Lectin histochemistry of the olfactory mucosa of Korean native cattle, Bos taurus coreanae

  • Background: The olfactory mucosa (OM) is crucial for odorant perception in the main olfactory system. The terminal carbohydrates of glycoconjugates influence chemoreception in the olfactory epithelium (OE). Objectives: The histological characteristics and glycoconjugate composition of the OM of Korean native cattle (Hanwoo, Bos taurus coreae) were examined to characterize their morphology and possible functions during postnatal development. Methods: The OM of neonate and adult Korean native cattle was evaluated using histological, immunohistochemical, and lectin histochemical methods. Results: Histologically, the OM in both neonates and adults consists of the olfactory epithelium and the lamina propria. Additionally, using periodic acid Schiff and Alcian blue (pH 2.5), the mucus specificity of the Bowman's gland duct and acini in the lamina propria was determined. Immunohistochemistry demonstrated that mature and immature olfactory sensory neurons of OEs express the olfactory marker protein and growth associated protein-43, respectively. Lectin histochemistry indicated that numerous glycoconjugates, including as N-acetylglucosamine, mannose, galactose, N-acetylgalactosamine, complex type N-glycan, and fucose groups, were expressed at varied levels in the different cell types in the OMs of neonates and adults at varying levels. According to our observations, the cattle possessed a well-developed olfactory system, and the expression patterns of glycoconjugates in neonatal and adult OMs varied considerably. Conclusions: This is the first study to describe the morphological assessment of the OM of Korean native cattle with a focus on lectin histochemistry. The findings suggest that glycoconjugates may play a role in olfactory chemoreception, and that their labeling properties may be closely related to OM development and maturity.

Ultrasonographic Appearance of Liver and Duodenum in Relation to Growth in Native Korean Cattle (한우에서 성장에 따른 간장 및 십이지장의 초음파상)

  • This study was performed to make fundamental data of ultrasonographic diagnosis for liver and duodenum disease in native Korean cattle. Ultrasonographic appearance of liver, caudal vena cava, portal vein, gall bladdr and duodenum according to the growth were determined from 6 to 13 months by monthly examinations in 9 native Korean cattle. The caudal vena cava was determined at 11th intercostal space by use of ultrasonography. The portal vein and gallbladder were determined at 10th intercostal space by use oof ultrasonography. The liver were determined at 12, 11 and 10 th intercostal space by use of ultrasonography. The liver, caudal vena cava, portal cein, gallbladder and duodenum were examinde in standing position. Ultrasonograms were obtainde with 3.5 or 5.0-MHz convex and 5.0-MHz sector transducer. The diameter of vena cava at the 6, 7, 8, 9, 10, 11, 12 and 13 months old age were 18.4, 20.6, 22.4, 25.1, 26.9, 27.3, 28.5 and 29.4 mm, respectively. The diameter of portal vein at the 6, 7, 8, 9, 10, 11, 12 and 13 months old age were 19.5, 20.6, 22.3, 24.5, 26.9, 28.1, 30.7 and 31.5 mm, respectively. The diameter of gall bladder at the 6, 7, 8, 9, 10, 11, 12 and 13 months old age were 63.2, 72.2, 75.1, 78.6, 80.5, 82.3, 84.4 and 91.1 mm, respectively. Cranial part of duodenum was identified near gall bladder by moving of hyperechoic ingesta. Descending duodenum paralleled with vertebrae is seen in the right upper flank and was adjacent to the abdominal wall, and was enveloped in the hyperechoic greater omentum, differentiating it from the jejunum and ileum. The diameter of cranial duodenum at the 6, 7, 8, 9, 10, 11, 12 and 13 months old age were 23.3, 27.9, 29.9, 32.2, 34.4, 34.5, 35.1 and 36.5 mm, respectively. The diameter of descending duodenum revealed smaller diameter than that of cranial duodenum. Calculi in gallbladder were observed by ultrasonography.

  • Ultrasonographic diagnosis of kidney and urinary bladder disease was performed in native Korea cattle. Ultrasonogaphic appearence of kidney and urinaryy baldder according to the growth were determined from 6 to 13 months by monthly examinations in 9 native Korean cattle. The kidney and urinary bladder were examined in standing position. The position, dimension, and structure of both kidneys were determined at 12 intercostal space and paralumbar fossa by use of ultrasonography. Ultrasonograms were obtained with 3.5 or 5.0-MHz sector transducer. The width and depth of the kidney, and the diameter of renal parenchyma and renal sinus were observed. Both kidneys were observedfrom the body surface until 8 months old age, but over the 9 months old age, only right kidney was easily accessible. The width of right kidney at the 6, 7, 8, 9, 10, 11, 12 and 13 months old age were 5.7, 6.3, 6.9, 7.6, 8.4, 9.3, 9.8 and 10.7 cm, respectively. The depth 5.8, 6.1 and 6.6 cm, respectively. The size of left kidney was similar to right kidney until 8 months old age. After the 9 months old age, only right kidney was easily accessible. The circumference of urinary bladder at the 6, 7, 8, 9, 10, 11, 12 and 13 months old age were 19.1, 21.7, 22.5, 23.0, 24.2, 25.3, 27.8 and 29.1 cm, respectively. Calculi in urinary bladder were observed by ultrasonography.

Identification of Stearoyl-CoA Desaturase (SCD) Gene Interactions in Korean Native Cattle Based on the Multifactor-dimensionality Reduction Method

  • Fat quality is determined by the composition of fatty acids. Genetic relationships between this composition and single nucleotide polymorphisms (SNPs) in the stearoyl-CoA desaturase1 (SCD1) gene were examined using 513 Korean native cattle. Single and epistatic effects of 7 SNP genetic variations were investigated, and the multifactor dimensionality reduction (MDR) method was used to investigate gene interactions in terms of oleic acid (C18:1), mono-unsaturated fatty acids (MUFAs) and marbling score (MS). The g.6850+77 A>G and g.14047 C>T SNP interactions were identified as the statistically optimal combination (C18:1, MUFAs and MS permutation p-values were 0.000, 0.000 and 0.001 respectively) of two-way gene interactions. The interaction effects of g.6850+77 A>G, g.10213 T>C and g.14047 C>T reflected the highest training-balanced accuracy (63.76%, 64.70% and 61.85% respectively) and was better than the individual effects for C18:1, MUFAs and MS. In addition, the superior genotype groups were AATTCC, AGTTCC, GGTCCC, AGTCCT, GGCCCT and AGCCTT. These results suggest that the selected SNP combination of the SCD1 gene and superior genotype groups can provide useful inferences for the improvement of the fatty acid composition in Korean native cattle.

Effects of Sawdust and Rice Hulls Litter on the Waste Management of Hanwoo (Korean Native Cattle) (톱밥이용 한우 분료처리에 관한 연구)

  • Litters such as sawdust and rice hulls are used to manage the waste excrements of Hanwoo(Korean Native Cattle). This experiment was conducted to investigate the adequate depth of bedding materials, and the effective substitutive rate of rice hulls and the recycled-dried sawdust mixed with cattle waste for sawdust from 1993 to 1994. The results were as follows; 1. The most adequate bedding depth of sawdust was 4cm and it was able to be utilized for 38 days. The cost of required sawdust was lower in order of 5, 15, 20 and 10cm in depth of bedding sawdust. 2. Rice hulls was able to be fully substituted for sawdust, but 50% substitution the most effective. 3. Recycled-dried sawdust mixed with cattle waste could be substituted less than 25% for sawdust.

The SNP of WBP1 is associated with heifer reproductive performance in the Korean native cattle Hanwoo

  • It is well documented that intensive selection in dairy cattle for economic value such as increased milk yield led to a decline in reproductive performance. Recent studies using genome-wide association studies (GWASs) discovered candidate genes involved in the lower fertility including embryo development and conception rates. However, the information, which showed a lower reproductive performance, is limited to dairy cattle, especially Holstein, and the candidate genes were not examined in the Korean native cattle Hanwoo which has been intensively selected and bred for meat in the last few decades. We selected the candidate genes WBP1 and PARM1 reported to be associated with cow and/or heifer conception in dairy cattle and analyzed the genotype because those genes have non-synonymous single nucleotide polymorphisms (SNPs). To determine the single base change, we used the high resolution melting (HRM) assay which is rapid and cost-effective for a small number of genes. We found that most heifers with higher conception (1: service per conception) have the AA genotype coding Threonine rather than Proline in the WBP1 gene. We did not detect an association for a SNP in PARM1 in our analysis. In conclusion, the genetic variation of WBP1 can be used as a selective marker gene to improve reproductive performance, and HRM assay can be used to identify common SNP genotypes rapidly and cost effectively.

Evaluation of Ultrasound for Prediction of Carcass Meat Yield and Meat Quality in Korean Native Cattle (Hanwoo)

  • Three hundred thirty five progeny testing steers of Korean beef cattle were evaluated ultrasonically for back fat thickness (BFT), longissimus muscle area (LMA) and intramuscular fat (IF) before slaughter. Class measurements associated with the Korean yield grade and quality grade were also obtained. Residual standard deviation between ultrasonic estimates and carcass measurements of BFT, LMA were 1.49 mm and $0.96cm^2$. The linear correlation coefficients (p<0.01) between ultrasonic estimates and carcass measurements of BFT, LMA and IF were 0.75, 0.57 and 0.67, respectively. Results for improving predictions of yield grade by four methods-the Korean yield grade index equation, fat depth alone, regression and decision tree methods were 75.4%, 79.6%, 64.3% and 81.4%, respectively. We conclude that the decision tree method can easily predict yield grade and is also useful for increasing prediction accuracy rate.

Reproduction and marketing plans for improving profitability of Korean native cattle (Hanwoo) farm (한우 농가 수익성 향상을 위한 번식 및 출하 계획)

  • Wholesale beef price is the critical factor for determining Korean native cattle, Hanwoo, farm's income in short-term. Wholesale beef price has seasonality due to high demand in Korean traditional holidays such as Korean thanksgiving day and lunar new year's day. Therefore, it is important to make reproduction and marketing plans for Korean Hanwoo farmers, in order to increase their farm income. However, there is no study available on changes in the expected farm income depending on reproduction and marketing schedules. This study analyzed the expected farm income per head depending on the monthly-based marketing schedules. The analysis was conducted based on the seasonality of wholesale beef price, reproduction efficiency, operating costs, relationship between carcass grade and slaughter age. The result shows that slaughter Hanwoo at the age of 29 months-old in August and January generating the highest expected farm income per head.

Prevalence of Johne's disease of Korean native cattle in Jeju Province, Korea (제주지역 한우의 요네병 감염실태 조사)

  • The bacterium causing Johne's disease (JD), Mycobacterium avium subspecies paratuberculosis (MAP), is distributed worldwide and causes progressive intestinal disease, also known as paratuberculosis, in domestic and exotic ruminants, including cattle. The subclinical form of this disease results in progressive weight loss, reduced milk production, lower slaughter value, and premature culling, with possible impacts on fertility and udder health. Eventually, infection can lead to the clinical form that manifests chronic diarrhea, emaciation, debilitation, and eventual death. JD is a major cause of economic loss in cattle industry. In this study, serological testing was conducted by ELISA to determine the prevalence of MAP in Korean native cattle of Jeju during the duration of 2010 to 2013. A total of 5,150 blood sera were collected in 111 farms and the samples were classified into four different age groups for MAP; group 1 (2 years or less), group 2 (2~4 years), group 3 (5~7 years), and group 4 (over 8 years). Overall seroprevalence of MAP was 5.2% (269/5,150), and annually from 2010 to 2013, they were 11.6% (106/915), 2.1% (10/474), 5.5% (89/1,627), and 3.0% (64/2,134), respectively. Also, the prevalence rates was determined by age groups: 48 of group 1 (17.8%), 154 of group 2 (57.3%), 48 of group 3 (17.8%), and 19 of group 4 (7.1%) were seropositive for MAP. Although seropositive samples were found in all age groups, the highest seroprevalence was found in group 2. This study showed that during the investigation the seroprevalence of MAP tended to decrease.

  • Some hematological and biochemical indices were assayed in Korean native cattle naturally harbouring Fasciola hepatica infection and compared with uninfected controls. Affected animals revealed reduction in total erythrocyte count and hemoglobin content, and increase in total leukocyte count. Infected cattle had significantly lower levels of serum total protein, serum albumin and magnesium. Significantly higher values were obtained for the serum enzymes aspartate aminotransferase and alanine aminotransferase.
