Influence of Dietary Beef Tallow and Dextrin on the Growth and Body Composition of Juvenile Far Eastern Catfish Silurus asotus (배합사료의 우지와 덱스트린이 메기(Silurus asotus)의 성장과 체조성에 미치는 영향)

  • This study investigated the utilization of beef tallow and dextrin as non-protein energy sources in the diet of juvenile far eastern catfish Silurus asotus. Three replicated groups of juveniles (initial mean weight of 3.6 g) were fed diets containing high levels of beef tallow or dextrin for six weeks. The final mean weight of fish fed on a 17% lipid diet with a high level of beef tallow was significantly higher than that of fish fed on a 9% lipid diet with a high level of dextrin (P=0.04). No significant differences were observed in the feed efficiency and protein efficiency ratio of fish fed on the experimental diets. Crude lipid content and the C18:0, C18:1n-9 and C18:2n-6 contents of whole-body fish were significantly affected by diet (P<0.05). The results of this study suggest that beef tallow is better than dextrin as a non-protein energy source for improving the growth of juvenile far eastern catfish, and that beef tallow could be economically used as an ingredient in practical feed.

  • Di-2-ethylhexyl phthalate (DEHP) is a widely used plasticizer known to be a suspected xenoestrogen and a causative agent of oxidative damage to the RBC cell membrane ir vitro. We evaluated the toxic effects of a scarcely documented aquatic environmental pollutant, DEHP, on selected haematological endpoints in the bagrid catfish, Pseudobagrus fulvidraco. Bagrid catfish were exposed to DEHP (300, 1,000 mg DEHP kg body weight$^{-1}$) through thrice intraperitoneal injection and effects were assessed in blood of the exposed organisms. Haematological property, serum organic and inorganic chemistry were monitored in blood of Bagrid catfish. DEHP exposed-fish showed erythropenia; low hematocrit (Ht) and hemoglobin (Hb) content and red blood cell count showed a significantly higher than in that of the control group. The treatment group showed a significantly lower concentration of serum total protein, cholesterol and triglyceride compared with those in the control group. The value of calcium and osmolality were significantly decreased in the DEHP treatment group, compared with the control group.

Comparison of Dietary Protein Levels on Growth Performance of Far Eastern Catfish Silurus asotus and Water Quality in Biofloc Technology and Flow-through Systems (바이오플락과 유수식 시스템에서 메기(Silurus asotus) 배합사료내 단백질 함량에 따른 성장도 및 수질 비교 연구)

  • Biofloc technology (BFT) systems provides an additional protein source for fish by converting waste in microbial flocs. Therefore, we aimed to determine the difference in the protein levels of the diet of Far Eastern catfish Silurus asotus reared in BFT and FW systems. We applied a two-way (six protein levels × two aquaculture systems) ANOVA design. We formulated six experimental diets to contain different protein levels (CPs)- 20%, 25%, 30%, 35%, 40% and 45%-referred to as the CP20, CP25, CP30, CP35, CP40 and CP45 diets, respectively. We randomly distributed 900 fish into 36 (18 each for BFT and FW), 300 L tanks (25 fish/tank) with three replications of each diet for 4 weeks. The weight gain (WG) and specific growth rate (SGR) considerably improved as the dietary protein level increased in the formulated diets. The BFT fish showed higher growth performance with any experimental diet than the FW fish. The growth performance of the fish fed the BFT-CP35 diet was higher than that of the fish fed the FW-CP45 diet. In conclusion, the cultivation of Far Eastern catfish (S. asotus) in BFTs can be carried out under reduced dietary protein levels.

Ultrastructural Changes of Oocyte in Korean Catfish, Silurus asotus (발달 단계에 따른 한국산 메기(Silurus asotus) 난모세포의 미세구조적 변화)

  • This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 1-year-old female catfish(Silurus asotus). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were observed by light and transmission electron microscope. All data were collected from October in 1992 to May in 1993. The size of the nucleoli and number of the yolk granules increased as the oocyte grew. Yolk granules were deposited in the oocyte as fluid. Due to the presence of large early and late maturing oocytes, their ovaries were large, transparent, granular, and greenish in color. As the percentages of fish in LMO and RO stage increased from March to April, mean of GSI values(19.95%) increased. Follicle cells such as granulosa cell and thecal cell change a squamous into cuboid shape in LPO and EMO stage. Processes, microvilli, from the granulosa cells and from the oocyte grow and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radiate becomes more compact and devoid of pore canals during oocyte maturation. The electrophoretic pattern of major band in mature stage was much thicker(21k, 24k, 32k, 45k, 67∼110k, 170k dalton) than that in previtellogenic phase.

The Toxic Effects of Parathion on Survival and Growth of Juvenile Bagrid Catfish (Pseudobagrus fulvidraco) (동자개 (Pseudobugrus fulvidraco) 치어의 생존 및 성장에 미치는 Parathion의 영향)

  • The toxic impact of parathion on bagrid catfish (Pseudobagrus fulvidraco) juvenile was examined 96 hr $LC_{50}$ value of parathion estimated as 1 90 mg/L. Chronically, bagrid catfish were exposed for 30 days to concentrations of parathion, i.e. 63, 95, 190 and $380\;{\mu}g/L$, representing 3.3, 5.0, 10.0 and $20.0{\%}$ of the 96 hr $LC_{50}$. Survival rate was significantly affected by the concentration over $63\;{\mu}g/L$ after 10 days. Growth rate, specific growth rate (SGR) and feed efficiency were reduced in time as concentration dependent manner. Growth rates of the exposed groups were significantly reduced as $28.06-60.63{\%}$ to the control fish, SGR was decresed as $1.50-4.72{\%}$ and feed efficiency was declined as $6.10-11.90{\%}$. It can be concluded that if p. lulvidraco is chronically exposed to parathion over $63\;{\mu}g/L$ concentration, the physiological damages might affect growth factors of the fish.

  • Transesterification of fat of Tra catfish with methanol in the presence of the KOH catalyst yields fatty acid methyl esters (FAME) and glycerol (GL). The effects of the reaction temperature and reaction time on rate constants and kinetic order were investigated. Three regions were observed. In the initial stage, the immiscibility of the Tra fat and methanol limited the reaction rate, hence this region was controlled by the mass transfer. Subsequent to this region, produced FAME like a co-solvent made the reaction mixture homogeneous, therefore the conversion rate increased rapidly so it was controlled by the kinetic parameters of the reaction until the equilibrium was approached in the final slow region. A second-order kinetic mechanism was proposed involving second regions for the forward reaction. The rate determining step for the overall KOH catalyzed-methanolysis of Tra fat was the conversion of triglycerides (TG) to diglycerides (DG). This rate constant was increased from 0.003 to $0.019min^{-1}$ when the reaction temperature was increased from 35 to $60^{\circ}C$. Its calculated activation energy was 14.379 ($kcal.mol^{-1}$).

Renal effect of experimental feeding of melamine and cyanuric acid in different concentrations on Japanese catfish (Silurus asotus) (멜라민과 cyanuric acid의 농도별 혼합투여에 따른 메기(Silurus asotus) 신장에서의 조직병리학적 소견)

  • The recent outbreak of renal failure in infants in China and in animals in USA and Europe has been determined to be caused by melamine adulterated in the food. In the course of the investigation, cyanuric acid was identified in addition to melamine in the offending food. Fish feeds were also recently found to be contaminated with melamine. The purpose of this study was to characterize the histopathological effect and toxicity potential of different concentrations of melamine and cyanuric acid in the kidney of Japanese catfish (Silurus asotus). The fish were administered melamine and cyanuric acid in combination at the concentrations of 12.5, 25, 50, 100 and 200 mg/kg/day for 3 days by single oral administration dissolved in carboxymethyl cellulose. The results showed that renal crystals were observed in renal tubules and collecting ducts at the concentration over 25 mg/kg dose group and the number of crystals in kidney were in proportion to the concentrations of melamine and cyanuric acid.

  • The Aeromonas strains were isolated from diseased cultured catfsh (Silurus asotus) in the ponds near by Gochang of Chunbuk. The present isolates were identified as A. hydrophila based on their biological and biochemica1 characteristics. The isolates of A. hydrophila were named GC-1, GC-2, GC-3, GC-4 and GC-5, Five strains were grew optimally at temperatures $35^{\circ}C,$ pH 7.5 and in 0.5 to l% NaCl. This bacterium(GC-1) was injected into health catfish in order to prove the causative agent of ascites and haemorrhagic ulcers. The symptoms in cat fish infected by this challenge method were observed to be very similar to the symptoms of a natural infection, but controls did not show any abnormal symptoms during the experimental period. At 24 post-injection, the red spot developed around the injection site and the haemorrhagic ulcers was extended near the gill and ventral fin. Five strains were tested for drug sensitivity by plate method. All strains were sensitivity to gentamicin and resistant to penicillin, streptomycin and tetracycline.

Ultrastructure of Egg Micropyles and Zona Radiata in Three Aquacultural Teleosts (양식산 경골어류 3종의 난문과 방사대의 미세구조)

  • Ultrastructure of the zona radiata and the micropyle of ripe eggs in the rainbow trout (Oncorhynchus mykiss), catfish (Silurus asotus) and loach (Misgurnus anguillicaudatus) were examined by light, scanning and transmission microscopes. The egg micropyle of rainbow trout and catfish consists of a funnel-shaped vestibule and a tapered canal transversing the zona radiata. The micropyle of rainbow trout and catfish showed the similar structure with flat pit and long canal. The micropylar wall showed the clockwise spiral structure in rainbow trout. The micropyle of the loach showed the type with a hollow pit leading into a short canal and the micropylar wall showed the counterclockwise spiral structure. There are numerous and various size pores at the surface area around the opening in every experimented fish. Interconnecting ridges were observed in the unfertilized eggs of every fish.

  • Viability, early growth and sex ratio of induced gynogenetic diploid bagrid catfish (Leiocassis ussuriensis, Siluriformes) were examined. Survival rates of gynogenetic diploids and normal bi-parental diploids up to 180 days post hatching were not significantly different (P>0.05), and consequently resulted the mean percent survivals of 76% (gynogens) and 78% (bi-parental progeny) at the end of growth trial. Growth in both body weight and total length of gynogenetic diploids was also similar to those of normal diploid controls (P>0.05). Gynogenetic bagrid catfish displayed a similar level of ovarian development when compared to bi-parental diploid females, as judged at 10 weeks post hatching. Sex ratio of bi-parental progeny was normal 1:1 whereas all-femaleness was observed in gynogenetic progeny (P<0.05), suggesting that the genetic sex determination mechanism of this species should be XX-XY based female homogamety.
