0 was formed in soils mixed with horticultural substrate and decomposed granite at 2 : 1 (v : v). On the other hand, sporophyte was not formed in soils containing peatmoss except for the one with peatmoss and decomposed granite at 2 : 1 (v : v).

Establishment of Pathogenicity Test Method for Macrophomina phaseolina Causing Soybean Charcoal Rot (콩균핵마름병균에 대한 병원성 검정법 확립)

  • The establishment of a laboratory assay and a greenhouse assay was conducted for evaluating the pathogenicity of Macrophomina phaseolina causing soybean charcoal rot established. In the laboratory assay, microsclerotia and hyphae were used as inoculum. In the laboratory assays using microsclerotia as an inoculum, disease incidences of M. phaseolina NSW17-108 and HSM17-034 were higher at 35℃ than 25℃. Of the two isolates NSW17-108 and HSM17-034, the disease incidence of HSM17-034 isolated from diseased sesame is higher than that of NSW17-108 isolated from diseased soybean. When the mycelia of M. phaseolina were used as an inoculum, the disease incidence of NSW17-108 and HSM17-034 at 35℃ exceeded 80% even after only 5 days of inoculation. Even at 25℃, furthermore, that of HSM17-034 exceeded 80% 5 days later. In the pathogenicity assays at a greenhouse, toothpicks where microsclerotia were produced or microsclerotia harvested from potato dextrose agar medium were used as an inoculum. In all greenhouse assays, M. phaseolina NSW17-108 and HSM17-034 showed 40-60% of disease incidences 35-65 days after inoculation with the pathogen, depending on the inoculation method. Between the two isolates, the pathogenicity of HSM17-034 was stronger than that of NSW17-108, and this result was consistent with laboratory assay results. Since the laboratory and greenhouse test methods tested in this study have different advantages and disadvantages depending on each test method, it is thought that the test method that can meet the purpose of the study should be selected and used.

A Study on the Traditional Dyeing of Korean buddhist Monk′s Robe (한국승복 염색에 대한 연구)

  • The philological consideration of Korean Buddhist Monk's robe and its reviving have been investigated in traditional way. The configuration and colour of Korean Buddhist monk's robes have been properly adapted for an period, territory, and atmospheric phenomena. In case of Korea, introduced Buddhism from China realize modern robe from under the influence of fusion of Chinese ornament culture and Korean traditional ornament culture. As a result of this modern robe has been consisted of a Buddhist monk's robe and a cope. The modem robes has been significantly affected by industrialization, contrary to ancient time taxed robes as public imposts. At present be familiar in mass production we have a preference for the easier way in making and even color forming. In this paper it is focused on dyeing part, which recognized its important in latest time, at first declined according to appearance of chemical dye, evaluated its convenience in use. That is, it is increased natural dye, people begin with recognition of difference of physical properties for dress dyed with chemical pigment and that with natural pigment. In consequence, I have presented that both making colour of gray using traditional method, and three demolished-color prescribed by Buddhist law. Of course, though it become generally know that dyeing of robes occupied significant part of traditional natural dyeing. But in case of religion, it also is important to know symbolical meaning involved in its colour. Most of Buddhism-believers or even Buddhists who actually be dress in robe are without knowing the meaning of colour, or its revealing method. There, I have considered mating three colour of demolished- color included blue, black, and red, and also represented dyeing method of Buddhist monk's robe using both charcoal and ink-spick. From religious font of view, as robes contains involving of blessing, I have an intention of improvement way in dyeing and succession and development of traditional culture through consideration of traditional dyeing method. And I have also intended to restoration the value of robe's colour, disappeared by the time. moreover, I have shown that dyeing with natural pigment is profitable for both environment and sanitary aspect.

Adsorption Characteristics of Charcoals of Major Korean Wood Species and Wood-based Materials (국산 주요 수종 및 목질재료 탄화물의 흡착 특성)

  • This research was to investigate the adsorption characteristics of charcoals of major Korean wood species (thinned trees) and wood-based materials. As carbonization time and temperature increased, methylene-blue adsorption (MBA) of charcoals of thinned trees and wood-based materials increased. Pinus koraiensis charcoal showed the highest MBA. MBA of softwoods was higher than that of hardwoods. There was a negative correlation between air-dried specific gravity before carbonization and MBA of carbonized thinned trees. The MBA of wood-based materials showed similar value, but wood-based materials in MBA was lower than thinned woods. Surface area and total pore volume of thinned trees and wood-based materials increased as carbonization temperature increased. The species showing highest MBA appeared to have the highest surface area at the carbonization temperature of 600℃ as well. There was a positive correlation between surface area, total pore volume and MBA of charcoals. The charcoal of wood-based materials generally exhibited micro pores.

  • A Korean fairy tale, "I live on my fortune" is discussed and interpreted from the perspective of analytical psychology. Analytical psychology understands fairy tales are stories in which the workings of archetypes of unconsciousness are represented symbolically. Therefore they are intended to achieve psychic wholeness compensating collective consciousness. The fairy tale is found in almost every region of Korea. There are several parallel stories that share similar motifs, such as Samgongbonpoori, a mythological story and a tale on the king Mooh. The father in the story asked his three daughters whose fortune they lived on. The youngest was expelled by her father for she answered, "I live on my own fortune." Then she came into a humble cottage in the mountain marrying a poor charcoal maker and she found gold there. She became rich, made the charcoal maker a decent learned man and seeked for and took care of her parents who had become beggars. This fairy tale is interpreted as a story about individuation process of a woman who integrated 'inferior' aspects and unconscious animus through actively accepting the suffering from being expelled and commitment to the unconscious world, and also about how rigid collective psyche, under patriarchal system, changes toward wholeness through the workings of the unconscious.

The Effect of Culture Methods and Plant Growth Regulators on Bulblet Formation and Growth in Scale Segment Culture of Fritillaria thunbergii Miq. (패모 인편 배양시 자구 형성과 비대에 미치는 배양 방법과 생장 조절제의 처리 효과)

  • This experiment was carried out to establish a year-round production system of pathogen-free stock through micropropagation in Fritillaria thunbergii as medicinal bulbous plant. The effect of different types of culture method and plant growth regulators, activated charcoal and mannitol on bulblet formation and subsequent growth were investigated. The MS solid medium containing 1. 0 mg/L kinetin and 0. 3 mg/L NAA was effective on bulblet formation and propagation rate compared to liquid and suspension culture. Addition of activated charcoal at 0. 01% to 0. 1% in the medium promoted bulbing of cultured bulblets and shoot formation. Addition of 1% to 2% mainnitol in MS medium was effective on the formation of bulblet and subsequent growth of bulblets compared to control. In addition of inhibitors, $10{\sim}100\;mg/L$ B-9 and Chloromequat had effective to stimulate bulblet growth but their effects were not so much as mannitol treatment. ABA treatment had detrimental effect on survival rate of explant and bulblet formation.
