Bovine fecal microbiota is important for host health and its composition can be affected by various factors, such as diet, age, species, breed, regions, and environments. The objective of this study was to evaluate the impact of diet and gender on fecal microbiota in Korean native Hanwoo cattle. The 16S rRNA gene amplicon sequencing of fecal microbiota was conducted from 44 Hanwoo cattle divided into four groups: (1) 11 heifers fed an oat hay plus total mixed ration (TMR) diet for breeding (HOTB), (2) 11 heifers fed an early fattening TMR diet (HEFT), (3) 11 steers fed the early fattening TMR diet (SEFT), and (4) 11 steers fed the late fattening TMR diet (SLFT). Firmicutes and Bacteroidota were the first and second most dominant phyla in all the samples, respectively. The Firmicutes/Bacteroidota (F/B) ratio associated with feed efficiency was significantly greater in the SLFT group than in the other groups. At the genus level, Romboutsia, Paeniclostridium, and Turicibacter were the most abundant in the SLFT while Akkermansia, Bacteroides, and Monoglobus were the most abundant in the HOTB group. Although the same early fattening TMR diet was fed to Hanwoo heifers and steers, Marvinbryantia and Coprococcus were the most abundant in the HEFT group while Alistipes and Ruminococcus were the most abundant in the SEFT group. Shannon and Simpson diversity indices were significantly lower in the SLFT group than in the other groups. Distribution of fecal microbiota and functional genetic profiles were significantly different among the four treatment groups. The present study demonstrates that different diets and genders can affect fecal microbiota and the F/B ratio may be associated with feed efficiency in Hanwoo cattle. Our results may help develop strategies to improve gut health and productivity through manipulation of fecal microbiota using the appropriate diet considering Hanwoo cattle gender.
In this research dairy cattle barn facilities what are 4,198 houses hold over 50 heads were surveyed by scale and province. Full-time farms hold over 50 heads breed total 344,514 heads. Each of Farms holds 50 to 99 heads were 79.8 percent and breed average 82.1 heads. Dairy cattle barns were constructed september 1995 averagely. Each of barns have $1,740.0\;m^2$ scale. The construction type of dairy cattle barn was almost litter barn type 84.0%, freestyle type 5.1%, mooring+litter ground type 17.3% and other types 4.4%. The litter barn type was popular in small farms. But in large farms, freestyle type was popular than small farms. The construction type of dairy cattle barn was almost litter barn type 84.0%, freestyle type 5.1%, moohng+ltter ground type 17.3% and other types 4.4%. Type of dairy cattle robotic milking system was pipeline 41.5%, herringbone 22.8% and tandem 35.8%. The pipeline type was popular in small farms which have 50~99 heads. But in large farms which have over 200 heads, tandem type was popular than small farms. Proportion of floor type of dairy cattle barn was almost litter type 94.9%. Scraper type was popular in large farms than in small farms. Proportion of roof type of dairy cattle barn was slate 32.5%, vinyl 16.3%, sunlight 11.1%, panel 10.9, zinc plate 8.8 and steel plate 8.3%. Roof type was lots of slate type before 1995. But vinyl type is increasing after 1995. Proportion of wall type of dairy cattle barn was almost open type 83.3% and winch-curtain 26.8%. Utilization period of dairy cattle barn was 9.2 years about milker, 7.9 years about automatic feeder, 9.2 years about waterer and 10.4 years about electric facilities. In this results, there were lots of improvements about automatic feeder.

Sixty Four fresh and 142 frozen embryos of dairy cattle were transferred to synchronized dairy, beef or Korean Native Cattle nonsurgically at National Animal Breeding Institute from 1985 to 1990. The results obtained were as follows ; 1. The pregnancy rate of fresh embryos(39.1%) was higher than that of frozen embryos(32.4%) and average pregnancy rate was 34.5%. 2. The pregnancy rate of grade 1 embryos was higher than that of grade 2 embryos for both fresh(41.3% vs 33.3%) and frozen embryos(35.4% vs 25.6%). 3. The pregnancy rate according to development stage of fresh embryos was increased with maturity as 29.2%, 33.3%, 50.0% and 54.5% for morula, early blastocyst, blastocyst and expanded blastocyst, respectively. For frozen embryos, the pregnancy rate of blastocyst(44.4%) was higher than those of morula(31.3%) and early blastocyst(28.0%). 4. The pregnancy rate according go recipient-donor synchrony for fresh embryos was higher when the recipients exhibited estrus 1 day earlier than the donors(43.8%) than when the recipients exhibited estrus 1 day later than the donors(38.1%) or when the recipients and donors exhibited estrus at the same time(37.0%). For forzen embryos, the pregnancy rate was decreased when the recipients and donors exhibited estrus at the same time(37.9%), when the recipients exhibited estrus 1 day later than the donors(32.0%) and when the recipients exhibited estrus 1 day earlier than the donors(23.5%), in sequence. 5. The pregnancy rate of heifers was higher than that of cows for both fresh(50.5% vs 37.9%) and frozen embryos(39.7% vs 25.7%). 6. The pregnancy rate according to recipient breed for fresh embryos was higher in dairy cattle(42.1%) and beef cattle(40.%) than in Korean Native Cattle(33.3%). For frozen embryos, the pregnancy rate was decreased beef cattle(39.1%), dairy cattle(30.3%) and Korean Native Cattle(14.3%), in sequence. 7. The pregnancy rate according to equilibrium steps of glycerol and freezing rate was higher when transferred after 3-steps equilibrium and freezing by the rate of $0.3^{\circ}C$/min from $-6^{\circ}C$ to $-35^{\circ}C$ and $0.1^{\circ}C$/min to $-38^{\circ}C$(39.4%) than when transferred after 6-steps equilibrium and freezing by the rate of $0.5^{\circ}C/min$ from $-6^{\circ}C$ to $-30^{\circ}C$(30.3%).
Korea, located in East Asia in the northern hemisphere, is experiencing severe climate changes. Specifically, the heat stress caused by global warming is negatively affecting the dairy sector, including milk production and reproductive performance, as the major dairy cattle Holstein-Friesian is particularly susceptible to heat stress. Here, we collected artificial insemination and pregnancy data of the Holstein and the Jersey cows from a dairy farm from 2014 to 2021 and analyzed the association between the conception rate and the temperature-humidity index, calculated using the data from the closest official weather station. As the temperature-humidity index threshold increased, the conception rate gradually decreased. However, this decrease was steeper in the Holstein breed than in the Jersey one at a temperature-humidity index threshold of 75. To evaluate the effects of heat stress on the oocyte quality, we examined the nuclear and cytoplasmic maturation of Holstein (n = 158, obtained from six animals) and Jersey oocytes (n = 123, obtained from six animals), obtained by ovum pick-up. There were no differences in the nuclear maturation between the different conditions (heat stress: 40.5℃, non- heat stress: 37.5℃) or breeds, although the Holstein oocytes seemed to have a lower metaphase II development (p = 0.0521) after in vitro maturation under heat stress conditions. However, we found that the Holstein metaphase II oocytes exposed to heat stress presented more reactive oxygen species and a peripheral distribution of the mitochondria, compared to those of the Jersey cattle. Here, we show that weather information from local meteorological stations can be used to calculate the temperature-humidity index threshold at which heat stress influences the conception rate, and that the Jersey cows are more tolerant to heat stress in terms of their conception rate at a temperature-humidity index over 75. The lower fertility of the Holstein cows is likely attributed to impaired cytoplasmic maturation induced by heat stress. Thus, the Jersey cows can be a good breed for the sustainability of dairy farms for addressing climate changes in South Korea, as they are more resistant to hyperthermia.

Objective: Although Hanwoo has been selected as the superior commercial beef cattle breed in Korea, Chikso (Korean brindle cattle) is still recognized as a valuable breed for beef production. The aim of this study was to compare the meat quality, fatty acid composition and aroma volatiles of beef from Chikso and Hanwoo steers maintained under identical feed management, as information regarding these characteristics is still limited. Methods: A total of 19 carcasses with a quality grade of 1 were selected, and strip loin (longissimus lumborum) cuts were collected from 11 Hanwoo carcasses and 8 Chikso carcasses. Meat quality and aroma analyses were performed at day four postmortem. Results: Though Hanwoo strip loin tended to have higher fat content (15.37%) than Chikso (12.01%), no significant differences were observed. Meat pH, water-holding capacity, cooking loss, shear force value, instrumental surface color (Commission International De L'eclairage $L^{\star}$, $a^{\star}$, $b^{\star}$, chroma, and hue angle) and fatty acid composition were not significantly different. Roasted Chikso beef released more intense aroma than roasted Hanwoo beef based on the total area units of identified volatiles. Among identified volatiles, the amounts of toluene, heptanal, octanal, and nonanal were higher in roasted Chikso beef than in roasted Hanwoo beef. In addition, the aroma pattern of the roasted beef from these breeds was well-discriminated by electronic nose. Conclusion: No distinct differences were found in terms of meat quality between Hanwoo and Chikso beef in this study. However, the aroma pattern and volatiles of roasted Hanwoo and Chikso beef were different according to instrumental analysis.
Due to the large amount of beef imported from the USA to Korea, Korean consumers have become increasingly interested in the country of origin since it can affect market prices. Previously, Bos indicus and Bos taurus-specific markers were developed for the purpose of cattle breed identification, specifically discrimination of Australian beef. In this study, six SNP markers derived from Illumina 50K bovine SNP chip data were used for the discrimination between Korean cattle (Hanwoo) and imported beef from USA. PCR-RFLP genotyping methods were also developed, which indicates that these markers can be applied relatively easily compared to other markers. Taking into account a discrimination rate of 55% based on MC1R marker between Hanwoo and imported beef from USA, two additional markers, SNPs 23803 and 34776, were ideal and resulted in probability of identification of 0.942 and probability of misjudgment of 0.03. Therefore, the markers developed in this study can greatly contribute to the correct discrimination between beef from USA and Hanwoo beef.
As an initial step toward a better understanding of the genome structure of Korean cattle (Hanwoo breed) and initiation of the framework for genomic research in this bovine, the bacterial artificial chromosome (BAC) end sequencing of 21,024 clones was recently completed. Among these clones, BAC End Sequences (BESs) of 20,158 clones with high quality sequences (Phred score ${\geq}20$, average BES equaled 620 bp and totaled 23,585,814 bp), after editing sequencing results by eliminating vector sequences, were used initially to compare sequence homology with the known bovine chromosomal DNA sequence by using BLASTN analysis. Blast analysis of the BESs against the NCBI Genome database for Bos taurus (Build 2.1) indicated that the BESs from 13,201 clones matched bovine contig sequences with significant blast hits (E<$e^{-40}$), including 7,075 single-end hits and 6,126 paired-end hits. Finally, a total of 5,105 clones of the Korean cattle BAC clones with paired-end hits, including 4,053 clones from the primary analysis and 1,052 clones from the secondary analysis, were mapped to the bovine chromosome with very high accuracy.
To estimate the genetic characteristics and cumulative power of discrimination (CPD) existing among Hanwoo (Korean cattle) and exotic foreign population (Angus, Herford, Charolais, Holstein) we used a total of 414 genomic DNAs from five breeds population (Hanwoo, Angus, Hereford, Charolais, Holstein). Genetic characteristics indices including mean allele number among loci, unbiased heterozygosity ($h_i$) within locus and polymorphic information content (PIC) and unbiased average heterozygosity (H) among loci in four breeds were calculated using the generated allele frequencies by each marker. The mean allele numbers for all loci ranged between 5 and 7 while heterozygosity (H) ranged from 0.75 (HW) to 0.64 (HF) among loci and across breeds heterozygosity (H) was 0.69. The generated unbiased average heterozygosity among loci in each breed was integrated to the global formula of CPD resulting in 99.71 % within the populations. The genetic variation of HW (Hanwoo) showed highest estimates among the analyzed breeds.
This study was deal with the development of breed-specific DNA marker which is able to identify Hanwoo and European cattle breeds(Non-Hanwoo) meat. Genetic differentiation between Korean cattle(Hanwoo) and European cattle breeds was examined by Random Amplified Polymorphic DNA(RAPD) analysis. The RAPD patterns were identical among Non-Hanwoo, such as Holstein, Hereford, Aberdeen Angus, Brown Swiss, Limousin or Simmental, but the above pattern was different from that of Hanwoo. All bands detected in the Hanwoo samples were observed in Non-Hanwoo cattle samples, but one of the common bands found in samples was not detected in the Hanwoo samples. The band(1.4kb) may be useful as a marker for identifying a meat of Hanwoo from imported cattle meat. Actually, the detection of the DNA marker was tested by DNA analysis with 929 samples which were prepared from bloods of 673 Hanwoo cattles and 141 Holstein cattles, from 115 imported cattle meats. The DNA marker was absent in 644 of 673 Hanwoo cattles (96%) but present in 245 of 256 Non-Hanwoo cattles (95%). These results show that the DNA marker is effective to characterize Hanwoo and Non-Hanwoo meat by its detection. This DNA marker, however, was not useful in detecting unwanted crossbreeding between two cattle breeds, because the band pattern in hybrid cattle shows one of two band patterns in Hanwoo and Non-Hanwoo.

Background: Research on loci influencing milk production traits of dairy cattle is one of the main topics of investigation in livestock. Many genomic regions and polymorphisms associated with dairy production have been reported worldwide. In this context, the purpose of this study was to identify candidate loci associated with milk yield in Argentinean dairy cattle. A database of candidate genes and single nucleotide polymorphisms (SNPs) for milk production and composition was developed. Thirty-nine SNPs belonging to 22 candidate genes were genotyped on 1643 animals (Holstein and Holstein x Jersey). The genotypes obtained were subjected to association studies considering the whole population and discriminating the population by Holstein breed percentage. Phenotypic data consisted of milk production values recorded during the first lactation of 1156 Holstein and 462 Holstein ${\times}$ Jersey cows from 18 dairy farms located in the central dairy area of Argentina. From these records, 305-day cumulative milk production values were predicted. Results: Eight SNPs (rs43375517, rs29004488, rs132812135, rs137651874, rs109191047, rs135164815, rs43706485, and rs41255693), located on six Bos taurus autosomes (BTA4, BTA6, BTA19, BTA20, BTA22, and BTA26), showed suggestive associations with 305-day cumulative milk production (under Benjamini-Hochberg procedure with a false discovery rate of 0.1). Two of those SNPs (rs43375517 and rs135164815) were significantly associated with milk production (Bonferroni adjusted p-values < 0.05) when considering the Holstein population. Conclusions: The results obtained are consistent with previously reported associations in other Holstein populations. Furthermore, the SNPs found to influence bovine milk production in this study may be used as possible candidate SNPs for marker-assisted selection programs in Argentinean dairy cattle.
