Development of Efficient Screening Methods for Melon Plants Resistant to Fusarium oxysporum f. sp. melonis (멜론 덩굴쪼김병에 대한 효율적인 저항성 검정법 개발)

  • This study was conducted to establish an efficient screening system to identify melon resistant to Fusarium oxysporum f. sp. melonis. F. oyxsporum f. sp. melonis GR was isolated from infected melon plants collected at Goryeong and identified as F. oxysporum f. sp. melonis based on morphological characteristics, molecular analyses, and host-specificity tests on cucurbits including melon, oriental melon, cucumber, and watermelon. In addition, the GR isolate was determined as race 1 based on resistance responses of melon differentials to the fungus. To select optimized medium for mass production of inoculum of F. oxysporum f. sp. melonis GR, six media were tested. The fungus produced the most spores (microconidia) in V8-juice broth. Resistance degrees to the GR isolate of 22 commercial melon cultivars and 6 rootstocks for melon plants were investigated. All tested rootstocks showed no symptoms of Fusarium wilt. Among the tested melon cultivars, only three cultivars were susceptible and the other cultivars displayed moderate to high resistance to the GR isolate. For further study, six melon cultivars (Redqueen, Summercool, Superseji, Asiapapaya, Eolukpapaya, and Asiahwanggeum) showing different degrees of resistance to the fungus were selected. The development of Fusarium wilt on the cultivars was tested according to several conditions such as plant growth stage, root wounding, dipping period of roots in spore suspension, inoculum concentration, and incubation temperature to develop the disease. On the basis of the test results, we suggest that an efficient screening method for melon plants resistant to F. oxysporum f. sp. melonis is to remove soil from roots of seven-day-old melon seedlings, to dip the seedlings without cutting in s pore s uspension of $3{\times}10^5conidia/mL$ for 30 min, to transplant the inoculated seedlings to plastic pots with horticulture nursery media, and then to cultivate the plants in a growth room at 25 to $28^{\circ}C$ for about 3 weeks with 12-hour light per day.

Effects of Ozone-Water Washing on the Quality of Melon (오존수 세척이 포장 참외의 품질에 미치는 영향)

  • For increasing shelf-life, melons with different washing and packaging were evaluated during storage at room temperature. Hardness and weight of melon was decreased during storage period and, after 12 days, severe rotten or decay appeared on melons except melon with ozone-water. Organoleptic test on color, appearance, taste and texture showed maximum values on melons eith ozone-water washing. Particularly, spots have been shown on all packaged melons in the later storage, except melons washed with ozone-water. According to these results, ozone-water washing was effective for keeping the high quality of melons.

  • Investigation on oriental melon was carried out for 30 minutes starting at 7 PM every day from March 21 to May 24 to find out the effect of light emitting diode on seedling quality, grafting, growth and flowering of oriental melon. According to the result of the investigation, plant height was longer in Blue, Infrared, Red+Blue and Red treatment and leaf number was higher in Blue, Red+Blue and Infrared treatment than those of control. No big difference was identified between control and Yellow, Green, Ultraviolet treatments. Grafting rate was high in Green, Red+Blue and Green treatment. The number of flower every week in control was nine, the number was almost 1 higher in White and Ultraviolet A treatments, but it was 1 to 4 lower in the rest of treatments. The number of female flowers of control was 10, however, it was 21 in Infrared treatment, 17 in White, 15 in Ultraviolet, 13 in Red+lnfrared, 12 in Blue and Red+Blue, 11 in Yellow and 8 in Green.

Establishment of Pre-Harvest Residue Limit for Buprofezin and Penthiopyrad during Cultivation of Oriental melon (Cucumis melon var. makuwa) (참외(Cucumis melon var. makuwa)에 대한 Buprofezin 및 Penthiopyrad의 생산단계 잔류허용기준 설정)

  • The present work was aimed to determine the pre-harvest residue limits (PHRLs) and the safety management of commonly used pesticides namely buprofezin and penthiopyrad on oriental melon (Cucumis melon var. makuwa). In this study, the buprofezin (diluted two thousand fold) and penthiopyrad (diluted four thousand fold) were sprayed single time on oriental melon in the cultivation areas Sangju (site 1) and Sungju (site 2). Oriental melon were randomly collected from the both areas at the end of 0 (2 hours after pesticides spaying), 1, 2, 3, 5, 7, 9 and 10 days. For analysis, each samples were partitioned twice (80 and 70 mL) with dichloromethane and purified by florisil SPE cartridge. Finally, the residual amounts of both pesticides in all samples were analyzed using gas chromatography/nitrogen phosphorus detector (GC/NPD). In this study, the method limit of quantification (MLOQ) for both buprofezin and penthiopyrad in oriental melon was found to be $0.01mg\;kg^{-1}$ and their recovery levels were 91.1~98.6% and 90.0~104.6%, respectively. Further, the calculated biological half-life for buprofezin and penthiopyrad in oriental melon were 3.9 and 3.5, and 3.0 and 2.7 days in site 1 and 2, respectively. The results of this study found that the PHRLs for buprofezin and penthiopyrad were 4.24 and $2.31mg\;kg^{-1}$, respectively at 10 days before harvest. Consequently, the present study suggest that the residual amounts of both pesticides will be lower than the maximum residue limits (MRLs) when oriental melon is harvested.

SNP Marker Development for Purity Test of Oriental Melon and Melon (멜론 및 참외 순도 검정을 위한 SNP 마커 개발 및 F1 종자 순도 검정)

  • Field screening method has been commonly used for purity test of $F_1$ hybrid seeds in melon and oriental melon. However, as this method takes a lot of time and cost, molecular marker-based purity test is necessary. To develop molecular markers for purity test, thirty pairs of SNP (single nucleotide polymorphism) primers were obtained from melon EST sequences, and 10 polymorphic markers showing HRM (high resolution melting) polymorphisms between parents of two melon cultivars and one oriental melon cultivar were selected. Blind tests were performed to validate usefulness of the selected markers for purity test. Blind test results showed that HRM genotypes were matched with the expected identity of individual sample, $F_1$ hybrid, male or female parents. Three HRM-based SNP markers were converted to CAPS markers for general use which is favor to breeders. We expect that SNP markers developed in this study will be useful for purity test of $F_1$ hybrid seeds in melon and oriental melon.

Beneficial Effect of Heat Fans on Quality and Yield of Korean Melon Cultivated in Greenhouses at Winter Season (히터팬 처리가 저온기 하우스 참외의 품질 및 수량에 미치는 긍정적 영향)

  • The purpose of this study was to investigate the changes of environmental conditions and the quality and yield of melon fruit by heat fan operation in greenhouses at winter season. The average daily temperature inside the tunnels during January 1 to 31, 2017 was $0.9^{\circ}C$ higher than that of the control $17.8^{\circ}C$. The air flow rate of heater fan treatment was 4.8 times higher than the control (untreated $0.05m{\cdot}s^{-1}$) at 20cm above the ground where the korean melon grew. The temperature of the heater pan was $5.6^{\circ}C$ higher than that of the untreated at $35.3^{\circ}C$ and the relative humidity was 8.1% lower than that of the untreated at 39.1%. The flowering rate of the heater fan treatment was 96%, 5% higher than the control. The number of first harvest days of heater fan treatment was shortened by 4 days than that of untreated treatment. Fruit quality and marketable fruit yield increased by 3.4% and 38% compared to untreated respectively, the heater fan treatment increased the temperature inside the greenhouse and air flow rete, which were beneficial for growing the korean melon in greenhouses at winter season.

Quality Characteristics and Volatile Flavor Compounds of Oriental Melon Wine Using Freeze Concentration (동결농축 참외와인의 품질 특성과 휘발성 향기 성분)

  • In this study, the physicochemical properties, antioxidant capacities, and volatile flavor compounds of oriental melon wine prepared by freeze concentration after heat treatment (HA), ascorbic acid treatment (AAT), and heat and ascorbic acid treatment (HAAT) were investigated. During fermentation period, the melon wine by HAAT showed greater reduction of soluble solids and reducing sugar contents compared to other treatments. In addition, the melon wine treated with HAAT also showed a higher L value and lower browning index compared to other treatments. After aging, free sugar including fructose, and organic acids including citric acid, succinic acid, and malic acid were detected in all samples. For antioxidant activities and contents, HAAT treated wine showed greater antioxidant activities and total phenolic contents than those of others. In GC/MS analysis, a total of 33 volatile flavor compounds were identified. In the principal component analysis of volatile flavor compounds, principal components 1 and 2 represented 88.15% of the whole date distribution and showed opposite tendencies. Taken together, HAAT enhanced the antioxidant activities and sensory properties of oriental melon wine. Moreover, freeze concentration gave the different volatile flavor characteristics in oriental melon wine.

Monitoring of the Changes in Volatile Flavor Components in Oriental Melon Wine Using SPME (SPME를 이용한 참외와인의 휘발성 향기성분의 모니터링)

  • This study was conducted to investigate changes in the main volatile flavor components of oriental melon during the process of alcohol fermentation via SPME (solid phase micro extraction). The flavor components of oriental melon were shown to have mainly included melon and green flavors. The green flavor was identified to be nonanal, 1-butanol, 1-octen-2-ol and benzene, and its relative concentration was shown to be 16.66%. The nonanal concentration was shown to have been reduced among the green-flavor components, but no significant change in remaining components was observed. Mainly, sweet flavor tended to increase, and the relative concentration of benzene was particularly shown to have increase by 25.58%, accounting for the highest relative concentration. The amount of green-flavor components, except for 1-butanol, was shown to have significantly decrease after alcohol fermentation. Then, no component of green-flavor, which causes an offensive smell, was found during fermentation and aging. Meanwhile, the volatile flavor components, which are consist of acids, were shown to have been produced during alcohol fermentation. In particular, octanoic acid, which causes off-flavor, was shown to be 60.99%, a very high relative concentration during the aging stage. In addition, acetic acid with a pungent sour flavor tended to be produced. A further study on the improvement of flavor in the production of oriental melon wine is required.

Characteristics of Reflective Light over Red and Black Plastic Mulch, and Effect on the Quality and Yield of the Oriental Melon and Tomato (적색 비닐멀칭과 흑색 비닐멀칭의 반사광 특성과 참외와 토마토의 품질과 수량에 미치는 영향)

  • BACKGROUND: Plastic mulches widely used in raised-bed culture mainly to conserve water, control weeds and raise soil temperature. The most widely used plastic mulch colour is black. Reflective red and far-red light can affect on growth and yield of various vegetable crops. Objectives of this study were to investigate the characteristics of reflective light of black and red plastic mulches, and to evaluate the reflective red and far-red light on the quality characteristics and yield of the Oriental melon (Cucumis. Melo L.) and tomato (Lycopersicon esculentum L.). METHODS AND RESULTS: Oriental melon and tomato were cultivated over the reflective red and black plastic mulches in plastic house. Reflected red and far-red light over the red plastic mulch were 2.6 times higher than those of black plastic mulch. Red to F-Red ratio of black plastic mulch, red plastic mulch and sunlight were 1.14, 0.93 and 1.16 respectively. Intensity of reflected red and far-red light over red plastic mulch were highest at surface height of 30 cm. The higher the height of the surface decrease the intensity of far-red light. Accordingly, Red to F-Red ratio were increased. Reflective red plastic mulch increased the weight of fruit and content of sugar in Oriental melon and tomato. CONCLUSION(s): Yield of Oriental melon over reflective red plastic mulch was higher than that of black plastic mulch. These results suggested that reflected red and far-red light over the red plastic mulch affected allocation of photosynthate in growing Oriental melon.

The Establishment of Critical Ranges of Inorganic Nutrition Contents in Leaves of Net Melon(Cucumis melo L.) in Protected Cultivation (시설재배 멜론 엽의 적정양분함량 설정)

  • This study was carried out to elucidate the relationship between yields and inorganic nutrient contents, and then establish of critical range of inorganic nutrient contents in leaves of net melon in protected cultivation in Naju from 2004 to 2006. In considering the yields and nutrient contents of net melon, the critical ranges of macro-nutrient contents in leaves of net melon at 25 days after transplanting were in N 3.4~4.8%, P 0.4~0.7%, K 3.4~5.1%, Ca 2.7~4.1% and Mg 0.6~0.8%, respectably. The critical ranges of micronutrient content, such as Fe, Mn, Zn, Cu and B, were not found the regular trend with different growth stages. However, the critical range of micronutrient contents in leaves of net melon at 25 days after transplanting were in Fe 102~164, Mn 52~116, Zn 28~98, Cu 7~15 and B $36{\sim}72mg\;kg^{-1}$, respectably. Finally, these results might be used at the indicator for critical nutrient contents for diagnosis of nutritional disorder in net melon in protected cultivation.
