  • Objectives : The aim of this experiments is to provide an objective differentiation of ginseng, Korean and Chinese cultivated wild ginseng, and natural wild ginseng through components analysis of different parts of ginseng. Methods : Comparative analyses of ginsenoside-$Rg_3$, ginsenoside-$Rh_2$, and ginsenosides $Rb_1$ and $Rg_1$ from the root, stem, and leaves of ginseng, Korean and Chinese cultivated wild ginseng, and natural wild ginseng were conducted using HPLC. Results : 1. For content comparison of leaves, ginseng showed highest content of ginsenoside $Rg_1$ than other samples. Natural wild ginseng showed relatively high content of ginsenosides $Rg_1$ and $Rb_1$ than other samples. 2. For content comparison of the stem, ginseng and 10 years old Chinese cultivated wild ginseng didn't contain ginsenoside $Rb_1$. Natural wild ginseng showed higher content of ginsenosides $Rg_1$ and $Rb_1$ than other samples. 3. For content comparison of the root, ginsenoside $Rh_2$ was found only in 5 and 10 years old Korean cultivated wild ginseng. 4. Distribution of contents by the parts of ginseng was similar in ginseng and Chinese cultivated wild ginseng. Conclusions : Above experiment data can be an important indicator for the identification of ginseng, Korean and Chinese cultivated wild ginseng, and natural wild ginseng.

  • The ginsenoside content was compared with wild simulated ginseng (Panax ginseng) collected every season at 11 wild simulated ginseng plantations in Korea. As a result, the total saponin of 7 years old wild simulated ginseng showed the highest content of 4.5% in spring sampling wild simulated ginseng, 2.0% in summer sampling wild simulated ginseng, 1.2% in winter sampling wild simulated ginseng and 1.0% in autumn sampling wild simulated ginseng. And also, the total saponin of 10 years old wild simulated ginseng showed the highest content of 3.9% in spring sampling wild simulated ginseng, summer sampling wild simulated ginseng (1.8%), winter sampling wild simulated ginseng (1.6%) and autumn sampling wild simulated ginseng (0.6%). Therefore, the total saponin of spring sampling wild simulated ginseng was about 4.5 - 6.5 times higher than that of autumn sampling wild simulated ginseng regardless of cultivation period.

  • Objectives: This study aimed to examine the changes in ingredients according to its cultivation method by examining the content of saponin and non-saponin components of ginseng. Methods: Ginseng saponin component analysis was compared and reviewed using the high-performance liquid chromatography method, and acidic polysaccharide component was measured using the carbazole sulfuric acid method. Results: The comparative analysis of ginseng saponin content of 4 and 6 years old fresh ginseng showed the following results. According to the cultivation method, upland field cultivation fresh ginseng showed higher average content of crude saponin than paddy field cultivation fresh ginseng. Whereas, paddy field cultivation fresh ginseng showed higher average content of total saponin than upland field cultivation fresh ginseng. Ginsenoside Rb1 showed higher content of paddy field cultivation fresh ginseng than upland field cultivation fresh ginseng in 6 years old ginseng. However, it showed higher content of upland field cultivation fresh ginseng than paddy field cultivation fresh ginseng in 4 years old ginseng. Additionally, ginsenoside Rg1 showed higher content of paddy field cultivation fresh ginseng than upland field cultivation fresh ginseng in 6 years old ginseng, whereas upland field cultivation fresh ginseng showed higher content of paddy field cultivation fresh ginseng in 4 years old ginseng. The effect on the content of ginseng saponins and acidic polysaccharides according to paddy field and upland field cultivation methods is considered to be small. Conclusions: The paddy field cultivation method, which is more efficient in production cost due to depletion of farmland and long-distance cultivation, is considered to be an economical cultivation method.

  • In the browning reaction of Korean ginseng, it appears that enzymatic and non-enzymatic browning reaction occurred in the initial stage of heating fresh ginseng at low temperature, and then non-enzymatic browning reaction followed in the drying period after heating. Activation energy of the browning reaction for red ginseng was about 9.0 kcal/mol. Browning reaction of red ginseng was accede- rated with an increase in steaming time, and a great extent of browning reaction occurred between 60-90 min of steaming at 10$0^{\circ}C$. Browning pigments of red ginseng were mostly water soluble subset.

  • A pathogenesis-related protein (PgPR5) gene that isolated from the leaf of Panax ginseng was characterized. The ORF is 756 bp with a deduced amino acid sequence of 251 residues. The calculated molecular mass of the matured protein is approximately 27.5 kDa with a predicated isoelectric point of 7.80. A GenBank BlastX search revealed that the deduced amino acid of PgPR5 shares highest sequence similarity to PR5 of Actinidia deliciosa (80% identity, 87% similarity). PgPR5 has a C-terminal and N-terminal signal peptide, suggesting that it is a vacuolar secreted protein. The expression of PgPR5 under various environmental stresses was analyzed at different time points using real-time PCR. Our results reveal that PgPR5 is induced by salt stress, chilling stress, heavy metal, UV, and pathogen infection. These results suggest that the PgPR5 could play a role in the molecular defence response of ginseng to abiotic and pathogen attack. This is the first report of the isolation of PR5 gene from the P. ginseng.

  • Cinnamoyl-CoA reductase (CCR, EC 1.2.1.44) catalyses the reduction of cinnamic acid CoA esters into their corresponding aldehydes, the first step of the phenylpropanoid pathway specially dedicated to monolignol biosynthesis. A cDNA clones encoding CCR have been isolated from Panax ginseng C.A. Meyer and its expression was investigated in response to abiotic stresses. The cDNA, designated PgCCR which is 865 nucleotides long and has an open reading frame of 590 bp with a deduced amino acid sequence of 176 residues. The PgCCR encoded protein possesses substantial homology with CCRs isolated and cloned from other sources; the highest identity (51.8%) was observed with CCR from Tomato (Lycopersicon esculentum). Under various stress conditions, expression patterns of the PgCCR were highly induced in adventitious and hairy roots by several abiotic stresses. These results indicated that PgCCR plays protective role against diverse environmental stresses.
