Comparison of RNA Interference-mediated Gene Silencing and T-DNA Integration Techniques for Gene Function Analysis in Chinese Cabbage (RNA Interference 및 T-DNA Integration 방법에 의한 배추 기능유전자 Silencing 효과 비교)

  • To compare RNA interference-mediated gene silencing technique and T-DNA integration for gene function analysis in Chinese cabbage, BrSAMS-knockout (KO) line and BrSAMS-knockdown (KD) line were used. The KO line had lost the function of a Brassica rapa S-adenosylmethionine synthetase (BrSAMS) gene by T-DNA insertion and the KD line had shown down-regulated BrSAMS genes' expression by dsRNA cleavage. From microarray results of the KO and KD lines, genes linked to SAMS such as sterol, sucrose, homogalacturonan biosynthesis and glutaredoxin-related protein, serine/threonine protein kinase, and gibberellin-responsive protein showed distinct differences in their expression levels. Even though one BrSAMS gene in the KO line was broken by T-DNA insertion, gene expression pattern of that line did not show remarkable differences compared to wild type control. However, the KD line obtained by RNAi technique showed prominent difference in its gene expression. Besides, change of polyamine and ethylene synthesis genes directly associated with BrSAMS was displayed much more in the KD line. In the microarray analysis of the KO line, BrSAMS function could not be clearly defined because of BrSAMS redundancy due to the genome triplication events in Brassicaceae. In conclusion, we supposed that gene knock-down method by RNAi silencing is more effective than knock-out method by T-DNA insertion for gene function analysis of polyploidy crops such as Chinese cabbage.

Anti-obesity effect of Amomum taso-ko ethanol extract in 3T3-L1 adipocytes (3T3-L1 지방세포에서 초과 에탄올 추출물의 항비만 효과)

  • Amomum tsao-ko used as a traditional oriental herbal medicine, is indigenous to several Asia countries. In this study, we investigated anti-obesity activity of the ethanol extract of Amomum Taso-ko (A. tsao-ko). The ethanol extract of A. tsao-ko inhibited adipocyte differentiation using Oil Red O assay in 3T3-L1 cells. Inhibitory effect of the ethanol extract of A. tsao-ko on adipogenesis was modulated by down-regulation adipogenic transcriptional factor such as peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), CCAAT-enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$) and suppressed expression of fatty acid synthase, aP2, and resistin. We demonstrated that A. tsao-ko significantly inhibited adipogenesis and reduced $PPAR{\gamma}$ and $C/EBP{\alpha}$ expression in a dose-dependent manner. These results suggest that A. tsao-ko has an anti-obesity effect by inhibition of adipogenic transcription factor and adipocyte-specific genes in 3T3-L1 cells.

The Development of Citation Indicators of Korean Medical Journals (한국 의학학술지 인용지표 개발 연구)

  • The study investigated the citation indicators and the citation analysis data developed in the KoMCI(Korean Medical Citation Index) project. With the full understanding of the current level of citation rates of Korean medical journals by Korean medical journals. and of the characteristics and problems associated with the KoMCI citation indicators, it is possible to further develop or modify citation indicators which will better represent the citation patterns of Korean medical journals. The highest impact factor reported in the KoMCI 2000 : Korean Medical Journal Citation Reports, which covered 69 Korean medical journals published in 2000 is 0.424 and the average is 0.182. It is because only 8.5% of references cited in Korean medical journal articles is to the Korean journal articles, The journal self-citation rates are very high (usually higher than 50%) due to the fact that there are only a few Korean journals published in the same subject area. The KoMCI impact factors of two Korean SCI journals for which SCI JCR reported the SCI 2000 impact factors are about 1/3 of the SCI impact factors. It is because SCI is based on the citations received from 5,900 journals whereas KoMCI is from 69 journals.

Growth Rate of Transgenic Pigs and Size of Pig Hearts for Xenotransplantation to Cynomolgus Monkey

  • To compensate for the critical shortage of human organs for allotransplantation, xenotransplantation studies using genetically modified pigs are being performed in Korea. Two types of pigs that are used are ${\alpha}1,3$-galactosyltransferase gene knockout (GalT KO) pigs and GalT KO+hCD46 (human complement regulatory protein) pigs. The present study measured the gestation time, birth weight, daily growth rate, and heart weight of both kinds of transgenic minipigs. The gestation period for both types of pigs was 117~119 days. There was no difference in the body weight of GalT KO (-/+) and GalT KO (-/-) piglets, but GalT KO+hCD46 ($-^{hCD46+}/+$) pigs were significantly heavier at birth than were GalT KO+hCD46 ($-^{hCD46+}/-^{hCD46+}$) pigs. During the first 10 weeks of life, the daily weight gain of GalT KO+hCD46 ($-^{hCD46+}/-^{CD46+}$) piglets, which are considered the optimal type for xenotransplantation, was 0.19 kg. The weight of hearts from GalT KO piglets up to two months of age was affected more by body weight than by age. Transgenic pigs showed no differences in gestation period or reproductive ability compared with normal pigs. These results comprise basic data that may be used in xenotransplantation studies and transgenic animal production in Korea.

Understanding of Yon-Ko-Jeon Culture : The Phenomenon of Women's Alienation in Yon-Ko-Jeon (연고전 문화 읽기: 연고전과 여성 소외현상)

  • By examining the constitutive characteristics of Yon-Ko-Jeon and analyzing the lived meanings and experiencing essence of students, this study details the students' awareness, responses, and strategies with regards to the phenomenon of women's alienation from Yon-Ko-Jeon. Students from the two colleges were divided into two groups- one which was aware of women's alienation from Yon-Ko-Jeon, and another group that was unaware of such a phenomenon. Students without awareness tended to perceive the alienation of women from Yon-Ko-Jeon as a natural phenomenon. On the other hand, the members who had awareness tended to see it as a phenomenon that was either justified, unequal, or in need of change. In the latter group, those who saw women's alienation as a justified phenomenon responded with tolerance and adopted a bystander strategy. Conversely, those who saw it as an unequal phenomenon responded with rejection and instead chose to adopt anti-/alternative Yon-Ko-Jeon strategies. The last group, which perceived women's alienation as a phenomenon in need of change, adopted correcting responses as well as intercollegiate solidarity and cooperation strategies. A generalization of the findings reveals that the students' experiencing essence of the phenomenon extends from critical thinking to awareness, response, and strategy in a step-by-step and sequential process. Through this study, it was found that the students' experiencing essence all follow a continuous thinking pattern, as illustrated in the diagram provided.

A study on improvement plan of Yon-Ko-Jeon as a gender equality sports event using the IPA (IPA기법을 활용한 성 평등적 스포츠 이벤트로서 연고전의 개선방안)

  • The purpose of this study is to check awareness of 'the phenomenon of women's alienation in Yon-Ko-Jeon' to women students, and improve the gender equality for Yon-Ko-Jeon through IPA(Importance Performance Analysis). For this study, 322 women undergraduates at Yonsei University and Korea University, from sophomore to senior students, are selected and asked to be surveyed. At first, a majority of women students from two universities didn't recognize the lack of women sports and athletes in Yon-Ko-Jeon. Moreover, they didn't think about women's alienation in Yon-Ko-Jeon as well as improvement for the issue seriously. It shows the life style of gender in the sport. Second, as a result of IPA, improvement plan of Yon-Ko-Jeon on gender equality shows it needs to reflect member's opinion and introduce women sports clubs first. Recently, the improvement for the Yon-Ko-Joen is the top priority because the event doesn't reflect member's opinion. Furthermore, it shows that Yon-Ko-Jeon needs to introduce the competition of women's club sports. These are what the event has to improve.

Korean Ironic Expression Detector (한국어 반어 표현 탐지기)

  • Despite the increasing importance of irony and sarcasm detection in the field of natural language processing, research on the Korean language is relatively scarce compared to other languages. This study aims to experiment with various models for irony detection in Korean text. The study conducted irony detection experiments using KoBERT, a BERT-based model, and ChatGPT. For KoBERT, two methods of additional training on sentiment data were applied (Transfer Learning and MultiTask Learning). Additionally, for ChatGPT, the Few-Shot Learning technique was applied by increasing the number of example sentences entered as prompts. The results of the experiments showed that the Transfer Learning and MultiTask Learning models, which were trained with additional sentiment data, outperformed the baseline model without additional sentiment data. On the other hand, ChatGPT exhibited significantly lower performance compared to KoBERT, and increasing the number of example sentences did not lead to a noticeable improvement in performance. In conclusion, this study suggests that a model based on KoBERT is more suitable for irony detection than ChatGPT, and it highlights the potential contribution of additional training on sentiment data to improve irony detection performance.

Dietary ellagic acid blocks inflammation-associated atherosclerotic plaque formation in cholesterol-fed apoE-deficient mice

  • BACKGROUND/OBJECTIVES: Atherosclerosis particularly due to high circulating level of low-density lipoprotein is a major cause of cardiovascular diseases. Ellagic acid is a natural polyphenolic compound rich in pomegranates and berries. Our previous study showed that ellagic acid improved functionality of reverse cholesterol transport in murine model of atherosclerosis. The aim of this study is to investigate whether ellagic acid inhibited inflammation-associated atherosclerotic plaque formation in cholesterol-fed apolipoprotein E (apoE)-knockout (KO) mice. MATERIALS/METHODS: Wild type mice and apoE-KO mice were fed a cholesterol-rich Paigen diet for 10 weeks to induce severe atherosclerosis. Concurrently, 10 mg/kg ellagic acid was orally administered to the apoE-KO mice. Plaque lesion formation and lipid deposition were examined by staining with hematoxylin and eosin, Sudan IV and oil red O. RESULTS: The plasma leukocyte profile of cholesterol-fed mice was not altered by apoE deficiency. Oral administration of ellagic acid attenuated plaque lesion formation and lipid deposition in the aorta tree of apoE-KO mice. Ellagic acid substantially reduced plasma levels of soluble vascular cell adhesion molecule and interferon-γ in Paigen diet-fed apoE-KO mice. When 10 mg/kg ellagic acid was administered to cholesterol-fed apoE-KO mice, the levels of CD68 and MCP-1 were strongly reduced in aorta vessels. The protein expression level of nitric oxide synthase-2 (NOS2) in the aorta was highly enhanced by supplementation of ellagic acid to apoE-KO mice, but the expression level of heme oxygenase-1 (HO-1) in the aorta was reduced. Furthermore, ellagic acid diminished the increased aorta expression of the inflammatory adhesion molecules in cholesterol-fed apoE-KO mice. The treatment of ellagic acid inhibited the scavenger receptor-B1 expression in the aorta of apoE-KO mice, while the cholesterol efflux-related transporters were not significantly changed. CONCLUSION: These results suggest that ellagic acid may be an atheroprotective compound by attenuating apoE deficiency-induced vascular inflammation and reducing atherosclerotic plaque lesion formation.
