Serological Survey of Leptospiral Antibody in Dairy and Korean Native Cattle (Leptospira 속균(屬菌)에 대한 한우(韓牛) 및 유우(乳牛)의 혈청항체조사(血淸抗體調査))

  • A serological survey for antibodies to Leptospira (L.) interrogans in dairy and Korean native cattle in Gyeongbuk area was performed using 6 different living antigens, which were L. icterohaemorrhagiae(RGA), L. canicola(Hond Utrecht IV), L. autumnalis(Akiyami A), L. australis(Ballico), L. pomona(Pomona) and L. hebdomadis(Hebdomadis), by microscopic agglutination test. In the microscopic agglutination test, greater than partial agglutination at a serum dilution of 1:300 or over was recorded as positive. In the dairy cattle(Holstein), four(4.7%) of 86 sera from 13 dairy farms were positive for L. canicola and L. pomona antibodies. In the Korean native cattle, four(3.2%) of 124 sera from the slaughter house in Taegu city were positive for L. canicola, L. icterohaemorrhagiae and L. pomona antibodies. Among the positive sera, L, canicola was dominated in this experiment.

Relationship between Single Nucleotide Polymorphisms in the Peroxisome Proliferator-Activated Receptor Gamma Gene and Fatty Acid Composition in Korean Native Cattle

  • The peroxisome proliferator-activated receptor gamma ($PPAR{\gamma}$) gene plays an important role in the biosynthesis process controlled by a number of fatty acid transcription factors. This study investigates the relationships between 130 single-nucleotide polymorphisms (SNPs) in the $PPAR{\gamma}$ gene and the fatty acid composition of muscle fat in the commercial population of Korean native cattle. We identified 38 SNPs and verified relationships between 3 SNPs (g.1159-71208 A>G, g.42555-29812 G>A, and g.72362 G>T) and the fatty acid composition of commercial Korean native cattle (n = 513). Cattle with the AA genotype of g.1159-71208 A>G and the GG genotype of g.42555-29812 G>A and g.72362 G>T had higher levels of monounsaturated fatty acids and carcass traits (p<0.05). The results revealed that the 3 identified SNPs in the $PPAR{\gamma}$ gene affected fatty acid composition and carcass traits, suggesting that these 3 SNPs may improve the flavor and quality of beef in commercial Korean native cattle.

A Genetic Marker for the Korean Native Cattle (Hanwoo) Found by an Arbitrarily Primed-Polymerase Chain Reaction (AP-PCR)

  • In order to develop a specific genetic marker for the Korean native cattle (Hanwoo), an arbitrarily-primed polymerase chain reaction (AP-PCR) analysis of 6 different cattle breeds was attempted. Eight different arbitrary primers, each longer than 20-mer nucleotides, were used. In comparison to the AP-PCR patterns, several distinctive DNA bands that are specific for a certain breed were detected. When the primer Kpn-X was employed, a 280bp DNA fragment was found to be specific only for Hanwoo. In an individual analysis of Hanwoo, this AP-PCR marker was observed in 123 head of cattle among the 153 that were tested (80.4%). Nucleotide sequencing revealed that this fragment has a short microsatellite sequence of tandem repeat, $A(G)_{1-2}\;(C)_{1-3}AGAG$. According to the analysis of AP-PCR band patterns, Hanwoo was discovered to be genetically most closely-related with Holstein among the various cattle breeds.

Use of Frequencies of Micronuclei as Biological Dosimetry in Korean Native Cattle and Goat Lymphocytes after Irradiation in vitro (한우 및 한국재래산양 유래 말초혈액 림프구의 미소핵을 이용한 방사선 피폭의 생물학적 선량측정)

  • The frequencies of gamma-ray-induced micronuclei (MN) in cytokinesis-blocked (CB) lymphocytes at several doses were measured in three donors of Korean native cattle and Korean native goat. Measurements performed after irradiation showed a dose-related increases in MN frequency in each of the donors studied. When analysed by linear-quadratic model the line of best fit was cattle : y : 0.1016D +$0.0118D^2$+0.0147, goat : y = 0.1353D +$0.0043D^2$+0.0087 (y : number of MN/CB cells and D = irradiation dose in Gy). The relative sensitivity of goat lymphocytes compared with cattle lymphocytes was estimated by best fitting linear-quadratic model based on the radiation-induced MN data over the range from 0 Gy to 4 Gy. In the case of MN frequency with 0.05, 0.1, 0.2, 0.4 and 0.8, the relative sensitivities of goat lymphocytes were 1.106. 1.166. 1.140, 1.069 and 0.976 respectively. Our in vitro radiobiological study confirmed that the cytogenetic response obtained in blood from cattle and goat can be utilized for application in environmental studies.

  • Blood samples were obtained from Korean native cattle and dairy cattle of Holstein species in the slaughter house and methylene blue color tests were performed for the detection of the inherited susceptibitity to hemolysis. Glucose-6-phosphate dehydrogenase activities expressed as the optical density obtained by methylene blue color test were the highest as 0.54 in male Korean cattle, 0.62 in female Korean cattle and 0.72 in dairy cattle of Holstein species. Percent hemolysis, packed cell volume and plasma protein contents were measured and compaired with relation to the results of methylene blue color test and no correlation were observed in each.

  • Enteritis occured in a herd of Korean native cattle in Gyeonbug district in February 1977. of 40 cattle ranging in age from 15 to 20 months, 17 had been sick for the past six days with main clinical findings of feces containing mucus and blood. Among affected cattle, feces of 8 cattle were examined bacteriologically and parasitologically to investigate the causative agent. The disease was diagnosed as bovine coccidiosis caused by Eimeria zurnii based on the detection of $o{\ddot{o}}cysts$ and one of cattle was infected E. bukidnonensis, too.

An outbreak of bovine coccidiosis in Korean native cattle (한우 콕시듐병의 발생)

  • Authors observed an outbreak of bovine coccidiosis at a farm stock in Gyeongnam, Korea, which were diagnosed on the ground of clinical and fecal examination with the following results. Twenty one of thirty four Korean native cattles evacuate feces containing blood and mucus or evacuate entirely of clots of fresh and red blood. On fecal examination showed eimeria zuernii. The patients were recovered with sulfamethazine and amprolium for 5 days. From these results the outbreak was considered to be typical bovine coccidiosis.

A histological study on the development of scapula in the Korean native cattle (한우 태자의 견갑골 발생에 관한 조직학적 연구)

  • This study was undertaken to establish the sequence of development of ages and its time of the fetal endochondral ossification in the scapula of the Korean native cattle. This study was also designed to confirm through histological observation the earliest stages of both chondrification and ossification. Thirty eight scapulae, a series of embryos and fetuses from the pregnant Korean native cattle ranging from 11 to 110mm in crown-rump (C-R) length, were used. The following results were obtained. The ossification center was observed in the supra- and infra- spinous fossa in the 5th group (CRL 51-60mm), that was markedly ossified in the 6th group (CRL 61~70mm) by Alizarin red S stain. The chondrogenic center of scapula was observed in the 1st group (CRL 11~20mm). The primary ossification center was presented in the 4th group (CRL 41~50mm). In the 5th group(CRL 51~60mm), the endochondral ossification progressed actively. Alcianophility was markedly increased in the interterritorial matrix in the 3rd group (CRL 31~40mm. However this reaction was markedly decreased in the interterritorial matrix the adjacent portion to the marrow cavity and trabecula in the 5th group (CRL 51~60mm).

Epitheliogenesis imperfecta in a bovine fetus of Korean native cattle (한우에서의 불완전상피발생)

  • In this study, we describe a case of epitheliogenesis imperfecta (EI) observed in the fetus of Korean native cattle. The fetus had multifocal areas of skin defect, especially on the distal portions of the four limbs, and the affected areas were bright-red and glistening. Histopathologically, these areas were characterized by complete absence of squamous epithelium, infiltration of inflammatory cells into the dermis, atrophy of hair follicles, sebaceous and sweat glands. To the best of our knowledge, this is the first report of epitheliogenesis imperfecta in Korean native cattle.

  • Ultrasonographic diagnosis of genital disease and early pregnancy diagnosis was performed in Korean native cattle. The size of ovarian follicle in preovulation, luteal stage and follicular cyst was 18.9, 9.2 and 27.6 mm, respectively, and the thickness of follicular wall was 2.3, 1.8 and 2.8 mm, respectively. The size of corpus luteums in formation stage, activity stage, regression stage, cystic corpora lutea and luteal cyst was 6.2, 11.3, 8.6, 26.7 and 25.9 mm, respectively. The thickness of luteal wall in cystic corpora lutea and luteal cyst was 8.4 and 4.9 mm, respectively. The size of embryo or fetus on day 25, 27, 30, 35, 40, 45 and 50 was 0.8, 0.9, 1.3, 1.5, 2.2, 2.8 and 3.8 cm, respectively. The size of amniotic vesicle on day 25, 27 and 30 was 1.2, 2.1 and 3,0 cm, respectively. The diameter of pregnant uterus on day 25 and 27 was 7.0 and 7.8 cm, respectively. It was concluded that the ultrasonographci values determined in this study can be used as references for the treatment of genital disease and early pregnancy diagnosis in Korean native cattle.
