Biochemical Changes in the Hemolymph of the Larvae of Thecodiplosis japonensis Uchi. et Inouye (솔잎혹파리 유충 체액의 생화학적 변화)

  • The concentration of amino acids, total nitrogen, trehalose, lipids and the activities of respiratory, acid$\cdot$alkaline phosphatase, glutamic oxalozcetic transaminase and glutamic pyruvic transaminase during larval stage in Pine leaf gall midge, Thecodiplosis janensis Uchi. et Inouye were measured using Paper chromatographic method, micro-Kjeldahl method, Thin layer chromatographic method, Warburg's manometric method, Bessey-Lowry method and Reitman-Frankel method, respectively. Healthy specimens )yore chosen as samples of each larval stages; alrva in gall and larva in soil. Amino acids present in the alcoholic extracts were alanine, glutamic acid, glycine, histidine, methionine, proline, threonine, tryptophan and valine. The total nitrogen concentration reached to 31.348mg/g during the larva in gall and the larval stage in soil of the value was decreased to 29.027mg/g. The hemolymph sugar, trehalose value for larva in soil was about two times of the value for larva in gall. Total lipid, phospholipid,monoacylglycerol, triacylglycerol, sterol, free fatty acid and ester cholesterol were identified at larval stages in gall and soil. Triacylglycerol concentration reached high level in contrast with other lipid contents during larvae in gall and larva in soil. Free fatty acid, sterol except decreased lipids during larval stage in soil. Endogenous respiration, succinate of respiratory activities decreased at larval stage in soil compare with larva in gall. The activities of acid phosphatase decreased larval stage in soil but the activities of alkaline phosphatase increased remarkably. The activities of glutamic oxaloacetic transaminase and glutamic pyruvic transaminase reached high level of the larva in gall.

Effects of amino acid composition in pig diet on odorous compounds and microbial characteristics of swine excreta

  • Background: Major amino acids in pig diets are Lys, Met, Thr, and Trp, but little is known about the requirements for the other essential amino acids, especially on odorous compounds and microbial characteristics in feces of growing-finishing pigs. To this end, different levels of amino acid composition added to diets to investigate the effects of amino acid composition on microbial characteristics and odorous compounds concentration. Methods: A total eight (n = 8) barrows (Landrace ${\times}$ Yorkshire ${\times}$ Duroc) with an average bodyweight of $89.38{\pm}3.3kg$ were individually fed diets formulated by Korean Feeding Standards 2007 (old version) or 2012 (updated with ideal protein concept) in metabolism crates with two replication. After 15-day adaptation period, fresh faecal samples were collected directly from pigs every week for 4 weeks and analysed for total volatile fatty acids (VFA), phenols and indoles by using gas chromatography. The nitrogen was determined by Kjeldahl method. Bacterial communities were detected by using a 454 FLX titanium pyrosequencing system. Results: Level of VFA tended to be greater in 2012 than 2007 group. Among VFAs, 2012 group had greater (p < 0.05) level of short chain fatty acids (SCFA) than control.Concentration of odorous compounds in feces was also affected by amino acid composition in pig diet. Levels of ammonium and indoles tended to be higher in 2012 group when compared with 2007 group.Concentration of phenols, p-cresol, biochemical oxygen demand, and total Kjeldahl nitrogen, however, were lower (P < 0.05) in 2012 treatment group compare to 2007. The proportion of Firmicute phylum were decreased, while the Bacteriodetes phylum proportion increased and bacterial genera includingCoprococcus, Bacillus, and Bacteroides increased (p < 0.05) in 2012 compare to 2007 group. Conclusion: Results from our current study indicates that well balanced amino acid composition reduces odor by modulating the gut microbial community. Administration of pig diet formulated with the ideal protein concept may help improve gut fermentation as well as reduce the odor causing compounds in pig manure.

  • Although the seafood cooking drips were the byproducts from the fishery industry and being wasted, it had many nutrients including proteins. In this study, the effect of a gamma irradiation on the cooking drips from Hizikia fusiformis, Enteroctopus dofleni and Thunnus thynnus were investigated. The cooking drips were extracted with 70% ethanol solution, and the extracts were analysed for the protein concentration by three different methods of Lowry, BCA and Kjeldahl. The extracts were irradiated with different doses and the protein contents were compared with respect to the absorbed doses. Total content of the proteins was increased with increasing irradiation dose. The change of protein pattern in the irradiated cooking drips was also confirmed by SDS-PAGE analysis. These results shown that the proteins in cooking drips could be unfolded or aggregated by the irradiation. Therefore, gamma irradiation could be considered as an effective method for extracting useful proteins.

  • Analysis of mineral nutrients in plant is required for evaluating diagnosis of plant nutritional status. Pretreatment procedure for the analysis of plant can be varied depending on elements to be analyzed. Wet-digestion is suitable for total nitrogen, phosphate and cations, however, digestion solution including nitric acid is not suitable for nitrogen analysis. Incineration procedure is required to analyze chloride, silicate and total sulfur. After digestion, total nitrogen is analyzed by Kjeldahl method, and phosphate is detected at 470nm by colorimetric analysis with ammonium meta vanadate. Cations and micro elements are determined by titration or colorimetry, also, these elements can be measured by Atomic absorption spectrometer (AAS) or Inductively coupled plasma spectrometer (ICP).

Protein, Ca, Mg and P Intakes of Breast-fed Infants during Lactation (모유영양아의 수유기간별 단백질, 칼슘, 마그네슘과 인 섭취량)

  • This study was longitudinally conducted to evaluate the intakes of protein, Ca, Mg and P of exclusively breast-fed infants compared with the Recommended Dietary Allowances (RDA) for Korean infants. Twenty Korean lactating women and their infants during the first 3 months of lactation in Incheon area were participated. Protein, Ca and Mg, and P contents in the milk were determined using semimicro Kjeldahl (N ${\times}$ 6.38) , atomic absorption spectrophotometer and colorimeter, respectively, and also the milk consumption of the infants was measured by the test-weighing method. Protein contents of the milk were 1.96, 1.63, 1.51, 1.25 and 1.16 g/100 ml, and protein intakes of the breast-fed infants were 9.00, 9.85, 9.17, 8.97 and 7.76 g/day at 7, 15, 30, 60 and 90 days postpartum. The average protein intake per body weight of the breast-fed infants was 1.84 g/kg/day. The average intakes of Ca, Mg, P were 172.1 mg/day, 15.2 mg/day and 91.4 mg/day, respectively, and the average Ca/P ratio was 1.91. There was positive correlation between protein and Ca, protein and p, and Ca and P contents while negative correlation between Mg and P, The body weight of breast-fed infants increased normally from 3.6 $\pm$ 0.41 g at birth to three month during lactation. It is suggested that the breast-fed infants in Incheon area consume almost adequately protein, Ca and P from the milk compared with RDA for Korean infants.

  • The concentration of total protein during the metamorphosis of the Indian meal moth, Plodia interpunctella $H\ddot{u}bner$ was measured using micro-Kjeldahl method by Oser(1965). Healthy specimens were chosen as samples at each developmental stages: the 1st instar larva, the 3rd inster larva, final instar larva, pupae 2, 6, 10 days and the adult 1 day. The total protein concentration decreased gradually until the two day pupal stage and increased at the six day pupal stage. In the one day dault stage, the concentration reached a maximum. In conclusion, changes of total protein concentation through the course of the physiological cycle form a U-shaped curve when graphed against the histolysis of larval organs and the histogenesis of adult organs. In view of controlling pest, the U-shaped pattern of physiological activity indicates that control will be the most effective at the time of the most physiological weakness manifested at the two day pupal stage.

Considerations of Acid Decomposition System for the Analysis of Heavy Metals in Packaging-grade Paper (포장용지류에서의 중금속 분석을 위한 산분해 전처리 방법의 탐색)

  • The fibrous raw materials in packaging-grade paper production in Korea were mainly obtained from waste paper. The use of recycled paper has both positive and negative impacts in papermaking process. The primary positive impacts are the environmental protection and manufacturing cost reduction, and the negative impacts are the quality reduction in paper quality and the accumulation of heavy metals and other pollutants in wet- and dry-end process. This study was carried out to consider the optimum acid decomposition system with the highest recovery rate for the analysis of heavy metals in packaging-grade paper. The open digestion system using Kjeldahl apparatus and the closed digestion system using microwave oven for decomposing the organic materials in paper were compared. In both open and closed digestion method, the combination of nitric acid, hydrochloric acid and hydrogen peroxide showed higher recovery rate than using only nitric acid alone because the presence of Cl- ions in hydrochloric acid stabilizes ligand formation with metal ions. KOCC was observed to have the highest heavy metal content among the recycled paper samples. The heavy metal contents decomposed with the closed digestion system were relatively higher than with open digestion system.

Daily influent variation for dynamic modeling of wastewater treatment plants

  • Wastewater treatment plants (WWTPs) with activated sludge system are widely used throughout the most common technologies in the world. Most treatment plants require optimization of certain treatment processes using dynamic modeling. A lot of examples of dynamic simulations require reliable data base of diurnal variation of the inflow and typical concentrations of parameters such as Chemical Oxygen Demand (COD), Total Kjeldahl Nitrogen (TKN), etc. Such detailed data are not available, which leads to problemsin the application of dynamic simulations. In many examples of plants, continuous flow measurements are only performed after the primary clarifier, whereas measurements from influent to the plant are missing, as is the case with the examples in this paper. In some cases, a simpler, faster and cheaper way can be applied to determine influent variations, such as the "HSG-Sim" method ("Hochschulgruppe Simulation"). "Hochschulgruppe Simulation" is a group of researchers from Germany, Austria, Switzerland, Luxembourg, Netherlands and Poland (see http://www.hsgsim.org). This paper presents a model for generating daily variations of inflow and concentration of municipal wastewater quality parameters, applied to several existing WWTPs in Bosnia and Herzegovina (B&H). The main goal of the applied method is to generate realistic influent data of the existing plants in B&H, in terms of flow and quality, without any prior comprehensive survey and measurements at the site. The examples of plants show the influence of overflow facilities on the dynamics of input flow and quality of wastewater, and a strong influence of the problems of the sewerage systems.

Relationship between Molecular Structure Characteristics of Feed Proteins and Protein In vitro Digestibility and Solubility

  • The nutritional value of feed proteins and their utilization by livestock are related not only to the chemical composition but also to the structure of feed proteins, but few studies thus far have investigated the relationship between the structure of feed proteins and their solubility as well as digestibility in monogastric animals. To address this question we analyzed soybean meal, fish meal, corn distiller's dried grains with solubles, corn gluten meal, and feather meal by Fourier transform infrared (FTIR) spectroscopy to determine the protein molecular spectral band characteristics for amides I and II as well as ${\alpha}$-helices and ${\beta}$-sheets and their ratios. Protein solubility and in vitro digestibility were measured with the Kjeldahl method using 0.2% KOH solution and the pepsin-pancreatin two-step enzymatic method, respectively. We found that all measured spectral band intensities (height and area) of feed proteins were correlated with their the in vitro digestibility and solubility ($p{\leq}0.003$); moreover, the relatively quantitative amounts of ${\alpha}$-helices, random coils, and ${\alpha}$-helix to ${\beta}$-sheet ratio in protein secondary structures were positively correlated with protein in vitro digestibility and solubility ($p{\leq}0.004$). On the other hand, the percentage of ${\beta}$-sheet structures was negatively correlated with protein in vitro digestibility (p<0.001) and solubility (p = 0.002). These results demonstrate that the molecular structure characteristics of feed proteins are closely related to their in vitro digestibility at 28 h and solubility. Furthermore, the ${\alpha}$-helix-to-${\beta}$-sheet ratio can be used to predict the nutritional value of feed proteins.

Ammonia Microdiffusion and Colorimetic Method for Determining Nitrogen in Plant Tissues (암모니아 확산 및 발생에 의한 식물조직의 질소분석 방법)

  • Ammonia microdiffusion method and colorimetric measurement are described for the nitrogen determination. The diffusion of ammonia could be successfully induced by using a microdiffusion cell. It is a simple and rapid technique, which is suitable for transforming the nitrogen in digests into $NH_4CI$ for the colorimetric N determination with ammonia color reagent. Above 99% of N recovery were obtained with microdiffusion up to 15 hours. The coloration method of collected $NH_4CI$ for the colorimetric N determination was also estabilshed with a scanning in U.V. spectrophotometer. Under the proposed coloration method (0.5 mL of sample digest, 4.0 mL of $H_2O$ and 0.5 mL of ammonia color reagent), a maximal absorbance was observed at 410 nm. The kinetic measurement of absorbance showed a high stability from 5 to 45 minutes after color development. Absorbance was directly proportional to the amount of $NH_4^+-N$ present. The microdiffusion-ammonia coloration method was successfully applied to the nitrogen determination in the forms of protein-N or total -N in plant tissue. Comparing with Kjeldahl distillation method, the values obtained with described method were slightly higher and more reliable.
